Phytochemicals Phytic Acid Which Reduces Risk and Treatment of Colorectal Cancer

By Kyle J. Norton

Phytic acid, an organic compound found much in healthy food sources may have a potential effect in reducing risk and treatment of colorectal cancer, some studies suggested.

Foods that containing high levels of phytic acid including nuts, sesame seeds, soybeans, wheat, pumpkin, beans, almonds,...
By Kyle J. Norton

Colorectal cancer also is known as bowel cancer, is a medical and chronic condition characterized by the abnormal proliferation of cells in the colon, rectum, or vermiform appendix due to the alternation of DNA with unidentified origin.

Colorectal cancer (CRC) is relatively very common cancer in the Western world. The cancer is considered a slowly growing and progressing cancer in a predictable way.

Bowl is divided in 2 parts, the first part of the bowel, the small bowl, is involved with the digestion and absorption of food and the  2nd part, the large bowel which consist the the colon and rectum, is involved in absorption of water from the small bowel contents and broken down of certain materials in the feces into substances of which some of them to be reabsorbed and reused by the body.

Why colorectal cancer is more susceptible to the  West than other parts of the world. Some researchers suggested it may a result of the Western diet. People who the diet with high saturated and trans fats, red meat, processed foods, and fewer fruits and vegetables, whole grain are associated with a substantially higher risk to colorectal cancer, compared to traditional diet.

Dr. Alan Moss, the lead author in the cancer program at The University of Melbourne, a diet that contains high levels of red meat is associated with risk of CRC.

The risk of colorectal is increased proportionally to the increased intake of red meat. In other words, the redder consumed the higher the risk of CRC.

And, processed meats result from salting, smoking, fermenting, or curing the meat, and common examples include ham, bacon, and sausage are also in the list of elevated the risk of colorectal cancer.

In the comparison of the “Western” and “prudent” dietary patterns on the incidence of CRC, researchers showed that people who follow the Western diet have a 31% increased risk of the disease onset, compared to a 14% reduced risk of CRC in people who follow the prudent diet with high fruits and vegetables, fewer red meat and processed foods.

These results suggested, regardless to present the colorectal cancer risk, change of diet pattern today, your risk the cancer is continuously lower, compared to before of the change.

Dr, Shafie NH, the lead scientists in the researchers at the cancer program team at the Universiti Putra Malaysia in the examine whether the phytic acid, a natural dietary ingredient and an essential component of nutritional diets effect in colorectal cancer risk showed that

* Phytic acid exerted a strongly inhibitory colorectal cancer activity by increasing the function of proteins (up-regulation of Bax)associated with program cells death and (down-regulation of Bcl-xl) anti-cancer cells proliferation.
* Furthermore, when exposed to IP6 (phytic acid), the phytochemical up-regulates of caspase-3 and -8 expressions and activation of both caspases may also contribute to the apoptotic cell death of human colorectal adenocarcinoma HT-29 cells.

The results suggested, combined with t rice bran IP6 (phytic acid) induced apoptosis, by regulating the pro- and anti-apoptotic markers.

Moreover, in the reaffirming the significant role of phytic acid in inhibition of colorectal cancer, Dr. Saad N, the team leader and colleagues launched an investigation of expression changes of β-catenin and cyclooxygenase-2 (COX-2) and cell proliferation in the adenoma-carcinoma sequence after treatment with rice bran PA by immunocytochemistry.

The experiment includes a total of 72 male Sprague-Dawley rats divided into 6 equal groups with 12 rats in each group.

Cancer induction by two intraperitoneal injections of azoxymethane (AOM) was given at 15 mg/kg body weight over a 2-week period. During the post-initiation phase, two different concentrations of PA, 0.2% (w/v) and 0.5% (w/v) were administered in the diet.

Before treatment with PA, rats induced cancer showed that β-catenin, a marker of alternation of gene transcription, COX-2 expressions associated with cancer cells proliferation and cell proliferation of Ki-67 associated to a significant contribution in colonic cancer progression.

And, a significant positive expression and correlation were found between COX-2 expression and proliferation and the total β-catenin also demonstrated a significant positive linear relationship with total COX-2.

After application phytic acid, researchers found that the expression changes of β-catenin and cyclooxygenase-2 (COX-2) and cell proliferation in the adenoma-carcinoma sequence were totally inhibited by rice bran PA.

