Phytochemical Indicaxanthin Blocks the Formation of Plague Against the Onset of Atherosclerosis

Atherosclerosis is a medical condition caused by plaque accumulated on the wall of arteries, leading to the narrowing of the artery and reducing oxygen risk blood flow to the heart and brain.

Depending on the location of the blockage, if the reduced blood flow occurred in the brain, it will cause the death of the brain cells, leading to stroke.

However, if the reduced blood flow occurred in the heart, it will cause damage to the heart tissues, leading to heart disease.

Most common disease associated with atherosclerosis is coronary artery disease. Plaques are made up of cholesterol, calcium and other chemical substances in the artery


Most people with atherosclerosis are asymptomatic at the early stage.

Some researchers suggested that atherosclerosis is a lipoprotein-driven disease. Therefore, people with high blood cholesterol are at an increased risk of atherosclerosis. Overuse of calcium supplement is also found to elevated the condition risk, particularly in the elderly.

Other risk factors that have bee found epidemiologically by studies are high blood pressure, triglycerides, smoking, insulin resistance, obesity or diabetes and inflammatory diseases, such as arthritis, lupus or infections.

Most cases of atherosclerosis can be reversed by making a change of lifestyle and moderate exercise.
Dr. Singh RB, the lead scientist wrote, "Adherence to diet and exercise was obtained through questionnaires and information obtained was quantified into a formula. After 24 weeks, the overall score of diet and exercises were significantly higher in group A(intervention diet( than in group B(control). There were a significant decrease in serum total cholesterol (13.3%), low-density lipoprotein cholesterol (16.9%), triglycerides (19.2%), fasting blood glucose (19.5%) and blood pressures (11.5/6.2 mm Hg) in the intervention group compared with initial levels and changes in group B".

Indicaxanthin is phytochemicals in the class of red and yellow indole-derived pigments of betacyanins, belonging to the group of betalains, found abundantly in beets, chard, etc.

With an aim to find a natural compound for the treatment of atherosclerosis, researchers at the Università di Palermo investigated the effect of phytochemical indicaxanthin (Ild) a major compound found in beetroot on human atheromas.

7-Ketocholesterol (7-KC)-induced the development of human atheromas, according to the kinetic measurements within 24 h, before injection of indicaxanthin showed the early formation of intracellular reactive oxygen species over basal levels, preceding NADPH oxidase-4 (NOX-4) over-expression and elevation of cytosolic Ca²⁺, involved in reactive oxygen species (ROS) production. and associated with the progressive depletion of antioxidant activity of total thiols. 

Furthermore, 7-KC- also induced the expression of NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) associated with the production of proinflammatory cytokines and innate and adaptive immune response.


Where 7-Ketocholesterol (7-KC) induced apoptosis of macrophages has been found to elevated the plaque progression and endoplasmic reticulum stress caused by secretion of pro-inflammatory, procoagulant, and proteolytic molecules by macrophages.

Believe it or not, Co-incubation of 2·5 μm of indicaxanthin completely prevented 7-KC-induced pro-apoptotic events.

In other words, indicaxanthin protects the macrophages integrity against apoptosis, thus reducing the risk of plaque accumulated on the wall of the arteries in facilitating the onset of atherosclerosis.

Based on the findings, researchers concluded, "The effects of Ind may be ascribed to inhibition of NOX-4 basal activity and over-expression, inhibition of NF-κB activation, maintaining cell redox balance and Ca homeostasis, with the prevention of mitochondrial damage and consequently apoptosis".


Taken altogether, phytochemical indicaxanthin (Ild) found in beetroot may be considered supplements for the prevention and treatment of atherosclerosis, pending to the confirmation of larger sample size and multicenter human study.

Intake of indicaxanthin (Ild) in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Scientists may have found a kitchen pungent spice for the treatment of rheumatoid arthritis with no side effects, according to studies.

Rheumatoid Arthritis is a chronic disorder characterized by inflammation of the flexible (synovial) joints and tissues and organs in the body.

The disease affects more women than men and generally occurs after the ages of 40, causing the diminished quality of life of many elders.

According to the statistics provided by the CDC, musculoskeletal disorders (MSDs) affects over 52 millions of adults in the US alone, including 294,000 children under age 18 with some form of arthritis or rheumatic conditions.

Rheumatoid arthritis was found to induce bone loss through elevating bone resorption without increasing bone formation. 

