Herbal Angelica Sinensis, A Promising Anti-Glioblastoma Drugs

By Kyle J. Norton

Cancers are the class of chronic and medical conditions characterized by cell growth disorderly and uncontrollably that start on the inner lining of the tissue of an organ due to the alternation of cell DNA.

At the advanced stage, most cancer cells can travel a distance away from the original site to infect other healthy tissue and organs through blood and fluids circulation.

Glioblastoma is one of the most aggressive cancers that begins within the brain, caused mostly by the abnormal astrocytic cells and dead cells (necrosis). The disease is considered one of the most aggressive cancers induced by the alternation of cells originated from the brain, including the cerebral hemispheres.

According to statistics, most patients diagnosed with cancer die within 12 to 15 months. Less than 5% of patients are survived longer than five years.

If you are male you are at 1.5 higher risks of glioblastoma, compared to the female's counterpart.

The causes of glioblastoma are unknown, some researchers suggested that genetic preposition and environmental factors, previous radiotherapy, a weakened immune system may be some risk factors associated with the onset of the disease.

However, most cases of GBM are diagnosed at an older age with a median age of 64. Some researchers suggested that aging with the change of body function may be one of the major risk factors that cause the incidence.

Most patients with GBM experience symptoms of brain swelling caused by the overexpression of the fluid surrounding the tumor.

Conventionally, due to the nature of rapid growth, surgery is necessary to relieve the brain pressure and remove the tumor as much as possible, followed by radiation and chemotherapy.

Furthermore, the condition also shares the common symptoms of other types of cancer such as gastrointestinal discomforts accompanied by personality changes, and symptoms similar to those of a stroke.

On finding a natural ingredient for the prevention and treatment of neurological diseases, scientists examined the effects of Angelica Sinensis on glioblastoma multiforme (GBM).

Angelica or Dang Gui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to tested analysis, Angelica sinensis accelerates apoptosis by their key substances influencing factors of apoptosis pathways, including
* Biochemical stress associated with cell death such as oxidative stress in the induction of cytotoxicity.

* Induced DNA damage to the cancer cells by enhancing the tumor suppressor protein such as p53.

* Lack of growth factors involved in cell proliferative and differentiation.

Based on the results, researchers suggest, "our review puts emphasis on the potential of natural products in GBM treatment by regulating multiple malignant cancer-related pathways solving pending problems such as reducing toxicity and side effect".

Taken altogether, Dandelion may be considered a remedy for the treatment of glioblastoma multiforme (GBM). pending to the confirmation of the larger sample size and multicenter human study.

Herbal Devil's Club Kills Human Pancreatic Cancer Cells In Vitro

By Kyle J. Norton

Pancreatic cancer is a chronic condition characterized by cell growth irregularly in the tissues of the pancreas.

Most cases of pancreatic cancer start in the cell on the surface of the inner lining of the pancreatic tissue, before penetrating into the deeper layers to form a large and invasive tumor.

At the early stage, patients with pancreatic cancer may be asymptomatic. However, at the later stage, besides the common symptoms shared by all types of cancer such as unintended weight loss, loss of appetite, fatigue and gastrointestinal discomforts, patients with the condition may also experience localized symptoms such as internal bleeding and severe pain due to the oversize tumor which suppresses the nerve cells and blood vessels.

The pancreas cancer was found to be the site of metastasis from renal cell cancer, lung, colon and breast cancers.

According to statistics, every year, approximately, over 55,000 adults in the United States are diagnosed with pancreatic cancer, accounting for 3% of all cancers and 7% of all cancer deaths.

The survival rate of pancreatic cancer even diagnosed the early is very low compared to other types of cancer.

The 5-year survival rate including all stages of people with pancreatic cancer is 8%, compared to 32% of cancer detected at an early stage.

Scientists do not know the exact causes of pancreatic cancers, but they do know several risk factors that can make a person more likely to get the disease, including the genetic and acquired genetic mutation of p16 and TP53 and KRAS, BRAF, and DPC4 (SMAD4).

