New: The Natural CD4+ T cells Inhibitor in Immunoregulatory Activity

Arnica may have a therapeutic effect in enhanced immune function, some scientists suggested.

The nuclear factor NF-κB pathway is considered as a prototypical proinflammatory signaling pathway and IL-12 is an immune response signaling pathway in response to antigenic stimulation.

Furthermore, in contrast to general belief, application of Arnica tinctures exerted a effect in suppressed contact hypersensitivity to sensitizer trinitrochlorobenzene and activation of dendritic cells.

These result indicated that herbal Anise inhibits induction of contact hypersensitivity through its anti-inflammatorily immunosuppressive effect via immune regulation.

In impairing protein and DNA synthesis, injection of helenalin also displayed a deregulated and unwanted T cell-mediated immuneresponses, through induced apoptosis in activated CD4+ T cells,  by triggering the mitochondrial pathway of apoptosis.

CD4+ T cells expression is a displaying CD4 molecules on their surface induced by glycoprotein found on the surface of immune cells in sending signals to T helper cells to initiate immune pro inflammatory response to the infectious site.

More precisely, helanalin deactivated production of CD4+ T cells through inhibition of proliferation by G2/M cell cycle arrest accompanied by the accumulation of cell cycle regulator proteins.

Further examination also found that helenalin injection enhanced function of p53 in inhibited CD4+ T cells cycle division through its ability as a tumor suppression as well as increased expression of reactive ogygen species in facilitated cytotoxicity.

Moreover, helenalin also de activated CD4+ T cells expression by suppressed nuclear translocation of NFATc2 with function in response to the activation of CD4+ T cells.

Dr. Berges C1, the lead author, at the end of the experiment said, "helenalin can be defined as a new immunosuppressive compound suited for the treatment of deregulated and unwanted T cell-mediated immune responses".

The evidences of the collection suggested that Arnica and its bioactive compound helenalin may be considered as functional herb in improved immune function. However, intake of Arnica extract and supplement should be taken with extreme care as toxicity was found in case of overdose.

Better Than Chemo Cisplatin in Inhibited Onset and Gene Expression in Induction of Colon Cancer, According Online Medical Literature

In the study to re confirm the use of technique in detected the early stage of colon cancer through examine the expression of aberrant crypt foci (ACF) and colon rectal mucosal epithelial cell proliferation researchers at the the University of São Paulo conducted a experiment of 50 male Wistar rats randomly divided into 5 groups, as indicated below

* Groups G1 to G4 were given 4 injections of the carcinogen 1,2-dimethylhydrazine (DMH)

* The G2 group received Lychnophora ericoides (LE) extracts for 6 wk.

Observation at the 42th day after the first injections of DMH for the neoplasic induction, found that Arnica treatment group (LE), including G2, 3 and 4 demonstrated a statistically significant decrease in the number of aberrant crypt foci (ACF) and an reduction of cell proliferation induced by DMH.

These indications suggested that Arnica consisted a protective influence on the process of colon carcinogenesis, through suppressing both the initiation and the promotion of colonic carcinogenesis.

Furthermore, in the reexamine the cytotoxic activity of phytochemical compound isolated from Arnica, against COLO 320, a human colorectal cancer cell line using the microculture tetrazolium (MTT) assay, researcher found that isolated flavonoids showed moderate to low cytotoxicity, in compared to the reference compound cisplatin at 2.9 microM.

Cisplatin is a conventional chemo medication used to treat a number of cancers.

In 2 hours of injection, sesquiterpene lactones, helenalin at dose of 1.0 microM, possessing both the reactive alpha-methylene-gamma-lactone moiety and a reactive alpha,beta-unsubstituted cyclopentenone ring, displayed the strongest cytotoxicity against the tested colon cell line in compared to other compounds.

In compared to tested GLC4, a human small cell lung carcinoma cell line, COLO 320 was more sensitive to many flavonoids (especially for flavones), but more resistant to the cytotoxic effect of the sesquiterpene lactones. 

Interestingly, in the evaluation of Arnica effect in deregulation of c-myb expression involved in the development of human tumors, especially certain types of leukemia and breast and colon cancer, researchers indicated that application of sesquiterpene lactones isolated from Arnica suppress Myb-dependent gene expression with potential anti-colon cancer activity with the IC50 values were in a range between 0.7 and >30 μM.

C-myb is a members of terminal domain with function in regulated the expression of hematopoiesis,  cell proliferation, and differentiation in tumorigenesis.

In compared to other isolated bioactive compounds, furanoheliangolide goyazensolide and the pseudoguaianolide helenalin acetate with IC50 = 0.6 and 0.7 μM, respectively, represented the most active inhibitors of c-Myb dependent gene expression.

Observed by the control measurements for cell viability (MTS assay), researchers also found that c-Myb dependent gene expression is not a function of cytotoxicity/unspecific cell damage.

All these results indicated that Arnica processed a significant effect in prevented early onset, progression and treatment of colon cancer through it bioactive compounds in inhibited onset of aberrant crypt foci (ACF) and colon rectal mucosal epithelial cell and colon cancer cells proliferation and gene expression in  risk of colon cancer.

