Common Fig or Fig (Ficus carica) Gobbles Up Glioblastoma GBM Cells in Vitro

By Kyle J. Norton

The brain and spinal cord made up the nervous system. Brain and Spinal Cord tumors originating are not very common. Most primary tumors are caused by abnormal cell growth that surrounds and supports neurons, with a small number may be caused by gene defect from exposure to radiation or toxic chemicals. Benign tumors are noncancerous, and malignant tumors are cancerous.

More than 190.000 people in the US are diagnosed with a brain tumor each year alone.

Brain cancer is a medical and chronic condition characterized by cell growth disorderly in the brain tissue.

Glioblastoma or glioblastoma (also called GBM) caused mostly by the abnormal astrocytic cells and dead cells (necrosis) is considered one of the most aggressive cancers induced by the alternation of cells originated from the brain, including the cerebral hemispheres.

Most patients with GBM experience symptoms of brain swelling caused by the overexpression of the fluid surrounding the tumor.

Conventionally, due to the nature of rapid growth, surgery is necessary to relieve the brain pressure and remove the tumor as much as possible, followed by radiation and chemotherapy.

According to the statistics, in the US, the incidence affects mostly in the elderly with the diagnosed rate of 3.19 per 100,000 persons. The disease is rare in children.

GBM is deadly, patients with GBM have a median survival of less than one year and less than 2% of patients survive three years or more.

Fig is a species of F. Carica, belonging to the family Moraceae. It is a deciduous shrub, native to Southeast Asia. The tree can grow to a height of 33 ft and the fruit can be as large as 2.0 inches.

On finding natural compounds for the treatment of neurological diseases, researchers examined the cancer suppressive effect of Ficus carica (fig) latex (FCL) on glioblastoma (GBM).

On T98G, U-138 MG, and U-87 MG GBM cell lines treated by the combination of FCL and  FCL-temozolomide (TMZ), according to the tested differentiation,
* FCL causes cell death in GBM cells with different responses to FCL-temozolomide (TMZ), an oral chemotherapy drug.

* The cell death was attributed synergistically increased in combination with TMZ

* More importantly, the effect of FCL on the modulation of let-7d expression by suppressing proliferation and cell migration may be an important underlying mechanism of the anti-invasive effect of this extract.

Taken altogether, common fig or fig (Ficus carica)  used in a combination with chemo drug may be considered a remedy for the treatment of glioblastoma (GBM), pending to the confirmation of the larger sample size and multicenter human study.

Broccoli, the Natural Antioxidants

By Kyle J. Norton

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid and cell damage.

Antioxidants can either produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

However, in people with a weakened immune system, intake dietary sources of antioxidants may be necessary.

There are 3 main types of antioxidants found in nature, including
* Antioxidant enzymes produced by the host, including superoxide dismutase (SOD), glutathione peroxidase, glutathione reductase, and catalases.

* Antioxidants vitamins, that may be injected from the dietary sources such as fruits and vegetables, including vitamins A, C, D, and E. Most antioxidant vitamins are considered free radical scavengers that gobble up free radicals before they can induce a cascade of damage.

* Antioxidant Phytochemicals found in the plant including second metabolites.

Antioxidants keep our bodies functioning properly by reducing the risk of diseases associated with oxidative stress.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily to donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of diseases associated with oxidative stress, researchers examined the phytochemical composition of 3 different purple sprouting broccoli varieties according to with their production cycle: EEP (Extra Early), EP (Early), and LP (Late) antioxidant activity.

The leaves of EP variety seems the richest in total phenolic acids, whereas the number of anthocyanins was higher in LP variety.

The levels of glucosinolates were higher in purple broccoli than in green cultivars and also higher than other purple broccolis grown under different climate conditions.

Additionally, EEP, EP, and LP purple sprouting broccoli varieties grown under the Mediterranean climate. showed a number of health-promoting nutrients and natural antioxidants.

The vitamin C, phenolic compounds, and glucosinolates in these purple varieties were higher than in traditionally grown green broccoli and other purple broccolis grown under different climate conditions.

Taken altogether, broccoli varieties may be considered antioxidant supplements for the prevention of oxidative stress disease, pending to the confirmation of the larger sample size and multicenter human study.

Beta-Sitosterol Reduces the Onset of Gouty Arthritis in Vivo

By Kyle J. Norton

Arthritis is a general form of the inflammatory disease associated with the inflammation of one or more joints.

The most common symptoms are joint pain and stiffness found mostly in the aging population.

The most common caused of arthritis is a joint injury that did not heal properly, metabolic abnormalities caused by overexpression of uric acid and infection-induced bacteria or virus.

However, there are some cases of arthritis caused by autoimmunity, a condition in which the immune system mistakenly attacks the joint tissues, such as in rheumatoid arthritis and systemic lupus erythematosus.

According to the information provided by the Arthritis Foundation, in the US, approximately 54 million adults are living with the condition.