Other prevalent risk factors, including long-term use of tobacco, exposure to certain chemicals, overweight, and obesity, and unpreventable risk factors of family history, gender, the increase of age and ethnicity are associated with the early onset of pancreatic cancer.

Devil's Club  (DC) is a large shrub, genus Oplopanax, belonging to the family Aralioideae, native to the cool moist forests of western North America.

The herb has been used in traditional and herbal medicine as Adaptogen, Antipyretic, Pain Reliever, Antitussive, Antibacterial agent and to treat Arthritis, Diabetes, Colds, Cough, Sore throat, Chest congestion, Stomach pain, Constipation, Gall stones, Influenza, Cancer, etc.

On finding a potential compound for the treatment of cancer, researchers examined the effects of DC 70% ethanol extract alone, or in combination with cisplatin, gemcitabine, and paclitaxel on pancreatic endocrine HP62 and pancreatic ductal carcinoma PANC-1 and BxPC-3 cells.

According to the tested differentiation,
* DC extract inhibited proliferation of HP62 with IC50 (50% inhibition concentration) at 0.037±0.002% (v/v), PANC-1 at 0.0058 ± 0.0004% and BxPC-3 at 0.021 ± 0.003%.

* DC at 0.0033% combined with 1 nM of paclitaxel showed inhibition synergy on PANC-1 cells with a combination index of 0.44.

*  Increased cytochrome C, claspin, cIAP-2, and HTRA2/Omi apoptosis-related markers were upregulated in DC-treated HP62 and PANC-1.

Collectively, researchers suggested, "DC acts through targeting the intrinsic mitochondrial apoptosis pathway in the pancreatic cancer cells.

And, "The high antiproliferation potency of DC on PANC-1 is potentially useful as an adjunct therapy for treating pancreatic cancer, which is known for developing resistance to conventional chemotherapeutics".

Taken altogether, Devil's Club may be considered a remedy for the treatment of pancreatic cancer, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Elder (Sambucus), A Natural and Potential Anti-Glycation Remedy

By Kyle J. Norton

Advanced glycation end is a by-product formed by protein or fat combine with sugar caused by cooking meat at high temperatures.

The products have been found to affect metabolic health.

The most common causes of AGEs are from the diet, particularly in people who follow the Western diet with high saturated and trans fat, a lot of red meats and processed foods.

Low levels of AGEs may not be a problem, however, overexpression of AGEs can induce oxidative stress, leading to inflammation and damage proteins, lipid and sometimes alternation of cells DNA.

In other words, AGEs initiate the production and free radicals and free radical's chain reaction that can not be stopped until all electrons in the outer ring paired or inhibited by antioxidants.

Diabetes mellitus, chronic renal failure, neurodegenerative diseases, osteoarthritis and also non-diabetic atherosclerosis and chronic heart failure are some of the most common forms of chronic diseases associated with oxidative stress-induced by AGEs.

Some researchers suggested that modern life byproducts AGEs are the results of the promotion of the processed food diet.

Dr. Chetan Sharma, Amarjeet Kaur, the lead scientist in the assessment of AGEs, wrote, "As modern diets are largely heat-processed, they are more prone to contain high levels of advanced glycation end products (AGEs)".

Elder or Common Elder, is a shrub or small tree of the genus Sambucus of 30 species, belonging to the family Adoxaceae, native to the temperate-to-subtropical regions of both the Northern Hemisphere and the Southern Hemisphere.

The herb has been used in traditional and herbal medicine to treat constipation, colic, diarrhea, colds, flu, bronchial and upper-respiratory, irritation of the skin, rheumatism, etc.

On findings a natural compound for the treatment of cancer, researchers analyzed the anti-glycation activity of chemical compounds found in Sambucus formosana (Elder)

According to chemical analysis,
* Chlorogenic acid, rutin, isoquercetrin, nictoflorin, astragalin, and quercetin were quantified in linearity range of 10-100 (μg/mL) with a correlation coefficient of greater than 0.996.

