Green Coffee, The Beverage Which Processes Anti Obesity Activity in Vitro, Scientists Reveal

Coffee may be considered as a function beverage in reducing obesity prevalence and management, some scientists suggested.

According to the information of the World Health Organization, in 2016, approximately, 650 million adults were obese and 39% of adults aged 18 years and over (39% of men and 40% of women) were overweight.

Believe or not, overall, about 13% of the world's adult population (11% of men and 15% of women) were obese in 2016. And the prevalence of obesity has increased at an alarming rate due to 30 years of economic prosperity in the Far East.

Sadly, the organization wrote, "The worldwide prevalence of obesity nearly tripled between 1975 and 2016".

Dr. Janssen I, in the study of "The public health burden of obesity in Canada" wrote, " At present, there are approximately 7 million obese adults and 600 000 obese school-aged children in Canada. Prevalence estimates based on waist circumference are even more troubling as they indicate that 37% of adults and 13% of youth are abdominally obese".

Truly, obesity in Canada not only is a major contributor to morbidity and mortality but also places a large economic burden on the country.

The doctor continued, " In 2006 obesity accounted for $3.9 billion in direct health care costs (e.g., hospitalizations, medications, physician and emergency room visits) and $3.2 billion in indirect costs (e.g., costs related to disability and lost productivity due to illness or premature death)".

Coffee, a popular and social beverage all over the world, particularly in the West, is a drink made from roasted bean from the Coffea plant, native to tropical Africa and Madagascar.

According to the joint study led by the Huanggang Normal University, the health benefits of coffee studies over last 10 years, expressed a significant improvement of risk and treatment of obesity in decreased lipid accumulation in cells through regulating the cell cycle during fat metabolism.

Additionally, coffee consumption induced changes in transcription factors in fat deposit decreased body weight and visceral fat form of animal and humans studies.

Furthermore, coffee influenced gut microbiota in obese animals and humans also played an important role in the use as a functional and integrated food in reducing risk and treatment of obesity.

The study of obesity-induced mice that fed with the following patterns only HFD or HFD with GCBE at 50, 100, and 200 mg/kg for four weeks, researchers found that
* Mice treated with green coffee injection show a significantly decreased body weight gain, liver weight, and white adipose tissue weights through the regulation of adipose tissue lipolysis hormones, such as adiponectin and leptin.

* Mice treated with green coffee injection also demonstrate a decreased mRNA expression levels of adipogenesis and adipocyte metabolis related genes in adipose tissues and the liver and decrease the corresponding protein expression.

* GCBE treated mice had a lower fat mass with decreased relative body weight and fat mass, compared to HFD alone fed mice.

These results suggested that green coffee intake exerted a strong activity in the regulation of adipogenesis and lipid metabolism which are found to associate to enhance weight gain.

According to the information collected, Dr. Choi BK, the lead author said, "GCBE has a potential anti-obesity effect with lowering body fat accumulation by regulating adipogenesis and lipid metabolism-related genes and proteins in WAT and liver".

More importantly, in the study of male day mice fed a standard diet containing GCBE and its principal constituents, namely, caffeine and chlorogenic acid, for 14 days, researchers at the Oryza Oil & Fat Chemical Co,, Ltd, indicated that
* GCBE with 0.5% and 1% reduce visceral fat content and body weight.

* Caffeine and chlorogenic acid show a tendency to reduce visceral fat and body weight.

* Oral administration of GCBE (100 and 200 mg/kg. day) for 13 days shows a tendency to reduce hepatic TG in mice.

* Chlorogenic acid (60 mg/kg. day) reduces hepatic TG level.

The findings displayed a strong effect of coffee and its phytochemicals in reducing weight gain and fat accumulation by inhibition of fat absorption and activation of fat metabolism in the liver, therefore the beverage may be considered a functional and integrated food against risk and treatment of obesity.

Cruciferous Vegetables - The Best Whole Food Medicine That Process Anti Acute and Chronic Inflammatory Potency, Scientists Say

Intake of cruciferous vegetable may be inversely correlated with circulating of proinflammatory levels of in women, some scientists suggested.

Inflammation is a natural immune response involving the stimulated production of pro-inflammatory activity with a target to protect our body against foreign invasions such as virus and bacteria after injury and damage.

In other words, pro-inflammatory cytokines are activated through systematic immune response to the damage cells which release chemicals including histamine, bradykinin, and prostaglandins to protect the injure area or location against infection, leading to blood vessels to leak fluid into the tissues, causing swelling and other symptoms such as pain, swelling, and redness.

However, in most cases, over-response of the immune system can lead to over expressions of certain pro-inflammatory cytokines, depending to the injured location such as Interleukin (IL)-1 and tumor necrosis factor (TNF) which can lead shock and death of healthy cells in the infectious location. 

In most cases, long-term overexpression of pro-inflammatory cytokines can lead to chronic inflammatory disease as the immune system no longer can kill the invasive agents and heal the damage caused by acute infection.