Believe it or not, almost 300,000 babies and children have arthritis or a rheumatic condition.

Gout, a painful and recurrent condition mostly affected one joint is a type of arthritis caused by uric acid building up in the blood that induces joint inflammation.

Epidemiologically, high levels of uric acid found in the body that leads to the forming of crystals are the culprit, caused by the inability of the kidney to get rid of uric acid.

Most cases of gout are found in patients who follow the diet with a high intake of meats and seafood and foods containing higher levels of purine such as alcoholic beverages, especially beer, and drinks sweetened with fruit sugar (fructose).

Reduced intake of the aforementioned products has been found to reduce the onset of recurrence of the condition in many previous patients.

Believe it or not, most patients with gout and recurrent gout also associated with medical conditions of hypertension, renal insufficiency, and hypertriglyceridemia.

Gouty arthritis is a form of inflammatory arthritis associated with high levels of uric acid in the blood that lead to sudden, severe episodes of pain, tenderness, redness, warmth, and swelling of the joints.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of arthritis, researchers examined the effects of ethanolic extract of L. trichocarpha, its ethyl acetate fraction and its main bioactive compounds on gouty arthritis by countering hyperuricemia and inflammation.

According to the results of an experimental model of gouty arthritis,
* Crude ethanolic extract and its ethyl acetate fraction showed significant urate-lowering effects.

* LTE was also able to significantly inhibit liver xanthine oxidase (XOD) activity in vivo at the dose of 250mg/kg.
\
* LTE, LTA, lupeol, β-sitosterol, lychnopholide, eremantholide, luteolin, and apigenin showed strong inhibitory activity against monosodium urate crystals-induced paw edema in mice.

Based on the findings, researchers said, "Ethanolic extract of Lychnophora trichocarpha and some of its bioactive compounds may be promising agents for the treatment of gouty arthritis since they possess both anti-hyperuricemic and anti-inflammatory properties.".
Taken altogether, beta-Sitosterol found abundantly in Lychnophora trichocarpha may be considered a supplement for the prevention and treatment of gouty arthritis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Omega-3 Fatty Acids, the Potent Lipid Inflammatory Mediators in Vitro and Vivo

By Kyle J. Norton

Inflammation is a natural response by the immune system to protect the body against the invasion of stimuli or foreign pathogens.

Acute inflammation is a short-term condition caused by tissue injury, leading to pain, redness, swelling, and heat to the affected area. depending on the size of the injury. Some patients with acute inflammation may also experience the symptom of loss of function.

In acute inflammation, the white blood cells on the first line of defense of the immune system after sensing the danger of an infection in the body through a message from the brain stimulate the production of platelets to cover the site of injury or damage and inflammatory cytokines to kill off all pathogens before they can cause harm to the body.

Most cases of infection are stopped at the acute phase. The wound or injury is slowly recovered.

However, people with a weakened immune system, acute inflammatory progression cannot be stopped due to the inability of the immune system to destroy all invaders, which may lead to low-grade chronic inflammation.

Lipids mediators, derived from the omega-6 polyunsaturated fatty acids (PUFA) are potent enhancers of innate and adaptive immune activation in numerous inflammatory disorders.

Dr. Bennett M, the lead scientist in the examing "Lipid Mediators in Inflammation" wrote, "Lipids are potent signaling molecules that regulate a multitude of cellular responses, including cell growth and death and inflammation/infection, via receptor-mediated pathways. Derived from polyunsaturated fatty acids (PUFAs), such as arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), each lipid displays unique properties, thus making their role in inflammation distinct from that of other lipids derived from the same PUFA".

In other words, each individual acid arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), each lipid displays unique properties which are derived from polyunsaturated fatty acids (PUFAs), plays a selective role in maintaining cellular process depending on the types of fatty acid.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of inflammatory diseases, researchers examined the effects of n3-PUFA DHA and its derived specialized pro-resolving lipid mediators (SPMs) MaR1, RvD1, and RvD2, on TNF-α inhibition of intestinal sugar and glutamine uptake.

Based on the differentiation,
* DHA blocked TNF-α-induced inhibition of α-methyl-D-glucose (αMG) uptake, an analog of glucose and SGLT1, a glucose transporter, expression in the apical membrane of Caco-2 cells.

* Compared to DHA, SPMs showed the same preventive effect at concentrations 1000 times lower.

* In diet-induced obese (DIO) mice, oral gavage of SPMs, MaR1 reversed the up-regulation of pro-inflammatory cytokines found in the intestinal mucosa of these mice without inducing a reduced intestinal transport of αMG and SGLT1 expression in the DIO mice.

* In Caco-2 cells, EPA, DHA, and the DHA-derived SPMs. also prevent the TNF-α in the inhibition of glutamine uptake.

