Lycopene Protects the Kidney and Heart Cells Against Nephrotoxicity and Cardiotoxicity

Nephrotoxicity is toxicity in the kidneys associated with reduced function of the kidney due to toxic chemicals, and medications.

The kidneys are essential bean shape organ of our body, located just below the rib cage, one on each side of your spine.

Kidneys play a critical role in filtering blood with billions of glomeruli by separating the cells of blood from plasma and reabsorb back substances, and secret waste such as urea and ammonium are stored into pelvis then into the ureter to the bladder.

Furthermore, kidneys also process the functions that regulate electrolytes, blood pressure, maintains acid-base balance, produces hormone calcitriol, renin, and erythropoietin, etc. 

In the urinary system, the kidney also reabsorbed back substances into the blood, while others waste, such as urea and ammonium are stored into pelvis then into the ureter to the bladder.

Most common symptoms of nephrotoxicity decreased urine output, fluid retention, causing swelling in your legs, ankles or feet, weakness, fatigue, and irregular heartbeat.

In severe cases, patients may also experience symptoms of shortness of breath, and confusion.

Cardiotoxicity, on the other hand, is toxicity in the heart associated with reduced heart muscle capacity in the blood circulation due to toxic chemicals, and medications.

The heart, a four-chambered, hollow muscle and double acting pump that is located in the chest between the lungs is a muscular organ in the human body processed the function that pumps blood through the blood vessels of the circulatory system to nourish the body and assist in the removal of metabolic wastes.

According to the statistic provided by the American Heart Foundation, on average, an American dies of CVD every 38 seconds. In 2016, cardiovascular diseases cause the death of 2,303 deaths each day, in the US.
Compared to nephrotoxicity, patients with cardiotoxicity may have symptoms of fatigue, shortness of breath on exertion, worsening to shortness of breath at rest, discomfort lying on your back and welling of the ankles.

Lycopene is a phytochemical in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.

On finding a potential compound which processes cardio and nephroprotective effects, researchers examined the effects of lycopene against the toxic effects of Aflatoxin B1(AFB1) exposure in kidney and heart of experimental rats.

A total of 42 healthy three-month-old male Wistar-Albino rats were randomly divided into six experimental groups including 7 rats in each as following control group, lycopene (5 mg/kg/day, orally for 15 days) group, AFB1 (0.5 mg/kg/day, orally for 7 days) group, AFB1 (1.5 mg/kg/day, orally for 3 days) group, AFB1 (0.5 mg/kg/day, orally for 7 days) + lycopene (5 mg/kg/day, orally for 15 days) group and AFB1(1.5 mg/kg/day, orally for 3 days) + lycopene (5 mg/kg/day, orally for 15 days) group.

Compared to AFB1 and according to the tested analysis, the elevated levels of malondialdehyde (MDA) levels associated with oxidative stress markers were significantly decreased in the lycopene treatment group.


The reduced levels of antioxidant enzymes in the AFB1 group such as glutathione (GSH), glutathione-S-transferase (GST), catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and glucose-6-phosphate-dehydrogenase (G6PD) activities were increased substantially in kidney and heart tissues upon the injection of lycopene.

Furthermore, the non-enzymatic antioxidant system in AFB treated rats that cause the rise of oxidative stress in the rat kidney and heart were also inhibited by administration of lycopene.

Moreover, lycopene also inhibited the increased urea, creatinine levels, and reduced-sodium concentrations in plasma of AFB1 treated rats.

Based on the findings, researchers said, "There was lycopene showed protection against AF induced nephrotoxicity and cardiotoxicity".

Taken altogether, lycopene found in tomato may be considered supplements for the prevention and treatment of osteoporotic fractures, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Bromelain Speeds Up Healing of Ecchymosis

Ecchymosis is a discoloration of the skin caused by the bleeding underneath the skin, typically in bruising.

Most cases of ecchymosis are results in which the blood vessels near the surface of the skin burst, particularly in the impact of an injury.

In other words, ecchymosis is associated with blood vessel leaks, and the blood is accumulated under the skin.

