Angelica Sinensis In the Treatment of Mammary Gland Hyperplasia in Vivo

Mammary gland hyperplasia or hyperplasia of the breast is the benign overgrowth of mammary ductal epithelial cells and interstitial fibers.

Believe it or not, patients with mammary gland hyperplasia are found in 99.3% of the total patients with breast-related diseases, in which 5–10% have the potential to develop breast cancer.

Hyperplasia is the enlargement organ or tissue caused by an increase in the reproduction rate of its cells.

Hyperplasia of the breast is considered an early stage of breast cancer.

According to the statistics, atypical ductal hyperplasia (ADH) is the common form of hyperplasia of the breast found in about 5% to 20% of the diseases.

Although atypical ductal hyperplasia (ADH) is benign, the progression of the condition has been found to induce ductal carcinoma in situ (DCIS) and invasive carcinoma.

Mammary gland hyperplasia doesn't cause symptoms, However, some patients may experience a lump or pain.

Conventionally, most cases of mammary gland hyperplasia are treated by surgery that removes the abnormal cell growth.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

On finding a natural compound for the treatment of diseases associated with the breast, researchers examined the effect of acupuncture combined with medication (Rule Granule) on mammary gland hyperplasia in an animal model.

A total of 55 female SD rats were randomized into blank control group (n=10), model group (n=10), acupuncture group (n=11), medication group (n=10), and acupuncture plus medication group (n=9), as follow

* The MGH model was established by muscular injection of E2 benzoate(0.5 mg/kg, once daily for 20 days), followed by injection of P (5 mg/kg) into the lateral muscle of the hind-limb, once daily for 5 days, and the rats of the control group were treated by muscular injection of normal saline at the same site and in the same procedures.

* Acupoint group A composed of bilateral Tianzhong (SI 11), Ganshu (BL 18) and Zusanli (ST 36), and group B composed of Tanzhong (CV 17), and bilateral Wuyi (ST 15) and Hegu (LI 4) were alternatively punctured with filiform needles and stimulated swiftly by twirling the needle in each acupoint for 20-30 sec, once daily for 30 days.

* Rats of the medication group and acupuncture +medication group were treated by gavage of Rule Granule fluid containing Baishao (Paeonia Iactiflora Pall), Danggui (Angelica Sinensis), etc. (1.5 mL/100 g for each rat) and those of the other groups treated by gavage of distilled water (1.5 mL/100 g). The treatment was given once daily for a continuous 30 d. At the end of the experiments,

According to the results of the analysis, the hyperplasia of the mammary gland was found to improve significantly. The serum PRL and E2 contents and the expression of prolactin receptor (PRLR), estrogen receptor (ER) and progesterone receptor (PR) was significantly decreased, and the content of P was notably increased in the acupuncture, medication, and acupuncture plus medication groups relevant to the model group.

Furthermore, acupuncture plus medication were significantly superior to those of both simple medication and simple acupuncture in down-regulating serum PRL and E2 contents and PRLR, ER and PR immunoactivity, as well as in up-regulating serum P content.

Additionally, no significant differences were found between the medication and acupuncture groups. 

Based on the findings, researchers said, "The therapeutic effects of acupuncture plus Rule Granule are better than those of simple acupuncture and simple Rule Granule".

Taken altogether, Herbal Angelica used a combination with other herbal medicine and acupuncture
for the treatment of mammary gland hyperplasia, pending to larger sample size and multicenter human study.
