Beta-Sitosterol Inhibits the Risk of Chronic Inflammation in Vivo

By Kyle J. Norton

Inflammation is the natural response of the immune system macrophages, the first line of defense in stimulating the production of inflammatory cytokines to protect our body against the infection caused by foreign microorganisms during the acute phase of body damage and injury.

Truly, the main focus of inflammation is to repair damage and injury.

Most cases of infection associated with inflammation are stopped at the acute phase. The wound or injury is slowly recovered.

However, overexpression of inflammatory cytokines in most cases can cause improper healing with scar formation or the death of nearby cells.

Any inflammation that lasts more than 3 months is considered chronic inflammation.

Chronic inflammation is the result of the immune system's inability to kill off all the microorganisms participated in the acute phase of infection, leading to the forming of autoimmune and chronic inflammatory diseases such as RA and diabetes.

Inflammation also can be caused by overexpression of free radicals compared to the antioxidant enzymes in the body. This result can lead to an imbalance of the ratio of the levels of free radicals and antioxidants, the major cause of oxidative stress.

However, people with a weakened immune system, acute inflammatory progression cannot be stopped due to the inability of the immune system to destroy all invaders, which may lead to low-grade chronic inflammation.

Epidemiologically, diseases associated with chronic inflammation are asthma, chronic peptic ulcer, tuberculosis, rheumatoid arthritis, periodontitis, inflammatory bowel inflammatory disease (ulcerative colitis and Crohn's disease), sinusitis and hepatitis.

Anti-inflammation is the process that inhibits inflammation or swelling without affecting the immune property to curb foreign pathogens.

Conventionally, the most common use of anti-inflammatory medicines is over counter aspirin, ibuprofen (Advil, Motrin, Nuprin) and other prescription NSAIDs.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of infectious diseases, researchers examined the topical anti-inflammatory effect of the chloroform extract from Achillea ageratum (Asteraceae) and of stigmasterol and beta-sitosterol on mouse ears edema.

According to the chemical analysis, on 12-0-tetradecanoylphorbol acetate (TPA)-induced mouse ear edema,* All the doses assayed (1, 3 and 5 mg/ear) significantly reduced the edema (50%, 66%, and 82%, respectively),

* The isolated sterols stigmasterol and beta-sitosterol (with doses of 0.5 mg/ear) had similar effects as those of extract with doses of 1 and 3 mg (59% and 65% respectively).

Furthermore, In the chronic model, the anti-inflammatory effect generally more moderate mice,
* The highest dose of the extract decreased the edema reduction to 26% with the highest dose of the extract applied, compared to the compounds the effect that decreased to 36% with stigmasterol, and 40.6% with beta-sitosterol.

* Moreover, Myeloperoxidase activity (MPO) associated with oxidative stress was reduced by the extract and the compounds in the acute model,

Based on the evidence, researchers said, "These results indicate that the chloroform extract of Achillea ageratum and some of its components stigmasterol and beta-sitosterol are more effective as topical anti-inflammatory agents in acute than in the chronic process and their action is markedly influenced by the inhibition of neutrophil migration into inflamed tissue".

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of acute and chronic inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Ashwagandha Protects the Neurons Against Excitotoxicity

By Kyle J. Norton

A neuron or nerve cell made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral nerves that play an essential role to process and transmit information.

The function of neurons can be classified into 3 types
* The sensory neurons found within the nervous system that convert external stimuli from the organism's environment into internal electrical impulses.

In other words, sensory neurons are activated by sensory input from the environment, including physical input of sound, touch, heat, or light.

*The motor neurons transmit impulses from the spinal cord to skeletal and smooth muscles,. In other words, motor neurons directly control all of the muscle movements.

* The interneurons connect the motor and sensory neurons. In other words, the interneurons transfer signals between sensory and motor neurons.

