Beta Carotene Inhibits the Stress Markers in Patients with Cystic Fibrosis (CF)

By Kyle J. Norton

Cystic fibrosis (CF) is the most common fatal genetic disease affecting mostly the lung in children and young adults, leading to difficulty breathing and coughing up mucus due to frequent lung infections.

However, the condition has also been found in the pancreas, liver, kidneys, and intestine.

According to the Cystic Fibrosis Foundation Patient Registry, more than 30,000 people are living with cystic fibrosis in the US.

Believe it or not, most cases of CF are diagnosed by age 2.

Most common symptoms are depending on the location of cystic fibrosis including
sinus infections, poor growth, fatty stool, clubbing of the fingers and toes.

In male adults, the condition also induces infertility as a result of the blockage or absence of the sperm canal.

Conventionally, as of today, there is no cure for CF. Most cases of CF caused by lung infection are treated intravenously, inhaled, or by mouth.

In severe cases, a lung transplant may be the only option.

Although most cases of CF are associated with a genetic preposition, environment, lifestyle, and age are also 2 main factors that increase the risk of CF.

According to the Dove Medical Press Limited, Life coaching including making lifestyle change associated with absorption of nutrition in people with cystic fibrosis may have a strong impact in loosening mucus in your airways strengthening the heart and lungs and improving quality of life, including drinking plenty of fluids, quit smoking, moderate exercise and increasing hand-washing to protect against infection.

Dr. Karin Bæk Knudsen, the lead scientist at Dove Medical Press Limited wrote, "Cystic fibrosis (CF) is a chronic, life-shortening disease with a significant treatment burden. To support young adults with CF in their everyday life, we previously conducted a life coaching feasibility trial (published elsewhere)".

And, "Life coaching is an intervention that is valued for those who feel challenged by their CF disease. Coaching programs should be designed to include the participants when they feel a need for coaching and are open to change. Screening parameters to identify persons who will most likely benefit from life coaching are needed".

β-Carotene is a phytochemical in the class of Carotenes, belonging to the group of Carotenoids (tetraterpenoids), found abundantly in dark, leafy greens and red, orange and yellow fruits and vegetable.

On finding a potential compound for the treatment of lung disease researchers examined the efficacy of oral beta-carotene supplementation for the correction of an oxidant-antioxidant imbalance in cystic fibrosis (CF).

The study included 24-patients with cystic fibrosis and 14 healthy controls in which 13 CF-patients were allocated to a CF-supplementation group received 1 mg beta-carotene/kg BW/d up to body weight (BW) of 50 kg, patients with a BW greater 50 kg received 50 mg beta-carotene/d for 12 weeks, following by a 12 weeks administration of 10 mg beta-carotene/d. to all patients of the CF-supplementation comapred to placebos of 11 CF-patients. with consumption of starch.

Based on the tested analysis, before the administration of beta carotene, plasma beta-carotene concentrations of CF patients were significantly lower than those of age-matched controls.

However, beta-carotene concentrations of the blood of the CF-supplementation group increased rapidly and reached a value of 0.6 mmol/l after 12 weeks of in the supplementation without affecting the levels of retinol concentrations which is the lower normal range.

Additionally, Levels of antioxdant enzymes were significantly increased and oxidative stress markers were decreased substantially compared to control.

Based on the findings, researchers said, "Thus oral beta-carotene supplementation is effective in the normalizing status of beta-carotene and malondialdehyde in CF patients.".

In other words, improving the circulation of the beta carotene in the blood may be potential for the treatment of cystic fibrosis (CF)

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Indicaxanthin, the Free Radical Scavenging Antioxidant

By Kyle J. Norton

Free radical scavengers are antioxidants that gobble up free radicals before they can cause a chain of reaction.

Most common types of free radical scavengers are catalase, glutathione peroxidase, superoxide dismutase (SOD), vitamin A, C, D, and E.

Free radicals are highly unstable molecules. They can be everywhere. Internally, free radicals are produced in our body during the process of cell metabolism. Externally, free radicals can enter our body from the environment sources, such as the air we breathe, the food we eat and the water we drink.

In other words, any chemical compounds or molecules which process oxygen atom can be free radical after chemical interaction.

