Gamma-Linolenic Acid Inhibits of Colonic Inflammation in Vivo

By Kyle J. Norton

Colonic inflammation is a condition associated with inflammation of the colon of the digestive system.

Inflammation is a natural immune response involving the stimulated production of pro-inflammatory activity with a target to protect our body against foreign invasions such as virus and bacteria after injury and damage.

In other words, pro-inflammatory cytokines are activated through systematic immune response to the damage cells which release chemicals including histamine, bradykinin, and prostaglandins to protect the injure area or location against infection, leading to blood vessels to leak fluid into the tissues, causing swelling and other symptoms such as pain, swelling, and redness.

Inflammation can be classified into
* Acute inflammation occurs quickly or within the hours and lasts a few days or weeks and the damaged tissues are healed themselves after all pathogens are killed off.

However, if the immune system can not completely eradicate all invaders in the acute phase of infection, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

* Chronic inflammation is a low-grade inflammation caused by the inability of the immune system to eliminate the invasive pathogens that occurred in the acute phase of infection.

Any inflammation that lasts more than 8 weeks is considered chronic inflammation.

Believe it or not, worldwide, 3 of 5 death are results from chronic inflammatory diseases like stroke, chronic respiratory diseases, heart disorders, cancer, obesity, and diabetes.

In the US, chronic inflammatory diseases affect more than 50 million Americans each year.

Diseases associated with low-grade inflammation include diabetes and cardiovascular.

Gamma-linolenic acid is a phytochemical in the group of Omega-3, 6,9 fatty acids, belongings to the class of Lipids, found abundantly in evening primrose, borage, blackcurrant, etc.

On finding a potential plant that processes anti-colonic diseases activity researchers examined the relationship between the high-fat, high-sugar diet (HFHSD) and alpha-linolenic acid (ALA) on mouse colitis induced by the administration of trinitrobenzene sulfonic acid (TNBS).

The study included female BABL/c mice which were fed with HFHSD for the 9 weeks. At the same time, ALA treatment (150 or 300 mg/kg) was administered on a daily basis. At the end of the 9 weeks, experimental colitis was induced by the intra-colonic administration of trinitrobenzene sulfonic acid (TNBS).

According to the results of the analysis,
* Before treatment, TNBS induced severe clinical manifestations of colitis and histological damage.
* Low-ALA (150 mg/kg) administration profoundly ameliorated TNBS-induced clinical manifestations, bodyweight loss, spleen weight loss, and histological damage.

* Interestingly, high-ALA (300 mg/kg) administration did not ameliorate colitis and even exacerbated the symptoms.

* HFHSD consumption assisted TNBS in changing IL-12, IFN-γ, IL-2, and IL-17A, the pro-inflammatory cytokines in the liver and these changes were recovered through low-ALA.

* Moreover, HFHSD had a significant impact on the total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and triglyceride (TG), which related to the increased risk of hyperlipidemia

Collectively, researchers said, "HFHSD exacerbated the TNBS-induced colitis via the Th1/Th17 (inflammatory) pathway. The Low-ALA (150 mg/kg) exhibited protective effects against the TNBS-induced colitis via the Th1/Th2/Th17 (anti-inflammatory) pathway".

Taken altogether, gamma-linolenic acid may be considered a remedy for the prevention and treatment of colonic inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Dandelion(Taraxacum officinale), the Potent Antioxidant Remedy

By Kyle J. Norton

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid and cell damage.

There are 3 main types of antioxidants found in nature, including
* Antioxidant enzymes produced by the host, including superoxide dismutase (SOD), glutathione peroxidase, glutathione reductase, and catalases.

* Antioxidants vitamins, that may be injected from the dietary sources such as fruits and vegetables, including vitamins A, C, D, and E. Most antioxidant vitamins are considered free radical scavengers that gobble up free radicals before they can induce a cascade of damage.

* Antioxidant Phytochemicals found in the plant including second metabolites.

Antioxidants keep our bodies functioning properly by reducing the risk of diseases associated with oxidative stress.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily to donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.

Antioxidants, on the other hand, are stable molecules with a unique function in the inhibition of oxidation.

Dandelion is a herbaceous perennial plant, genus Taraxacum, belongings to the family Asteraceae, native to temperate regions of the world. It has been used in traditional and herbal medicine to treat gout, eczema, acne, gall bladder, kidney, liver and urinary disorders, hypoglycemia, dyspepsia with constipation, edema, blood pressure, and heart weakness, chronic joint, skin diseases, etc.