* Furthermore, six phenolics with the relative standard deviation ranging from 1.7-3.1%.showed a high recovery (86.5-93.1%) and good reproducibility.

* Methanol extract showed the greatest values of total phenolic content (93.1 mg gallic acid equivalent/g).

* Additionally, the methanol extract revealed the best antioxidant capacity based on the DPPH scavenging activity and anti-glycation activity (IC50 was observed at 97.1 and 77.9 μg/mL, respectively).

Taken altogether, Elder processes a number of antioxidants may be considered an anti-glycation activity, pending to the confirmation of the larger sample size and multicenter human study.

Cruciferous Vegetables Processes Anti Glioblastoma Progression in Vivo

By Kyle J. Norton

Glioblastoma is one of the most aggressive cancers that begins within the brain.

Most cases of GBM are diagnosed at an older age with a median age of 64.

Although the disease does not have some specific symptoms, it shares the common symptoms of other types of cancer such as gastrointestinal discomforts accompanied by personality changes, and symptoms similar to those of a stroke.

According to the statistics, in the US, the incidence affects mostly in the elderly with the diagnosed rate of 3.19 per 100,000 persons. The disease is rare in children.

GBM is deadly, patients with GBM have a median survival of less than one year and less than 2% of patients survive three years or more.

Cruciferous vegetables are a group of vegetables, belongings to the family Brassicaceae (also called Cruciferae) with many genera, species, and cultivated all over the world in suitable climate for commercial profits, including cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts, and green leafy vegetables.

On finding potential and natural foods that process neuroprotective property, researchers investigated the protective effect of sulforaphane (SFN), an isothiocyanate (ITC) derived from cruciferous vegetables against glioblastoma (GBM) progression.

According to the tested analysis, the application of SFN in GBM showed to induce apoptosis and to inhibit both growth and invasion of GBM cells [78].

Furthermore, SFN can overcome the chemoresistance of tumor cells.

The efficacy of SFN against the progression of glioblastoma (GBM) was attributed to the phytochemicals that can cross the blood-brain barrier (BBB) to reach the tumor mass by inhibiting the functions of MMP-9 in GBM in mediating the BBB alternation.

Furthermore, SFN also demonstrated a strong activity in the induction of cancer cell apoptosis and anti-cancer cell proliferation and the protection of healthy neurons.

Moreover, SFN also induces apoptosis of GBM cells by increasing ROS levels in the cancer cells and inhibiting cancer cell migration and/or invasion.

Combination with chemo drug, SFN improved the function of the drug and reduce the chemoresistance of GBM without conferring significant toxicity to normal cells.

Based on the findings, researchers said, " ...the results of these studies support further investigations using SFN in animal models of GBM.

Taken altogether, dietary cruciferous vegetables may be considered functional remedies for the prevention and treatment of GBM, pending to the confirmation of the larger sample size and multicenter human study.

Cranberry, the Anti-Alzheimer's disease Functional Remedy

By Kyle J. Norton

Dementia is a neuropsychiatric disorder as a result of cognitive impairment and behavioral disturbances. The behavioral and psychological symptoms of dementia (BPSD) are common, contributing to caregiver burden and premature institutionalization.

About 5-8% of all people over the age of 65 have some forms of dementia, and this number doubles every five years above that age.

Dementia is the loss of mental ability that is severe enough to interfere with people's every life.

Alzheimer's disease (AD) is a neurodegenerative disease occurred mostly in the older population characterized by progressive brain cell death that happens over time.

Alzheimer's disease worsens over time and it is fatal.

American typical diet contains a high amount of saturated and trans fat, artificial ingredients with fewer fruits and vegetable which can lead to dementia and other kinds of diseases.

As the disease progression, the brain tissue of the patient with AD has fewer and fewer nerve cells and connections.