Dr. Dinarello CA, the lead author in the study of pro and anti-inflammatory cytokines said, " Some cytokines act to make disease worse (proinflammatory), whereas others serve to reduce inflammation and promote healing (anti-inflammatory). Attention also has focused on blocking cytokines, which are harmful to the host, particularly during overwhelming infection".

This explanation showed that under normal conditions, some anti-inflammatory cytokines may have a strong effect in reducing inflammation in the body daily to speed up the healing, caused by various events.

Cruciferous vegetables are the group of vegetable belongings to the family Brassicaceae, including cauliflower, cabbage, cress, bok choy, broccoli etc.

In the study to investigate whether higher intakes of cruciferous vegetables or their constituents can decrease the levels of inflammation in humans, researchers conducted an experiment to evaluate associations of vegetable intake with a panel of inflammatory and oxidative stress markers among 1,005 middle-aged Chinese women.

* The differences in concentrations of inflammatory biomarkers between extreme quintiles of cruciferous vegetable intake were 12.66% for TNF-α (Ptrend=0.01), 18.18% for IL-1β (Ptrend=0.02), and 24.68% for IL-6 (Ptrend=0.02).

* Inverse association was found for intakes of all vegetables combined but not for noncruciferous vegetables

* Levels of the urinary oxidative stress markers F2-isoprostanes and their major metabolite, 2,3-dinor-5,6-dihydro-15-F2t-IsoP, were not associated with intakes of cruciferous vegetables or all vegetables combined.

These results suggested that daily intake of cruciferous vegetables may have a profound effect in reducing the risk of overexpression of inflammation in inducing chronic inflammatory diseases.

Dr. Jiang Y, the lead author, after taking into account co and con-founders said, " he previously observed health benefits of cruciferous vegetable consumption may be partly associated with the anti-inflammatory effects of these vegetables".

 Sulforaphane (SFN) and Diallyl disulfide(DAFDs), the naturally occurring isothiocyanate found in cruciferous vegetables, in the highly metastatic B16F-10 melanoma cells, both in vivo and in vitro models exerted a significant effect in killing cancer cells through a change of tumor cells membrane blebbing, DNA condensation, and also by nuclear DNA fragmentation.

Additional differentiation suggested that apoptosis of cancer cells induced by Sulforaphane (SFN) and Diallyl disulfide(DAFDs) was attributed to the involvement of proinflammatory cytokines, namely tumor necrosis factor-alpha (TNF-α), interleukin (IL)-1β, IL-6, IL-12p40, and granulocyte-macrophage-colony-stimulating factor (GM-CSF).

Dr. Pratheeshkumar P, the lead author of the study suggested, DADS induces caspase-dependent apoptosis through an induced cell death pathway in B16F-10 melanoma cells by activating...... and suppressing pro-inflammatory cytokines.

Other phytochemical found the vegetables, such as 3,3'-diindolylmethane (DIM), a derivative of indole-3-carbinol, also expressed anti proinflammatory effect through interleukin (IL)-1beta, IL-6 and tumor necrosis factor.

In fact, chemical compounds found in the vegetable inhibited the pro-inflammatory cytokines in the expression of chronic diseases and cancers, involved in other aspects, such as environment airborne particulate pollutants, according to the East Campus - Laurentian University.

However, due to the limitation of above studies, further data collection on large example size and multi-centers studies performed with human consumption of the whole food during the course of the disease will be necessary to complete the picture of garlic anti-cancer possibilities.

Arnica may have substantial and sustainable effect in reducing risk, progression, and treatment of leukemia, some scientists suggested.

Leukemia is a medical condition characterized by cells growth uncontrollable and disorderly produced by the body's blood-forming tissues, including the bone marrow and the lymphatic system. In most cases, it is the result of abnormal white blood cells, which don't function properly.

Further analysis, injection of helenalin inhibited HL-60 through activating the protein kinase C (PKC) inhibitor with a function to act as ruboxistaurin in signaling molecules that link to multiple cellular processes of cancer.

Moreover, researchers also found that helenalin also inhibits HL-60 proliferation through activating the extracellular signal-regulated kinase (ERK) inhibitors which are used in some cancer therapy in controlling diverse tumor cellular processes such as proliferation, survival, and motility.

Protein kinase C (PKC) is a family of protein kinase enzymes with function in controlling the expression of other proteins through certain mechanisms.

These result postulated that helenalin isolated from Arnica inhibits the expression of human promyelocytic leukemia HL-60 cell line through many different pathways involved several aspects in tumor cellular differentiation.

More importantly, in the different aspect of the study, Dr. Dirsch VM and colleagues found that sesquiterpene lactones also exerted a significant cytotoxic effect in triggering leukemia cell apoptosis.

After considering all other factors, the study strongly insisted, "Spent coffee grounds (SCG), rich in dietary fiber can be fermented by colon microbiota producing short-chain fatty acids (SCFAs) with the ability to prevent inflammation".

