#Quercetin Isolated from #Onion Inhibits Parameters Associated with the Onset of #Oralcancer, Researchers Say

Oral cancer is a medical condition of malignant cell growth in the tissue located in the oral cavity, including the tissue of the lips or the tongue, cheek lining, floor of the mouth, gums (gingiva), and the roof of the mouth (palate).
The most common types of oral cancer are
* Squamous cell carcinoma
Squamous cell carcinoma is one of the most common types of oral cancer accounting for over 90% of all cases and arises from the squamous cell layer in the lining of the oral cavity and oropharynx.

* Verrucous carcinoma
The low-grade and rarely spread verrucous carcinoma accounts for only approximately 5% of all cases. The cancer can spread deeply into the surrounding tissue.

* Minor salivary gland cancers
The cancer originated from the salivary gland and accounted for less than 1% of all cases.

According to the statistics, approximately, 53,000 Americans will be diagnosed with oral or oropharyngeal cancer in 2019.

The disease also causes the death of over 9,750 patients, roughly 1 person per hour, 24 hours per day.

The grades of oral cancer are depending on the tendency of spreading. Low-grade cancers usually grow more slowly and are less likely to spread while high-grade cancer indicates otherwise.

The 5-year survival rate of the localized stage of oral cancer is 84%. However, If cancer has spread to surrounding tissues or organs and/or the regional lymph nodes, the overall 5-year survival rate is 65%.

The survival rate is even lower (39%) If cancer has spread to a distant part of the body,

The onion is a plant in the genus Allium, belongings to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild, or sweet.

In the urgency to find a potential compound for the treatment of oral cancer, researchers investigated the potential effects of quercetin, a principal flavonoid compound in onions' anti-oral cancer activities.

On SAS human oral cancer cells, quercetin significantly reduced wound closure of SAS cells in culture plates after 12- and 24-h treatments.

Quercetin inhibited the expression and activity of matrix metalloproteinase (MMP)-2 that promotes cancer cell growth and proliferation and (MMP)-9 is involved in cancer cells' angiogenesis and metastasis.

Additionally, quercetin also inhibited the proteins associated with the promotion of forming new blood vessels in SAS human oral cancer cells.


Furthermore, quercetin also blocked the expression of enzymes associated with the pathogenesis of many solid tumors through its inflammatory activation.
Moreover, proteins involved in cellular interactions with the extracellular matrix that cause tumor initiation, progression, and metastasis were also inhibited by the injection of quercetin.

More importantly, quercetin also exhibited the gene expression of tumor suppression such as P53 and apoptosis.

In other words, quercetin protects the oral cells by blocking the proteins involved in the signaling expression of tumor initiation, progression, and metastasis such as Ras homolog gene family, member A (RhoA) and protein kinase C (PKC).

Taken altogether, onion may be considered a remedy for the prevention and treatment of oral cancer, pending the confirmation of the larger sample size and multicenter human study.

Lung cancer is a medical condition characterized by cell growth disorderly and uncontrollably of lung tissues. At the late stage, the cancerous cells may travel a distance from the original site to infect other organs and tissues.
In the study to investigate the anti-lung cancer effect of Epigallocatechin-3-gallate (EGCg), the major polyphenolic compound present in green tea, after taking into account other confounders, researchers at the joint study lead by the Kyushu University of Health & Welfare, suggested that

1. The phytochemical-induced apoptosis of lung cancer through suppression of cell division, in cell culture
2. In  A549 human non-small-cell lung cancer cell line, EGCg inhibited cancer cell proliferation by interfering with mRNA expression in the modulation of apoptotic cell death.

3. Additionally, application of 100 μM EGCg for 24 h. exhibited cancerous morphological changes, thus reducing cell proliferation.

The study also revealed that the efficacy of EGCg inhibited B-cell lymphoma-extra large (Bcl-xL) through the expression of mRNA and suppression of genes in related to cell death.


Further analysis of Epigallocatechin-3-gallate (EGCg) in inhibited gene expression in risk of lung cancer by binding HIF-1α, in promoted cell proliferation and anchorage-independent growth, researchers launched a study to elucidate the role of microRNA in the EGCG inhibition of tobacco carcinogen-induced lung tumors in A/J mice found that 26 potential targeted genes of the 21 microRNAs. with the mRNA expression profiles may have some implications for increased lung cancer risk.

Interestingly, the application of EGCg, indeed, demonstrated a significant effect in improved cell regulation and reduced cell proliferation through many mechanisms, including the regulation of

1. AKT pathway in promoted survival and growth in response to extracellular signals.
2. NF-κB expression in cellular responses to stimuli such as stress, cytokines, free radicals, heavy metals, ultraviolet irradiation and
3. MAP kinases, involved a variety of fundamental cellular processes such as proliferation, differentiation, motility, stress response, apoptosis, and survival.

