Bromelain Found in Pineapple Suppresses the Symptoms and Onset of Osteoarthritis (OA)

Osteoarthritis is an inflammation of joints caused by the depletion of the cartilage on the ends of the bones due to the wear and tear over time.

The disease is one most common form of arthritis that affects millions of people worldwide.

According to the statistics, in the US, approximately 10% of men and 13% in women aged 60 years or older have osteoarthritis (OA).

Most common risk factors found in patients with OA are older adults, female gender, overweight and obesity, knee injury, repetitive use of joints, low bone density.

Besides pain on the joints, other most common symptoms of OA include tenderness, stiff, low flexibility, grating sensation. 

Conventionally, treatment of OA is focused on managing symptoms, by reducing pain, stiffness and swelling through the use of pain killers, improving joint mobility and flexibility and maintaining a healthy weight by exercise.

Some researchers suggested that the high prevalence of OA in the US has become a burden to the health care cost and economic burden.

Dr.Bitton R, the lead scientist at the Health Plan of Nevada wrote, "approximately one-third of direct OA expenditures are allocated for medications, much of which goes toward pain-related agents. Hospitalization costs comprise nearly half of direct costs, although these expenditures are consumed by only 5% of OA patients who undergo knee or hip replacement surgery".

And, "Indirect costs for OA are also high, largely a result of work-related losses and home-care costs".

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural compound for diseases associated with inflammation, researchers investigated the effects of bromelain in mild-to-moderate knee OA patients.

The randomized, single-blind, active-controlled pilot study included 40 knee OA patients randomized to receive oral bromelain (500 mg/day) or diclofenac (100 mg/day). 

At week 4, both bromelain and diclofenac treated groups showed an improvement of total the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and pain subscales compared to the baseline.

According to the tested assays, bromelain and diclofenac groups exerted similar effects after 4 weeks.

At week 16 of bromelain treatment, patients had improved total WOMAC scores (12.2 versus 25.5), pain subscales (2.4 versus 5.6), stiffness subscales (0.8 versus 2.0), and function subscales (9.1 versus 17.9), and physical component of SF-36 (73.3 versus 65.4) as compared with baseline values.

In other words, bromelain reduced symptoms and improved quality of life in OA patients by inhibiting the levels of oxidative stress and overexpression of pro-inflammatory cytokines at week 16. 

Based on the results, researchers after taking other factors into account wrote, "Our results show the inhibitory effect of enzyme therapy on collagen-induced arthritis in rats, as well as the efficacy of cyclosporin".

Taken altogether, bromelain may be considered supplements for the prevention and treatment of osteoarthritis (OA), pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Plant-Based Bioactive Compound 3,3'-Diindolylmethane Kills Lung Cancer Cells in Vivo

Lung cancer is a medical and chronic condition associated with the alternation of the cells in the lung tissue.

Most cases of lung cancer start in the cell on the surface of the inner lining of lung tissue before striking into deeper layers.

At the early stage, due to the small size of the tumor, most patients are asymptomatic. However, at the advanced stage, besides the common symptoms shared with other types of cancer, the patients may also experience symptoms of difficult breathing, persistent cough and hoarseness, and chest pain.

Some patients may also vomit blood caused by the oversize tumor suppressing on the nearby blood vessels.

Lung cancer is diagnosed mostly in the elderly, particularly in people over 65 years of age.
The mortality rate of lung cancer is higher than the colon, breast, and prostate cancers combined.

According to the statistics, In Canada, the 5-year net survival for lung cancer is 17%.

In other words, on average, only 17% of people diagnosed with lung cancer will survive for at least 5 years after their diagnosis.

However, lung cancer at the early localized stage has 5 years of the survival rate of 60%.

Although there are many risk factors associated with the onset of lung cancer, smoking is considered epidemiologically a major cause of the disease onset.

Dr. Ozlü T, the lead scientist in the examination of smoking and lung cancer risk, "The association between cigarettes and lung cancer has been proven by large cohort studies. Tobacco use has been reported to be the main cause of 90% of male and 79% of female lung cancers. 90% of deaths from lung cancer are estimated to be due to smoking. The risk of lung cancer development is 20-40 times higher in lifelong smokers compared to non-smokers".

And, "After smoking cessation, the cumulative death risk from lung cancer decreases. Patients who continue smoking experience greater difficulties during cancer treatment. Stopping smoking may prolong survival in cancer patients, and also decreases the risk of recurrent pulmonary carcinoma".