The most common diseases associated with neuron dysfunction are amyotrophic lateral sclerosis (ALS). Lou Gehrig's disease or classical motor neuron disease that disrupts signals to all voluntary muscles on both sides of the body.

Excitotoxicity is the pathological process associated with neurons damage and killed by the overactivations of receptors for the excitatory neurotransmitter glutamate, such as the NMDA receptor and AMPA receptor.

Where NMDAR (The N-methyl-D-aspartate receptor) is a glutamate receptor and ion channel protein found in nerve cells and glutamate is the anion of glutamic acid which plays a critical role as a neurotransmitter.

Excitotoxicity has been found to induce, neurodegenerative diseases, including multiple sclerosis, Alzheimer's disease, amyotrophic lateral sclerosis (ALS), Parkinson's disease.

Most cases of excitotoxicity have been found to be caused by spinal cord injury, stroke, traumatic brain injury,...

On finding a potent compound that processes the neuroprotective effect, researchers examined the effects of Withania somnifera anti excitotoxicity that lead to neurons' damage or killed.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

According to tested differentiation, Withanone isolated from Withania somnifera showed a significant activity to protect the neurons against NMDA-induced excitotoxicity in retinoic acid, differentiated Neuro2a cells.

Furthermore, the pretreatment of Withanone at 5, 10 and 20 μM doses were found to inhibit the NMDA induce- significant neuron cell death through several parameters associated with the accumulation of intracellular Ca2+,  generation of reactive oxygen species (ROS), loss of mitochondrial membrane potential.

Additionally, Withanone also improved the ratio of Bax/Bcl-2 ratio, the release of cell cytochrome c and lipid peroxidation involved in neuron DNA damage and the death of neurons.

Based on the findings, researchers said," All these parameters were attenuated with various doses of Withanone pre-treatment. These results suggest that Withanone may serve as a potential neuroprotective agent".

Taken altogether, Ashwagandha processed a high amount of antioxidant Withanone may be considered a functional herb for the prevention and treatment of neurological diseases including Parkinson's disease, Alzheimer's disease, pending to the result of larger sample size and multicenter study

Omega 3 Fatty Acids Inhibit Inflammatory Allergic Diseases

By Kyle J. Norton

Chronic allergic airways disease (AAD) is a group of the disease characterized by airway inflammation and airway hyperresponsiveness (AHR).

Airway inflammation is the hallmark of chronic obstructive pulmonary disease (COPD) associated with low-grade inflammation caused by persistent production of proinflammatory cytokines by the immune system in the airway.

On the other hand, airway hyperresponsiveness (AHR) is a cardinal feature of asthma associated with the response of the immune system against pathogens.

Most common symptoms of coughing, wheezing, and shortness of breath in reactive airway disease (AAD) may not be due to asthma or other chronic airways conditions.

Conventionally, reactive airway inflammatory disease is treated with medication to control symptoms such as long-term use of an inhaler to relieve attacks and improved quality of life through avoidance of irritants.

Asthma is a chronic inflammatory disease affecting the airway of the lung with recurring symptoms, such as wheezing, chest tightness, shortness of breath, and coughing. The disease affects people of all ages, and mostly starts during childhood.

During the asthma attacks, patients may experience symptoms of difficult breathing, and severe wheezing in the breath and coughs that won't stop.

According to the statistic provided by the Centers for Disease Control and Prevention (CDC), 1 in 13 people in the US has asthma.

In other words, more than 26 million Americans have asthma, with half of the patients are children.

If you experience symptoms of a cough which often worse at night or early in the morning, wheezing in your breath, accompanied by chest tightness and shortness of breath, you may be at the early stage of asthma. Please check with your doctor to rule out the possibility.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of chronic inflammatory diseases, researchers examined the causal relationship between decreased intake of fish oil in modernized diets and an increasing number of individuals with asthma or other allergic diseases.

