#Phytochemical Piperine Inhibits Angiogenesis of #Cancer Cells, Scientists Say

Piperine, a phytochemical alkaloid in the class of organosulfur compound may have a significant effect in blocking the energy that is necessary for cancer cells proliferation and survival, some scientists suggested.
Angiogenesis is the formation of new blood vessels in the body for healing and reproduction.

However, in invasive cancers, the formation of a new blood vessel from pre-existing vessels stimulated by cancer chemicals may be deadly to the host.

These new blood vessels provide the nutrients and fluids for cancer growth and metastasis.

Cancer is a class of diseases, characterized by cell growth irregularly that starts in the surface of the inner lining of tissue or organs. Patients at the beginning of cancer onset do not experience any symptoms.

At the later stage of the progression, the cancerous cell may travel a distance away from the primary site to infect other healthy tissues and organs. If cancer has grown large enough, it may affect other nearby healthy tissue and nerve cells, causing localized symptoms and persistently severe pain.

Angiogenesis occurs totally depends on a net balance of angiogenic factors and antiangiogenic factors, which are secreted by both tumor cells and host-infiltrating cells. If the tumor's angiogenic factors win, the new blood vessels are formed.

Some researchers suggested if we can produce a drug that cuts off the forming of the new blood vessel which supplies energy to cancer, we may cure cancer.

However, according to the epidemiological studies, antiangiogenic therapy in the inhibition of the growth of the tumor works best in combination with conventional cytotoxic chemotherapy.


Dr. Roberta WC Pang, the lead author in the investigation "Clinical Implications of Angiogenesis in Cancers" wrote, "Recently, a monoclonal antibody against vascular endothelial growth factor, which is one of the most potent angiogenic factors, has been approved for clinical use in colorectal cancer patients after a clinical trial confirmed that combining the antibody with standard chemotherapy regimen could prolong patient survival".


We can only hope that in a distant future, a single miracle drug can have an antiangiogenic potency to block off all cancers proliferation.

Piperine is a phytochemical alkaloid in the class of organosulfur compounds, found abundantly in white and black pepper, long pepper, etc.

A research team at the Dalhousie University in the investigation of piperine, a major alkaloid constituent of black pepper physiological actions in inhibition of cancer cell proliferation including the killing of cancer cells launched a study to examine the angiogenetic effect of piperine in tumor progression.

Injection of piperine inhibited the proliferation and G(1)/S transition of human umbilical vein endothelial cells (HUVECs) without causing cell death.


In vitro, piperine also inhibited HUVEC migration and tubule formation and collagen-induced angiogenic activity in rat aorta explants and breast cancer cell-induced angiogenesis in chick embryos.

Importantly, piperine inhibited phosphorylation of Ser 473 and Thr 308 residues of Akt (protein kinase B), which is a key regulator in the induction of cancer cells survival and angiogenetic proliferation.

The findings reconfirmed the function of piperine as an angiogenesis inhibitor for use in cancer treatment.

Additionally, the finding of the phytochemical which processes a strong effect in improving the function of curcumin, in the suppression of angiogenesis by inhibiting the curcumin's lack of water solubility, and relatively low bioavailability.

After multiple approaches were sought to overcome these limitations, researchers finally discovered natural curcumin analogs, namely piperine.

Piperine in the co-administration with curcumin enhanced the functions of curcumin in the suppression of angiogenesis without reducing the antitumor activity.


Taken together, piperine may be considered a functional supplement used for the prevention of cancers and an adjunction therapy by cutting the supply of energy of cancer that is life and death for cancer colonization, depending on the further study of large sample size and multi-center in human consumption.

Peyronie's disease is a rare condition caused by plaque, that forms inside the penis, leading to a bent erect penis, depending on where the plaque is located.

Patients with Peyronie's disease has been found to affect sexual desire due to a physical condition associated with painful sexual intercourse and erectile dysfunction.

Researchers do not know the exact causes of Peyronie's disease. However, according to the epidemiological studies, researchers suggested that injury that causes bleeding inside the penis may induce the built-up of scars, particularly in men with the genetic preposition of certain genes.

Most cases of pain in Peyronie’s disease are mild and often do not require treatment, however, over counter medicine may be needed occasionally.

Believe or not, most mild cases of men with Peyronie's disease do not require treatment as the condition has been found to go away within a year. In severe cases, surgery may be necessary to remove the scar tissues in the penis that cause the condition.


Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant has been used as an herb in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, psoriasis, diabetes, etc.

Researchers to find a natural compound for the treatment of Peyronie's disease investigated various antioxidants, including bilberry in PD treatment which significantly increase the likelihood of therapeutic success.