Taken all together, there is no doubt that phytic acid can be considered a functional supplement in the prevention of colorectal cancer and combined with primary therapy for the treatment of the disease with inducing any side effects.

However, further data collection large example size and multi-centers studies performed with human consumption of the whole food or its bioactive compounds during the course of the disease will be necessary to complete the picture of phytic acid anti-colorectal cancer possibilities.

Cruciferous Vegetables, The Functional Food May Process Anti Bladder Cancer Potency

By Kyle J. Norton

Cruciferous vegetables may process a significant effect in the reducing risk and treatment of bladder cancer, some scientists suggested.

The bladder is a hollow elastic organ in the center of the lower abdomen that collects urine from the kidneys and excreted them through the urethra.

Bladder cancer is a medical and chronic condition caused by cell growth irregularly and disorderly in the bladder tissue as a result of cell DNA alternation.

Most cases of primary bladder cancer start on the surface of the inner lining of the bladder. At the cancer progress, the cancerous cell can penetrate deeper in the bladder tissue and travel a distance away to infect other healthy tissue and organs.

According to the statistic, men are 4 times more likely than women to be diagnosed with the disease. and 90% of people with bladder cancer are older than 55, and the average age people are diagnosed with bladder cancer is 73.

Cancer causes 17,000 deaths (12,500 men and 4,500 women) every year.

Although there are many risk factors associated with the onset of bladder cancer, long-term cigarette smoking use is considered a significant cause of the disease. Smokers are 4 to 7 times more likely to develop bladder cancer than nonsmokers.

However, most of the cigarette smoker has never developed bladder cancer.

Dr. Neal D Freedman, the lead author in the concerns of bladder cancer widespread in men launched an investigation to evaluate the cigarette smoking in the risk of bladder cancer.

The study included a total of 81,394 men and 86,134 of women who complete the NIH-AARP cohort lifestyle questionnaire and followed from 1995 through Dec 31, 2006.

During 4,518,938 years of follow-up, incident bladder cancer occurred in 3,896 men (144.0/100,000 person-years) and 627 women (34.5/100,000 person-years).

Former smokers and current smokers were found to have higher risks of bladder cancer than never smokers.

The summary risk estimate for current smoking followed from 1963–1987) was 2.94.

The risk of bladder cancer of ever smoking in our study was 0.50 in men and 0.52  in women.

The findings not only suggested that the risk of bladder cancer not only increases in the current smoker but also elevates in previous smokers.

Cruciferous vegetables are the group of vegetables belonging to the family Brassicaceae, including cauliflower, cabbage, cress, bok choy, broccoli etc.

Its chemical constituents of cruciferous vegetables include glucosinolates, dithiolthiones, indoles, glucoraphanin, s-methyl cysteine sulfoxide, isothiocyanates, indole-3-carbinol, etc.

In the reaffirming the epidemiologic evidence suggests diets rich in cruciferous vegetables, particularly broccoli, are associated with lower bladder cancer risk, researchers at The Ohio State University conducted an evaluation to test the mechanisms whereby broccoli and broccoli sprout extracts and pure isothiocyanates inhibit normal, noninvasive (RT4), and invasive (J82, UMUC3) human urothelial cell viability.

Sulforaphane (IC(50) = 5.66 ± 1.2 μM) and erucin (IC(50) = 8.79 ± 1.3 μM), the major compounds found in the broccoli and broccoli sprout are the most potent inhibitors without harming to the normal cells.

Further analysis suggested that the efficacy of the tested veggies are attributed to the inhibition of survivin, which plays an important role in the protection of cell death, epidermal growth factor receptor (EGFR), a growth factor receptor that induces cell differentiation and proliferation upon and human epidermal growth factor receptor 2 (HER2/neu) in the initiation of cancer development, G(2) /M cell cycle accumulation, and apoptosis.

In a murine UMUC3 xenograft model, injection of diets containing 4% broccoli sprouts, or 2% broccoli sprout isothiocyanate extract; or gavaged pure sulforaphane or erucin (each at 295 μmol/kg, similar to dietary exposure) showed a tumor weight reduction of 42%, 42%, 33% and 58%, respectively.

These results showed that broccoli and broccoli sprout has a strong effect in targeting bladder cancer prevention.

Moreover, in the reconfirmation of the effects of Sulforaphane (SF) inhibits bladder cancer in vivo, researchers at the Roswell Park Cancer Institute conducted an investigation SF effect in both human bladder cells in vitro and mouse bladder tissue in vivo.

