Tocopherols Inhibits the Onset of Colon Cancer in Vitro

The colon also is known as the large bowel or large intestine, formed parts of the digestive system in our body.

The colon consists of four parts: descending colon, ascending colon, transverse colon, and sigmoid colon.

The main functions of the colon are to reabsorb fluids and process waste products from the body and prepare for its elimination.

Colon cancer is a chronic condition caused by cell growth irregularly and disorderly in the colon due to the alternation of DNA.

Most cases of colon cancer start in the cells on the surface of the inner lining of the colon. The stage of colon cancer is depended on how far the alternated cells have spread in the colon.

Patients with the early stage of colon cancer, may not experience any symptoms. But as the tumor spread and expanse, it may suppress the nearby tissue and nerve, causing symptoms of locally severe pain and discomforts.

Although there are many risk factors associated with the early onset of colon cancer, a poor diet high in saturated fat and trans fat, red meat and processed foods have been found to play a major role in the development and recurrence of colon cancer in the Western world.

According to the statistic by the American Cancer Society, in the US, there are
97,220 new cases of colon cancer diagnosed in 2018. The lifetime risk of developing colorectal cancer is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

The causes of colon cancer are unknown but certain risk factors, such as genetic preposition and chronic inflammation induced by colonic diseases are associated with the onset of the disease.

According to the statistics provided by the Canadian Cancer Society, In Canada, the 5-year net survival for colorectal cancer is 65%.

In other words, out of 100 people diagnosed with colon cancer, 65% of people will survive for at least 5 years after their diagnosis.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of chronic diseases, researchers examined the effect of co-treatment of γ-tocopherol and 5-FU in enhancing the efficacy of chemotherapy against HT-29 colon cancer cells.

According to the analysis,
* The IC50 values for 5-FU and γ-tocopherol were 21.8  ± 2.5 and 14.4 ± 2.6 μM, respectively, and also this combination therapeutic increased the percentage of apoptotic cells from 35% ± 2% to 40% ±  4% (P  <  .05).
* Incubation HT-29 colon cells with combined concentrations of two drugs also caused significant accumulation of cells in the subGsubG1 phase.

After talking about other factors into account, researchers said, "Our results presented the combination therapy with 5-FU and γ-tocopherol as a novel therapeutic approach, which can enhance the efficacy of chemotherapy".

Taken altogether, tocopherols may be considered an adjunct therapy in the treatment of colon cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