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

Researchers in the concern about the risk of inflammatory conditions of the lung such as chronic obstructive pulmonary disease (COPD) that contributes to the onset of lung cancer risk, particularly lung squamous cell carcinoma (LSCC) examined the chemopreventive effects of BioResponse diindolylmethane (DIM).

The study included a mouse model of inflammation-driven LSCC induced by N-nitroso-trischloroethylurea (NTCU) and enhanced by lipopolysaccharide (LPS), a potent pro-inflammatory agent contained in tobacco and tobacco smoke.

Before treatment of DIM, mice treated with the combination of NTCU and LPS had a 9-fold increase in the number of bronchioles with LSC.

Compared to mice treated with LPS alone, mice treated with NTCU plus LPS showed significantly increased expression of the pro-inflammatory cytokines, accompanied by the increased expression of proteins associated with the production of pro-inflammation and cancer cells initiation.

Pretreatment of DIM at a concentration of 0 μmol/g diet or 2,460 ppm) to mice treated with NTCU plus LPS reduced the incidence of LSCC by 2-fold and suppressed activation/expression of proinflammatory and procarcinogenic proteins without affecting the expression of cytokines and p53, a tumor suppressive gene.

In other words, DIM blocked the development of LSCC through cytotoxicity and gene associated with tumor suppression.

In order to reveal more information about DIM anti-lung cancer property, researchers looked into the anticancer efficacy of 1,1-bis (3'-indolyl)-1-(p-biphenyl) methane (DIM-C-pPhC₆H₅) inhalation delivery alone and in combination with i.v. docetaxel in a murine model.

According to assays, the lung weight reduction in mice treated with the drug combination was 64% compared with 40% and 47% in mice treated with DIM-C-pPhC₆H₅ aerosol and docetaxel alone, respectively.

The combination treatment inhibited the lung cancer by decreased expression of proteins associated with the pro-inflammatory cytokines, cancer cells survival, and proliferation through stimulating the production of the new blood vessels.

Furthermore, the combination also increased expression of genes involved autophagy in the tumor formation, compared to the tumor cell counts from single-agent treatment and control groups.

Dr. Ichite N, the lead scientist said, "These results suggest that DIM-C-pPhC₆H₅ aerosol enhanced the anticancer activity of docetaxel in a lung cancer model by activating multiple signaling pathways".

Taken altogether, 3,3'-Diindolylmethane may be considered a supplement for the prevention and combined with primary medicine for the treatment of lung cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Plantbased Bioactive Compound Sinigrin, the Anti Fungal Infection Bioactive Compound

A fungus is a primitive organism, including mushrooms, mold, and mildew.found in the air, in the soil, on plants, and in water.

Fungi reproduce through tiny spores in the air if you inhale the spores, it may spread to cause a fungal infection that often starts in the lungs or on the skin. 

People with a weakened immune system are more likely to infected by fungi, including the fungus Aspergillus which cause lung infection.

Similar to other bacterias, there are millions of different fungal species on Earth, but only about 300 are known to infect people.

Conventionally, fungal infection is treated with antibiotics, including topical antifungal ointments or oral medications, depending on the severity of the disease.

If you are currently taking antibiotics for the treatment of fungal infection, please make sure that you know all possible side effects for preventive measure.

Most common symptoms of skin fungal infection are red and cracking or peeling skin accompanied by itching, depending on the location of affected skin.

In a serious case of fungal lung infection, accumulated fungus fibers, blood clots, and white blood cells may appear in the lungs or sinuses, leading to coughing up blood or fever, chest pain, and difficulty breathing, depending to the size of the accumulation.

In the seeking a natural compound for the treatment of fungal infection, researchers analyzed the antifungal infection of the roots of twelve plant spades from bath mycotrophic (hosts of YAM fungi) and non‐mycotropic (non‐hosts) families against fungus Cladosporium cucumerimun Ellis & Arch.

2 species from the Brassicaceae such as Chenopodiaceae (Spinacea oleracea L., Beta vulgaris L.)  showed a significant effect on fungus compared to Brassica kaber (DC.) Wheeler, Brassica napus L., Brassica campestris L., Thlaspi arvense L., Raphanus raphasnistrum L. which showed a limit activity.

According to the TLC bioassay, Chloroform extracts of B. Kaber roots appeared to have the greatest quantity of extractable antifungal compounds.

Three antifungal compounds in chloroform extracts from B. Kaber roots were shown to be derived from glucosinolates, containing high amount sinigrin.

The finding strongly supports the use of sinigrin for the treatment of fungal infection.

