Devil's club (Oplopanax horridus) Gobbles Up Human Colorectal Cancer Cells in Vitro

The colon-rectum formed a part of the digestive system is involved in the absorption of water from the small bowel contents and broken down of certain materials in the feces into substances of which some of them to be reabsorbed and reused by the body.

Colorectal cancer is a medical condition caused by cell growth irregularly in the tissue of the colon or rectum due to the alternation of cells DNA.

Colon cancer is relatively very common and slowly growing and progressing in a predictable way.

Most cases of primary colon cancer start in the cell on the surface of the inner lining of the colon. At the early stage. most patients with colon cancer do not experience any symptoms.

Rectal cancer is the slowest growing cancer. Therefore, most cases of colorectal cancer are diagnosed at the advanced stage in the older population.

According to the statistics provided by the American Cancer Society, in the US, colorectal cancer is the third most common cancer diagnosed in both men and women compared to other types of cancer.

The lifetime risk of colorectal cancer is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

Approximately, 140,000 cases of colorectal cancer were diagnosed in the US in 2018. Cancer also causes the death of over 50, 000 people, representing 8.3% of total cancer death.

Colorectal cancer is relatively very common and slowly growing and progressing cancer.

The disease is also the third most commonly diagnosed cancer in developed countries, including the US and Canada.

Being overweight or obese. smoking, physical inactivity, excessive alcohol drinking, the increase in age, family and personal history, genetic preposition are some of the prevalent risks of the disease.

Devil's Club is a large shrub, genus Oplopanax, belonging to the family Aralioideae, native to the cool moist forests of western North America. 

The herb has been used in traditional and herbal medicine as Adaptogen, Antipyretic, Pain Reliever, Antitussive, Antibacterial agent and to treat Arthritis, Diabetes, Colds, Cough, Sore throat, Chest congestion, Stomach pain, Constipation, Gall stones, Influenza, Cancer, etc.

On finding a potential compound for the treatment of cancer, researchers examined the low-toxic dichloromethane fraction of Oplopanax elatus, (OED) from the genus Oplopanax anticancer effects on colorectal cancer.

According to the tested analysis,
* The 5 μg/ml of OED combined with 40 μM of irinotecan possessed significant synergetic inhibition on SW-480 cells with a combination index (CI) of 0.56.

* The percentage of apoptotic cells was significantly increased from 69.57% (40 μM of irinotecan) or 72.7% (5 μg/ml of OED) to 95.6% after treatment of OED combined with irinotecan (OCI).

In other words, OED and irinotecan exerted a strong synergistic apoptotic effect against the tested cells by activating the protein associated with pro-apoptosis.

Based on the evidence, researchers said, "OED could enhance antiproliferative effects of irinotecan on colorectal cancer cells, which was related to the induction of apoptosis and regulations of activity of caspase-3".

Taken altogether, Devil's club may be considered a remedy for the treatment of colorectal cancer, pending to the confirmation of the larger sample size and multicenter human study.