In the elderly, age-related to the changes in the brain include atrophy (shrinking) of certain parts of the brain, inflammation associated with overexpression of free radicals in the induction of neuro cytotoxicity that affects the energy production within cells and long-term accumulated of plaques in the neurons caused by unhealthy diet are many prevalent risk factors that elevate the early onset and progression of the disease.

In free radical theory, free radical overexpression causes the reduced levels of antioxidant enzymes produced by the elderly initiates the onset of oxidative stress that may attack and damage lipids, proteins, and DNA, leading to irreversible brain cells damage and development of AD.

β-amyloid (Aβ) accumulated in the neurons, the main component of the amyloid plaques in the brains have been found in patients with Alzheimer's disease.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of dementia, researchers investigated the cranberry extract (CBE) rich in polyphenols that delay paralysis induced by alleviating β-amyloid (Aβ) toxicity Alzheimer's disease (AD) in C. elegans model.

According to the experimental results, researchers suggested that CBE showed a strong anti-AD activity by inhibiting the level of Aβ toxicity and promoting memory health, the two major pathological parameters in AD.

C. elegans AD model treated with CBE exerted significantly preventive and therapeutic protocols.

Based on the findings, researchers said, "CBE prominently showed the preventive efficacy, providing a basis for further investigation of these effects in mammals".

Taken altogether, cranberry may be considered a remedy for the management of AD, pending to the confirmation of the larger sample size and multicenter human study.

Antioxidant Lycopene Protects Kidney Cells Against Renal Damage

By Kyle J. Norton

The kidneys, a pair of bean-shaped organs on either side of your spine are the second detoxed organ after liver by removing wastes or toxins in the blood come from the normal breakdown of active muscle and from the food you eat.

Kidney damage is a condition caused by the build-up of waste products and excess fluid in the body.

Depending on the stage of kidney damage, patients may experience symptoms of weakness, shortness of breath, lethargy, swelling, and confusion.

If the kidney can not remove extra potassium from the bloodstream, the condition can lead to abnormal heart rhythms and sudden death.

Kidney diseases can be classified into 2 types
* Acute kidney disease is a sudden episode of kidney failure or kidney damage associated with reduced kidney function in keeping the right balance of fluid in the body, that lasts a few hours or a few days.

* Chronic kidney disease (CKD) is a condition of kidney damage, leading to a decreased level of kidney function that lasts more than three months.

Conventionally, most cases of the renal damage caused by toxins are treated with cisplatin often associated with an acute kidney injury.

On CKD, treatment is totally depending on the causes of the disease including high blood medications, lowering high blood cholesterol and pain relievers.

Dr. Ryoma, the lead scientist in the study "The association between changes in lifestyle behaviors and the incidence of chronic kidney disease (CKD) in middle-aged and older men", wrote, "with respect to habitual moderate exercise and late-night dinner, maintaining an unhealthy lifestyle resulted in a significantly higher odds ratio (OR) for the incidence of CKD".

And, "the lack of habitual moderate exercise, late-night dinner, and bedtime snacking may increase the risk of CKD".

Lycopene is a phytochemical found in tomato in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.

With an aim to find a potent ingredient for the protection of the kidney function, researchers examined protective effects of lycopene against the toxic effects of Aflatoxin B1(AFB1) exposure in the kidney of experimental rats.

The study included 42 healthy three-month-old male Wistar-Albino rats that were randomly divided into six experimental groups including 7 rats in each, as follows: control group, lycopene (5 mg/kg/day, orally for 15 days) group, AFB1 (0.5 mg/kg/day, orally for 7 days) group, AFB1 (1.5 mg/kg/day, orally for 3 days) group, AFB1 (0.5 mg/kg/day, orally for 7 days) + lycopene (5 mg/kg/day, orally for 15 days) group and AFB1(1.5 mg/kg/day, orally for 3 days) + lycopene (5 mg/kg/day, orally for 15 days) group.

