Beta-Sitosterol in the inhibition of Mammary Gland Cancer in Vivo

Cancer is a class of chronic diseases associated with irregular cell growth in the tissues of the affected organ.

The mammary gland located in the breasts of females is responsible for lactation, or the production of milk.

Both males and females have glandular tissue within the breasts; however, in females, the glandular tissue begins to develop after puberty in response to estrogen release.
A mammary tumor is a medical and chronic condition originated from the milk-producing gland.

Most cases of the mammary cancer are first found in the cell on the surface of the inner lining of the milk-producing gland, before penetrating deeper into the layers.

Epidemiologically, mammary cancer has been found to spread to the colon and the liver.

Most common risk factors of mammary cancer are similar to those of breast cancer, including, aging, physical inactivity, being obese or overweight, family and personal history, having a dense breast, radiation therapy, pregnancy history, genetic proposition, particularly in women who had used drug diethylstilbestrol (DES).

Conventionally, treatment of mammary gland cancer is totally dependent on the stage and grade of cancer, including surgery, radiation therapy, chemotherapy, or a combination.


Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of cancer, researchers examined the anti-cancer effects of beta-sitosterol (BS), a plant-derived sterol on mammary gland cancer in albino Wistar rats.

Selected rats induced by a single i.v. injection of MNU followed by β-sitosterol supplementation therapy for 115 days, were randomized and divided into four groups with 8 animals each. Group I (sham control 1 % CMC in normal saline p.o.); Group II (toxic control, MNU 47 mg/kg, i.v); Group III (MNU 47 mg/kg, i.v + β-sitosterol, 10 mg/kg, p.o); Group IV (MNU 47 mg/kg, i.v + β-sitosterol, 20 mg/kg, p.o).

According to the experimental analysis,
* Treatment with β-sitosterol inhibited the onset of mammary gland cancer evidenced by the decrease in the alveolar bud and lobule score in the whole mount of the mammary gland.

* Furthermore, β-sitosterol inhibited the overexpression of oxidative stress through synchronizing lipid and enzymatic antioxidant defense.

* Moreover, β-sitosterol demonstrated showed a significant decrease in the saturated and unsaturated fatty acid.

* Additionally, β-sitosterol treatment inhibited the protein gene product 9.5 (PGP9.5) overexpression, a marker for tumors in a dose-dependent manner, compared to the MNU treatment group.

Based on the evidence, researchers said, "β-sitosterol afforded significant protection against the deleterious effects of MNU".
Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of mammary gland cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

The order of clot lysis activity was found to be: Streptokinase > Chloroform fractions > Methanol (crude) extract > Hydro-methanol fractions > Ethyl acetate fractions > n-hexane fractions > Water.

The Lower effects of other extracts might be associated with the lack of bio-active components and/or insufficient quantities in the extract.

Also, in vivo, T. Orientalis and B. monnieri also demonstrated clot dissolving property

Based on the findings, researchers concluded, " ..., active component(s) of T. orientalis and B. monnieri for clot lysis could lead the plants for their therapeutic uses" and ",,,. these plants could be incorporated as a thrombolytic agent for the improvement of the patients suffering from atherothrombotic diseases".

Taken altogether, wild cabbage may be considered a functional food for the prevention of stroke, pending to the confirmation of the larger sample size and multicenter human study.

Inflammation is the natural response of the immune system macrophages, the first line of defense in stimulating the production of inflammatory cytokines to protect our body against the infection caused by foreign microorganisms during the acute phase of body damage and injury.

Truly, the main focus of inflammation is to repair damage and injury.

Most cases of infection associated with inflammation are stopped at the acute phase. The wound or injury is slowly recovered.

Female BALB/c mice selected to the investigation were randomly divided into five groups with 10 mice in each group: (1) control group (saline), (2) LPS group, (3) LPS+dexamethasone (LPS+Dex, 2mg/kg, administered by gavage), (4) LPS+WETMHM (5 g/kg, administered by gavage), (5) LPS+WETMHM (10 g/kg, administered by gavage).

According to the results from the tested analysis, treatment with the WETMHM exhibited a number of mechanisms against LPS-induced inflammation, including the

* Attenuating inflammation cell numbers in the BALF

* Decreasing the protein levels of lung PI3K/Akt/mTOR, and 

* Improving the antioxidant activity of  SOD.

* Inhibiting MPO activity associated with inflammation

Furthermore, the histological studies also demonstrated that WETMHM substantially inhibited LPS-induced neutrophils in lung tissue.

Based on the results, researchers suggest, " The results indicated that the WETMHM had a protective effect on LPS-induced ALI in mice".


Taken altogether, dandelion may be considered a remedy for the treatment of diseases associated with inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Turmeric Postpones the Onset of Age-Related Disease

Age-related diseases are medical conditions associated with the increase of age.

Most recently age-related diseases in the elderly are cardiovascular diseases.

Cardiovascular diseases are a group of conditions triggered by the abnormal function of the heart and blood vessels abnormality.

The 3 most common forms of cardiovascular disease in North America are coronary artery disease, peripheral artery disease, and carotid artery disease caused by plaques build-up of plaque in the arteries that supply blood to the heart, the arms and legs, and the brain, respectively.

*Cancer
Cancer is a group of medical and chronic diseases characterized by cell growth disorderly and uncontrollably in the tissue of specific organs in the body.

Over time, the alternative cells induced tumor may become progressively worse and damage other healthy tissues, depending on the grade of the tumor.

*Rheumatoid Arthritis is a chronic disorder as a result of inflammation, affecting mostly the flexible (synovial) joints and tissues and organs in the body. The disease affects more women than men and generally occurs after the ages of 40, causing the diminished quality of life of many elders(1).

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

In the urgency to discover a natural ingredient for the prevention of chronic inflammatory diseases, researchers examined the effects of curcumin on aging and age-related diseases.

According to the tested analysis, injection of curcumin showed a strong activity against low-grade inflammation driven by oxygen stress in the induction of the increased level of pro-inflammatory cytokines such as IL-1, IL-6, and TNF-α.

Furthermore, the anti-aging-related diseases involved in low-grade inflammation of curcumin were attributed to the inhibiting genes activated by the transcription factor NF-κB. 

In other words, curcumin slowed down the aging process and postponed the onset of age-related diseases by blocking the NF-κB-dependent inflammation

Based on the findings, researchers said, " We consider the possibility of the spice curcumin, a powerful antioxidant and anti-inflammatory agent possibly capable of improving the health status of the elderly".

Taken altogether, turmeric processed a high amount of bioactive compound curcumin may be considered supplements for the prevention of aging related-diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
