Healthy Veggie Broccoli Inhibits the Onset of Oxidative Stress

By Kyle J. Norton

Oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily to donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid and cell damage.

Antioxidants can either produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

There is no single cause of oxidative stress. Some researchers suggested that a number of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Sadly, a prolonged period of oxidative stress presented in the human body can cause oxidative damage to biomolecules, (lipids, proteins, DNA) and the death of healthy cells through cytotoxicity.

Certain chronic diseases are found to be associated with oxidative stress, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The vegetable has large flower heads, usually green in color and the mass of flower heads is surrounded by leaves and evolved from a wild cabbage plant on the continent of Europe.

On finding a potential plant that processes anti-oxidative activity researchers examined the efficacy of sulforaphane found in broccoli in protecting alcohol-induced hepatic injury in vivo.

The study included selected male C57BL/6 mice which were orally administrated with broccoli sprout extract (BSE) containing sulforaphane [7.6, 25.2, and 50.4 mg/kg of body weight (bw)] once a day for 14 days.

On the 13th day, mice were challenged with alcohol (5 g/kg of bw) every 12 h for 3 times, which increased malondialdehyde (MDA) levels (4.44 ± 1.24 nmol/mg of protein, p < 0.01) of oxidative stress in the liver.

According to the results of the experiment, the administration of low-, medium-, and high-dose BSE markedly reversed the decrease of antioxidant capacity.

More precisely, BSE increased the levels of antioxidant enzymes such as glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and glutathione S-transferase (GST)  in the liver, compared to control.

The efficacy in protecting the liver oxidative stress was attributed to the activation of phase 2 enzymes via nuclear erythroid-2-related factor 2 (Nrf2).

Most importantly, BSE also significantly attenuated the levels of the endoplasmic reticulum (ER)-stress-specific proteins, a defense system which protects cells from ER stress such as glucose-regulated protein 78 (GRP78), activating transcription factor 6, and protein kinase RNA (PKR)-like ER kinase (PERK),

Collectively, researchers said, "These results indicate that BSE protects the liver against alcohol challenge via upregulating antioxidant capacity and downregulating ER stress".

Taken altogether, broccoli may be considered a functional food for the prevention and treatment of diseases associated with oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Moronic Acid, A Promising Antidiabetic Antioxidant

By Kyle J. Norton

Diabetes is a chronic low-grade inflammatory condition caused by insufficient insulin entering the bloodstream to regulate glucose.

In other words, diabetes occurs when cells in the pancreas dying off or receptor sites clogged up by fat and cholesterol. In some cases, diabetes is also caused by allergic reactions of cells in the immune system.

Inflammation is a natural defense of the immune system that protects the body against the insult from the foreign invasion.

Acute inflammation occurs quickly or within the hours and lasts a few days or weeks and the damaged tissues are healed themselves after all pathogens are killed off.

However, if the immune system can not completely eradicate all invaders in the acute phase of infection, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

Chronic inflammation is a low-grade inflammation caused by the inability of the immune system to eliminate the invasive pathogens that occurred in the acute phase of infection.

Any inflammation that lasts more than 8 weeks is considered chronic inflammation.

Believe it or not, worldwide, 3 of 5 death are results from chronic inflammatory diseases like stroke, chronic respiratory diseases, heart disorders, cancer, obesity, and diabetes.

In the US, chronic inflammatory diseases affect more than 50 million Americans each year.

Sub-chronic diabetic treatment is a process associated with the treatment of diabetes that lasts only in a moderate or intermediate duration.

Moronic acid is a phytochemical in the subclass of Triterpenoid, belonging to the group of Terpenes found in extracted from Rhus javanica, Mistletoe, etc.

On finding a potential phytochemical for the treatment of diabetes, researchers examined the ix derivatives (1-6) of moronic acid that were semi-synthesized and their in vitro protein tyrosine phosphatase 1B (PTP-1B) inhibition activity in diabetes.
* In rat induced diabetes, derivatives 2 (IC50=10.8 ± 0.5 μM) and 6 (IC50=7.5 ± 0.1 μM) displayed the most potent inhibitory activity.

* The derivatives decreased plasma glucose levels during all the experiments in vitro and vivo.