On finding a natural ingredient for the prevention and treatment of diseases involved in oxidative stress, scientists examined the n-hexane soluble compounds from aerial parts of dandelion (Taraxacum officinale Weber ex F.H. Wigg) antioxidant activity.

Where the n-hexane soluble and insoluble fraction showed a strong antioxidant property such as decreasing the mortality rate caused by degenerative and infectious diseases with an IC50 value of 78 µg/ml and 34 µg/ml, according to the study by the Universitas Gadjah Mada, Yogyakarta.

In chemical analysis, n-hexane (unpolar) fraction showed the presence of 30 biologically active compounds. Phytol [14.7% of total ion current (TIC)], lupeol (14.5% of TIC), taraxasteryl acetate (11.4% of TIC), β-sitosterol (10.3% of TIC), α-amyrin (9.0% of TIC), β-amyrin (8.3% of TIC), and cycloartenol acetate (5.8% of TIC).

In other words, all chemical compounds found in n- (unpolar) fraction work synergistically to protect the cells against alternation induced by overexpression of free radicals.

Moreover, the unipolar fraction exhibited an antioxidant activity comparable to those of 46.7 mmol Trolox equivalents/g extract (determined by 1,1-diphenyl-2-picrylhydrazyl method).

According to the tested differentiation, in vitro, compared to control values obtained from cells infected without treatment, treatment of dandelion showed a low level of HIV-1 replication.

Furthermore, the treatment of dandelion also exerted a strong effect on the viral transcriptase activity in a dose-dependent manner.

In other words, the dandelion extract has potent inhibitory activity against HIV-1 replication and reverse transcriptase activity to induce infection.

Based on the results, researchers suggest, "This fraction demonstrated insignificant antimicrobial activity and can be used in cosmetic and pharmaceutical industries".

Taken altogether, Dandelion may be considered an antioxidant remedy for the protection oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Cruciferous Vegetables Show an Inverse Association with the Risk of Coronary Heart Disease

By Kyle J. Norton

Heart diseases are a medical condition associated with any disruption of blood vessels and heart function.

There are 5 types of heart diseases, including
* Heart disease affected by the arteries to the heart, such as coronary artery heart disease.

* heart disease affected by heart valves functions in regulating blood flow such as valvular heart disease.

* heart disease affected electrical conduction such as arrhythmias.

* Heart disease affected by heart muscles such as cardiomyopathy. And

* Congenital heart disease caused by heart defect at birth

According to the statistics, heart disease is the leading cause of death in the US, approximately 610,000 people die of heart disease every year.

Coronary heart disease (CHD) kills over 370,000 people annually.

Furthermore, the most common heart disease in North America is CHD, the type of heart disease caused by plaque buildup on artery walls, leading to narrow, thickens and stiffens arteries. Some researchers suggested that the causes of plaque build-up on the arterial wall were the results of several factors including an unhealthy diet, lack of exercise, being overweight and smoking.

What causes plaques building up on the arterial is debatable. However, researchers suggested that the Western diet is associated with a significantly increased risk of coronary artery disease.

Dr. Venegas-Pino DE in the classification of the effect of diet on heart diseases said," the western-diet-fed male ApoE-/-: Ins2+/Akita mouse, with profound cardiovascular disease involving extensive atherosclerosis, coronary artery disease and myocardial infarct resulting in shortened lifespan(4)".

The most common symptoms of coronary artery disease are angina, the condition characterized by chest pain or discomfort and shortness of breath as a result of the weakening of heart muscle which can not pump enough blood to the body's needs.

Over time, the prolonged weakened heart muscle can lead to complications of heart failure, irregular heartbeat, or arrhythmia.

Cruciferous vegetables are a group of vegetables, belongings to the family Brassicaceae (also called Cruciferae) with many genera, species, and cultivated all over the world in suitable climate for commercial profits, including cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts, and green leafy vegetables.

On finding a potential remedy for the prevention of plaque accumulated on the arterial wall, researchers investigated the association of intake of glucosinolates with the risk of coronary heart disease (CHD).

A total of 74,241 women in the Nurses' Health Study (NHS; 1984-2012), 94,163 women in the NHSII (1991-2013), and 42,170 men in the Health Professionals Follow-Up Study (1986-2012), who were free of cardiovascular disease and cancer at baseline were given Glucosinolate intake that was assessed using validated semi-quantitative food frequency questionnaires at baseline and updated every 2-4 years during follow-up.