Application of SF activates NF-E2 related factor-2 (Nrf2), which is a well-recognized chemopreventive target and activates the Nrf2-regulated cytoprotective signaling pathway in response to oxidative stress.

Comparison between wild-type mice and mice without Nrf2 shows that Nrf2 activation is required by SF for inhibition of Androgen-binding protein (ABP)-induced DNA damage.

Additionally, Nrf2 activation by SF in the bladder occurs primarily in the epithelium, which is the principal site of bladder cancer development.

Accompanied with the result of SF-enriched broccoli sprout extracts strongly inhibits N-butyl-N-(4-hydroxybutyl)nitrosamine-induced bladder cancer development, suggest that SF is a highly promising agent for bladder cancer prevention and provides a mechanistic insight into the repeated epidemiological observation that consumption of broccoli is inversely associated with bladder cancer risk and mortality.

Taken altogether, cruciferous vegetables, including broccoli and broccoli sprout processed high amounts of bioactive compounds may be considered functional for the prevention and an adjunct therapy combined with primary medicine for the treatment of bladder cancer.

Ginger, the Potential Remedy for the Prevention and Treatment of Atherosclerosis with No Side Effects

By Kyle J Norton

Ginger may have a potential and profound effect in blocking the accumulation of plaques on the arterial wall in initiating the risk of atherosclerosis,  some scientists suggested.

Atherosclerosis is a medical condition associated with plaque building up in the arteries, leading to the narrowing the arterial function in the transportation of oxygenated blood to the heart and other blood of the body.

Plaques are substance made up from the accumulated fat, cholesterol, calcium, and other substances found in the blood, including cadmium.

Long-term accumulation of plaque can also cause hardening and stiffing arterial wall can lead to the blockage of the arteries and risk of heart disease depending on the location of the blockage.

Most people in the early stage of atherosclerosis do not experience any symptoms. However, as the condition progresses and the location affected, patients may develop chest pain, shortness of breath, sweating, and anxiety.

Most common causes of atherosclerosis are the imbalance of the ratio of bad cholesterol and good cholesterol in the blood, long-term smoking, obesity, increase in age, obesity and some medical conditions, such as diabetes and high blood pressure.

Therefore adding a portion of olive oil into your daily diet can reduce the risk of high blood cholesterol

Dr. Siasos G, the lead scientists in the heart disease team in the concerns of the smoking detrimental effect to many organ systems and is responsible for illnesses such as lung cancer, pneumonia, chronic obstructive pulmonary disease said, " smoking is an important modifiable risk factor for the development of cardiovascular disease,....via initiation and progression of atherosclerosis".

And, " variety of studies has proved that cigarette smoking induces oxidative stress, vascular inflammation, platelet coagulation, vascular dysfunction and impairs serum lipid profile in both current and chronic smokers, active and passive smokers and results in detrimental effects on the cardiovascular system".

The statement clearly indicated that cigarette smoking has a strong implication of the onset and progression of atherosclerosis.

Some researchers suggested that cigarette smoke contain a high amount of toxic chemicals that cause inflammation and formation of blood clot in the arteries and oxidation of low-density lipoprotein cholesterol.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine to treat dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

On finding a natural ingredient or whole food for the prevention and treatment, researchers at the joint study led by the University of Sistan and Baluchestan examined the potential protective effects of ginger in atherosclerosis and atherosclerotic cardiovascular disease.

The randomized, experimental, and controlled study included thirty healthy overweight women aged 20-30 years randomly divided into three equal groups, namely, ginger, ginger + high-intensity interval training (HIIT), and placebo + HIIT.

The training groups performed high-intensity interval exercises (i.e. 40-m maximal shuttle run) for ten consecutive weeks. The supplement groups daily took 3 g of ginger pills and the third group took the placebo.

The paired t-test revealed a significant decrease in the density of type 1 monocytes chemotactic protein (MCP-1) in HIIT + ginger (P = 0.026) and HIIT + placebo (P = 0.001) groups, an indication of reduced inflammation.

These results suggested that the relative and efficient role of HIIT combined with the ginger supplement in overweight women in inducing antioxidant/anti-inflammatory activity against atherosclerosis.

Furthermore, the investigation of the effects of air-dried ginger powder (0.1g/kg body weight, po, for 75 days) was carried out by the RNT Medical College, on experimentally induced atherosclerosis in rabbits by cholesterol feeding (0.3g/kg body weight, po).

