Dithiolthiones, the bioactive compound found abundantly in the cruciferous vegetables may have a potential effect on reducing the risk of neurodegenerative disease, some scientists found.
Dithiolthiones are phytochemicals in the class of Organosulfides, found abundantly in cruciferous vegetables, garden sorrel, horseradish, etc.

Neurodegenerative disease is a group of conditions that primarily affect the neurons in the brain, including Parkinson's disease (PD) and Alzheimer's disease.

In other words, neurodegenerative disease is caused by the progressive death of neurons in different regions of the nervous system.


The progressive loss of neurons induced neurological and neuropsychological signs and symptoms have also been found in comparison to the stage of disease onset of patients.

The causes of neurodegenerative diseases are unknown, however, researchers do know that the increase in age is one most consistent risk factor for developing a neurodegenerative disease, especially AD or PD, as the disease has been found over 60% in the population aged 65 and beyond compared to another group.

And this number is found to increase at an alarming rate. Dr. Serge Przedborski, the lead author in the investigation of the risk of neurodegenerative disease in elderly said, "it can be anticipated that, over the next generations, the proportion of elderly citizens will double, and, with this, possibly the proportion of persons suffering from some kind of neurodegenerative disorder".

Epidemiologically, geographically, hereditary viral, and toxicological exposure correlates, and deficiencies of elements such as calcium and selenium are some of the risk factors in the development of neurological diseases.


Also, people who follow the Western diet pattern are at an increased risk of the disease. According to the 2018 Alzheimer's Association International Conference, July 22-26, 2018 in Chicago, Illinois, the Western diet is known to include high consumption of red meats, processed meats, as well as fatty and fried foods. This dietary pattern has been linked to a greater risk of cognitive decline and dementia.

Sulforaphane (10 or 50mg/kg) was administered to C57BL/6 mice by oral gavage for 14 days (days 1-14), Memory impairment was induced by intraperitoneal injection of scopolamine (1mg/kg) for 7 days (days 8-14).

Mice receiving an injection of scopolamine showed impaired learning and memory retention and considerably decreased cholinergic system reactivity in the hippocampus and frontal cortex. The results were observed by a decreased acetylcholine (ACh) level and increased acetylcholinesterase (AChE) activity.

Where acetylcholine (ACh) is a neurotransmitter, associated with the function in sending signals to other cells [neurons, muscle cells, and gland cells] and acetylcholinesterase (AChE) is an enzyme that catalyzes the breakdown of acetylcholine.

Treatment of sulforaphane significantly attenuated the scopolamine-induced memory impairment and improved cholinergic system reactivity.

In other words, sulforaphane exerted a neuroprotective effect against the scopolamine in the induction of oxidative stress to the neurons. Sulforaphane at doses of 10 or 20μM increased the ACh level, decreased the AChE activity, and increased ChAT expression in scopolamine-treated primary cortical neurons.

Dr. Lee S, the lead scientist suggests "Sulforaphane might exert a significant neuroprotective effect on the cholinergic deficit and cognitive impairment".

Additionally, in the concern of no drug treatment of schematic/traumatic brain injury, Alzheimer's disease, and Parkinson's disease which share common characteristics such as oxidative stress, misfolded proteins, excitotoxicity, inflammation, and neuronal loss, researchers investigated the effect of sulforaphane against the mentioned diseases.

In several in vitro and in vivo studies isothiocyanate sulforaphane, derived from the hydrolysis of the glucosinolate glucoraphanin mainly present in Brassica vegetables, has demonstrated neuroprotective effects.

The efficacy was attributed to the activity of sulforaphane in activating the Nrf2/ARE pathway in modulating the expression of oxidative stress.

Dr. Tarozzi A, the lead author in the final report of the experiment wrote, " sulforaphane appears to be a promising compound with neuroprotective properties that may play an important role in preventing neurodegeneration".

More importantly, in dopaminergic neurotoxicity in mice induced by 6-hydroxydopamine (6-OHDA), the phytochemical showed a neuroprotective effect through its ability to enhance the antioxidant enzymes of the host and improve neuronal survival.

Neuropsychological assessment provides both general and specific information about current levels of cognitive performance. An average or composite score across multiple ability areas provides an overall index of how well a person functions cognitively at the current time.

Cognition is the process of the brain functioning associated with the attention of working memory, producing and comprehending language, learning, reasoning, problem-solving, and decision-making.