Furthermore, in the differentiation of the fungitoxicity of allyl‐isothiocyanate (AITC) vapor produced from pure sinigrin against Penicillium expansum, the causal agent of blue mold on pears, 5 mg L−1 AITC‐enriched atmosphere showed the best control of blue mold by exposing fruits for 24 h.

At 1 × 103 conidia mL−1to almost 80% at 1 × 106 conidia mL−1, AITC decreased the percentage of infected wounds, compared to >98% of wounds infected in untreated fruits.

Moreover, AITC treatments were effective up to 24 h after inoculation for Conference and 48 h for Kaiser. AITC treatments also controlled a thiabendazole‐resistant strain of P. expansum, reducing the incidence of the blue mold by 90% in both cultivars.

Dr. M. Mari, at the end of the experiment, wrote, "The use of AITC produced from pure sinigrin or from Brassica junceadefatted meal may be an economically viable alternative to synthetic fungicides against P. expansum".

Taken altogether, sinigrin may be considered a supplement for the prevention and treatment of fungal infection, pending to the confirmation of large sample size and multicenter human study.

However, intake of bioactive in the form of supplement should be taken with extreme care, as liver toxicity may be induced if overdosed.

Plant-based Chemical Betanin Kills Cancer Cells in Vitro

Cancer is the class of diseases associated with the cells growth disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.
The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Compared to all cancers, lung, breast, prostate, and colorectal cancers are the most common cancer in the US.

The causes of cancer are unknown. age, excessive alcohol drinking, exposure to cancer causing-substances, diseases associated with chronic inflammation,  unhealthy diet,  hormones therapy, use of immunosuppression are some of the prevalent factors involved the onset of cancer.

However, most people with some of the aforementioned risk factors have never developed cancer.

Some researchers suggested that the rise of cancers in the obese individual may correlate to the promotion of the Western diet over the past few decades.

Dr.Timothy J Key, the lead scientist wrote, "Diet-related factors are thought to account for about 30% of cancers in developed countries. Obesity increases the risk of cancers in the esophagus, colorectum, breast, endometrium, and kidney. Alcohol causes cancers of the oral cavity, pharynx, larynx, esophagus, and liver, and causes a small increase in the risk of breast cancer".

And, "Adequate intakes of fruit and vegetables probably lower the risk for several types of cancer, especially cancers of the gastrointestinal tract".

Betanin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

With an aim to find a natural ingredient which reduces the side effects of potent anticancer drugs e.g. doxorubicin (DOX) on the normal tissues, researchers examined betanin (BET) effect in the co-administration with them.

In the study to overcome the restrictions of BET, researchers designed a new pH-responsive nanocarrier via decoration of gelatin nanoparticles (GNPs) by (methoxy poly (ethylene glycol)-poly ((2-dimethylamino) ethyl methacrylate-co-itaconic acid) (PGNPs) for the DOX and BET loading capacity of 20.5% and 16.25%, respectively).

Injection of DOX@BET loaded PGNPs decreased the significant cell viability amount of breast cancer cells (MCF-7) compared to the treatment of the free form of DOX or BET. 

In other words, the newly designed combination kills induce a higher percentage of cell death compared to the use of DOX or BET individually.

Sadly, the study did not confirm the combination reduced the side effect caused by DOX  during the experiment.

Dr. Javadi B in the examination of herbal plants for the treatment of breast cancer showed that beetroot (Beta vulgris) induced cancer death by cytotoxicity without affecting the nearby healthy cells.

He continued, "additional pharmacological studies and clinical trials are needed to elucidate their molecular and cellular mechanisms of actions, frequency, and amount of consumption, possible adverse effects, and optimum preparation methods.".

Taken altogether, betanin used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of cancer, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Ginger, the Potential Anti Hyperlipidemia Functional Remedy, Researchers Find

Hyperlipidemia is abnormally high levels of cholesterol in the bloodstream. The syndrome is a member of the cluster conditions of metabolic syndrome, a leading cause of cardiovascular disease.

Cholesterol, a waxy substance produced either by the liver or food intake plays a critical role in the production of vitamin D, steroid hormones and the building of the cell walls.

Overexpression cholesterol is a medical condition which causes plaque accumulated on the arterial wall, leading to narrowing the arteries in transport the oxygenated blood to the parts of the body.

In severe cases, it can cause blood flow blockage, leading to chronic ailments depending on the location of the blood clot.

There are 2 types of blood cholesterol, the low-density lipoprotein (LDL) which carries the aforementioned functions and the high-density cholesterol (HDL) which carries cholesterol from the blood back to the liver for the future re-use.