In other words, Epigallocatechin-3-gallate (EGCg) effect reduced the risk and treatment of lung cancer and is attributed to the inhibition of the expression of RNA in transmitting information to proteins with the function to initiate cancer cell change, including cell proliferation and apoptosis.


The findings also suggested that green tea Epigallocatechin-3-gallate (EGCg) has a strong effect in reducing the risk lung cancer through both expressions of mutating and cancer-related genes.

More importantly, mutated gene expression has been identified as contributing to cancer development and progression, therefore by target the expression of such genes may help scientists in exploring the origins of abnormal gene expression compared to normal cells.

Other researchers suggested that targeting both DNA and RNA data to identify alternative expression of the mutated gene in patients of lung cancer may open the door by offering specific treatment only for such patients

More precisely, by identifying anti-lung genes in cancer cell patients, scientists may prescribe gene-targeting technology, specific medication, and treatment to stop cancer from growing and spreading.


Further differentiation of the effect of green tea extract on lung cancer risk, researchers at the University in Toruń filed the following report
1. Green tea extract catechins demonstrated a significant effect in the inhibition of lung cancer line A549 independent of doses applied and

2. Increased cell and marker of cell degradation.

Acute inflammation is a condition that lasts in the short term caused by an immune system response to the injury.

Symptoms of pain, redness, immobility (loss of function), swelling, and heat may occur within minutes or hours, depending on the size and penetration of the wound.

Overproduction of proinflammatory cytokines induced by the immune response may cause damage or death of the nearby healthy cell, leading to the formation of scars.

The symptoms may disappear sooner in people with a stronger immune system compared to the elderly and the wound is covered quickly in a few days.

However, if the wound is quite large, then some stitches may be needed and the wound recovery will go through 4 overlapped stages, including

* The stage of hemostasis is the process of the wound being closed by clotting.
* The second stage is involved in the production of inflammatory cytokines in the prevention of infection.
* The third stage is proliferation in which the wound is rebuilt with new tissues made up of collagen and extracellular matrix.
and

Betanin is phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

With an aim to discover a potential ingredient for the treatment of diseases associated with inflammation, researchers investigated the effects of the natural pigment betanin on oxidative stress and inflammation in the kidney of paraquat-treated rats.

Selected rats were injected with Paraquat intraperitoneally to induce renal damage, then randomly divided into four groups: a control group, a paraquat group, and two paraquat groups that were treated with betanin at 25 and 100 mg/kg/d three days before and two days after paraquat administration.

The betanin-treated group showed a significant effect in alleviating the paraquat-incurred acute kidney injury, evidenced by histological improvement, reducing serum and urine markers for kidney injury compared to the untreated group.

The bioactive compound also attenuated the inflammation induced by the injection of Paraquat observed by the reduced expression of inflammatory proteins.

Furthermore, betanin also inhibited the kidney's oxidative stress through its antioxidant activity.

Based on the finding, researchers said, "betanin may have a protective effect against paraquat-induced acute kidney damage. The mechanisms of the protection appear to be the inhibition of oxidative stress and inflammation".

Taken altogether, betanin may be considered a supplement for the prevention and treatment of diseases associated with inflammation, pending the confirmation of a larger sample size and multicenter human study.

Intake of betanin in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Memory impairment is a condition of partial or complete loss of memory, depending on the quality of the neuron that has been affected.
Memory impairment other than the physical impact is a progressive disease that can lead to permanent loss of memory due to the gradual loss of neurons in the brain.

The most common risk factors associated with memory loss in the general population are long-term use of certain medications, excessive alcohol drinking, smoking, drug abuse, depression, nutrient deficiency, stroke, and daily stress.

In elderly or aging-induced memory loss is associated with the deterioration of a region of the brain involved in the formation and retrieval of memories, the hippocampus, and the reduced hormones and protein that protect and repair brain cells and stimulate neural growth.

However, some researchers suggested the rise of memory impairment in the elderly probably is correlated to the promotion of the Western diet over the past few decades.

Dr. Scott E. Kanoski, the lead scientist in the review of the risk cognitive impairment in the Western diet consumption wrote, "Intake of saturated fats and simple carbohydrates, two of the primary components of a modern Western diet, is linked with the development of obesity and Alzheimer's Disease".

And, "Western diet intake is associated with cognitive impairment, with a specific emphasis on learning and memory functions that are dependent on the integrity of the hippocampus".


The results clearly stated if you are a Western diet follower, you are at a higher risk of memory impairment compared to healthy diet individuals.