According to the food-questionnaire returned
* During 4,824,001 person-years of follow-up, 8,010 cases of CHD were identified in the three cohorts.

* After adjustment for major lifestyle and dietary risk factors of CHD, weak but significant positive associations were observed for glucosinolates with CHD risk when comparing the top with bottom quintiles (hazard ratio [HR]:1.09; 95% CI: 1.01, 1.17; Ptrend<0.001).

* Higher intakes of three major subtypes of glucosinolates were consistently associated with a higher CHD risk, although the association for indolyl glucosinolates did not achieve statistical significance. Regarding cruciferous vegetable intake.

Moreover, participants who consumed one or more servings per week of Brussels sprouts (HR: 1.16; 95% CI: 1.06, 1.26; P<0.001) and cabbage (HR: 1.09; 95% CI: 1.02, 1.17; P=0.009) had a significantly higher CHD risk than those who consumed these cruciferous vegetables less than once per month.

Based on the results, researchers said, " dietary glucosinolate intake was associated with a slightly higher risk of CHD in US adults".

Taken altogether, cruciferous vegetables may be considered functional remedies for the prevention of coronary heart disease, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Ashwagandha Inhibits the Parameters Associated with Epilepsy in Vivo

By Kyle J. Norton

Epilepsy is a neurological disorder associated with sudden recurrent episodes of sensory disturbance, loss of consciousness, or convulsions, caused by abnormal electrical activity in the brain.

Approximately, 1.2 percent of U.S. people have active epilepsy.  More than 65 million people have epilepsy globally.

Additionally, about 1 in 26 people will develop epilepsy at some point during their lifetime.

The most common causes of epilepsy are low oxygen during birth, head injuries that occur during birth or from accidents during youth or adulthood, brain tumors, genetic mutation, infections such as meningitis or encephalitis.

The most common symptoms of epilepsy are temporary confusion, uncontrollable jerking movements of the arms and legs, loss of consciousness or awareness, and psychic symptoms such as fear, anxiety or deja vu.

Experimentally, melatonin, a hormone that regulates the sleep-wake cycle. has been found to attenuate seizures, and it is most effective in the treatment of juvenile intractable epilepsy.

Melatonin also processed additional benefits include improved physical, emotional, cognitive, and social functions.

On the other hand, melatonin has been shown to induce electroencephalographic abnormalities in patients with temporal lobe epilepsy and increase seizure activity in neurologically disabled children.

The hormone showed very low toxicity in clinical practice, according to the study by the Department of Pathophysiology, Medical University, PL 20-090 Lublin, Jaczewskiego 8, Poland(22).

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

In the urgency to discover a natural ingredient for the management of the neurological disorder, researchers investigated the effect of Withania somnifera (WS) root extract and withanolide A (WA) in the management of temporal lobe epilepsy.

According to the tested analysis, in the epileptic test group of rats, levels of antioxidants are found to substantial decrease compared to the increase of the levels of peroxide.

Oxidative stress caused by an imbalance of the ratio of antioxidants and free radicals in the tested rats, induced significant decrease levels of proteins associated with systemic inflammation, compared to the increase of pro-apoptotic proteins.

Moreover, the epileptic condition of the tested rats also showed to increase the muscarinic receptor binding, which leads to reduced function of transduction of cholinergic signals and increases mRNA expression involved of the inhibition of the cell growth in the cerebral cortex.

However, the administration of the WS reversed the altered muscarinic receptor expression, oxidative stress and resultant derailment in cell signaling in experimental rats.

Based on the findings, researchers said, "Withania somnifera and withanolide A play important roles in central muscarinic receptor functional balance and activation of the antioxidant system in the cerebral cortex in temporal lobe epilepsy".

Taken altogether, Ashwagandha may be considered supplements for the management of epilepsy, pending to the confirmation of the larger sample size and multicenter human study.

Omega 3 Fatty Acid Promotes Myocardial Ischemia-Reperfusion in Vivo

By Kyle J. Norton

Compared to acute brain infarction that is the medical condition characterized by the obstruction of the blood supply to the brain, causing local death of the brain tissue. The occurrence and extent of brain infarction are associated with three basic factors, including the site of arterial occlusion, the rapidity of arterial occlusion, and the presence or absence of collateral circulation.