The healthy ratio of total blood cholesterol is less than 4 LDL/ 1 HDL.

Most people diagnosed with high blood cholesterol substantially higher total cholesterol ratio.

Some researchers suggested that the widespread high blood cholesterol in North America is associated with the promotion of the Western diet over the past few decades.

Dr. Kamilah Ali, the lead scientist in the assessment of the Western diet risk of high blood cholesterol in an animal model, wrote, "mice on a C57BL/6 background after receiving a high fat-high cholesterol diet for 12 weeks. ApoD−/− mice had higher HDL-cholesterol levels (61±13-apoD−/− vs. 52±10-WT-males; 37±11-apoD−/− vs. 22±2 WT-female) than WT mice with sex-specific changes in total plasma levels of cholesterol and other lipids".

And, "Compared to WT, the HDL of apoD−/− mice showed an increase in large, lipid-rich HDL particles and according to size various quantities and sizes of LDL particles".

The results suggested that long-term users of the Western diet may be associated with the risk of hyperlipidemia.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.


The root has been used in traditional and Chinese medicine for the trerament dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.


Researchers on finding a natural compound for the treatment of hyperlipidemia investigated the effect of ginger extracts on alcohol-induced hyperlipidemia and associated myocardial damage.

The study included 24 Wistar male albino rats (250 ± 20 g) divided into four groups including, normal control (NC) (0.9% NaCl), ginger treated (Gt) (200 mg/Kg b.w.), alcohol-treated (At) (20% of 6g/kg b.w. alcohol), and alcohol along with Ginger treatment (At+Gt).

Before ginger administration, alcohol-induced myocardial damage rats showed an increased lipid profile except for high-density lipoprotein compared with control.

Ginger treatment significantly inhibited the alcohol-induced lipid profiles caused by the injection of alcohol except for high-density lipoproteins.

Furthermore, ginger treatment also inhibited the elevated cardiac biomarkers activity with alcohol intoxication. In other words, ginger treatment for 7-weeks significantly minimized the alcohol-induced myocardial damage.

Dr. Subbaiah GV, the lead scientist said, "ginger could protect alcohol-induced myocardial damage by suppression of hyperlipidemia and cardiac biomarkers".

In order to reveal more information about ginger anti hypolipidemic effects, researchers examined the hypolipidemic effects of a hydroethanolic ginger extract (GE) in the liver of rats fed a high-fat diet (HFD).

The study included male Wistar rats divided into the following 3 groups: (i) Rats fed a standard diet (3.5% fat), the control group; (ii) rats fed an HFD (33.5% fat); and (iii) rats fed an HFD treated with GE (250 mg·kg-1·day-1) for 5 weeks (HFD+GE).

GE improved lipid profile and attenuated the increase of plasma levels of glucose, insulin, and leptin in HFD rats.

In the depth analysis, researchers found that ginger antihyperlipidemic activity was attributed to the induction of a higher liver expression of proteins associated with a variety of metabolic processes including lipid and enhanced levels of adiponectin which plays a critical role in preventing dyslipidemia.

Taken altogether, ginger may be considered a functional remedy for the prevention and treatment of hyperlipidemia with no side effects, pending to the confirmation of large sample size and multicenter human study.

Bioactive Compound 3,3'-Diindolylmethane Protects the Neurons Integrityand Viability Against Autoimmune Encephalomyelitis,

Autoimmune encephalitis is a group of conditions caused by the body's immune system mistakenly attacks healthy brain cells.

Autoimmune encephalitis has been found to induce neurologic symptoms such as impaired memory and cognition, abnormal movements, seizures, and/or psychiatric symptoms such as aggression, inappropriate sexual behaviors, panic attacks.

Autoimmune encephalitis can be classified into
* Post-infectious encephalitis or acute disseminated encephalomyelitis (ADEM) are the conditions caused by an infection. In these cases, the immune system not only attacks the pathogens which cause infection it also destroys mistakenly the healthy cells in the brain.

Methylprednisolone is the first line of drug for the treatment of post-infectious encephalitis, if it failures, plasma exchange or intravenous immunoglobulin may be used.

* Other forms of autoimmune encephalitis are caused by the specific antibodies in the blood produced by the immune system that attack the brain cells. 

According to the Autoimmune Encephalitis Alliance, a number of options are available to treat AE, the first line option include including medication that decreases the inflammatory response to antibodies, and intravenous immunoglobulin (IVIG); to increase removal of antibodies, inhibit binding of the harmful antibodies.