Before administration of ginger powder, cholesterol feeding for 75 days led to the distinct development of atheroma in the aorta and coronary arteries of the rabbits.

The accumulation of plaque in the arteries was significantly inhibited by about 50% following ginger administration.

Moreover, ginger treatment also decreases lipid peroxidation and enhanced fibrinolytic activity associated to dissolve blood clots.

The findings indicated that ginger distinct protection from the development of atherosclerosis is attributed probably because of its free radical scavengings, prostaglandin inhibitory and fibri
properties.

Taken altogether, ginger may be considered a functional food for the prevention and an adjunct therapy for the treatment of atherosclerosis without inducing any adverse effects.

Tualang Honey May Process a Potential for the Prevention and Treatment of Osteosarcoma

By Kyle J. Norton

Scientists may have found a natural ingredient for the prevention and treatment of osteosarcoma without inducing any side effects, according to some studies suggested.

Osteosarcoma is a medical condition caused by the production of immature bone, affecting mostly younger population under the age of 25.

Male are more susceptible to the incidence, compared to female.

At the advanced stage, cancer cells can travel a distance away to infect other healthy tissue and organs, particularly, the lung.

There are 2 types of osteosarcoma.

A localized osteosarcoma is seen only in the bone. This type of osteosarcoma.
normally started on the surface of the tissues next to the bone, such as muscle, tendon, or fat.

A metastatic osteosarcoma is a tumor which has spread to other parts of the body.

Genetic mutation of gene p53, the tumor suppressed gene is the most common cause of osteosarcoma.

However, the disease can also be acquired, due to the alternation of cells DNA caused by long-term exposure to toxic chemicals and radiation therapy.

Some researchers suggested that the increase in age, gender, height, ethnicity and other forms of bone diseases are also the prevalent risk factors associated with the onset of osteosarcoma.

Dr. Julie D. K. McNairn, the lead author in the cancer team at the SUNY Upstate Medical University examined the connection of the inheritance of osteosarcoma and Paget’s disease of bone, said, "Pagetoid osteosarcoma is a complication of Paget’s disease of bone. Sarcomatous transformation is most often seen in severe, long-standing Paget’s disease".

And, "some researchers have shown loss of heterozygosity in a region of chromosome 18q in a relatively high percentage of studied patients affected with either Paget’s disease alone, in Pagetoid osteosarcoma, and in uncomplicated osteosarcoma".

These results clearly suggested that the alternation of certain genetic material found in the chromosome 18q, more specifically 18q21-22  is associated with the onset of Pagetoid osteosarcoma.

Honey is the miraculous product made by bees using nectar from flowers.

The rich golden liquid It is considered one of healthy sweet food for replacing the use of white sugar and artificial sweetener by many people.

With an aim to find a natural ingredient or whole food for the treatment of osteosarcoma without indiu=cuiing any side effects compared to the conventional treatment of surgery followed by the radio and chemotherapy, researchers investigated the antiproliferative activity of local honey (Tualang) on human osteosarcoma (HOS) cell lines.

Different concentrations of Tualang honey (1% - 20%) were applied on HOS cell lines for 3, 6, 12, 24, 48 and 72 hours.

Cellular apoptosis was done using the Annexin V-FITC Apoptosis Detection Kit.

By examining the morphological appearance after application of Tualang honey, HOS cell showed apoptotic cellular changes like becoming rounded, reduction in cell number, blebbed membrane and apoptotic nuclear changes like nuclear shrinkage, chromatin condensation, and fragmented nucleus.

In others words, Tualang honey incubated with the cancer cell line demonstrated a strong effect in inducing cell apoptosis through degradation.

The 50% inhibitory concentration (IC50) for HOS cell lines was found to be 3.5%.

The maximum inhibition of cell growth of ≥80% was at the concentration of 15%.

These results suggested that Tualang honey induces HOS cells apoptosis through suppressing the cancer cell viability and integrity.

Taken altogether, Tualang honey may be used as a functional food for the prevention and combined with the primary medicine for the treatment of osteosarcoma

However, further data collection large example size and multi-centers studies performed with human consumption of the whole food during the course of the disease will be necessary to complete the picture of Tualang honey anti-osteosarcoma possibilities.

Phytochemical Polysulfides, the Natural and Potential Chemo- Therapeutic Agent for colon Cancer with No Side Effects

By Kyle J. Norton

Phytochemical Polysulfides may be considered a functional supplement in reducing the risk and treatment of colon cancer.

