Herbal Angelica Inhibits the Onset of Osteonecrosis in Femoral Head (ONFH)

Osteonecrosis is a medical condition associated with the death of bone cells caused by the reduced blood flow.

Osteonecrosis of the femoral head (ONFH) is the endpoint of a variety of disease processes that decreased blood flow within the femoral head.

Patients with osteonecrosis at the early stage are asymptomatic. However, as the disease gets worse, progressive osteonecrosis can induce symptoms of pain.

Patients with long-term use of high-dose steroid medications and excessive alcohol intake are at the increased risk of the disease developing.

From the age perspective, people between the age of 30 to 50 are also at a higher risk of the disease.

Other risk factors associated with the disease prevalence are Injuries, that damage to nearby blood vessels and reduce blood flow to bones.

Diseases associated with the osteonecrosis are
* Pancreatitis
Pancreatitis is a condition of inflammation of the pancreas that affects the function of the pancreas in secreting digestive juices, or enzymes. Pancreatitis can be acute or chronic.

* Diabetes
Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream to regulate the glucose.

In other words, diabetes occurs when cells in the pancreas dying off or receptor sites clogged up by fat and cholesterol. In some cases, diabetes is also caused by allergic reactions of cells in the immune system.

* Systemic lupus erythematosus
The systemic lupus erythematosus (SLE) or Lupus is a chronic, autoimmune disease as a result of the development of autoantibodies that attack the systems and organs in the body.
* Sickle cell anemia

* Sickle cell anemia (SCD), a genetic preposition form of anemia is a condition associated with not enough healthy red blood cells to carry adequate oxygen throughout your body.

The red blood cells in sickle cell anemia patines are rigid and sticky with the shapes of sickles or crescent moons.


Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

On finding a potential herbal remedy for the prevention and treatment of bone disease, researchers examined the herbal formula Huo Xue Tong Luo capsule (HXTL capsule) containing Peach kernel, Safflower Carthamus, Angelica Sinensis, Ligusticum wallichii etc, effects on osteonecrosis of the femoral head (ONFH).

According to the results from the experiment, 
* HXTL capsule promoted osteogenesis in rat MSCs.

* HXTL capsule silences the endogenous lncRNA-Miat involved in vascular dysfunction to promote osteogenesis of rMSCs.

* Additionally, HXTL capsule significantly increased the occupancy of H3K27me3 and decreased H3K4me3 in promoter regions of Miat to improve blood circulation.

Based on the evidence, researchers said, "HXTL capsule can promote osteogenesis to ameliorate ONFH through inhibiting the transcriptional expression of Miat, at least partially".

Taken altogether, Herbal Angelica used in a combination with other herbal medicine may be considered remedies for the treatment of osteonecrosis of the femoral head (ONFH)., pending to larger sample size and multicenter human study.