More importantly, in the examined numbers of prospective and retrospective cohort studies, coffee consumption against relative risk of developing major depressive disorder in humans, caffeine and phytochemicals such as chlorogenic acid, ferulic acid and caffeic acid, all important constituents of coffee, showed to attenuated risk of depression through mediated inflammation, in vitro and in vivo.

Dr. Hall S, the lead author said, "The ability of bioactive coffee constituents to modulate the parameters of neuroinflammation has been shown with caffeine having strong antioxidant properties in vitro, chlorogenic acid and the caffeic acid having strong anti-inflammatory and antioxidant properties in vitro and ferulic acid having activities in vivo animal models of depression".

Including a portion of carrot in your daily diet may have a protective effect against the liver diseases,  some scientists suggested.

Hepatoprotection is the ability of certain agents which process the properties to reduce the injurious effects or to preserve the normal hepatic physiological mechanisms. In most cases, liver injury is the result of overexpression of toxin accumulation in the liver.

The exact causes of liver diseases are unknown. However, researchers believed that liver disease can be caused by genetic mutation gene inherited from the parents or by viruses and excessive alcohol drinking.

Some researchers also suggested that liver damage also associated with patients who are obese. 

Long-term use of certain medications, such as antibiotics, anticonvulsants, and antidepressants are also found to damage the liver.

Untreated liver damage can lead to liver scars, which in long run can increase the risk of liver failure, a life-threatening complication.

Besides symptoms of digestive discomforts, depending on the stage of the live damage, patients may also experience symptoms of jaundice on skin and eyes, abdominal pain and swelling, particularly, swelling in the legs and ankles and darkening urine color.

Carrot, a root vegetable with an orange color is a subspecies of Daucus carota, belongings to the family Apiaceae, native to Asian and Europe.

In the investigation of the liver protective activity of methanolic extract of Daucus carota (D. carota) seeds in experimental rats model, researchers at the Rawatpura Sarkar Institute of Pharmacy assigned randomly into 4 groups of rats (two test, standard and toxic control).

Two test groups received D. carota seeds extract (DCSE) at doses of 200 mg/kg and 400 mg/kg. 

The standard group received silymarin (25 mg/kg) and toxic control received only thioacetamide. Control group received the only vehicle. 

All animal were given thioacetamide 100 mg/kg s.c to induce oxidative stress through overproduction of ROS and reduced levels of antioxidant in the hosts.

At the 8th day, in observation of animal liver before treatment researchers found that levels of the antioxidant enzymes produced by the host, such as superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GRD), glutathione peroxidase (GPX), glutathione-S-transferase (GST) were decreased and levels of lipid peroxidation (LPO) were significantly higher compared to control.

However, rat group treated with methanolic extract of Daucus carota (D. carota) seeds showed a significant decrease of  serum glutamic pyruvic transaminase (SGPT), serum glutamic-oxaloacetic transaminase (SGOT) and alkaline phosphatase (ALP) which are associated to several expressions of liver diseases and increased levels of natural antioxidants produced by the body.

These results suggested that methanolic extract of Daucus carota (D. carota) seeds processes properties on inhibition of oxidative stress facilitated liver damage and stimulation of the host in the production of antioxidant from the hosts.

In other words, methanolic extract of Daucus carota (D. carota) seeds exerted liver protective effect by restoring the ratio of levels of reactive oxygen species and antioxidants in the tested animals.

Further analysis of the efficacy of carrot, mango, and wheat extracts against carbon tetrachloride (CCl4)-induced oxidative stress and hepatotoxicity, researchers at the Ain Shams University conducted an animal study including 4 albino rats.

Rats used in the experiment were divided randomly into 5 groups. Group 1 was fed a basal diet and group 2 was fed a basal diet and CCl4. Groups 3, 4, and 5 were treated with carrot, mango, and wheat extracts, respectively, in addition to a basal diet and CCl4.

Observation of the tested rats liver fed with the extract displayed a anti hepatocellular toxicity caused by injection CCL4 decreased significantly compared to before treatment.

According to assays, lipid profile and liver enzymes markers were decreased remarkably and total protein and high-density lipoprotein (HDL) increased dramatically.

Makers of oxidative stress expressed through levels of Malondialdehyde (MDA) were also decreased noticeably

These results indicated a strong effect of the extracts in reduced lipid peroxidation by increased levels of antioxidant in inhibition of the reactive oxygen species in the induction of oxidative stress.

Dr. Ebeid HM, the lead author after taking into account of other co and confounders said, "Microscopic examination of the treated rats exhibited a normal histopathological structure" and "carrot, mango, and wheat extracts could be used as nutraceuticals for the prophylaxis and treatment against hepatotoxicity and oxidative stress".

Taken together, carrot and its bioactive extracts used alone or combined with mango, and wheat extracts may be considered as functional food for reduced risk of liver diseases caused by overexpression of free radicals.

Intake of supplementation should be taken with exceptional care in the prevention of acute liver toxicity.

However, additional data collection on large example size and multi-centers studies performed with human consumption of the whole food during the course of the disease will be necessary to complete the picture of carrots liver protective possibilities.