Bilberry is a species of low-growing shrub in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, psoriasis, diabetes, etc.

With an aim to find a potential ingredient for the treatment of memory impairment, researchers investigated the effects of Vaccinium uliginosum L. (bilberry) on the learning and memory impairments induced by amyloid-β protein (AβP) 1-42.

The evaluation included selected ICR Swiss mice divided randomly into 4 groups: the control (Aβ40-1A), control with 5% bilberry group (Aβ40-1B), amyloid β protein 1-42 treated group (Aβ1-42A), and Aβ1-42 with 5% bilberry group (Aβ1-42B).

According to the tested assays, bilberry-added groups showed to decrease in the finding time of the hidden platform, compared to those of escape latencies of AβP to that of Aβ40-1.

Furthermore, the bilberry-added Aβ1-42B group also showed a significant (P<0.05) increase in probe crossing frequency compared to Aβ1-42A.

In the passive avoidance test, bilberry significantly (P<0.05) increased the time of staying in the lighted area compared to AD control.

These results suggested that bilberry improved memory and learning capability in chemically induced Alzheimer's disease in experimental animal models.


In the incidence of degenerative disorders including Alzheimer's disease (AD), rats consumed approximately 3.2 mg kg(-1)day (oral), of the anthocyanins significantly enhanced short-term memory, but not long-term memory in the inhibitory avoidance task.

Moreover, injection of approximately 3.2 mg kg(-1)day (oral), of the anthocyanins also induced an increase in the number of crossings in the first exposure to the open field.

However, in the comparison of the radial maze and elevated plus-maze, lyophilized berries (blueberry, bilberry) improved working memory in the radial maze, with significant differences observed during sessions 1-2 and 4, compared to no effects in the elevated plus-maze task.

The findings strongly indicated the positive effects of the administration of lyophilized Vaccinium berries (blueberry, bilberry) and anthocyanin in the prevention of memory deficits related to AD.

Taken altogether, bilberry-processed abundantly bioactive compound anthocyanin may be considered a functional remedy for the prevention and treatment of memory impairment, pending the validation of a larger sample size and multicenter human study.

Melanoma is a medical condition characterized by irregular cell growth of pigment-containing cells known as melanocytes. At the later stage, the cancerous cells may travel a distance away to infect other healthy organs and tissues.
According to the study, approximately 7,200 Canadians will be diagnosed with melanoma skin cancer.

Green tea, a precious drink processes a number of health benefits known to almost everyone in Asia and the Western world. The ubiquitin-conjugating enzyme(UBC13(E2)) is essential in the activation protein (NF- kappa B) in controls the transcription of DNA, cytokine production, and cell survival.

Furthermore, the phytochemicals also suppressed the TRAF6 E3 ubiquitin ligase activity of the tumor generated by TRAF6 in the regulation of important biological processes, particularly, in cell division and for the survival of the tumors.

Additionally, green tea bioactive EGCG also regulated the gene expression of proteins (IκBα and p-TAK1 e) in modulation and inactivated NF-κB-directed gene expression of inflammation, immunity, cell proliferation, differentiation, and survival in melanoma.


Dr. Zhang J, the lead author said, " EGCG is a novel E3 ubiquitin ligase inhibitor that could be used to target TRAF6 for chemotherapy or the prevention of melanoma".


Other researchers in the focus of Green tea polyphenol (-)-epigallocatechin-3-O-gallate (EGCG) effect in attenuated and treatment of melanoma in the gene implication via MicroRNAs (miRNAs, non-coding RNAs involved in various biological processes by regulating their target genes) also indicated that

Green tea polyphenol (-)-epigallocatechin-3-O-gallate (EGCG) inhibited melanoma tumor growth by activating 67-kDa laminin receptor (67LR) signaling which has a function in down-regulated tumor cell proliferation and tumor formation by inducing apoptosis.

The bioactive compound also up-regulated miRNA-let-7b expression in induced tumor suppressor function in decreased function of 67LR in enhanced invasive and metastatic melanoma cells.

Indeed, green tea EGCG also displayed a significant effect in promoting the function of let-7b in altering the traits, of malignant cells in melanoma by regulating the expression of HMGA2.with action as a DNA transcriptional regulator.


Green tea EGCG. also, induced expression of let-7b in inhibitions of oncoproteins and activation of tumor suppressor via tumor-over expressed 67LR in the activation of PP2A, a tumor suppressor. through adenylate cyclase/cAMP pathway in cell communication.

In comparing the effects of TNF-related apoptosis-inducing ligand (TRAIL), a protein with function in induced tumor cells apoptosis. and green tea bioactive EGCG for the treatment of groups of melanoma.
In the A375 melanoma cell line, the TRAIL application group demonstrated a strong effect in inhibition with an apoptosis rate was 11.8% at a dose of 150ng/mL compared to the green tea EGCG treatment group of 5%-7 %.