The discoloring of the skin is also involved in the clotting factors, that stop the bleeding so that the tissue starts healing.

The progression of ecchymosis is gone through a certain stage from red or purple to black or blue
 to brown and to yellow.

Certain medications have been found to induce ecchymosis including:
* Blood thinners, such as aspirin or warfarin 

*Antibiotics such as cephalexin, cefaclor, and cefuroxime-axetil can cause ecchymosis by inducing low platelet count

* Corticosteroids 
According to the study from the University College Hospital, easy bruising tended to be older (61 vs 52 yrs), on higher daily dosages (1,388 vs 1,067 micrograms), particularly in the long term inhaled corticosteroids users.

Most cases of ecchymosis usually heal on their own within two to three weeks.  Most common medications used for the treatment of ecchymosis are pain relievers.

Based on the results, 5 out of 7 randomized controlled trials reported improvement postprocedure ecchymosis in the bromelain-treated group.

Taken altogether, bromelain may be considered a remedy for the treatment of postprocedure ecchymosis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Cancer is a group of diseases associated with cells growing and multiplying disordered and uncontrollable in the tissue of the specific organ.

Because of less nervous in the kidney and location, patients in the early stage of renal cell carcinoma are asymptomatic. In other words, kidney tumor can grow without causing any pain and discomfort. Most common symptoms of kidney cancer are
* Abdominal pain that is dull and constant lasting for hours.

*Back pain that is dull and constant lasting for hours.

* Blood in the urine is a result of break off of the tumor in the tubules.

* Unintentional weight loss
Any intentional weight loss of over 10% is one of the most common symptoms of cancer.

*  Flank pain is a sensation of discomfort, distress, or agony in around the back and side of the abdomen.

* Swelling and enlargement of the abdomen happens to approximate 15% of all cases of renal cell carcinoma. 

*Anemia
If cancer has affected the function of erythropoietin in the production of hemoglobin.

* Varicocele enlargement is a result of the blockage of the left (right) testicular vein by tumor invasion of the left (right) renal vein.

*Hypertension is a result of secretion of renin by the tumor.

According to the tested analysis, curcumin markedly induced apoptosis in NVP-BEZ235-treated cells.

Co-treatment with curcumin and NVP-BEZ235 led to the down-regulation of Mcl-1 protein expression Involved in the regulation of apoptosis and survival of cancer cells but not mRNA expression in tested cells.

Furthermore, the co-administration also inhibited the protein associated with the anti-apoptotic activity of Bcl-2.

Interestingly, in colon cancer cell line analysis, Combined treatment with NVP-BEZ235 and curcumin reduced Bcl-2 expression in wild-type p53 HCT116 human colon carcinoma cells but not p53-null HCT116 cells.

In skin cell perspectively, NVP-BEZ235 combined with curcumin did not have a synergistic effect on normal human skin fibroblasts and normal human mesangial cells.

Based on the findings, researchers suggested that curcumin combined with the NVP-BEZ235 induced human renal carcinoma Caki cells apoptosis by regulating the protein associated with anti-apoptosis accompanied by blocking the activation of proliferative signaling pathways.

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of kidney cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Blood pressure is the force of blood pushing against the walls of the arteries as the heart pumps out blood. High blood pressure is a condition associated with abnormally high blood pressure in the arteries.

High blood pressure is also a condition of a cluster of metabolic syndrome involved in the risk of cardiovascular disease and diabetes.

If the blood pressure stays high over time, it not can damage the heart muscle but also the body in many ways.

According to the statistic from the CDC, 1 of 3 U.S. adults or about 75 million people is living with high blood pressure. Unfortunately, only about half (54%) of these people have their high blood pressure under control.

There is no single cause of high blood pressure,

And, "an elevation of plasma FAs may contribute to the development of hypertension via a process involving the elevation of Ca(2+) current density and alteration of channel kinetics in the vascular smooth muscle membrane".

The onion is a plant in the genus Allium, belongings to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures. 