If the first line option failure, immune suppressive medicine may be used as a second line of treatment, including Rituximab, CellCept, and Cytoxan (Cyclophosphamide).

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

With an aim to find a potential ingredient for the treatment of autoimmune encephalomyelitis, researchers evaluated the 3,3'-Diindolylmethane (DIM) effects on the regulation of activated T cells during the development of experimental autoimmune encephalomyelitis (EAE).

During the experiment, administration of DIM 10 days after EAE was effective in ameliorating disease parameters, including inflammation and central nervous system cellular infiltration.

According to the microRNA (miRNA) microarray analysis, post-treatment of DIM alter the expression of miRNA profile in brain infiltrating CD4(+) T cells, an indication of autoimmune dysfunction on EAE mice. 

Observed by the bioinformatics analysis, the efficacy of DIM-altered the miRNAs expression associated with pathways that halt cell cycle progression and promote apoptosis. 

Particularly, DIM impacted these cellular processes in activated T cells.

More precisely, DIM protected the brain against the immune dysfunction in attacking the brain cell in the forming of autoimmune encephalomyelitis, by activating and suppressing the micro genes- induced immune dysfunction of EAE.

Dr. the lead scientist, after taking into account co and confounders wrote, "these studies demonstrate that DIM post-treatment leads to the amelioration of EAE development by suppressing T-cell responses through the induction of select miRNAs that control cell cycle progression and mediate apoptosis".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of autoimmune encephalomyelitis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Tomato Improves Sleep Quality by Enhancing Levels of Melatonin

Sleep is a naturally recurring state of mind and body in human, In sleep, most functions of the brain are reduced including the sensory activity, voluntary muscles are inhibited.

In other words, sleep is the state in which human reduced interactions with the outside world.

The human sleep recurrently to reenergize their body so that when awake they can carry out a daily activity without feeling tired or weak.

In the brain, sleep helps to maintain the pathways associated with brain function such as learning and studying. A recent study suggested, sleep helps to remove toxins in the brain that was built up when  awake.

Believe it or not, we spend about one-third of our life sleeping. 

Sleep disorders are syndromes associated negatively with the changes in the sleep pattern, including insomnia, one of the most common causes of disease disorders in the US, found in 30% of adults, including 10% of chronic insomnia is a condition of difficulty falling asleep or staying asleep throughout the night.

According to the statistics, 50-70 million US adults have a sleep disorder including 37.9% reported unintentionally falling asleep during the day.

Sleep disorders have been found to have a strong implication on economic cost. Dr. Hillman DR, the lead scientist at the Sir Charles Gairdner Hospital wrote, "The overall cost of sleep disorders in Australia in 2004 (population: 20.1 million) was dollar 7494 million".

And, "The direct and indirect costs of sleep disorders are high. The total financial costs (independent of the cost of suffering) of dollar 4524 million represents 0.8% of Australian gross domestic product. The cost of suffering of dollar 2970 million is 1.4% of the total burden of disease in Australia".

Tomato is red, edible fruit, genus Solanum, belongs to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

On finding a natural and potential compound for the improvement of sleep quality, researchers investigated the effect of tomato intake before bed on obese postmenopausal women's urinary 6-sulphatoxymelatonin (aMT6s) level, a reliable surrogate biomarker reflecting the blood melatonin concentration and sleep quality.

The study is an 8-week open-label, randomized controlled dietary intervention trial included 36 subjects completed the entire trial.

In the tomato group, participants ate 250 g of beefsteak tomatoes 2 h before sleep for 8 weeks.

After 8 weeks, by comparing the blood and urine samples collected at the baseline and in the 8th week, researchers found at 8 weeks of the beefsteak intervention, improved all components of all components the PSQI compared to the Pittsburgh Sleep Quality Index (PSQI) before treatment.

Furthermore, tomatoes and beefsteak intervention also improved the levels of aMT6s level by 10-fold compared that of the control group.

In other words,  tomato combined with beefsteak improved the sleep quality of the participants.

These results strongly suggested that tomato enhanced the levels of melatonin in the beef substantially in the improvement of sleep quality.

Dr. Yang TH, the lead scientist at the Taipei Medical University wrote in the final report, "supplementation with beefsteak tomato before sleep can increase circulating melatonin and improve sleep quality in obese postmenopausal women".

Taken altogether, tomatoes combined with beefsteak may be considered treatment to improve sleep quality, pending to the confirmation of the larger sample size and multicenter human study.