Lipid peroxyl radicals are free radical generated from the peroxidation of polyunsaturated fatty acids.

Lipoperoxyl radical scavengers are antioxidants that slow or inhibit the chain reaction of the lipoperoxyl free radicals before they can cause oxidative stress

Oxidative stress is a result of overexpression of free radical-induced by a chain reaction that can not be stopped due to reduced levels of antioxidant enzymes produced by the body.

Why? overexpression of free radicals causes lipid peroxidation by oxidizing the polyunsaturated fatty acids and cholesterol through three distinct mechanisms, including free radical-mediated chain reaction, enzyme-mediated oxidation, and nonradical, non-enzymatic oxidation.

Over time, oxidative stress can damage the lipid, and protein and induce the alternation of healthy cell DNA.

Antioxidants, the stable molecules have been found to protect polyunsaturated fatty acids against peroxidation by scavenging peroxyl radicals.
Dr. Authors: Maria A., the lead author wrote, "Aerobic life is characterized by a steady formation of reactive oxygen species and free radicals, which is almost entirely counteracted by endogenous primary and secondary antioxidant systems".

And, "Maintenance of these systems is then imperative to ensure a continuous defense to cells and to avoid conditions known as oxidative stress. Apart from antioxidant vitamins, many compounds from the plant kingdom are now considered very helpful to maintain a proper cell redox balance".
In other words, antioxidants with free radical scavenging activity may protect the body against the damage of various cellular components.

Indicaxanthin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.

On finding a potential compound that processes free radical scavenging activity, researchers examined the effect of Indicaxanthin incubated with lipoperoxyl radicals generated in methyl linoleate methanol solution by 2,2'-azobis(2,4-dimethylvaleronitrile).

According to the tested analysis of the solution, indicaxanthin acted as a chain-terminating lipoperoxyl radical scavenger by inhibiting (2,4-dimethylvaleronitrile oxidation by inducing the clear-cut lag periods and the decrease of the propagation rate in the initiation of free radical chain reaction in a dose of concentration-dependent manner.

Moreover, the injection of dietary indicaxanthin also delayed the liposome oxidation remarkably delayed.
However, the phytochemical efficacy is not linearly related to the levels of concentration.

More precisely, indicaxanthin and alpha-tocopherol incorporated in liposomes exhibited cooperative synergetically antioxidant effects and reciprocal protective interactions.

Based on the findings, researchers said, "The extent of synergism decreased at the increase of the ratio (indicaxanthin)/(alpha-tocopherol). A potential antioxidant mechanism of indicaxanthin is discussed in the context of the chemistry of the molecule, and of the possible reactivity of a short-lived intermediate".

Taken altogether, indicaxanthin may be considered a free radical scavenger pending to the confirmation of the larger sample size and multicenter human study.

Carotenoids Process the Strong Impact In the Prevention and Treatment of Cystic Fibrosis (CF)

By Kyle J. Norton

Cystic fibrosis (CF) is the most common fatal genetic disease affecting mostly the lung in Canadian children and young adults, leading to difficulty breathing and coughing up mucus and frequent lung infections.

However, the condition has also been found in the pancreas, liver, kidneys, and intestine.

According to the Cystic Fibrosis Foundation Patient Registry, more than 30,000 people are living with cystic fibrosis in the US.

Cystic Fibrosis Canada in the investigation of the risk of CF in older adults wrote, "Today, the estimated median age of survival of CF patients in Canada is over 50 years, and the majority of patients are over 18 years of age. CF patients enjoy an improved quality of life, pursuing careers, and starting families of their own".

Lycopene is a phytochemical found in tomato in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

On finding a potential compound for the treatment of lung disease researchers examined the effects of carotenoids, the potent antioxidants on singlet oxygen and other toxic oxygen species in patients with cystic fibrosis (CF).

The study included a total of of 17 patients with cystic fibrosis (CF) and 10 normal children,

According to the assessment of plasma levels of four carotenoids, beta-carotene, alpha-carotene, lutein, and lycopene, levels of carotenoids in children with CF were significantly lower compared to normal control participants.

There was no significant difference in total cholesterol total carotene between groups. Futhermore, there was no correlation of fat absorption or measures of adiposity in children with CF comapred to control.