According to the tested differentiation, the pro-resolving mediators (SPM: protectins, resolvins, and maresins) generated from omega-3 fatty acids such as EPA and DHA via several enzymatic reactions showed a strong impact in the regulation of the airway eosinophilic inflammation and promotion of the resolution of inflammation in vivo.

The efficacy of the mediator in the inhibition of allergic diseases associated with inflammation was attributed to the improvement of the impairment of the biosynthesis of SPM.

The clinical lipidology wrote, "Defects in anti-inflammatory/pro-resolution lipid mediators can contribute to inflammatory disease severity. Anti-inflammatory/pro-resolution lipid mediators can be used to develop therapeutics that enhance rather than block the endogenous regulation of inflammation".

In other words, diseases associated with chronic inflammation in the lung might be a result of a resolution defect.

Taken altogether, Omega-3 fatty acids may be considered a functional food for the prevention and treatment of allergic diseases caused by SPM defect, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Red Cabbage May Process a Significant Antioxidant and Cytotoxic Activity

By Kyle J. Norton

Cytotoxicity is the process in the induction of cell death through toxins.

In other words, cell cytotoxicity is the process to use certain chemicals to destroy living cells.

In general, cytotoxicity can cause the death of healthy cells

In cancers, cytotoxicity can be benefits to humans by destroying the cancer cells and prevent them from proliferation.

For example, cytotoxicity induced by oxidative stress due to the imbalanced ratio of free radicals and antioxidant enzymes produced by the body can cause protein, lipid and cellular damage that leads to chronic low-grade inflammatory diseases and cancer.

More precisely, most cases of cancer cell cytotoxicity are associated with the stimulation of protein and pathways involved in either gene suppression or cell death programming.

The aim of the cytotoxic studies is to find the minimal quality that is essential for the successful development of pharmaceutical or cosmetic preparation that induces cellular toxicity for the treatment of cancer.

Before the preparation can be used in humans, bioassays associated with a number of different cell lines in vitro and Vivo have been used to assess the cytotoxicity of many herbal plants, including those of African medicinal plants.

However, stimulation of ROS and antioxidants production to induce cytotoxicity to the tumors can also cause cancer cell death without damage to nearby healthy cells.

Many herbal medicines and foods such as fennel (*) , Onion (*), and tomato(*) have been found not only to promote the chemo drug cytotoxic activity against target tumors, but also protect the healthy cells against the chemo drugs side effects.

With an aim to find a natural food for the treatment of cancers, researchers the roles of zeatin (2 mg/L) on antioxidants and cytotoxic activity in regenerated shoots of red cabbage.

Red cabbage is a species of Brassica oleracea belongings to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem of which is crowded with a mass of leaves.

Based on the results of differentiation,
* The extract of a plant treated by 2 mg/L zeatin gives the highest content of total phenolics (5.18 mg gallic acid equivalent/g dry weight) and flavonoids (1.52 mg rutin equivalent/g dry weight).

* Furthermore, various bioactive compounds in red cabbage are significantly enhanced with increasing zeatin concentration.

* In vitro shooting culture using 2 mg/L zeatin displayed higher antioxidant activity in all assays (DPPH, FRAP, and ABTS) compared to control with respective values of 68.12%, 73.28%, and 54.1%, respectively.

* Moreover, the extracts of red cabbage explant treated by 2 mg/L zeatin exhibited the strongest cytotoxic effect towards cancer cells compared to control.

Taken altogether, red cabbage combined with Zeatin may be considered a functional food that processes antioxidant and cytotoxic capacity, pending the confirmation of the larger sample size and multicenter human study.

Green bean Processes the Anti-Proliferation Against the Onset of Cancer

By Kyle J. Norton

Cancer is a class of diseases characterized by irregular cell growth that starts in the tissue of specific organs.

Most cases of cancer overtime after penetrating into deeper layers of the affected organs, the oversize tumor can sometimes spread to other organs in the body via lymph or blood, leading to secondary metastasis.

