Omega 3 Fatty Acids, the Natural Non-Opioid Analgesic Drug

Non-opioid analgesics are types of medicine used to disrupt the production of prostaglandin associated with pain and inflammation, including nonsteroidal anti-inflammatory drugs (NSAIDs), selective COX-2 inhibitors, and acetaminophen. NSAIDs inhibit cyclooxygenases (COX-1 and COX-2).

Pain relief is an action to remove pain, distress, oppression by blocking the pain-transmitting information from nerve cells to the brain neurons.

Inflammation is a systematic immune response during the acute phase of infection.

Macrophages, one the lymphocytes and the first-line defense of the immune system after sensing the damage of localized tissue, stimulate the production of proinflammatory cytokines to the site to protect the body against invasion of potential pathogens.

Acute pain is any pain that lasts less than 6 months. Most cases of acute pain are due to physical damage such as soft tissue damage of a sprained ankle or a paper cut. Acute pain is relatively more sharp and severe.

On the other hand, there are many risk factors associated with chronic pain. Some chronic pains can be caused by single factors such as diseases involved nerve damage to injuries that fail to heal properly. Others may be results from several factors including diseases associated with normal aging.

Acute inflammation is a natural response of the immune system to protect the human body against harmful stimuli and pathogens.

Acute inflammation usually receded after a few days or weeks depending on the size and damage. However, if all pathogens were not eradicated after 3 to 8 weeks, the immune system will compromise, leading to chronic low-grade inflammation, a leading cause of chronic illness.

The most common symptoms of acute inflammation occurred with a short period of time such as hours is pain, redness, immobility, swelling, and heat.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of pain, researchers examined the mechanism of pro-resolving lipid mediators (SPMs) on an active and time-dependent biosynthetic shift from pro-inflammatory to pro-resolution mediators,

SPMs are biosynthesized from the omega-3 fatty acids arachidonic acid (AA), eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA), or docosahexaenoic acid (DHA).

According to the tested analysis, isolated SPMs show a strong efficacy at very low doses to relieve rheumatic diseases and neuropathic pain, in experimental models showing no side effects compared to biological agents, opioids, corticoids, and gabapentinoids.

Furthermore, SPMs also act as an immunoresolvents to induce long-lasting analgesic and anti-inflammatory effects, including blocking pain.

Based on the findings, researchers said, "therefore, (pro-resolving lipid mediators (SPMs)) emerge as a new class of non-immunosuppressive and non-opioid analgesic drugs".

Taken altogether, Omega-3 fatty acids may be considered a non-opioid analgesic drugs for the prevention and treatment of diseases,  pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