However, levels of selected carotenoids were correlated negatively with serum IgG levels associated with the measurement of inflammation.

More importantly, researchers also found that the differences in plasma carotenoid levels between children with CF and normal children were involved in the rapid turnover of carotenoids, by quenching of toxic oxygen species in inflammatory states of CF.

The results strongly suggested that improving the concentration of carotenoids that reduces the implication of the toxic ROS expression may have an impact on the prevention and treatment of CF.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Lycopene, the Anti-diabetic Nephropathy Phytochemical, Researchers Find

By Kyle J. Norton

Diabetic nephropathy (diabetic kidney disease) is a complication of kidney damage in diabetes.

Long-term unmanaged hyperglycemia has been found to damage parts of the kidneys, leading to reduced kidney function in filter waste from the blood filters, resulting in a  'leaky' syndrome that allows protein into your urine.

Diabetic nephropathy is the 3 stages of kidney change in patients with diabetes. In this stage, patients may experience symptoms of abnormally elevated urinary albumin excretion, compared to the 15 and 300 μg/min in the baseline situation.

The progression of diabetic nephropathy that leads to an increased rate in albumin excretion is higher in patients with increased blood pressure.

The condition in the advanced stage can lead to symptoms of worsening blood pressure, protein in the urine, swelling of feet, ankles, hands or eyes, increased need to urinate, loss of appetite, nausea and vomiting, fatigue and persistent itching.

In severe cases, patients may also experience symptoms of confusion or difficulty concentrating.

More importantly, if high blood pressure is left uncontrol, diabetic nephropathy can induce persistent proteinuria >0.5 g/ 24 h that furtherly declines the renal function with the mean fall rate being around 1 ml/min/mo.

In other words, by treating hypertension and hyperglycemia in patients with diabetes, the progression of diabetics kidney disease may be slowed.

Some researchers suggested that by maintaining healthy blood glucose levels and keeping them within the target range and a change of healthful diet accompanied by moderate exercise, diabetic nephropathy can be control

Dr. Chijoke Onyenwenyi, "Kidney disease is common in patients with type 1 and type 2 diabetes mellitus and is associated with adverse health outcomes, including progression to end-stage renal disease. In the general population, adherence to a healthy lifestyle is known to reduce the risk of cardiovascular events and death".

And, "Among individuals with diabetic kidney disease, modifications in lifestyle factors, including diet, physical activity, smoking habits, and body mass index, represent a promising cost-effective therapeutic adjunct to pharmacologic treatment of kidney disease incidence and progression".

With an aim to find a potent compound for the treatment of diabetic complications, researchers examined the effects of lycopene, a carotenoid, on oxidative stress-induced the progression of diabetic nephropathy.

According to the tested assays, lycopene inhibited the menace ad oxidative stress and stress markers associated with advanced glycation end products (AGEs) both in vitro and in vivo.

Furthermore, lycopene decelerated the ribose induced AGE formation in the normal adult human kidney. (HK-2) cells and in rat kidneys thereby inhibiting the expression RAGE involved in the inflammatory response, leading to reduced levels of nuclear factor κB (NFκB) and matrix metalloproteinase 2 (MMP 2) expressions.

Where nuclear factor κB (NFκB) and matrix metalloproteinase 2 (MMP 2) expressions. are associated with cytokine production under oxidative stress.

Most importantly, nephropathy (DN) induced by the administration of ribose was totally inhibited by the injection of lycopene.

Based on the findings, researchers wrote, "lycopene provides protection against the development of diabetic nephropathy and ameliorates renal function by halting AGE-RAGE axis".

Taken altogether, lycopene found in tomato may be considered supplements for the protection against the risk of diabetic nephropathy in diabetics, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Green Peas, the Potential Anti Cancer Whole Food

By Kyle J. Norton

Cancer is a group of cell mutated diseases caused by the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining of the tissue of an organ. At early, most people with cancer are asymptomatic.

At the later stage, due to the size and the tumor, patients may experience the general cancer symptoms such as unintended weight loss, fatigue, gastrointestinal discomfort and loss of appetite and localized symptoms such as severe pain and bleeding.

