Tocopherols Promote Cytotoxicity in the Sensitive and Drug-Resistant Cancer Cells

Cancer is the class of diseases associated with the growth of the cells disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.
The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Compared to all cancers, lung, breast, prostate, and colorectal cancers are the most common cancer in the US.

Drug-resistant cancer is a type of cancer that process chemotherapy resistance ability.

In other words, rug-resistant cancer processes the ability to survive and grow despite anti-cancer therapies. In some cases, cancers can evolve resistance to multiple drugs, called multiple drug resistance.

Cancer that responds to treatment with anticancer drugs is called sensitive-drug cancer.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of cancer, researchers examined the effect of the encapsulated tocopheryl succinate (PTX) loaded with PSST micelles (PTX/PSST-M) which are designed to display synergistic functions in anticancer activities.

According to the analysis,
* PTX/PSST-M showed significantly increased cytotoxicity against PTX-resistant human ovarian cancer A2780/PTX cells for 48 h,
* The efficacy was attributed to the equivalent IC50 value of PTX that was reduced from 61.51 to 0.49 μmol/L.

* Other contributors to the cytotoxic activity also included reducing the mitochondrial transmembrane potential, ATP depletion, ROS production, and activation of apoptotic pathways.

Additionally, researchers also found that the administration of PTX/PSST-M significantly increased cell apoptosis/necrosis and cell cycle arrest at the G2/M phase in A2780/PTX cells.

After talking about other factors into account, researchers said, " These results demonstrate that the redox-responsive PSST micelles inhibit P-gp activity and have a good potential to effectively reverse PTX resistance in human ovarian carcinoma cells by activating intrinsic apoptotic pathways".

Taken altogether, tocopherols may be considered an adjunct therapy in the treatment of drug-resistant cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol, the Potential Anti-Metabolic Dysfunction Antioxidant

Metabolic syndrome is a group of conditions associated with an increased risk of cardiovascular disease and types 2 diabetes, including high blood pressure, sugar, cholesterol and triglyceride levels and excess body fat around the waist.

Epidemiological studies suggested if you have 3 of the above syndromes, you have metabolic syndrome.

According to statistics, metabolic syndrome is a growing prevalence in the Western world, In the US, 30% of Americans are living with metabolic syndrome,

In other words, 1 in 5 people in Canada has metabolic syndrome, affecting 19.1% of all Canadian adults.

The CDC statistic suggested, among US adults aged 18 years or older, the prevalence of metabolic syndrome rose by more than 35% from 25.3% to 34.2% according to the data from 1988–1994 to 2007–2012.

The exact causes of metabolic syndrome are unidentified. However, researchers suggested eating too many calories, particularly in a high-fat diet, physical inactivity, and excessive weight gain are some of the most common and prevalent factors in elevated the risk of metabolic syndrome in the Western world.


Most people with metabolic syndrome do not experience any symptoms, until the condition progression into a later stage, overtimes, leading to insulin resistance and plagues built-up on the wall of the arteries.

Other risk factors that increased the risk of metabolic syndrome include aging, race, family history, long-term hormone imbalance such as polycystic ovary syndrome (PCOS), gestational diabetes.

According to the results after 12 weeks of feeding,
* The high-fructose diet group showed an increase of visceral adiposity and plasma triglyceride concentration but decreased (P < .05) plasma adiponectin concentration associated with return cholesterol in the blood.

* β-sitosterol treatment prevented the high-fructose diet-induced visceral obesity, hypertriglyceridemia, and hypoadiponectinemia.

* Furthermore, β-sitosterol alone increased plasma adiponectin concentration and reduced plasma insulin concentration and homeostatic model assessment index.

Based on the results, researchers concluded, "β-sitosterol could be potentially used to prevent high-fructose diet-induced metabolic dysfunction".

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of diseases associated with metabolic dysfunction, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Muscle atrophy is a condition associated with a lack of physical activity, inducing loss of muscle mass due to the muscles weakening and shrinking.

A neuron or nerve cell made of a cell body (also called soma), dendrites, and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord, and peripheral nerves that play an essential role to process and transmit information.