Cognitive deficit is a medical condition, that affects a person's thinking, communication, understanding, or memory. The cognitive deficit can be acute or chronic.

Acute cognitive deficit is a short-term health problem caused by drug toxicity and withdrawal, electrolyte disturbance, lack of drugs, liver disease, infection, reduced sensor input, intracranial urinary retention/fecal impaction, or myocardial/metabolic/pulmonary. An acute cognitive deficit may be recovered if the cause is found.

Chronic cognitive deficit is a permanent medical condition caused by the gradual or permanent loss of neurons, leading to problems with language, ideomotor and constructive praxis, visual gnosis, spatial attention, learning abilities, and executive functions.

The double-blind, placebo-controlled, randomized, parallel-group, clinical trial included 50 community-dwelling, cognitively intact volunteers, > or = 60 years old, who reported no history of dementia or significant neurocognitive impairments.

Participants in the study were randomly assigned to receive either 32 ounces/day of a beverage containing 27% cranberry juice per volume (n = 25) or placebo (n = 25) for 6 weeks.

According to the neuropsychologic tests before and after treatment on participants' self-reported, there was a nonsignificant trend (X2(1) = 2.373, p = 0.123) on overall abilities to remember.

However, more than twice of many participants (37.5%) reported the overall ability to remember at the end of treatment compared to placebo controls (17.4%).

In other words, cranberry extract (CE) although can not improve overall neuropsychologic symptoms, enhances the short-term memory of dementia or significant neurocognitive impairments.

An atheroma, or atheromatous plaque ("plaque"), is an abnormal accumulation of plaque in the inner layer of the wall of an artery, found in most adults, a leading cause of atherosclerosis.

Atherosclerosis is a condition caused by long-term plaques built up on the wall of the arteries, leading to narrowing the arteries, a major cause of the cardiovascular disease (CVD)

CVD is a medical condition involved in blood vessels. CVD can be heart disease or stroke depending on the location of a blood clot which causes blockage of the blood flow in the artery.

According to the statistics provided by the Europeanean Hear Network, cardiovascular disease (CVD) causes 3.9 million deaths in Europe and over 1.8 million deaths in the European Union (EU) every year. In 2015, approximately, 11.3 million new cases of CVD in Europe and 6.1 million new cases of CVD in the EU were diagnosed.

Plaque is made up of fat, cholesterol, calcium, and other substances found in the blood.


Atheromatous plaques build up in the arteries can be classified into several stages, including

The study included the measurement of plasma levels of carotenoids and genotyped 20 single nucleotide polymorphisms (SNP) in SOD1, SOD2, SOD3, XRCC1, and OGG1 in 559 men with non-metastatic prostate cancer undergoing radical prostatectomy.
According to the results from tested assays, the serum of circulated carotenoid levels was inversely associated with the risk of high-grade prostate cancer, with odds ratios (OR) of 0.34 compared to the highest versus lowest quartiles.

In chemical analysis, the relative odds ratio of α-carotene,β-carotene, lycopene, and total carotenoids.was 0.31, .55, 34, and 0.37 respectively.

In other words, all major phytochemicals found in carotenoid work synergistically to enhance the efficacy of carotenoid against the risk of high-grade prostate cancer.


Furthermore, the presence of the SNPs rs25489 in XRCC1, rs699473 in SOD3 and rs1052133 in OGG1 genotypes also was found to modify the activity of α-carotene, β-carotene, and lycopene, respectively.

Believe it or not, the high serum of lycopene was found among men with Gleason score ≤ 3 + 4 and lower FGA (P = 0.04).


More precisely, the efficacy of carotenoids reduced the risk of prostate cancer, however, the risk of high-grade invasive prostate cancer are depending on the levels of lycopene.

Dr. Nordström T, the lead scientist said, "Higher lycopene level was associated with less genomic instability among men with the low-grade disease which is novel and supports the hypothesis that lycopene may inhibit the progression of prostate cancer early in its natural history".

Phytochemical allicin isolated from garlic may be used for the prevention and treatment of osteosarcoma with no side effects, some scientists suggested.

Osteosarcoma is a bone cancer found mostly in teenagers and people over age 60.

Most cases of osteosarcoma are associated with genetic syndromes. In other words, the disease is the result of the conditions associated with inherited mutated genes from the parent.

However, an unhealthy lifestyle such as body weight, physical activity, diet, tobacco, gender, age, height, ethnicity, and exposure to bone radiation also play a major role in many adult cancers.

