Herbal Angelica Protects the Neurons Against the Onset of Parkinson's Disease in Vivo

Neurological diseases are the medical conditions of brain disorder. Anything that involves the structural, biochemical or electrical abnormalities in the brain, spinal cord or other nerves, including brain damage are considered as neuro diseases.

The most common neurodegenerative diseases in the elderly are dementia. Depending on the brain regions that are damaged or dysfunction, dementia can be Parkinson's, Alzheimer's, and Huntington's disease.

Genetic mutation inherited from parents such as the NOTCH3 gene is the most common autosomal dominant inherited cause of stroke and vascular dementia.

Dementia is a neurodegeneration syndrome among the elderly, affecting memory, thinking, orientation, comprehension, calculation, learning capacity, language, and judgment over 47 million
of the worldwide population, mostly in the West.

Parkinson disease (PD) is a disabling, progressive condition induced symptoms of the olfactory deficit, sleep problems such as rapid eye movement behavior disorder, constipation and the more recently described male erectile dysfunction(456), due to the interruption of frontal-subcortical loops facilitated cognition and parallel the motor loop(457).

Contrary to common perception, many non-motor symptoms (NMS) also link to early onset of PD(459) and some may even predate the diagnosis of PD based on motor signs(458).

In the very early stage of PD, patients may not experience any symptoms. However, Parkinson's disease symptoms gradually worsen as the condition progresses.

As of today, researchers do not know why the neurons in the brain in some people are susceptible to gradually break down or die while others do not even they have similar health conditions and from the same family.

Epidemiologically aging, gender, long-term exposure to certain toxic chemicals and genetic deposition are some of the prevalent factors involved in the increased risk of the disease.

On finding a natural ingredient for the prevention and treatment of neurodegenerative diseases, scientists examined the investigated the neuroprotective effects of the triple herbal extract DA-9805 ((Moutan cortex, Angelica dahurica root, and Bupleurum root in a 1:1:1 mixture) in experimental models of Parkinson's disease (PD) by induced neurotoxic effects on dopaminergic neurons by inhibiting mitochondrial oxidative phosphorylation (OXPHOS) complex.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the results of the differentiation
In SH-SY5Y cells,
* DA-9805 ameliorated the suppression of tyrosine hydroxylase expression and mitochondrial damage on OXPHOS complex 1 activity,

* DA-9805 also inhibited the expression of mitochondrial membrane potential, reactive oxygen species (ROS) generation, and oxygen consumption rate.

In the MPTP-induced subacute PD model mice,

* Oral administration of DA-9805 recovered dopamine content as well as bradykinesia.

* DA-9805 protected against neuronal damage in the substantia nigra pars compacta (SNpc) and striatum.

In both vitro and vivo, DA-9805 normalized the phosphorylation of AKT that that promotes survival and growth of the neurons at S473 and T308 on the insulin signaling pathway and the expression of mitochondria-related genes.

Based on the results, researchers suggest, "triple herbal extract DA-9805 showed neuroprotective effects via alleviating mitochondria damage in experimental models of PD. We propose that DA-9805 may be a suitable candidate for disease-modifying therapeutics for PD".

Taken altogether, Dandelion may be considered an antioxidant remedy for the treatment of PD, pending to the confirmation of the larger sample size and multicenter human study.