Cinnamon is a spice derived from the inner bark of tree, native to South East Asia, of over 300 species of the genus Cinnamomum, belongings to the family Lauraceae, use in herbal and traditional medicine as anti-fungal and bacteria level, to improve reproductive organ, prevent flatulence and intestinal cramping, and to treat indigestion, diarrhea, bad breath, headache, migraine, etc.


Topical use of essential oil extracted from cinnamon may be the next generation whole herb medicine for prevention and treatment of menstrual pain and heavy menstrual bleeding.

The results of the investigation were reported by several respectable institutes and published on the data bases of numbers online medical literature, including PubMed.

According to the Eulji University, in the study of compared the effect of aromatherapy massage and acetaminophen on menstrual pain in Korean high school girls. cinnamon treatment group showed a significantly higher reduction of menstrual pain than in the acetaminophen group.

Acetaminophen or APAP is a over-counter and popular conventional medicine used to treat pain and fever with numbers of intolerant adverse effects.

In a randomized, double-blind trial, out of 114, group received Cinnamon (capsule containing 420 mg Cinnamon, TDS, n= 38) in 24 h significantly reduced the severity and duration of pain during menstruation and menstrual bleeding.


In primary dysmenorrhea, cinnamon expressed significantly over reduction of pain, menstrual bleeding, nausea and vomiting with primary dysmenorrhea without side effects.


Dr. Marzouk TM., the lead author at the Mansoura University said, " ... aromatherapy abdominal massage once daily for seven days prior to menstruation using the essential oils (cinnamon....) is effective in alleviating menstrual pain, its duration and excessive menstrual bleeding...." and "Aromatherapy can be provided as a nonpharmacological pain relief measure and as a part of nursing care given to girls suffering of dysmenorrhea, or excessive menstrual bleeding".

Further study of the effect of cinnamon extract on symptoms of dysmenorrhea and contractility in some smooth muscles, Dr. Alotaibi M. suggested, cinnamon may be used to alleviate menstrual pain associated with dysmenorrhoea or prevent unwanted uterine activity in early pregnancy.


However, intake of cinnamon should be taken with care as toxicity was found in case of overdoses.

* The most toxic compound was the flavone jaceosidin on GLC4

* Helenalin, possessing both the reactive alpha-methylene-gamma-lactone moiety and a reactive alpha,beta-unsubstituted cyclopentenone ring, displayed the strongest cytotoxicity on GLC4.

Flavonoids processed significantly antioxidant and anti-inflammatory activities, according to online literature were found abundantly in tea, citrus fruit, citrus fruit juices, berries, red wine,....

Moreover, in compared to all anethole modificatiions in vivo, S-valproate and S-diclofenac, exerted a strong anti growth of A549 cells. but not S-sulindac.

S-valproate and S-diclofenac significantly inhibited A549 or NCI-H1299 xenograft proliferation in nude mice, but had no effect on the nude mouse body weight.

Dr. Moody TW, the lead author, after taking into comparable tests of all ingredients through Western blot, said, "S-valproate and S-diclofenac inhibit the growth of NSCLC cells and reduce PGE(2) levels, they may prove beneficial in the chemoprevention and/or therapy of NSCLC".

3. Hyperlipoproteinemia
In a study of Fifteen patients with hyperlipoproteinemia (HLP), types IIA (n = 8), IIB (n = 3) and IV (n = 4) were given 40 g of heat prepared alfalfaseeds 3 times daily at mealtimes for 8 weeks with otherwise unchanged diet, conducted by Mölgaard J, von Schenck H, Olsson AG.(3), found that patients with type II HLP alfalfa treatment caused after 8 weeks a maximal lowering of pretreatment median values of total plasma cholesterol from 9.58 to 8.00 mmol/l (P less than 0.001) and low density lipoprotein (LDL) cholesterol from 7.69 to 6.33 mmol/l (P less than 0.01), which corresponds to decreases of 17% and 18%, respectively. Maximal decrease was 26% in total cholesterol and 30% in LDL cholesterol. In two patients with hypercholesterolemia the LDL cholesterol decreased less than 5%. Apolipoprotein B decreased in the same period from 2.17 to 1.43 g/l (P less than 0.05) in type II HLP, corresponding to 34% decrease, whereas apolipoprotein A-I did not change. Body weight increased slightly during the first 4 weeks of alfalfa treatment (P less than 0.001) probably because of the caloric content in the alfalfa seeds. After cessation of treatment, all lipoprotein concentrations returned to pretreatment levels. We conclude that alfalfa seeds can be added to the diet to help normalize serum cholesterol concentrations in patients with type II HLP.

In a study of five groups of 12-week-old female mice were per oral treated with vehicle (control), lyophilized AS (550 mg wt/kg BW), ASEA (ASEA, 25 mg/kg BW), coumestrol (CUM, 0.075 mg/kg BW) and tamoxifen (TAM, 0.375 mg/kg BW) as the positive control, conducted by Institute of Microbiology and Biochemistry, College of Life Science, National Taiwan University, Taipei, Taiwan (4), researchers found that alhalfa decreased the disease severity, increased survival and life span of the autoimmune-prone MRL-lpr/lpr mice, suggesting a potential of ASEA in the treatment of autoimmune diseases.

5. Anti-inflammatory activity