* Most importantly, The #2 had polar and Van der Waals interactions in both sites with Val49, Gln262, Met258, Phe182, Ala217, Ile219, and Gly259 and # 6 showed polar interaction with Gln262 and Van der Waals with Val49, Ile219, Gly259, Arg254, Ala27, Phe52, Met258, the proteins associated with the onset of diabetes.
Based on the findings, researchers said, "derivatives 2 and 6 have the potential for the development of antidiabetic agents".

Taken altogether, moronic acid may be considered a remedy for the treatment of diabetes in a short duration, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Pinto Bean (Phaseolus) Suppresses the Onset of Nasopharyngeal Cancer in Vitro

By Kyle J. Norton

The nasopharynx is the upper part of the pharynx, connecting with the nasal cavity above the soft palate that allows a person to breathe through the nose.

Cancer is a group of diseases caused by cell growth irregularly in the affected tissue caused by DNA alternation.

Nasopharyngeal cancer is a medical condition characterized by cells growth irregularly and disorderly in the tissues of the nasopharynx.

Most cases of nasopharyngeal cancer start in the cells on the surface of the inner lining of the nasopharynx. The early stage of nasopharyngeal cancer is asymptomatic due to the very small size of the tumor.

However, at the advanced stage, the oversize may suppress the nearby nerves and blood vessels, leading to severe throat pain and symptoms of a lump in your neck blood in the saliva, bloody discharged from your nose, particularly, persistent nasal congestion or ringing in your ears, accompanied with the general symptoms shared with other cancers.

According to the statistics provided by the American cancer society, nasopharyngeal cancer (NPC) is quite rare in the United States). The risk of cancer is less than one case for every 100,000 people.
Epidemiologically, some patients at this stage may also experience symptoms of frequent ear infections, hearing loss and headaches.

Pharynx cancer is originated in the tissue of the pharynx and classified
* The nasopharynx (the upper part of the throat behind the nose)
Cancer of the pharyngeal most commonly begins with malignant cells that line the nasopharynx, including nasal passages and auditory tubes join the remainder of the upper respiratory tract.

* The oropharynx (the middle part of the pharynx)
Cancer of the oropharynx also begins with the malignant cells that line the oropharynx, including the base of the tongue, the tonsils, the soft palate.

* The hypopharynx (the bottom part of the pharynx)
Cancer of the hypopharynx most commonly begins with the malignant squamous cell that lines the hypopharynx where the area of the larynx and esophagus meet. 90% of oropharyngeal neoplasms are squamous cell carcinoma.

On finding a natural ingredient for the prevention and treatment of diseases associated with oxidative stress, scientists examined the effects of purification of a glucosamine-binding antifungal lectin from Phaseolus vulgaris cv. Chinese pinto beans on cancers.

Pinto bean is also known as common bean, genus Phaseolus and the family Fabaceae. It has a mottled appearance of darker reddish-brown spots on a lighter brownish beige background, thus appearing painted or spotted.

According to tested analysis,
* Lectin at 30 μM showed strong antioxidant activity against the infectious agents including fungal species such as Phyllosticta citriasiana, Magnaporthe grisea, Bipolans maydis, Valsa mali, Mycosphaerella arachidicola in the induction of oxidative stress associated with proteins lipids and DNA damage.

* The lectin did not exert any discernible antiproliferative effects on breast cancer MCF-7 cells but was able to suppress proliferation of nasopharyngeal carcinoma HONE-1 cells, with an IC50 of 17.3 μM.

In other words, purification of a glucosamine-binding antifungal lectin from Phaseolus vulgaris cv. Chinese pinto beans exerted a significantly apoptotic activity against the viral induction of nasopharyngeal carcinoma HONE-1 cells.

Taken altogether, Pinto beans may process antioxidant and cytotoxic properties against nasopharyngeal cancer cells, pending to the confirmation of the larger sample size and multicenter human study.

Devil's claw (Harpagophytum procumbens) Processes Anti Inflammatory Bowel Disease (IBD) Potential

By Kyle J. Norton

Inflammatory Bowel Disease is a condition characterized by long term inflammation of the gut.

Most common types of IBD are
*Crohn’s disease (CD) is an inflammation of any part of the digestive tract. Most cases of CD affect the tail end of the small intestine.

* Ulcerative colitis is an inflammation of the large intestine.

The exact causes of inflammatory bowel disease are unknown. As of today, scientists do not know why, people with the same health conditions, are susceptible to the onset of the disease, while others do not.