The function of neurons can be classified into 3 types
* The sensory neurons found within the nervous system that convert external stimuli from the organism's environment into internal electrical impulses.

*The motor neurons transmit impulses from the spinal cord to skeletal and smooth muscles,. In other words, motor neurons directly control all of the muscle movements.

* UA treatment showed negative results of a decrease in BMD, trabecular bone volume fraction, and trabecular number, and an increase in trabecular separation in the SED group.

* No significant difference was observed in atrophic hindlimbs between SED and UA groups.

After talking about other factors into account, researchers said, "These results alone are insufficient to suggest that UA is a potential resistance exercise mimetic for atrophic skeletal muscle and weakened bone. However, this study will help determine the potential of UA as a resistance exercise mimetic".

Taken altogether, ursolic acid, may have an effect on muscle atrophy, pending the confirmation of the larger sample size and multicenter human study.

Intake of ursolic acid in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Queen Herb Chaste Tree Berry Shows a Valuable Chemotherapeutic Property for the Treatment of Cancers in Vitros, Researchers Say

Chaste tree berry is found to exhibit a significant anti-cancer activity, a recent study by renowned study advised.

Chaste tree berry is a species of Vitex agnus-castus, genus Vitex, belonging to the family Lamiaceae, native to the Mediterranean region, used in herbal medicine for thousands of years as an aphrodisiac herb and considered as Queen herb in treating menstrual problems and discomforts.

Cancer is a class of diseases characterized by cells growing and multiplying disordered and uncontrollable ways in our body. In the late stage, the cancer cells may travel away from the originated site

According to the Tianjin Institute for Biomedicinal Research, in the evaluated the apoptosis-inducing effect of chaste tree berry, using observation under a light microscope, flow cytometric analysis, and agarose gel electrophoresis, Vitexicarpin, a flavonoid found in chaste tree berry significantly inhibited proliferation of human cancer cells lines A2780(human ovarian carcinoma), HCT-15(human, colon, colorectal adenocarcinoma), HT-1080(fibrosarcoma cell line) and K562(Human chronic myeloid leukemia cells), depending to the concentration.

Dr. Wang HY the lead researcher in the study said, "Vitexicarpin induces apoptosis.... via mitochondria(in regulated cell metabolism)-controlled apoptotic pathway".


In support of the above analysis, the crude extract prepared with ethanol from dried ripened Vitex agnus-castus fruits exerted strong cytotoxic effects against cancer cell lines of human uterine cervical canal fibroblast (HCF), human embryo fibroblast (HE-21), ovarian cancer (MCF-7), cervical carcinoma (SKG-3a), breast carcinoma (SKOV-3), gastric signet ring carcinoma (KATO-III), colon carcinoma (COLO 201), and small cell lung carcinoma (Lu-134-A-H) cells.

The inhibition of the extract is attributed to the effect on cell death through apoptosis and increased intracellular oxidation in the tumor cells.

Other researchers suggested that the cytotoxic effects of Chaste tree berry may be involved various aspects, including the JNK (apoptosis) pathway. as indicated in the new native lignan VB6 study.

Taking together, there is no doubt, that chaste tree berry may be considered as a valuable chemotherapeutic agent for the treatment of various types of cancer, depending on further evaluation.

Coffee and Coffee's Caffeine May Reduce The Risk of Colon Cancer, Researchers Say

Good news for coffee drinkers, coffee consumption regularly is associated with reduced risk of colon cancer, a renowned institute study suggested.

Coffee, becoming a popular and social beverage all over the world, particularly in the West, is a drink made from roasted beans from the Coffea plant, native to tropical Africa and Madagascar.

Colon Cancer is a disease characterized by irregular cell growth in the colon, with the tendency in spreading to distant parts of the body, depending on the stage.

According to the Federico II University of Naples, the efficacy of coffee consumption in attenuated risk of the colon may be attributed to its bioactive components, such as polyphenols, melanoidins, carbohydrates, diterpenes,...

Dr. Vitaglione P, the lead author said, " ...metabolism and composition, both coffee chlorogenic acids and dietary fiber, including melanoidins, may reduce CRC risk, increasing colon motility and antioxidant status".