Proliferation is a process of cancer growth associated with an abnormally high rate of cancer cells division, leading to rapid overgrowth of the tumor.

Cancer cell proliferation by dividing unlimitedly, compared to normal cells that die after a certain cell cycle division.

More precisely, a primary tumor or original tumor that the tumor progression to yield a cancerous mass. Most cancers can not be detected until they formed a small grape-like lump. Cancer cells need 42 doublings before it can produce enough cancer cells to cause death.

According to the statistics provided by the National Cancer Institute, In 2018, approximately 1,735,350 cases of cancer were diagnosed in the US and the disease also caused the death of 609,640 Americans.

Epidemiologically, the odds of cancer incidence is 439.2 per 100,000 men and women per year based on 2011–2015 statistics.
Green bean is a genus of Phaseolus Sensu Stricto, containing 55 difference species, belong to the family Fabaceae and native to the New World tropics and grown in Europe for its attractive flowers and fleshy immature pods. It can be classified into two major groups, bush beans, and pole beans.

On finding a potent veggie for the prevention of cancer, researchers investigated the beans antiproliferative activity in tested cancer cell lines.

Based on the tested analysis, beans polyphenols, such as gallic acid, chlorogenic acid, epicatechin, myricetin, formononetin, caffeic acid, and kaempferol exerted a significant antioxidant expression against proteins, lipid and DNA damage in the induction of oxidative stress ranged from 1.568 ± 0.041 to 66.572 ± 3.197 mg.

Furthermore, bean extracts showed obtained from the non-pigmented samples inhibited the proliferation of the human epithelial colorectal adenocarcinoma (Caco-2) cells, human breast cancer cells MCF-7, and A549 NSCLC cell line used in the experiment.

More importantly, the qualitative and quantitative differences in phenolic composition between the groups of beans influenced antioxidant and proliferative efficacy biologically.

In other words, all beans groups exert significant anti-proliferation in the cancer cells depending on bean types but did not show significant differences in the biological activities within the same variety, before and after cooking.

Taken altogether, green beans may be used as an adjunct therapy for the prevention and treatment of cancer, pending to the confirmation of larger sample size and multicenter human study.

Phytochemical Omega-3 Fatty Acids Induce the Death of Ovarian Cancer Cells In Vitro

By Kyle J. Norton

Cancer is the class of chronic disease caused by irregular cell growth in the tissue or specific organ in the body due to the alternation of cell DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite, and fatigue but also localized symptoms depending on the affected tissues and organs.

The ovaries are oval-shaped ovum-producing reproductive organs formed part of women's reproductive system that connects to the fallopian tubes and attaches to the outer layer of the uterus via the ovarian ligament. Usually, the left and right ovaries take a turn to release an egg every month.

In other words, the main functions of the ovary play a critical role in producing oocytes (eggs) for fertilization and reproductive growth hormones, estrogen, and progesterone.

According to the statistics, ovarian cancer is one of the fifth most common cancer, accounting for more deaths than any other cancer of the female reproductive system in the US.

The lifetime risk of getting ovarian cancer in a woman is about 1 in 78.comapred to the lifetime of dying from ovarian cancer risk of 1 in 108.

The exact causes of ovarian cancer are not identified. However, epidemiological studies suggested that women who have endometriosis, genetic preposition such as BRCA1 and BRCA2 gene, acquired genetic mutation such as the TP53 tumor suppressor gene are susceptible to the substantial risk of the development of the disease.

Besides sharing the common symptoms of other types of cancer, most ovarian cancer patients may experience symptoms of discomfort in the pelvic area, changes in bowel habits, such as constipation. and a frequent need to urinate.

According to the statistics provided by the Canadian Cancer Society, in Canada, the 5-year net survival for ovarian cancer is 45%.

In other words, on average, 45% of women diagnosed with ovarian cancer will survive for at least 5 years.