A cross-sectional population-based study of 1042 patients with rheumatoid arthritis showed that RA patients had an increased risk of death from various causes.

Patient with rheumatoid arthritis (RA) often associates to valgus deformities of the feet and deformities of gait due to body function due to an effort to support the collapsing longitudinal arch of the foot.  The kinematic and kinetic gait changes in rheumatoid arthritis (RA) are the result of a significant reduction in joint motions, joint moments, such as decreased hip flexion-extension range, hip abduction, knee flexion-extension range,...

Long-term bone damage and the prolonged onset of the disease can induce deformities of hand and wrist and small peripheral joints (such as fingers and wrist) due to chronic inflammation.

The effects of muscle strength in patients with RA may contribute to the prevalence of functional limitations physically that can affect daily living and quality of life.

Furthermore, rheumatoid nodules, lump on the skin, closed to the joint have been found to affect the joints of patients with RA.

Moreover, morning stiffness, a marker of inflammatory activity that reflects functional disability and pain is a very common symptom of patients in early stage of rheumatoid arthritis.

Dr. Motl RW and Dr. McAuley E. in the assessment of chronic diseases said, " The rationale and the associated onset of chronic disease conditions that influence function, disability, and quality of life (QOL) are embedded in the "Graying of America""(29), including patients with RA.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Scientists in seeking a potent natural ingredient or compound for the treatment of rheumatoid arthritis investigated ginger's traditional use as an anti-inflammatory against RA in Chinese and Ayurvedic medicine.

In joint swelling in an animal model of rheumatoid arthritis and streptococcal cell, wall-induced researchers compared crude ginger extract and a fraction containing only gingerols and their derivatives.

According to the assays, both extracts were efficacious in preventing joint inflammation.

However, the crude dichloromethane extract, containing essential oils and more polar compounds, was more efficacious in preventing both joint inflammation and destruction, compared to the fraction.

The results strongly suggested the use of ginger extract for the prevention of joint inflammation and destruction in RA.

Dr. Funk JL and colleagues at the University of Arizona wrote, " these data document a very significant joint-protective effect of these ginger samples and suggest that nongingerol components are bioactive and can enhance the antiarthritic effects of the more widely studied gingerols.

In order to reveal more information about ginger anti-RA property, researchers evaluated the anti-inflammatory effects of ginger's other secondary metabolites, the essential oils (GEO), containing terpenes in female Lewis rats with SCW-induced arthritis.

Injection of GEO (28 mg/kg/d ip) prevented chronic joint inflammation, without altering the initial acute phase of joint swelling and granuloma formation at sites of SCW deposition in the liver.

Compared to pharmacologic doses of 17-β estradiol (200 or 600 μg/kg/d sc), GEO elicited the same pattern of anti-inflammatory activity by its phytoestrogenic activity.

Based on the findings, Dr.Funk JL, the lead scientist wrote again in the final report, "ginger's anti-inflammatory properties are not limited to the frequently studied phenolics, but may be attributable to the combined effects of both secondary metabolites, the pungent-tasting gingerols and as well as its aromatic essential oils".

Take all together, ginger processed a high amount of bioactive compounds and secondary metabolites may be considered a functional alternative for prevention and treatment of RA, pending to the confirmation of large sample size and multicenter human study.

Gut microbiota or gastrointestinal microbiota is the complex community of over 10 trillions microorganisms living in our digestive tracts.

The study included 5 experimental groups (n = 11) given different dietary treatments 7 days before slaughter: negative control (basal diet), positive control (basal diet + 9% chicory-inulin), basal diet + 4.1% Jerusalem artichoke, basal diet + 8.1% Jerusalem artichoke and basal diet + 12.2% Jerusalem artichoke.

Increasing levels of Jerusalem artichoke to entire male pigs reduced skatole, a mildly toxic compound in digesta from colon and in feces and level of Clostridium perfringens, type of bad bacterias in the gut in both colon and rectum.

Furthermore, injection of JA also increased the total amount of SCFA (linear, P < 0.05), acetic acid (linear, P < 0.05) and valerianic acid (linear, P < 0.01) produced by the friendly bacteria in your gut in feces.

JA improved intestinal microflora by reducing skatole levels through decreasing in C. perfringens and the increase in SCFA with a subsequent reduction in pH.