Colon cancer is a very slowly growing medical condition characterized by irregular cell growth in the colonic tissue.

At the advanced stage, cancerous cells can travel a distance away from the colon to infect other healthy tissue and organ through lymph and blood.

The colon is a part of the digestive system involved the absorption of water from the small bowel contents and broken down of certain materials of the feces into substances of which some of them to be reabsorbed and reused by the body.

According to the statistics,  in Canada, every year approximately over 17,000 new cases of colorectal cancer are diagnosed causing over 6500 death

Colorectal cancer ranks third overall in the number of both new cancer cases and cancer deaths in Canada.

The causes of colon cancer are unknown. however, people who are obese, follow the Western diet and long-term exposure to ionizing radiation due to occupation are some of the most prevalent risk factors that cause colon cancer.

The “Western” diet, characterized by high intakes of saturated fat and trans fat, red and processed meats, sweets, refined grains, and desserts increased the risk and recurrent risk of colon cancer, according to the study published on the Aug. 15 issue of the Journal of the American Medical Association.
.
Dr. the lead scientist in the comparison of diet pattern said, " Compared to the Western-Southern pattern, the fruit-vegetable and metropolitan patterns were associated with more healthful dietary behaviors (e.g., higher vegetable intake and lower red meat consumption), and demographic/lifestyle characteristics typically correlated with low colon cancer risk, for example, lower BMI, higher education, and higher NSAID use".

Therefore, by changing diet pattern from the Western diet to traditional diet with more fruits and vegetable, reducing weight and actively engaging in a healthy lifestyle, risk of colon cancer are decreased substantially.

If you have a sudden change of bowel habits such as diarrhea or constipation that last more than 3 to 4 weeks, persistent blood in the stool and abdominal cramps, gas or pain, accompanied fatigue with no reason and unexplained weight loss, please check with your doctor right the way to rule out the possibility of colon cancer.

Polysulfides are phytochemicals in a class of chemical compounds containing chains of sulfur atoms, belonging to the group ofOrganosulfides found abundantly in an oxidized product, including beer, wine, whiskey, garlic oil, etc.

In the finding, the new anti-colon cancer drug with the same effect of diallyl polysulfides are well known as anticancer drugs with no side effects, researchers at the University of the Saarland conducted an experiment to examine the effect of the combined coumarins with polysulfides in the form of di-coumarin polysulfides in HCT116 colorectal cancer cell line.

Application of combination reduced cell viability of cancer cells in a time and concentration-dependent manner.

The combined coumarins with polysulfides exerted apoptosis through the G(2)/M phase of the cell cycle.

The apoptosis activity was attributed to the effect of the combined coumarins with polysulfides in the activation of the proteins that program cell death.

In the comparison of these new coumarin compounds with the well-known diallyl polysulfides, researchers found that the coumarin disulfides are more active than the corresponding diallyl disulfides.

Dr. Saidu NE, the lead author in the final report said, "coumarin polysulfides also down-regulated the level of Cell Division Cycle 25C,(cdc25C), which also contributed to the arrest in the G(2)-phase of the cell cycle".

Futhermore, in the evaluation of the garlic-derived organosulfur compounds such as diallyl sulfide, a member of polysulfides, researchers revealed that the compound exerts a significant influence on cell viability, cell cycle arrest and induction of apoptosis in HCT116 human colon cancer cells.

Diallyl trisulfide is even more efficiently diallyl tetrasulfide treatment of HCT116 cells that lead to a reduced cell viability, cell cycle arrest, and apoptosis.

Additional analysis suggested that the efficacy of the compound was attributed to the function in inhibition of the redox state of the cell, and its anti-oxidants that prevent sulfide-induced apoptosis.

The growth arrest and induction of apoptosis are associated with a considerable reduction of the level of cdc25C similar to those of the previous study.

Taken together, the bioactive polysulfides may be considered a functional phytochemical for the prevention of colon cancer and combined with primary therapy for the treatment of colon cancer.

Phytochemical Tartaric Acid Improves the Digestive Absorption and Feces Secretion

By Kyle J. Norton

Tartaric acid, an organic phytochemical may process a significant effect in improving the digestive function in the absorption of nutrients and fluids from foods intake, some scientists found.

Phytochemicals, the natural chemical constituent, protect the plants against diseases and form their outer's color.