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

On finding a potent ingredient for the prevention of hypertension, researchers evaluated the effect of aqueous extract of garlic, white onion, and purple onion on subjects with blood pressure.

Elevated levels of an angiotensin-converting enzyme associated with the high blood pressure were inhibited by the aqueous extract of garlic, purple onion, and white onion in a concentration-dependent manner (0-4 mg/ml).

Additional differentiation also suggested that purple onion (IC50 = 0.59 mg/ml) had higher inhibitory effect on angiotensin-converting enzyme comapred to those of white onion (IC50 = 0.66 mg/ml) and purple onion (IC50 = 0.96 mg/ml).

Furthermore, all aqueous extracts showed a significant activity to scavenge 2,2- diphenyl-1-picrylhydrazyl (DPPH*) free radical and reduce Fe3+ to Fe2+ in a dose-dependent manner.

Based on the findings, researchers concluded, "a possible mechanism by which garlic, white onion, and purple onion exert... antihypertensive properties could be through the inhibitory effect on ACE,.. coupled with their ability to prevent lipid peroxidation in the pancreas and heart, which justify their strong antioxidant properties".

Taken altogether, onion used alone or combined with garlic may be considered a remedy for the prevention and treatment of hypertension, pending to the confirmation of the larger sample size and multicenter human study.

Phytochemical Lycopene Ameliorates Neuro-Oxidative Stress in Aging Degenerative Diseases

Oxidative stress in a condition caused by overexpression of free radicals and the depletion of antioxidant enzymes found in the host.

In other words, oxidative stress is a result of a long-term imbalance of the ratio of free radicals and antioxidants.

Antioxidants can be either produced by the liver or consumptions of antioxidant-rich fruits and vegetables.

Antioxidants are stable molecules that protect our body against free radical oxidation.

Free radicals are any chemical compounds which have an oxygen molecule that can become free radical after chemical interaction in the body

Most healthy individuals can produce enough amounts of antioxidant enzymes endogenously to counter the bombards of free radicals.

However, for people with a weakened immune system such as elderly and immature immune system may require the injection of antioxidants from the outside sources, particularly from the second metabolites.

Epidemiologically, oxidative stress has been found to promote a number of age-related conditions, including cardiovascular diseases CVDs, chronic obstructive pulmonary disease, chronic kidney disease, neurodegenerative diseases, and cancer.

Not all free radicals are bad in the moderate amounts, our body needs limit levels of some free radicals to function better, including lower blood pressure, relax blood vessels to move blood, such as nitric oxide NO.

Lycopene is a phytochemical in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.

On findings a phytochemical compound for the treatment of diseases associated with oxidative stress, scientists examined the effects of lycopene (LYC), a carotenoid pigment, on oxidative stress-induced cognitive defects.

According to the behavioral tests, chronic LYC supplementation (50 mg/kg body weight per day) alleviated d-galactose-induced CD-1 male mice cognitive impairments.

Furthermore, LYC ameliorated histopathological neuron damage and restored brain-derived levels of neurotrophic factor (BDNF), a neurotransmitter modulator associated with neuronal survival and growth, serves in the hippocampus of mice.

Moreover, LYC also significantly stimulated the production of antioxidant enzymes such as HO-1 and NQO-1 and reduced levels of inflammatory cytokines such as IL-1β and TNF-α in the d-galactose-treated mice serum.

The Immunohistochemical assay also suggested that LYC significantly restored the expression of glial cells inflammatory makers Iba-1 and GFAP that responses to proinflammatory expression.

More importantly, LYC protected the neuronal against oxidative damage through activation of Nrf2 signaling in the regulation of anti-inflammatory gene expression and inactivation of NF-κB translocation associated with pro-inflammatory expression in an H2O2-induced SH-SY5Y cell model.

Dr. Zhao B, the lead scientist wrote, "LYC could ameliorate oxidative stress-induced neuroinflammation and cognitive impairment possibly via mediating Nrf2/NF-κB transcriptional pathway".

Taken altogether, lycopene found in tomato may be considered supplements for the prevention and treatment of diseases associated with oxidative stress, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