Collectively, scientists said, " DHA and its SPMs as promising biomolecules to restore intestinal nutrients transport during intestinal inflammation".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the treatment of diseases associated with the impaired lipid inflammatory mediators, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol Inhibits the Parameters Associated with the Risk of Diabetes

By Kyle J. Norton

Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream to regulate the glucose.

In other words, diabetes occurs when cells in the pancreas dying off or receptor sites clogged up by fat and cholesterol. In some cases, diabetes is also caused by allergic reactions of cells in the immune system.

There are 2 major types of diabetes,
* Type 1 diabetes, also known as juvenile diabetes or insulin-dependent diabetes, the most common form diabetes found in children is a condition in which the pancreas does not produce any insulin. The most common form of type 1 diabetes is caused by genetic mutation and some viruses.

* Type 2 diabetes is a medical condition characterized by insufficient insulin entering the bloodstream to regulate the glucose due to either cell in pancreas dying off or receptor sites clogged up by fat and cholesterol.

The most common symptoms of type 2 diabetes are an increased thirst that won't stop with water intake, increased hunger.

Diabetic complications are any diseases associated with hyperglycemia if the blood glucose is not managed properly. Patients with diabetes may induce a number of complications including cardiovascular disease, neuropathy, nephropathy, retinopathy, and foot ulcer.

By managing your blood glucose properly and daily, the risk of diabetic complications can be reduced in diabetics by
*Taking your medication as directed.

*Keeping tight control of your blood sugar or managing the blood glucose in the required range

* Eating healthy with moderate exercise to protect your weight, blood pressure and cholesterol. Overweight, blood pressure and cholesterol are some of the conditions associated with increased risk of diabetes complications.

* Get regular checkups

* Quit smoking

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention of chronic diseases involved in oxidative stress, researchers investigated the anti-diabetes activity of beta-sitosterol (β-sitosterol) on insulin signaling molecules and glucose oxidation.

According to the differentiation of the adipose tissue of high-fat diet and sucrose-induced type-2 diabetic experimental rats given the dose of β-sitosterol (20 mg/kg b.wt, orally for 30 days), β-sitosterol normalized the altered levels of blood glucose, serum insulin in the tested rats.

Furthermore, the hormone testosterone, lipid profile, oxidative stress markers, antioxidant enzymes, insulin receptor (IR), and glucose transporter 4 (GLUT4) proteins associated with glucose uptake were also inhibited by the application of beta-sitosterol.

In other words, most parameters associated with the characteristics of diabetes were inhibited by the injection of the bioactive compound.

Based on the findings, researchers aid, "β-sitosterol improves glycemic control through activation of IR and GLUT4 in the adipose tissue of high fat and sucrose-induced type-2 diabetic rats".

Taken altogether, beta-Sitosterol may be considered a supplement for the treatment of diabetes, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

α-Tocopheryl succinate (α-TOS) Protects the Brain Against the Onset of Cerebral Malaria in Vivo

By Kyle J. Norton

The brain formed part of the central nervous system is the most complex organ made up of more than billions of nerve cells or neurons.

The functions of the brain are associated with the communication of trillions of connections called synapses in many specialized regions.

A neuron or nerve cell made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral
Neurological disorders are diseases of the central nervous system including the brain, spine and the nerves that connect them. The most common diseases associated with neurological disorders in the elderly are dementia.

Cerebral malaria is the most severe case of neurological complications caused by the infection with the Plasmodium falciparum malaria and parasite on peripheral blood smears.

The most common symptoms of cerebral malaria are fever, headache, body ache and progressively, delirium and coma.

However, in adults, cerebral malaria is part of a multi-organ disease.

In African children, seizures papilledema and retinal changes are less common and coma resolution is slower, compared to other races.

Conventionally, the first-line treatment of cerebral malaria is therapy with concentrations of anti-malarial drugs as quickly as possible.

Tocopherols are phytochemicals of which many have vitamin E activity, belongings to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of cerebral diseases, researchers examined the effects of α-Tocopheryl succinate (α-TOS), a derivative of vitamin E, synthesized by esterification of α-tocopherol for treatment of cerebral malaria.

On C57BL/6J mice infected with P. yoelii 17XL and P. berghei ANKA, a lethal strain of rodent malaria, treatment with several concentrations of α-TOS by intraperitoneal administration on 1, 3, 5, and 7 days post-infection (dpi) showed a strong effect on Plasmodium parasites by oxidative stress.

Moreover, α-TOS treatment decreased the parasitemia and increased the survival rate significantly in mice infected with both parasites.

Additionally, the intensity of Evans blue staining on brains taken from α-TOS-treated mice was weaker than that of untreated mice.

In other words, α-TOS inhibited the breakdown of BBB that allow the initiation and progression of cerebral malaria.

Collectively, researchers said, "These findings indicate that vitamin E derivatives like α-TOS might be a potential candidate for treatment drugs against malaria".

Taken altogether, tocopherols may be considered a supplement for the prevention and treatment of cerebral malaria, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