In human, the digestive system consists of a set of accessory organs of digestion and the gastrointestinal tract, involving in the breakdown of food into smaller and smaller components, until they can be absorbed and assimilated into the body.

There is no single cause of the digestive disorder. the disorder can be induced by multiple factors including bacterial infection, viral infection, inflammation, lactase deficiency, intake of some difficulty digesting certain foods.

In severe cases, the digestive disorder can be caused by poor circulation to the intestines or other organs and complication of other medical conditions such as muscle dysfunction, gallstones, stress, and side effects of certain medications including the anti-inflammatory drugs.

Other common diseases that cause digestive disorder also include irritable bowel syndrome and disease, heartburn.

Most common acute symptoms of digestive disorders are bloating, diarrhea, gas, stomach pain, and stomach cramps.

Lifestyle has been found to associate with the onset of digestive disorders. Dr. Hall EH, the lead scientist in the digestive disorders team in the investigation of the effects of environmental and lifestyle on disorders of the large and small intestine, wrote, "External factors including obesity, lack of physical exercise, and tobacco consumption also impact diseases of the luminal gastrointestinal (GI) tract" and : new treatment options for celiac disease reflect the complex interaction of diet, genetic factors, and the host immune response, the implications for treatment of many conditions of the large and small intestine that arise from environmental and lifestyle are as basic as ensuring adequate nutrition, regular exercise, and cessation of tobacco use".

This statement suggested, by following a healthy diet and lifestyle with moderate exercise is one of the best methods that reduce substantially the risk of digestive disorders.

Tartaric acid is a phytochemical of the organic acid found abundantly in nuts, apricots, apples, sunflower, avocado, grapes, etc.

In the study to examine the effects of D-tartaric acid (TA) on intestinal drug absorption under both in situ and in vitro experimental conditions, researchers found that
* TA improved paracellular drug transport by changes the integrity of the epithelial tight junctions in rat colon at low pH conditions.
* Application of TA (20 mM) increased the absorption of Rho123, a chemical compound and a dye in both ileum and colon but not in the jejunum.

* TA (20 mM) also increased the absorption of daunorubicin in the ileum, but TA (20 mM) did not change the expression level of P-glycoprotein (P-gp) which processes a function in pumping foreign substances out of cells.

* TA (20 mM) significantly inhibited excretion of i.v.-administered Rho123 and daunorubicin into the ileal lumen.

These results suggested that TA increases the intestinal absorption of P-gp substrates Rho123 and daunorubicin, a chemotherapy medication used to treat cancer, by modulating the P-gp function without changing the expression level of P-gp in the rat intestine.

Futhermore, in the evaluation of the new starch preparations produced by thermolysis of potato starch in the presence of inorganic (hydrochloric) and organic (citric and tartaric) acids under controlled conditions to examine whether the new starch effect in intestinal absorption.

Dextrin D3, obtained by heating starch acidified with hydrochloric and tartaric acids exerted a strong effect in breaking down foods into simpler chemical compounds. and absorbed by the digestive tract.

The presence of high concentration of the tartaric acid at had a significantly stronger influence on chemical breakdown of a compound due to reaction with water compared to on citric acid.

The results confirmed the possibility of applying dextrins, prepared under specific conditions, as soluble dietary fiber in the promotion of colon feces movement.

Taken altogether, the Tartaric acid may be considered a function supplement in improving the digestive system in breaking down foods for absorption of nutrients and fluids for the body need and
promoting the colon feces secretion.

Phytochemical Allyl isothiocyanate, the Potential Anti Bladder Cancer Functional Remedy

By Kyle J. Norton, Master of Nutrition

Allyl isothiocyanate, a phytochemical found abundantly in the horseradish may have a strong effect in reducing risk and treatment of bladder cancer, some studies suggested.

Human bladder is a hollow elastic organ in the center of the lower abdomen that collects urine from the kidneys and excreted them through the urethra.

Bladder cancer is a medical condition characterized by irregular cell growth in the bladder tissue due to the alternation of DNA.

Most cases of primary bladder cancer start on the cell in the surface of the inner lining. However, after growing large enough, the cancerous call can spread to other regions in the body through the circulation of fluids and blood.

According to the statistic. 90% of bladder cancer were diagnosed in people who are older than 55. and the average age people are diagnosed with bladder cancer is 73.