By comparing the effects of silymarin, ginkgo biloba, vitamin E, bilberry, topical diclofenac sodium, and pentoxifylline (PTX) in patients with PD, researchers found that by administration of a single oral antioxidant; two oral antioxidants; and three oral antioxidants; five oral antioxidants + local diclofenac; and five oral antioxidants + local diclofenac + PTX achieve better results when combining more than one agent.

According to the tested assays, the combination of more than a single compound reduced the symptoms in patients with PD, including the improvement and disappearance of penile pain; percentage of reduction in the volume of penile plaque; reduction in penile curvature; recovery of erectile function in patients with erectile dysfunction; increase in the International Index of Erectile Function score; and reduction of psychosexual impact.

Particularly, treatment of combined therapy including oral treatment with more than one antioxidant is greater when topical use of diclofenac gel and perilesional injection of PTX, compared to those of single conventional medicine.

In other words, combined natural or dietary antioxidants such as bilberry into the diet of patients with PD are more effective in inhibiting the symptoms compared to the use alone of any single ingredient therapy.


The final outcome of 6 months of treatment showed that combined antioxidant therapy is effective in treating PD.

Dr. Paulis G, the lead scientist, after taking other factors into account wrote in the final report, " Although several articles have already been published reporting the effectiveness of combined treatment in PD, this is the first study clearly proving how, as the number of substances used in treatment rises, a proportionally greater therapeutic effect is achieved."

Taken together, bilberry combined with other antioxidants or conventional medicines may be considered function remedies for the treatment of Peyronie's disease (PD), pending the validation of a larger sample size and multicenter human study.

Indole-3-carbinol may be the pharmaceutical target in the search for a chemo drug doe the treatment of breast cancer, some scientists suggested.

Breast cancer is a medical and chronic condition caused by cell growth irregularly and uncontrollably in the breast tissue. Furthermore, indole-3-carbinol cyclic tetrameric derivative CTet induces autophagy and inhibits breast cancer cell proliferation, researchers at the University of Urbino Carlo Bo conducted an investigation to examine the antitumoral activity of a mixture of tri- and tetrameric cyclic I3C derivatives (CTr/CTet) both in vitro (MCF-7 and MDA-MB-231 breast cancer cell lines.

Observation of the application on the tested cancer cell line suggested CTr/CTet mixture inhibits breast cancer cell proliferation (IC50 = 1.3 and 1.6 μg/ml in MCF-7 and MDAMB- 231, respectively) inducing the G0/1 cell cycle phase accumulation.

Moreover, in Vivo, the tumor growth of xenotransplanted was also inhibited by reducing the function of Cyclin E and its low molecular weight which is important in the transition from G1 to the S phase of the cell cycle that determines the initiation of DNA duplication.


Taken together, Indole-3-carbinol processed apoptotic and antiproliferative activity can be considered a functional supplement for the prevention and combined with primary medicine for the treatment of breast cancer.

However, further data collection large example size, and multi-center studies performed with human consumption of the phytochemical during the course of the disease will be necessary to complete the picture of Indole-3-carbinol anti-breast cancer possibilities.

#Phytochemical #Polysulfides Inhibit Pathways Associated with the Onset of #SkinCancer, Reserachers Suggest

Polysulfides are phytochemicals in a class of chemical compounds containing chains of sulfur atoms, belonging to the group of Organosulfides found abundantly in the oxidized product, including beer, wine, whiskey, garlic oil, etc.
Skin cancer is a medical condition of uncontrolled growth of abnormal skin cells and is often detected at an early stage.

In skin cancer progression, diallyl trisulfide (DATS) is more potent than mono- and disulfides against skin cancer, through inhibited cell growth of human melanoma A375 cells and basal cell carcinoma (BCC) cells by increasing the levels of intracellular reactive oxygen species (ROS) and DNA damage and by inducing G2/M arrest, endoplasmic reticulum (ER) stress, and mitochondria-mediated apoptosis(1).

The study by the National Taiwan University, also showed that Diallyl sulfide (DAS), diallyl disulfide (DADS), and diallyl trisulfide (DATS), extracted from crushed garlic, inhibited skin cancer cell growth through involvement in G(2)/M arrest and apoptosis via activation of the p53 pathway in response to the oxidative DNA damage(2).


The Chung Shan Medical University Hospital, study also insisted that DAS protects against ultraviolet B (UVB)-induced skin tumor formation through anti photocarcinogenesis effect accompanied by the down-regulation of cell-proliferative controls, involving thymine dimer, PCNA, apoptosis, transcription factors NF-κB, and of inflammatory responses involving COX-2, PGE2, and NO, and up-regulation of p53, p21/Cip1 to prevent DNA damage and facilitate DNA repair in hairless mice(3).