Myocardial ischemia injury is a medical condition caused by severe impairment of coronary blood supply induced damage or dysfunction of the cardiac tissue.

Myocardial reperfusion is the process to restore the coronary blood flow after a period of coronary blockage that leads to severely reduced or no oxygen .blood flow to the heart.

The condition severity is totally dependent on the intensity and duration of the coronary occlusion.

According to the statistics from the WHO, in 2008, 12% of all death or approximately 7,254,000 deaths worldwide have directly or indirectly caused by myocardial ischemia injury.

Atherosclerosis, one of the major causes of cardiovascular disease is a medical condition caused by the accumulation of plaque on the arterial wall. Over time, the condition can lead to narrowing the arteries or blocked arteries, leading to reduce oxygenated blood flow to the heart and other parts of the body.

Arteries are blood vessels carried the oxygen-rich blood to the heart before passing them to the tissues and organs.

Plague is made up of fat, cholesterol, calcium, and other substances found in the blood. In other words, cholesterol that cannot be returned to the liver for reuse may have a strong impact to induce plaque accumulation.

Coronary heart disease causes 1/4 total death in the worldwide population and 50% of the death associated with other forms of heart disease.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of post-stroke, researchers examined the cardioprotective effects of ω-3 polyunsaturated fatty acids (PUFAs) postconditioning against ischemia-reperfusion (I/R) injury.

The study included 60 Sprague-Dawley rats that were randomly divided into 4 groups (n = 15 for each) and used to generate the Langendorff isolated perfused rat heart model.

* The sham group received continuous perfusion of 150 min.

* The remaining three I/R-treated groups sequentially received 30-min perfusion, a 30-min cardioplegia, and 90-min reperfusion.

* The I/R-ischemic preconditioning (IP) group additionally received three cycles of 20-s reperfusion and 20-s coronary re-occlusion preceded the 90 min of reperfusion.
* The I/R-ω group were perfused with ω-3 PUFAs for 15 min before the 90 min of reperfusion.

According to the results of the study,
* Compared with the I/R group, the I/R-ω group had significantly reduced myocardial infarct size, reduced levels of lactate dehydrogenase and malondialdehyde associated with cardio-stress markers.

* Furthermore, Omega 3 fatty acids elevated superoxide dismutase level involved in antioxidant production and elevated rising (+dp/dtmax) and descending (-dp/dtmax) rate of left ventricular pressure against myocardial ischemia-injury.

* Moreover, The I/R-ω group had a significantly lower rate of mitochondrial damage in myocardial tissue compared with the I/R and I/R-IP groups.

In other words, Omega-3 fatty acids protect the heart tissues and muscles against damage after stroke.

Moreover, researchers said, " ω-3 PUFA postconditioning possesses good cardioprotective effects and may be developed into a therapeutic strategy for myocardial I/R injury".

Taken altogether, Omega-3 fatty acids may be considered a remedy for the treatment of myocardial ischemia injury, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Fermented Soy Processes Anti Inflammatory Cytokines Release

By Kyle J. Norton

Inflammation is a natural immune response involving the stimulated production of pro-inflammatory activity with a target to protect our body against foreign invasions such as virus and bacteria after injury and damage.

In the acute phase of infection, the immune white blood cells in the first line of defense stimulating the blood vessel constriction and palette accumulated to the injury area to cover the wound and the proteins associated with the production of the pro-inflammatory cytokines to kill off the invasive microorganisms.

In people with a strong immune system, during the acute phase of infection, the levels of pro-inflammatory cytokines are balanced by the expression of anti-inflammatory cytokines without affecting the immune activity.

However, in children, the elderly and people with the weakened immune system, overexpression of proinflammatory cytokines can damage the nearby cells and tissue, leading to the formation of scars.

Chronic inflammation is a medical disease caused by the immune inability to destroy the invaders in the acute phase of infection, leading to the immune system in the adaption of the change.

Over time, chronic inflammation can have a strongly negative impact on the affected tissues and organs.

However, in most cases, over-response of the immune system can lead to over expressions of certain pro-inflammatory cytokines, depending on the injured location such as Interleukin (IL)-1 and tumor necrosis factor (TNF) which can lead shock and death of healthy cells in the infectious location.

In most cases, long-term overexpression of pro-inflammatory cytokines can lead to chronic inflammatory disease as the immune system no longer can kill the invasive agents and heal the damage caused by acute infection.