Some researchers suggested that the disease may be caused by the immune response failing to kill off the invasion of infectious micro-organisms in the acute phase of digestive tissue injury and damage or immune dysfunction mistakenly attacking the digestive tract, a case of autoimmune disease.

Genetic mutation inherited from the parents such as NOD2, ATG16L1and IL23R involved in innate immunity, in the autophagy, and in the inflammatory response may elevate the risk of IBD

Furthermore, prolonged periods of imbalance of the ratio of good and bad bacteria in the colon, long-term intake of certain medicines, age, family history, ethnicity, parasite, unhealthy diet, and smoking are some prevalent risk factors that can lead to the onset of inflammatory bowel disease.

The most common symptoms of inflammatory bowel disease are severe diarrhea, abdominal pain, fatigue, and weight loss.

Believe it or not, if you are living in the Western world, you are also at a higher risk of the disease. For example, people who live in Western countries such as the United States and Europe have a higher IBD risk than those in other parts of the world.

Some researchers suggested an individual diet pattern may influence the risk of IBD.

According to the statistics provided by the CDC, inflammatory bowel disease Prevalence (IBD) affects approximately 1.3% of US adults (3 million)in 2015.

On finding a natural ingredient for the prevention and treatment of colonic diseases, scientists compared the antioxidant effects of Devil's claw and Aminosalicylates (5-ASA) in cell-free oxidant-generating systems and inflamed human colorectal biopsies.

Devil's claw also known as the wood spider, is a plant of genus Harpagophytum in the family of Pedaliaceae, native to South Africa.

The plant has been used as an herb in traditional and herbal medicine to treat fever, rheumatoid arthritis, skin, gallbladder, pancreas, stomach and kidneys conditions, etc.

According to tested analysis,
* Like 5-ASA, Devel's claw showed a strong effect in scavenged superoxide dose-dependently.

* Devil's claw demonstrated the decreased oxygen radical release, after incubation with the tested cell lines.

Based on the results, researchers suggest, "...devil's claw, ...merit(s) formal evaluation as novel therapies in inflammatory bowel disease.".

Taken altogether, Devil's claw may be considered a remedy for the treatment of inflammatory bowel disease, pending to the confirmation of the larger sample size and multicenter human study.

Tocopherols Inhibit the Onset of Seizure

By Kyle J. Norton

Epilepsy is a condition of a neurological disorder caused by abnormal electrical activity in the brain inducing sudden recurrent episodes of sensory disturbance and convulsions.

A seizure is a sudden, uncontrolled electrical disturbance in the brain, leading to changes in a person's behavior, movements or feelings, and levels of consciousness in the short term.

Patients with two or more seizures or a tendency of recurrent seizures are having epilepsy.

Seizures are not a disease in themselves. Instead, they are a symptom of many different diseases that can affect the brain. Some seizures may have unknown causes.

A neurological examination was done in a doctor’s office that can be helpful in the diagnosis of a seizure by examing the nervous functioning.

However, an EEG test or electroencephalogram that examines the electrical activity of the brain by looking into certain patterns on the EEG that orchestrates typical of epilepsy.

Seizures are classified into two groups
* Generalized seizures affect both sides of the brain, including
** Absence seizures that cause rapid blinking or a few seconds of staring into space.

 ** Tonic-clonic seizures.

** Lose consciousness.

* Focal seizures are located in just one area of the brain., including

** Simple focal seizures affect a small part of the brain.

** Complex focal seizures can make a person with epilepsy confused or dazed.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of epilepsy, researchers examined the vitamin E (as α-tocopherol, α-T) a neuroprotective and anti-inflammatory effect on epilepsy,

According to the analysis of rat seizures induced by single intraperitoneal injection of kainic acid and, starting from 3 h after status epilepticus, before treated with an intraperitoneal bolus of α-T (250 mg/kg b.w.; once a day) for 4 days.

Post-seizure α-T administration significantly reduced astrocytosis and microglial activation and decreased neuron degeneration and spine loss by lowering lipid peroxidation in the hippocampus.

These results suggested α-T administration exerted the anti-inflammatory and neuroprotective activity in kainic acid-induced epilepsy.

Furthermore, post-seizure treatment with α-T provides effective secondary prevention against post-seizure inflammation-induced brain damages and possibly against their epileptogenic effects.

Taken altogether, tocopherols may be considered an adjunct therapy in the treatment of seizure pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitostero, a Natural Free Radical Scavenging Antioxidant

By Kyle J. Norton

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.