Every year, the disease causes over 17,000 deaths and bladder cancer is the eighth most common cause of cancer death in men.

Out of the many causes of bladder cancer, researchers found that lifelong smoking, aging, and chronic bladder inflammation are some of the most common prevalent risk factors of bladder cancer cases found in the Western world.

Dr. Neal D Freedman, the lead author in the study "Association between smoking and risk of bladder cancer among men and women" clearly stated, "the population attributable risk of bladder cancer for tobacco smoking is 50–65% in men and 20–30% in women and that current cigarette smoking triples bladder cancer risk relative to never smoking".

" Over the last 30 years, incidence rates have remained stable in the United States (men: 123.8/100,000 person-years to 142.2/100,000 person-years; women: 32.5/100,000 person-years to 33.2/100,000 person-years)".

The study suggested if you quit smoking today, re\gardless of now many years that you have been smoking, your risk bladder cancer is reduced compared to those do not.

In chronic bladder inflammation patients, Dr. Michaud DS, in the review the relation among patients in Egypt with a chronic bladder infection and inflammation in the risk bladder cancer suggested inflammation is likely to have an important role in bladder carcinogenesis in developed countries probably associated with the genetic susceptibility to inflammatory pathways that cause persistent inflammation in damaging the cells in the bladder.

In some cases, the persistent inflammation can lead to the alternation of the cells DNA.

Allyl isothiocyanate is phytochemical containing sulfur in the class of organosulfur compound, found abundantly in horseradish, mustard, wasabi, etc.

In the study to reaffirm the effect of Allyl isothiocyanate (AITC)in the inhibition of the urinary bladder cancer growth and progression, researchers at the Department of Chemoprevention.launched
an investigation to examine the effect of the phytochemical used combination with the conventional medicine Celecoxib, a selective Cox-2 inhibitor against bladder cancer in the animal model.

Injection of the combination of AITC and celecoxib at low dose levels (AITC at 1 mg/kg and celecoxib at 10 mg/kg) in a syngeneic rat model of orthotopic bladder cancer showed to increased or perhaps synergistic inhibition of bladder cancer growth and muscle invasion, compared with each agent used alone.

Furthermore, the combination regime was also more effective than every single agent in inhibiting microvessel formation and stimulating microvessel maturation in the promotion of the growth of cancer in the tumor tissues.

The results of the efficacy of the combination was associated with the inhibition of the levels of prostaglandin E2 which not only plays an important role in the promotion of tumor growth but also is a  key downstream signaling molecule of Cox-2, caspase activation in the induction of cancer cell proliferation in solid tumors and downregulation of vascular endothelial growth factor in facilitating the growth of new blood vessels to the tumor tissues.

Dr. the lead author said, at the final report said, "ITC and celecoxib complement each other for inhibition of bladder cancer and provide a novel combination approach for potential use for prevention or treatment of human bladder cancer".

Additionally, in the human bladder cancer UM-UC-3 cells or rat bladder cancer AY-27 cells injection of NAC-AITC at 15 μM results in significant inhibition of cell growth and proliferation, together with cell cycle arrest and apoptosis.

Where AITC is excreted in urine mainly as N-acetyl-S-(N-allylthiocarbamoyl) cysteine, more commonly known as the N-acetylcysteine conjugate (NAC-AITC).

In an orthotopic rat bladder cancer model, NAC-AITC administered orally at 10 μmol/kg body wt inhibits cancer growth by 40% and muscle invasion by 49%.

The anticancer activity of NAC-AITC is associated with the modulation of several important molecular targets, including downregulation of both α-tubulin and β-tubulin which are correlated with increased tumorigenesis, tumor invasiveness, activation of caspase-3 in the promotion of cancer apoptosis and downregulation of vascular endothelial growth factor in the establishment of new blood vessels for the bladder cancer to grow and proliferate..

These results are coincidently similar to those shown previously for AITC and are consistent with the understanding that NAC-AITC is a carrier of AITC.

After taking into account of all co and confounds and the result of the previous study, Dr.Bhattacharya A the lead author once again said, "comparison of the pharmacokinetic and physical properties of NAC-AITC with those of AITC suggests that NAC-AITC is superior to AITC for potential use for prevention and therapy of bladder cancer".

Taken altogether, Phytochemical Allyl isothiocyanate may be considered a functional phytochemical in the prevention of bladder cancer and combined with primary therapy for the treatment of bladder cancer, depending to the large and multi-center in the human study.