Fermented soy products are diverse varieties of soy food that are made from fermented soybeans, including miso, natto, tempeh, and soy sauce.

On finding a potential plant that processes anti-inflammatory activity researchers examined the effect of health benefits of QCAN® Plus-an easily available fermented soy drink.

The study included lean (n = 10) and obese (n = 10) subjects, during pre-soy (weeks -2, -1, and 0), on-soy (weeks 1, 2, 3, and 4), and post-soy (weeks 6, 8, 10, and 12) periods.

According to the results of the analysis,
* The lipid peroxidation associated with total cholesterol and low-density lipoprotein (LDL) cholesterol levels were significantly reduced in both lean and obese individuals during on-fermented soy treatment.

* Cytokines associated with inflammation such as platelet-derived growth factor (PDGF) AA and AB/BB were significantly lowered on-soy compared with pre-soy (P ≤ .05) in lean subjects.

* PDGF AA, IL-1RA, and GMCSF associated with insulin resistance, lipoperoxidation and inflammation were significantly reduced on-soy (P ≤ .05) in obese subjects, respectively.

* Q-CAN Plus was safe.
Collectively, researchers said, "Q-CAN Plus ensures its safety for consumption and highlights its hypolipidemic and suppressive effect on certain cytokines".

Taken altogether,  QCAN®  may be considered a functional food for the prevention of the initiation of cytokines, pending to the confirmation of the larger sample size and multicenter human study.

Dandelion(Taraxacum officinale), the Potential Antimicrobial Remedy

By Kyle J. Norton

Microbial organisms refer to microscopic organisms that exist as unicellular, multicellular, or cell clusters.

Microorganisms are widespread in nature and are beneficial to life, and most of them are unharmful to humans.

Antimicrobial activity is an action that kills microorganisms or stops their growth. In humans, these actions are carried out daily by the immune system.

Infection is a processive condition caused by the battle of the invasion of microorganisms and the host immune white blood cells. Microorganism during the battle might produce toxins which can lead to the initiation of infection-related transmissible disease or communicable disease.

In acute infections, the systematic immune system responds properly to the invasive pathogen, leading to a brief battle after which the immune system is always victorious.

However, chronic infections occur when the immune cannot kill off the invasive stimuli in the acute phase of infection, leading to chronic infectious diseases.

The most common diseases associated with microbial infection include
* Diseases caused by bacteria are totally dependent on the types of bacterias, such as pneumonia, caused by Streptococcus and Pseudomonas, and foodborne illnesses caused by bacteria such as Shigella, Campylobacter, and Salmonella.

* Diseases caused by the virus include smallpox. the common cold and different types of flu. measles, mumps, rubella, chicken pox, and shingles. hepatitis. herpes and cold sores. polio. rabies, Ebola and Hanta fever.

* Diseases caused by parasites are malaria, trichomoniasis, giardiasis, cryptosporidiosis, and toxoplasmosis.

* Diseases caused by fungi aspergillosis usually occurs in patients with lung diseases or weakened immune systems and Candidiasis is caused by the yeast Candida.

Depending on the types of microbial infection, most common symptoms are fever, diarrhea, fatigue, muscle aches, and coughing

On finding a natural ingredient for the prevention and treatment of infectious diseases, scientists examined the Taraxacum officinale, for the synthesis of silver nanoparticles (TOL-AgNPs) anti-microbial activity, ranging between 5 and 30 nm, with an average particle size of about 15 nm.

According to the tested differentiation, the TOL-AgNPs (at 20 μg/mL concentration) showed significant antibacterial activity with significant growth inhibition of phytopathogens X. axonopodis (22.0 ± 0.84 mm) and P. syringae (19.5 ± 0.66 mm).

Furthermore, the synthesized AgNPs had higher antibacterial activity in comparison with commercial AgNPs.

Moreover, nanoformulations with tetracycline showed better broad-spectrum efficiency to control phytopathogens.

Most importantly, AgNPs also possessed significant antioxidant potential and The TOL-AgNPs showed a high cytotoxic effect against human liver cancer cells (HepG2).

Based on the results, researchers suggest, "dandelion-mediated AgNPs synthesis can represent a novel approach to develop effective antimicrobial...drugs with a cheap and eco-friendly nature.".

Taken altogether, Dandelion may be considered a remedy for the protection against microbial infection, pending to the confirmation of the larger sample size and multicenter human study.