The exact causes of cancer are not identified. However, most prevalent factors associated with the cancer onset include age, family history, long-term exposure to certain cancer-induced chemicals, use of tobacco and excessive alcohol drinking,..... the list goes on and on, depending on the types of cancer.

Some researchers suggested that the most common risk factor that causes cancer in the Western world may be associated with widespread obesity.

Dr. Karen Basen-Engquist, in the evaluation of obesity risk of cancer, said, "In the United States, approximately 85,000 new cancer cases per year are related to obesity. Recent research has found that as the body mass index increases by 5 kg/m2, cancer mortality increases by 10%".

And, "High priorities for future research involve additional work on the underlying mechanisms, and trials to examine the effect of lifestyle behavior change and weight loss interventions on cancer and intermediate biomarkers".

The findings strong indicated the risk of cancer is increased substantially in obese.

Green peas are species of Pisum Sativum belongings to the family Fabaceae and native to western Asia from the Mediterranean Sea to the Himalaya Mountains. The pea is a green, pod-shaped vegetable and a cool-season crop grown in many parts of the world.

On finding a potent ingredient for the treatment of cancer, researchers investigated the impact of green pea (GP) extracts anti-DNA alternative activity.

According to the studies,
*  The extracts of P. sativum have been found to be pharmacologically active inducing anticancer effects.

* Compared to the effect of Bowman-Birk trypsin-chymotrypsin inhibitor, a potential cancer chemopreventive agent, with the protease inhibitors, rTl1B and rTl2B, P. sativum seed showed similar significant activity.

* In human colorectal adenocarcinoma HT-29, P. sativum extracted lectins from Egypt exhibited the antiproliferative property to liver cancer cell line.

* In MCF-7 (breast), HepG-2 (liver), HEP-2 (larynx), and HCT-116 (colon). an extracted lectin from leaves and buds of P. sativum demonstrated cytotoxic property.

* Recently, the seed extracts also showed the cytotoxic effects on LS174 (colon), MDA-MB-453 (breast), A594 (lung), and K562 (blood) cell lines.

Taken altogether, green peas may be considered a functional food for the prevention of cancer,  pending to the confirmation of the larger sample size and multicenter human study.

Cranberry Promotes the Cardio-Metabolic Health

By Kyle J. Norton

Cardiometabolic risk is a condition that estimates the possibilities of developing atherosclerotic cardiovascular (CV) disease and diabetes mellitus.

Cardioprotection is a process including the use of substances associated with the preserve of the function and viability of cardiac muscle tissue by the reintroduction of oxygenic blood flow back to the tissues.

Cardiovascular disease (CVD) is a group of conditions caused by the underlying damage of blood vessels and the heart.

More precisely, CVD can be coronary artery diseases (CAD) such as angina, myocardial infarction (commonly known as a heart attack) or stroke, depending on the location of the blockage or disruption,

According to the statistics provided by the American Heart Association, approximately 84 million people in this US living with some form of cardiovascular disease. The condition has also been found to cause the death of about 2,200 people a day.

Atherosclerosis is a medical condition caused by plaque accumulated on the wall of arteries, leading to the narrowing of the artery and reducing oxygen risk blood flow to the heart and brain.

Depending on the location of the blockage, if the reduced blood flow occurred in the brain, it will cause the death of the brain cells, leading to stroke.

However, if the reduced blood flow occurred in the heart, it will cause damage to the heart tissues, leading to heart disease.

Most common disease associated with atherosclerosis is coronary artery disease. Plaques are made up of cholesterol, calcium and other chemical substances in the artery

Most people with atherosclerosis are asymptomatic at the early stage.

Diabetes is a medical condition characterized by insufficient insulins in the bloodstream. or insulins in the bloodstream are not used properly to convert glucose to energy.

Epidemiologically, diabetes is caused by either the beta cells in the pancreas died-off or insulin receptors with a function that use insulin to convert glucose to energy clogged-up by fat or cholesterol.

In a healthy individual, after eating, the rise of blood glucose triggers the production of insulin by the pancreas into bloodstream After you eat, your blood sugar (glucose) rises. This rise in glucose triggers your pancreas to release insulin into the bloodstream that travels through the blood to your body's cells before converting glucose into energy or store it to use later.