On finding a potential phytochemical for the treatment of cancer, researchers investigated the effect of OM-3FAs on epithelial ovarian cancer cell lines.

According to the results of 3 epithelial ovarian cancer cell lines (SKOV-3 [p53 null], TOV-21G [wt p53] and OVCAR-3 [mutant p53]) and one immortalized ovarian surface epithelial cell line (IOSE-29 [wt p53]) treated with OM-3FAs, OM-3FAs showed a statistically significant inhibitory effect in all four cell lines.

Furthermore, in the p53 null and mutant cell lines, SKOV-3 and OVCAR-3, the application of OM-3FAs showed inhibitory activity without inducing apoptosis and down-regulated expression of VEGF.

However, the apoptosis and/or VEGF down-regulation was strongly evident in the wt p53 cell lines TOV-21G and IOSE-29.

Collectively, researchers said, "under the influence of OM-3FAs, there are definitive growth-suppressive mechanisms at work and that the biologic effects of OM-3FAs may in part be mediated by the p53 status".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the treatment of ovarian cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Omega 3 Fatty Acid, a Natural Source of Inflammation Relief

By Kyle J. Norton

Inflammation is a systematic immune response during the acute phase of infection.

Macrophages, one the lymphocytes and the first-line defense of the immune system after sensing the damage of localized tissue, stimulate the production of proinflammatory cytokines to the site to protect the body against invasion of potential pathogens.

Acute inflammation is a natural response of the immune system to protect the human body against harmful stimuli and pathogens.

Acute information is the first phase of injection.

Acute inflammation usually receded after a few days or weeks depending on the size and damage. However, if all pathogens were not eradicated after 3 to 8 weeks, the immune system will compromise, leading to chronic low-grade inflammation, a leading cause of chronic illness.

The most common symptoms of acute inflammation occurred with a short period of time such as in hours with pain, redness, immobility, swelling, and heat.

Conventionally, depending on the types of acute inflammation, non-steroidal anti-inflammatory drugs (NSAIDs) may be prescribed to alleviate the pain by counteracting an enzyme that contributes to inflammation.

Chronic inflammation is a result of a prolonged period of inflammation that lasts for several months and even years caused by the inability of the immune system to kill off all harmful microorganisms during the acute phase of infection.

Chronic inflammation can also be caused by an autoimmune disorder in which the immune system continuously attacks normal and healthy tissue, by mistaking them for pathogens.

Low-grade inflammation is a different type of chronic inflammation characterized by a production of the inflammatory cytokines in a low-grade state.

On finding a potential phytochemical for the treatment of infectious diseases, researchers investigated the protective effects of omega-3 polyunsaturated fatty acids (ω-3 PUFAs) against hepatic steatosis-induced inflammation and its potential molecular mechanisms in hepatocyte of large yellow croaker (Larmichthys crocea).

According to the experimental results, ω-3 PUFAs exerted anti hepatic steatosis-induced inflammation activity.

The SIRT1 gene associated with cellular regulation that decreases in the hepatic steatosis-induced inflammation was increased by ω-3 PUFAs.

Furthermore, the Nuclear factor-kappa B (NF-κB) is a ubiquitous transcription factor that is involved in inflammatory and immune responses were significantly decreased after ω-3 PUFAs treatments compared to the palmitic acid stimulation group.

In other words, ω-3 PUFAs alleviate hepatic steatosis-induced inflammation through Sirt1-mediated nuclear translocation of NF-κB p65 subunit in hepatocytes of large yellow croaker.

Based on the findings, researchers concluded, "The present study provides important insight into the mechanisms of the protective effects of ω-3 PUFAs, providing theory bases for alleviating the hepatic steatosis induced inflammation of farmed fish, thereby offering great benefits to the aquaculture industry and fish consumers".

Taken altogether, Omega-3 fatty acids may be considered a functional food for the prevention and treatment of diseases involved in inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