According to the tested analysis, injection of Aflatoxin B1(AFB1) showed a significant effect on the kidney oxidative stress marker observed by the significant increase of malondialdehyde (MDA) levels.

Levels of antioxidant enzymes produced by the host such as glutathione (GSH), glutathione-S-transferase (GST), catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and glucose-6-phosphate-dehydrogenase (G6PD) were also decreased in AFB1-rats kidney and heart tissues.

Additonally levels urea, creatinine levels of sodium concentrations in plasma of AFB1 treated rats were also increased substantially.

More precisely, Aflatoxin B1(AFB1) resulted in a significant reduction in the activities of antioxidant enzymes and non-enzymatic antioxidant system in AF treated rats observed by the elevated levels of MDA, compared to the control group.

In other words, Aflatoxin B1(AFB1)  exhibited some parameters associated with kidney damage and injury.

However, all these parameters were significantly inhibited or normalized by the administration of lycopene.

Based on the results, researchers said, " lycopene showed protection against AF induced nephrotoxicity...".

Taken altogether, lycopene found in tomato may be considered supplements for the prevention and treatment of renal damage, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Moronic acid, the Antihyperglycemic Antioxidant

By Kyle J. Norton

Antihyperglycemic activity is the process that promotes a hypoglycemic effect against the progression of hyperglycemia.

Oral antihyperglycemic agents are the medications used to lower glucose levels in the blood in the treatment of diabetes mellitus.

Hyperglycemia, a medical condition of abnormally high blood glucose and a hallmark of diabetes, is a major concern, in people with both type 1 and type 2 diabetes.

Most cases of hyperglycemia occurred in patients who do not manage their diabetes properly.

Untreated hyperglycemia has been found to induce ketoacidosis characterized by the symptoms shortness of breath, breath that smells fruity and gastrointestinal discomforts such as nausea, vomiting, and dry mouth.

Ketoacidosis is a life-threatening condition caused by your body's inability to make enough insulin.

Most cases of hyperglycemia can be controlled by the following diabetes guidance.

By eating a healthy diet high in fiber, such as vegetables, fruits, nuts, legumes (beans, peas, and lentils), whole-wheat flour and wheat bran, accompanied by healthy fish, patients with type 1 and 2 diabetes can control their blood glucose without worrying the onset of hyperglycemia.

The most common diseases associated with untreated hyperglycemia are cardiovascular diseases,
nerve damage (neuropathy) and kidney damage (diabetic nephropathy) or kidney failure and damage to the blood vessels of the retina (diabetic retinopathy), potentially leading to blindness.

Moronic acid is a phytochemical in the subclass of Triterpenoid, belonging to the group of Terpenes found in extracted from Rhus javanica, Mistletoe, etc.

On finding a potential phytochemical for the treatment of high blood glucose, researchers examined the effects of Morolic (1) and moronic (2) acids, the main constituents of acetonic extract from Phoradendron reichenbachianum (Loranthaceae), a medicinal plant on hyperglycemia.

On mice induced diabetic model,
* Daily-administered morolic (1) and moronic (2) acids (50 mg/kg) significantly lowered the blood glucose levels at 60% since the first day until the tenth day after treatment than the untreated group (p<0.05).

* Both compounds diminished plasmatic concentration of cholesterol (CHO) and triglycerides (TG), returning them to normal levels (p<0.05).

* Pretreatment with 50 mg/kg of each compound induced significant antihyperglycemic effect after glucose and sucrose loading (2 g/kg) compared with the control group (p<0.05).

* Moreover, in vitro studies showed that compounds 1 and 2 induced inhibition of 11β-HSD 1 associated with hepatic glucose output activity at 10 μM.

Based on the findings, researchers said, "moronlic and moronic acids have shown sustained antidiabetic and antihyperglycemic action possibly mediated by an insulin sensitization with consequent changes of glucose, cholesterol, and triglycerides, in part mediated by inhibition of 11β-HSD 1".

Taken altogether, moronic acids may be considered supplements for the treatment of hyperglycemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