In other words, insulin released by the pancreas joints with the Insulin receptors on the outer part of a cell that binds insulin in the blood to convert glucose to energy.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of cardiometabolic health, researchers examined the effect of Vaccinium berry fruit in modulating gut microbiota function and cardiometabolic risk factors.

In cardiometabolic health, cranberry constituents and their bioactive catabolites improved the
serum lipid profiles reduced the levels of blood pressure.

Furthermore, the endothelial functions associated with vascular relaxation and contraction, glucoregulation involved in the regulation of glucose metabolism. and a variety of biomarkers associated with inflammation and oxidative stress in the induction of CVD and diabetes were also inhibited by the cranberry constituents and their bioactive catabolites

Based on the findings, researchers said, " These results warrant further research, particularly studies dedicated to the elucidation of dose-response relations, pharmacokinetic/metabolomics profiles, and relevant biomarkers of action with the use of fully characterized cranberry products.".

Taken altogether, cranberry may be considered a remedy for the improvement of the cardiometabolic health, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Turmeric Processes Anti Colorectal Cancer Activity

By Kyle J. Norton

Colorectum including the large intestine and the rectum is the second part of the digestive system involved in the absorption of water from the small bowel contents and breaks down of certain materials in the feces into substances of which some of them to be reabsorbed and reused by the body.

Colorectal cancer is a medical condition caused by irregular cell growth in the tissue of the colon or rectum.

Most cases of colorectal cancer start in the cells on the surface of the inner lining in the colon and rectum, before penetrating into deeper layers to form a tumor through proliferation.

Colorectal cancer is relatively very common and slowly growing and progresses cancer in a predictable way.

According to the statistics provided by the American Cancer Society, excluding skin cancers, colorectal cancer is the third most common cancer diagnosed in both men and women in the United States.

In 2019, approximately 101,420 new cases of colon cancer and 44,180 new cases of rectal cancer will be diagnosed in the US.

The 5-year survival rate of colorectal cancer diagnosed at the localized stage is 90%. Sadly, only 39% of patients are diagnosed at this early stage.

The exact causes of colorectal cancer are unknown. Epidemiologically, the most common risk factors associated with the onset of the disease are
* Age
The disease is more common in people over the age of 50.  Regular colonoscopy after age of 50 is valuable to detect the disease in the early stage.

*  Bowl inflammatory diseases
A prolonged period of inflammatory diseases of the colon, such as ulcerative colitis and Crohn’s disease, can increase your risk of colon cancer due to long term inflammation of the lining of the colon.

* Family History of colorectal cancer
People with parents, siblings, children who have had colorectal cancer are more likely to develop colon cancer.

* Inherited Syndromes
Genetic syndromes passed through generations can increase the risk of developing colon cancer.

However, most people with some of the aforementioned prevalent factors have never developed colorectal cancer.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, etc.

With an aim to find a potential compound for the treatment of colorectal cancer, researchers investigated the effects of curcumin nanoformulations anti-cancer activity by overcoming the limitations of the compound.

Application of the nanoformulations not only displayed a greater anticancer activity compared to free curcumin but also extended at the same time non-toxic to the normal cells.

Furthermore, curcumin also demonstrated a strong anti colorectal cancers effect in the regulation of multiple cell signaling pathways associated with tumor growth and anti-apoptosis.

Moreover, curcumin also promoted the expression of tumor suppressor gene which is crucial in cancer development and progression.

The joint study led by the Ahvaz Jundishapur University of Medical Sciences in the examination of curcumin, a bioactive substance originating from turmeric root anticancer properties showed that curcumin inhibited the expression of a gene associated with involved in tumor initiation and progression. In other words, through inhibition of this gene, curcumin reduced the risk of tumor metastasis.

Additionally, by inhibiting the insulin and insulin-like growth factor-1 receptor curcumin showed a greater decrease in the proliferation and migration of chemoresistant colorectal cancer cells.

Based on the results, Dr. Hosseini SA, the lead scientist said, "These results suggest the possible therapeutic effects of curcumin in adjuvant therapy in metastatic colorectal cancer".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of colorectal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
