Angelica Sinensis, A Natural and Potential Anti Liver Fibrosis Herbal Medicine

Fibrosis is a medical condition caused by the excessive formation of fibrous connective tissue in an organ or tissue.

The liver is the largest internal organ that not only plays an essential role in cleansing the blood coming from the digestive system before passing them to other parts of the body but also secrets cholesterol to aid digestive function, produce steroid hormones and vitamin D and build strong cell walls.

Hepatic fibrosis is a condition characterized by overly exuberant wound healing causing excessive connective tissue builds up in the liver. In other words, hepatic fibrosis is a portion of the liver which was affected by scars formation.

The severe case of hepatic fibrosis can lead to cirrhosis, affecting the liver function in filtering the blood. In the US, over 4.5 million people are affected by liver cirrhosis, leading to the death of 12/100,000 of the population every year.

The disease in most cases is associated with the infection of the hepatitis virus. However, not all people with the virus will develop liver fibrosis.

Since liver fibrosis is a very slowly progressive disease, people who carry the disease may not experience any symptoms until the advanced stage.

Truly, people who consume excessive alcohol and carry the hepatitis virus may not develop the diseases until their later life.

The exact causes of liver fibrosis are unknown. However, certain risk factors such as repeated liver injury caused by a virus infection, and a blockage in the bile ducts are found to elaborate on the onset of liver fibrosis.

Liver fibrosis can also be induced by lactose intolerance, fructose, artificial sweeteners and other digestive disorder such as Crohn's disease, ulcerative colitis, celiac disease.

Long-term untreated liver fibrosis can lead to cirrhosis, a condition of the severe scarring of the liver and life-threatening liver disease.

Symptoms of liver fibrosis include appetite loss, difficulty thinking clearly, fluid in the abdomen, bleeding in the intestines, unexplained weight loss, weakness,..... and poor blood clotting.


On finding a natural ingredient for the prevention and treatment of liver diseases, scientists examined the effects of Levistilide A (C24H28O4, molecular weight = 380.48) derived from Angelica sinensis (Danggui) on liver fibrosis relating to angiogenesis.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to tested analysis of rats that were subcutaneously injected with CCl4 twice a week for six weeks to duplicate the liver fibrosis model and then treated with 10 mL/kg of normal saline, 4 mg/kg of sorafenib, and 3 and 6 mg/kg of Levistilide A for three weeks from the fourth week,
* Levistilide A attenuated LX-2 cell activation and HHSEC proliferation, inhibited the HHSECs to form tube-like structures in the formation of blood vessels and decreased the number of functional vessels in transgenic zebrafish.

* Furthermore, Levistilide A reduced collagen deposition in the induction of liver scar and the number of new microvessels ameliorated sinusoid capillarization and downregulated the expression of CD31, VEGF, and VEGF-R2 in forming new blood vessels.

In other words, Levistilide A can inhibit liver fibrosis through antiangiogenesis by alleviating sinusoid capillarization via the VEGF signaling pathway in the promotion of the new vessel development.

Based on the results, researchers suggest, "Levistilide A can inhibit liver fibrosis through antiangiogenesis by alleviating sinusoid capillarization via the vascular endothelial growth factor signaling pathway".

Taken altogether, Angelica may be considered a remedy for the treatment of liver fibrosis, pending to the confirmation of the larger sample size and multicenter human study.