Interestingly, in combined TRAIL and green tea EGCG treatment groups, the inhibited rate increased sufficiently to 48.9 - 59.1%

Further analysis also found that EGCG's effectiveness in the treatment of melanoma was attributed to the activity of caspase-3 rather than caspase-8 in the mediation of programmed cell death (apoptosis).

The findings suggest that by blocking the sequence of gene expression of the tumor, green tea polyphenol (-)-epigallocatechin-3-O-gallate (EGCG) may be considered an adjunct combined with TRAIL for the prevention and treatment of melanoma.

#Coffee and #CoffeeCaffeine Process AntiGenotoxic Effects Against Cells' DNA Mutation in Vivos, Researchers Say

Coffee and coffee caffeine may have a profound and positive effect in reducing the risk of genotoxicity, caused by environmental toxins, a coffee production company opinionated.

Coffee, a popular and social beverage all over the world, particularly in the West, is a drink made from roasted beans from the Coffea plant, native to tropical Africa and Madagascar.

In rat liver and primary hepatocytes, regular intake of coffee daily illustrated an enormous effect on cellular antioxidant defenses through regulating exhibited enzymes involved in mRNA and protein expression.

The study also suggested that coffee and coffee caffeine through the expression of detoxifying enzymes GSTs and AKR(in cellular detoxification) also demonstrated significant activity in reduced risk of genotoxicity and cytotoxicity caused by the injection of aflatoxin B1 (AFB1).

Dr. Cavin C, the lead author said, "The coffee-mediated stimulation of the Nrf2-ARE pathway resulting in increased endogenous defense mechanisms against electrophilic but also oxidative insults further support that coffee may be associated with protection against various types of chemical stresses".

Other, in the evaluated L5178Y mouse lymphoma cell line to assess the modulatory effects of coffee against genotoxicity induced by N-methyl-N-nitro-N-nitrosoguanidine (MNNG), researchers found that
1. 3 doses of instant coffee caffeine injection displayed a decrease in genotoxicity following the co-treatment of mouse lymphoma cells.

2. Coffee caffeine pre-treatment or co-treatment exhibited significant antigenotoxic effects against MNNG(an environmental carcinogen that causes DNA mutation) expression.

3. Both caffeinated and decaffeinated instant coffee demonstrated a similar effect against genotoxicity in the experiment samples.

Dr. Abraham SK, the lead author said, " Results from three or five independent experiments demonstrated significant protective effects of caffeinated instant coffee against MNNG-induced DNA damage in the comet assay, mutation at the Tk locus, and chromosomal damage in the cytokinesis-block micronucleus test".

In support of the above analysis, the Swiss Federal Institute of Technology (ETH), in the study of the protective effects of coffee against somatic mutation and mitotic recombination induced by chemo drugs, cyclophosphamide (CPH)(, mitomycin C (MMC) and urethane (URE), using standard (ST) and high bioactivation (HB) crosses of the wing spot test in Drosophila melanogaster assays, suggested that in dose depending, coffee injection showed a strong effect on the genotoxicity of URE in the ST and the genetically susceptible HB cross as well as the genotoxicity caused by MMC application.

Dr. Abraham SK, the lead researcher said, "The magnitude of the protective effects of coffee against these three genotoxins was independent of the genotype of the larvae used for treatment, that is it was independent of the bioactivation capacity of these larvae".

The collective information showed definitive support for the use of coffee and coffee caffeine as a functional food in reduced risk against genotoxic expression, including side effects of chemo medicine used in the treatment of multi types of cancers.

Chemotherapy is a type of therapy that uses of drugs to destroy cancer cells by keeping them from proliferation and migration.

In 2017, an estimated 15,270 children and adolescents ages 0 to 19 were diagnosed with cancer, and 1,790 died of the disease.

Conventionally, chemo drugs used for chemotherapy are powerful for the treatment of cancers, however, they also can cause damage to nearby healthy cells, leading to serious adverse effects.

Cancer is a class of chronic diseases caused by cell growth and multiplies disorderedly and uncontrollably in the body.

Depending on the type and grade of cancer, some cancers can be progressively worse and damage other healthy tissues. Sometimes cancers can spread to other tissues and organs in the body via lymph or blood.

Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.

On finding a potential compound that processes chemo-activity, researchers examined the effect of Indicaxanthin as a natural chemo-active and chemopreventive agent.

According to the tested analysis, the bioactive compound showed significant activity in neuromodulation and inflammatory regulation that prevent cellular damage.