The expression of oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of diseases associated with oxidative stress, researchers elucidated in vitro antioxidant and antimicrobial potential of Schimpera arabica extract and fractions (2-Hexadecen-1-ol and beta-sitosterol).

According to the tested assays,
* Among all the extracts, 2-Hexadecen-1-ol and beta-sitosterol were the major constituents of crude extract and exhibited significant antioxidant potential as well antimicrobial activity, compared to all other fractions that only exhibited a fair degree of antioxidant potential.

* In the free radical assay, 93% DPPH was scavenged by 100 μg ml-1 of the ethyl acetate fraction; i

* In ABTS assay chelation was 86·2 and 88% in ferric chloride reducing power assay.

Based on the findings, researchers said, "S. arabica contains a good amount of natural antioxidants of medicinal importance. However, further investigations are needed to characterize the further activity of 2-Hexadecen-1-ol and beta-sitosterol in vivo.".

Taken altogether, beta-Sitosterol found abundantly in S. arabica may be considered an antioxidant supplement, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Angelica Inhibits the Parameters Associated with Anemia in Vivo

By Kyle J. Norton

Anemia is a condition of deficiency of hemoglobin, leading to not enough red blood cells in the body. The function of hemoglobin is to carry oxygen from the lung to the body' s cells.

Depletion of the stored iron can cause reduced hemoglobin production, leading to iron deficiency anemia.

Iron deficiency is simply as too little iron in the body for the production of hemoglobin.

Anemia occurs when there isn't enough hemoglobin to transport oxygen to the body cells and organs need, leading to symptoms of irritability, fatigue, shortness of breath, low blood pressure, etc.

Approximately 2 out of 1000 people in Canada are considered an iron deficiency.

Anemia can be classified into acute and chronic anemia
* Anemia due to chronic disease is a type of blood disorder as a result affected by one or more chronic medical condition(s), causing reduced red blood cell production.
** Microcytic, hypochromic anemia
Microcytic, hypochromic anemia is chronic anemia caused by many conditions including iron deficiency, Beta-Thalassemia trait, lead poisoning, chronic illness or inflammation, and sideroblastic anemia (Korones & Cohen, 1997; Wu et al., 2002)

** Macrocytic anemia
Macrocytic anemia is a condition of not having enough healthy red blood cells in the body to transport oxygen and nutrients to the body tissue and organ as a result of the insufficiency of vitamin B12.

** Normocytic anemia
Normocytic anemia is a common disease, occurring to men and women over 85 years old as a result of low red blood cell volume as a result of a decreased production of normal-sized red blood cells, and increased destruction or loss of red blood cells, deficiency of vitamin B6, B12, etc.

** Hemolytic anemia
Hemolytic anemia is a condition of not enough red blood cells in the blood, as a result of the premature destruction due to abnormal breaking down of red blood cells (hemolysis) either in the blood vessels or elsewhere in the body.

* Acute anemia denotes is short term drops of red blood red cell production. Most of the time, it is caused by heavy internal bleeding or blood loss due to injury. It is a serious problem, in most cases, it requires medical attention immediately.

On finding a natural ingredient for the prevention and treatment of diseases associated with red blood cells, scientists examined the effects of ganggui buxue tang (DGBX, Angelicae Sinensis, Radix Astragali and Rhizoma Coptidis) on anemia.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the results of immune-induced AA mice used as an animal model, and the immunosuppressive agent, Ciclosporin A (CsA), was used as a positive control that was treated by the decoction,
* DGBX treatment significantly increased the production of whole blood cells in peripheral blood (PB); inhibit the expansion of Th1 and Th17 cells in the tested animals.

* Furthermore, DGBX increased the differentiation of Th2 and Tregs cells, regulated the expression levels of T-bet, GATA-3, RORγ in the immune cells, and pro-inflammatory cytokines, and decrease the expression levels of key molecules in the Jak/Stat signaling pathway involved in the process of immunity.

Collectively, DGBX can regulate the differentiation of T lymphocytes that orchestrates the immunosuppressive and hematogenic functions on AA mice against the onset of anemia.

Based on the results, researchers suggest, "DGBX might be a good candidate for inclusion in a randomized study for AA with more data on the possible side effects and doses used in humans".

Taken altogether, herbal angelica alone or combination with other herbal medicines may be considered anti-anemic remedy, pending to the confirmation of the larger sample size and multicenter human study.