Taken altogether, artichoke may be considered a functional formula for the improvement of the intestinal microflora, with no side effects, pending to the validation of larger sample size and multicenter human study.

The study included 80 overweight women completed the randomized crossover study randomly assigned to consumed dried sea buckthorn berries (SBs), sea buckthorn oil (SBo), sea buckthorn phenolics ethanol extract mixed with maltodextrin (SBe+MD) (1:1), or frozen bilberries.for 30 days.

By comparing the metabolic profile that reflected higher cardiometabolic risk(group B) at baseline, and participants who had a lower-risk profile (group A), researchers showed that bilberries caused beneficial changes in serum lipids and lipoproteins in group B, whereas the opposite was true in group A.

In other words, bilberry exerted a protective effect against people with cardiometabolic risk without causing harm to people with lower risk profiles.

In order to reveal more information about bilberry inhibition of cardiovascular risk factors researchers investigated regular consumption of bilberries effects on CVD risk reduction, such as decreasing low-density lipoprotein cholesterol (LDL-C) and triglycerides (TGs) and increasing high-density lipoprotein cholesterol (HDL-C).

The study involved women (n=25) and men (n=11) who consumed 150 g of frozen stored bilberries 3 times a week for 6 weeks.

During the experiment, the consumption of bilberries led to a decrease in total cholesterol, LDL-C, TG, glucose, albumin, γ-glutamyltransferase, and a positive increase in HDL-C (P=.044). In both women and men.

However, in men, additionally, favorable changes were observed in total cholesterol, glucose, albumin, aspartate aminotransferase, γ-glutamyltransferase, and HDL-C.

Dr. the lead scientist said, "the regular intake of bilberries can be important to reduce CVD risk, by decreasing LDL-C/TG and increasing HDL-C".

Taken altogether, bilberry may be considered a functioning remedy for the inhibition of cardiometabolic risk factors and prevention of CVD, pending to the validation of larger sample size and multicenter human study.

Phytochemical 3,3'-Diindolylmethane, the Natural Treatment for Breast cancer in Vivo and Human Studies

Cancer is a class of diseases in which a group of cells growing and multiplying disorderly and uncontrollably in our body.

Breast cancer (malignant breast neoplasm) is a medical condition characterized by irregular cells growth in the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk. 

There are also rare cases that breast cancer started in other areas of the breast. 

In 2010, over 250,000 new cases of breast cancer were diagnosed in women in the US and the risk of getting invasive breast cancer during the lifetime of a woman is 1/8.

In the early stage of breast cancer, patients may experience symptoms of the lump (mass) in the breast. and in the armpit (lymph nodes), nipple discharge, inverted or retracted nipple, scaly or pitted skin on nipple,  persistent tenderness of the breast and unusual breast pain or discomfort.

DIM combined with Herceptin enhances the effectiveness of Herceptin by significantly reducing cancer cell viability, and increasing apoptosis-induction and inhibition of colony formation, compared to the use of the chemo drug alone.

The combination also induced cancer cell death by inhibiting the expression of proteins associated with the production of proinflammatory cytokines, cancer cell division and proliferation and migration.

Furthermore, the combination also significantly exhibited the expression of miR-200, which plays an essential in blocking tumor cell invasion and metastasis and reduction of FoxM1 expression in the promotion of cancer cell proliferation.

Dr. Ahmad A, the lead scientist said, "DIM-plus Herceptin therapy could be translated to the clinic as a therapeutic modality to improve treatment outcome of patients with breast cancer, particularly for the patients whose tumors express high levels of HER-2/neu".

Moreover, in order to know more information about DIM anti-breast cancer activity, researchers investigated the bioactive compound against the transcription factor Forkhead Box M1 (FoxM1) associated with aggressive human carcinomas, including breast cancer.

3,3'-diindolylmethane (DIM), a nontoxic dietary chemopreventive agent showed to inhibit the FoxM1 in various breast cancer cell lines, according to the observation of gene transfection, real-time reverse transcription-PCR, and Western blotting.

Similar to the aforementioned results, DIM combined with Taxotere enhanced Taxotere-induced growth inhibition of breast cancer cells and decreased the invasive capacity of breast cancer cells compared to treatment alone.

Most importantly, the combination treatment also significantly inhibited tumor growth in severe immunodeficiency (SCID) mice,

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of breast cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