#Fennel Improves the Levels of Blood #Cholesterols, According to Studies

Scientists may have found a natural whole food with the potential to treat hyperlipidemia without inducing any side effects, some studies suggested.

Cholesterol, the natural ingredient produced in the liver plays an important role in stimulating the production of vitamin D, steroid hormones, and bile acids and maintaining healthy cell membranes.
Hyperlipidemia is a condition characterized by abnormally elevated levels of cholesterol in the plasma of the blood.

The causes of hyperlipidemia are unknown. However, an unhealthy diet with a lot of unsaturated fat and trans fat is associated with the early onset of the disease. Foods such as cheese egg yolks fried and processed foods and red meat have been found to increase blood cholesterol, and long-term intake of such foods in great amount have been found to increase the risk of hyperlipidemia, epidemiological studies found.

The Beni-Suef University, in the study to determine the development of obesity in response to a high-fat diet (HFD) an Egyptian Herbal mixture formulation (HMF), including fennel, showed, "feeding an HFD diet significantly increased final body weight, triglycerides (TG), total cholesterol, & LDL concentration compared with controls, while significantly decreasing HD".

Hyperlipidemia is a lifelong condition that is treatable.

Dr. Robert B. Kelly at the Fairview Hospital/Cleveland Clinic Family Medicine Residency Program in the article "The most beneficial changes result from reducing intake of saturated and trans fats; increasing intake of polyunsaturated and monounsaturated fats; fortifying foods with plant stanols or sterols; isocalorically adding tree nuts to the diet; consuming one or two alcoholic drinks per day; and adopting a Portfolio, Mediterranean, low-carbohydrate, or low-fat diet. Smaller but still beneficial effects result from reducing intake of dietary cholesterol, increasing intake of soluble fiber and soy protein, and eating fatty marine fish or taking marine-derived omega-3 fatty acid supplements".



Therefore changing the diet pattern and engaging in regular physical exercise are the fundamental
options that not only can prevent the onset of the condition, particularly in people who are at a higher risk of hyperlipidemia but also treat high blood cholesterol in patients who already have the condition.

Fennel (Foeniculum vulgare) is a plant species of a genus, belonging to Apiaceae (Umbelliferae), native to the Mediterranean.

The plant has been used in traditional and herbal medicine as a warming, carminative, antispasmodic, antidepressant agent to stimulate the appetite, ease indigestion, soothe coughing, reduce intestinal spasms, regulate the menstrual cycle, and relieve PMS.

In the study to examine the effect of the aqueous extract of fennel in the reduced risk of hyperlipidemia, researchers conducted an experiment in Triton WR-1339-induced hyperlipidemia in mice.

Mice treated with the extract at different doses display a significant reduction of cholesterol, triglycerides, LDL-cholesterol, and apolipoprotein-B decreased by 40%, 23%, 61%, and 61%, respectively.

Further analysis, the application also increased the levels of HDL-cholesterol and apolipoprotein A1 by 85% and 58%, respectively

The Mashhad University of Medical Sciences also conducted a study to assess the effects of Fennel (Foeniculum vulgare Mill.) on lipid profiles, in sixty eligible postmenopausal women, randomly assigned into the fennel and placebo groups.

Measurement of total blood cholesterol, cholesterol fractions, and triglycerides was tested at the baseline, and after a three-month follow-up.

At the end of 3 months, blood profiles withdrawn from the women suggested
* There are no significant difference in triglyceride (P = 0.679), total cholesterol (P = 0.103), low-density lipoprotein cholesterol(LDL-C; P = 0.146) and high-density lipoprotein cholesterol (HDL-C; P = 0.266) levels between the two groups.

* Fennel treated group demonstrates a significant borderline improvement for HDL-C and triglyceride, compared to the placebo group.

In additional differentiation of the process of the double-blind, randomized, and placebo-controlled trial, researchers suggested that longer durations, higher doses, and larger sample sizes are recommended to validate the results.

These results indicated that the intake of foods for the treatment of conditions or diseases must be on a long-term basis. The time of 3 months may be too short for foods to exert their potential effects.


Taken together, fennel may be considered a functional food for the prevention and treatment of hyperlipidemia. Intake of fennel should be increased gradually for the food to reach its antihyperlipidemic stage, a longer duration may be needed. In other words, please be patient.

There are many risk factors that liver problems. Most cases of liver disease are diagnosed in the advanced stage, it takes years to develop.

There is no single cause of liver disease. Researchers suggested that viruses such as hepatitis A, B, and C, long-term excessive alcohol drinking, and genetic mutation inherited from the parents are the major risk factors associated with the early onset of the diseases.