However, phytochemicals found in coffee may be different that depends on the amount and type of compounds ingested with the beverage, the variety of coffee used, the roasting degree, the type of brewing method as well as the serving size.


Additionally, in the investigated the protective action of coffee against Caco-2 human colon carcinoma cells. researchers filed the following results
1. Even with low concentrations, coffee (<5%) inhibited the proliferation of Caco-2 cells without affecting normal cell viability.
2. . Increasing the duration of coffee bean roasting showed a significant in reduction the mutative risk of gene KRAS expression.

3 Coffee reduces risk colon cancer through suppressed miR-30c and miR-96, in promoted cell proliferation, migration and invasion, and enhanced tumor growth.

The finding evidence suggested that coffee and coffee caffeine may have a profound effect in reduced risk of cancer linearly.

Further differentiation of delphinidin in cancer risk insisted that the chemical constituents also restained cancer cell invasion through reduced expression of inhibited serum- and vascular endothelium growth factor(VEGF) in induction of making new blood vessel to provide nutrients and oxygen for proliferation and cancer progression.

Vascular endothelial growth factor (VEGF) is a potent stimulator of angiogenesis, associated with unfavorable clinical characteristics by promoting proliferation secreted by various human cancers.
Moreover, researchers also indicated that the efficacy of anti-cancer cell activity of delphinidin was attributed to the function of phytochemical in triggering extracellular signal-regulated kinases (ERKs) in regulated cancer cell cycle division, including meiosis, mitosis, and post mitosis.


In-depth analysis also discovered that delphinidin isolated from eggplant also activated the independence of nitric oxide pathway in suppression of cancer cell progression by initiated cell cycle arrest in the G(0)/G(1) phase.

In humans, gelatinases are expressed by matrix metalloproteinases MMP2 and MMP9.
Importantly, gallic acid also ameliorated cancer progression through inhibition of the matrix metalloproteinase(MMP)-2 and MMP-9 same as the effectiveness as delphinidin involved in the anti-angiogenetic activity.


Taken together, eggplant with abundant bioactive chemical compounds such as gallic acid and anthocyanins may be used as a functional food for reduced risk and treatment of fibrosarcoma, depending to further investigation.

Beetroots Inhibit the Early Onset of Insulin Resistances and Type 2 Diabetes, Researchers Say

Beet juice containing phytochemicals may alter postprandial glucose and insulin responses in prediabetics, some scientists suggested.

Insulin resistance, a condition of not providing enough insulin to counter the rise of blood glucose is considered as the early progression of onset of diabetes.

According to The American Diabetes Association (ADA), people with insulin resistance and prediabetes are associated with the early onset of type 2 diabetes as a result of unhealthy diet and physical inactivity.


And, 150 million people worldwide and 43 million people in the US are currently suffered from type 2 diabetes mellitus or metabolic syndrome.

In broader terms, insulin resistance is considered a major factor that can lead to type 2 diabetes, gestational diabetes, and prediabetes.


The actual cause of insulin resistance is debatable, some researchers suggested genetic mutation, aging and ethnicity play important roles in the development of insulin sensitivity, And people such as obesity, excess body weight, high blood cholesterol, and pressure, too much belly fat, a lack of exercise, smoking are susceptible to increase risk of insulin resistance.

Moreover, in the homeostasis model assessment-insulin resistance index, dietary nitrate from beet juice also demonstrated a strong effect in lowering insulin resistance in test subjects. compared to control through regulation of glucose homeostasis and insulin signaling pathway.

Dietary nitrate from beet juice also improved lipid homeostasis, inflammation, and endothelial function.

Dr. Peter C, the lead researcher said, "Betalains, polyphenols and dietary nitrate found in the beetroot juice may each contribute to the observed differences in the postprandial insulin concentration".

Interestingly, in comparison the effect of ingredients of combined 100 ml black tea, beetroot juice or control (water) in 16 obese, insulin-resistant males, researchers revealed that beetroot juice plus black tea treated groups exhibited a decreased postprandial peripheral vascular resistance (VR) in resistance vessels, but lesser than black tea, in compared to other groups.