According to the statistics, in the US, osteosarcoma is rare and approximately 900 new cases of osteosarcoma are reported each year. and children and teens younger than 20 account for half of the numbers.

Dr. James J. Morrow, the lead scientist in the study of the genetic risk of osteosarcoma wrote, "Osteosarcoma is the most common primary malignancy of bone, typically presenting in the first or second decade of life"

And, " Increased risk of osteosarcoma is associated with a number of well-defined genetic syndromes: hereditary retinoblastoma (germline mutation of the Rb gene), Li-Fraumeni syndrome (germline mutation of the p53 gene), Bloom syndrome (germline mutation of the RECQL2 gene), Werner syndrome (germline mutation of the RECQL3 gene), and Rothmund-Thomson syndrome (germline mutation of the RECQL4 gene)"


These results strongly suggested that young teenagers with genetic syndrome are more likely to be infected by the disease, compared to others.

Allicin is a phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in onion and garlic.

With an aim to find a natural ingredient for the treatment of osteosarcoma with no side effects, researchers at the Wenling City Chinese Medicine Hospital looked into the anti-osteosarcoma activity of the main biologically active compound derived from garlic.


The combined treatment significantly suppressed the invasion, motility, and colony formation ability and activated the protein associated with a pro-apoptotic activity, compared to the lone treatment group.

Furthermore, in vivo. artesunate and allicin also inhibited tumor growth and cell proliferation

Dr.Jiang W, the lead scientist said, "Artesunate and allicin in combination exert synergistic effects on osteosarcoma cell proliferation and apoptosis".

Moreover, in examining the effects of diallyl sulfide (DAS) derived from allicin in vitro and in vivo on the proliferation of human osteosarcoma MG-63 cells, researchers found that in vivo, the nude mice MG-63 cells tumor-bearing model after twenty-one days of treatment with DAS inhibit proliferation of MG-63 cells in a time- and concentration-dependent manner.

DSA groups (different concentrations) showed significantly reduced expression levels of VEGF mRNA, associated with stimulating the formation of blood vessels. compared with those in the control group.

At the dose of 20 and 40 μg/mL, cell migration was significantly lower in the DAS groups (91.4±8.3 and 81.8±7.4, respectively) compared to the control group (150.4±14.7, both).

Also at the dose of 20 and 40 μg/mL, DAS also decreased the microvessel density (MVD) of MG-63 tumor-bearing tissue.

Taken together, phytochemical allicin may be considered a bioactive compound for the prevention and combined with the primary therapy for the treatment of osteosarcoma.

However, at the later stage of the tumor progression, if undetected, the large size of the tumor can suppress and break the nearby small blood vessels, leading to internal bleeding and severe if the tumor also suppresses the nearby nerve cells.

Primary cancer cells at this advanced stage can also travel a distance away from the original site to affect other healthy tissue and organs, leading to secondary metastasis.

Pancreatic cancer is a medical and chronic condition characterized by irregular cell growth in the tissue of the pancreas.

In the early stage, pancreatic cancer starts in the cells on the surface of the inner lining of pancreatic tissue. At the advanced stage, cancerous cells of the pancreas can travel a distance away to invade other healthy tissues and organs.

According to the tested analysis, the extract significantly increased the cells' spherical structure with double-layer membranes degraded by autophagy through cleaning out damaged cells in normal cells.

However, in human pancreatic cancer (PC) cells, withanolide extract also blocked the degradation of autophagy by inhibiting SNARE protein in the the-mediated fusion of autophagosomes and lysosomes which contains degradative enzymes enclosed in a membrane.

In other words, in human pancreatic cancer (PC) cells, the extract promotes the cancer cell degradation by autophagy to induce apoptosis.

More precisely, WA induced ER stress-mediated apoptosis. by promoting the accumulation of ubiquitinated proteins, associated with degradation via the proteasome, a protein involved in degrading unneeded or damaged proteins.

In vivo, combining WA with a series of ER stress aggravators enhanced apoptosis synergistically in tumor growth in PC xenografts.

Based on the findings, researchers said, "simultaneous suppression of 2 key intracellular protein degradation systems rendered PC cells vulnerable to ER stress, which may represent an avenue for new therapeutic combinations for this disease".

Taken together, Ashwagandha may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of pancreatic cancer, pending the result of larger sample size and multicenter study