Some researchers suggested overweight and obese and congenital diseases such as Hemochromatosis and Wilson’s disease also are prevalent risk factors for liver damage and diseases.

Broccoli is a mustard/cabbage plant, belonging to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of liver diseases, researchers examined the effect of combined therapy with Indian mustard and broccoli on rats' liver-induced hepatotoxicity by long-term vincristine (VCR)

According to the results of rats selected for the study induced liver toxicity by
* Vincristine (VCR) treated group showed signs of hepatotoxicity, including altered liver architecture and higher serum levels of liver stress markers ALT and AST.

* Vincristine (VCR) treated group also showed an impaired antioxidant potential and higher mRNA levels of IL-12 and IL-4, which are markers of apoptosis in rat livers.

In other words, VCR treatment-induced hepatocyte apoptosis by stimulating the levels of apoptotic proteins and pathways.

Most importantly, the treatment of individual administration of mustard or broccoli partially ameliorated all these responses, compared to maximum improvement in the combined therapy of both extracts.

Based on the results, researchers wrote, " Thus, the long-term administration of VCR is hepatotoxic and induces apoptosis; however, the combined therapy of both extracts mitigated these effects"
Taken altogether, broccoli may be considered a liver protective supplement, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#6-Gingerol Found in #Ginger Significantly Reduces Cell Viability of Human ColonCancer Cells, Reserachers Suggest

Scientists may have found a kitchen spice that has the potential to treat colon cancer, according to studies.
Colon cancer is a medicinal condition associated with irregular cell growth in the colon tissues.

According to the statistics provided by Stat-Canada, 1 in 16 Canadian men and 1 in 18 women will develop colorectal cancer over their lifetime. Cancer is the third leading cause of death behind prostate and lung cancer.

Every year, approximately 26,,000 Canadians are diagnosed with colorectal cancer, representing 13% of all new cancer cases.

Colon cancer is a slow-growth cancer, therefore, most colon cancer is detected in the later stage after experiencing symptoms of blood in your stool or an ongoing change in bowel habits. In other words, only 39% of colorectal cancer is detected in the early stage.

The 5-year survival rate of localized-stage colorectal cancer is 90%.


The cause of colon cancer is a result of the alternation of cell DNA. Researchers do not why? people with the same health conditions and family history, are susceptible to the onset of the disease, while others do not.

Although there are many factors associated with the onset of colon cancer, aging may be one of the most prevalent risks. It probably is due to the disease taken over 10 to 15 years before the patients experience some related symptoms. In other words, according to epidemiological studies, the majority of colorectal cancers occur in people older than 50.

"70% of colorectal cancer is diagnosed in patients over 65. Evaluation of the influence of age and comorbidity on survival and cause of death in a non-selected population".

And, "In colon cancer patients age and comorbidity predict survival. This represents possible bias or a reduced survival benefit of treatment, and is an indication that colon cancer is not the prognosis-defining illness in the majority of patients".

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers on the finding of a bioactive compound to replace the chemo medication which induces serious side effects to cancer patients examined 6-Gingerol, a natural component of ginger, anti-colon cancer activity.

Injection of 6-gingerol treatment significantly reduced the cell viability of human colon cancer cell, LoVo, in a dose-dependent manner.

Furthermore, 6-gingerol induced significant G2/M phase arrest and had a slight influence on the sub-G1 phase in LoVo cells, according to the flow cytometric analysis and observed by the reduced levels of a protein involved cancer cell cycle.

Moreover, 6-gingerol also induced cancer cell apoptosis by elevating the functions of levels of tumor-suppressive genes and exhibiting cytotoxicity through elevated intracellular reactive oxygen species (ROS).

These results strongly suggested that 6-gingerol should be beneficial to the treatment of colon cancer, according to Dr. Lin CB and colleagues.

In order to understand more the effect of ginger on colon cancer, researchers investigated a novel prodrug of aspirin, [6]-gingerol aspirinate (GAS) against colon cancer.

GAS exerted enhanced anti-cancer properties in vitro and superior gastroprotective effects in mice. by suppressing the enzymes involved mediation of tumor repopulation.

Importantly, GAS was also able to survive stomach acid and decompose in intestinal linings or after absorption simultaneously release ASA and [6]-gingerol.

Dr. Yingdong Zhu, the lead scientist said in the final report, "Our results demonstrate the enhanced anticancer properties along with gastroprotective effects of GAS, suggesting that GAS can be a therapeutic equivalent for ASA in inflammatory and proliferative diseases without the deleterious effects on stomach mucosa".

Taken altogether, ginger processed as a highly active bio-compound may be considered a functional remedy for the prevention and combined with primary medicine for the treatment of colon cancer with no side effects, pending the confirmation of large sample size and multicenter human study.
