#Healthyfood #Carrots Inhibit the Onset of Inflammatory Related Diseases Naturally, Researchers Suggest

By Kyle J. Norton

Carrots may have a profound and positive effect on reduced risk, progression, and treatment of inflammatory-related diseases, some scientists suggested.
The results of the studies were conducted by a number of best institutes including the National Institute of Nutrition, Jamai Osmania, and published in online medical literature.

Inflammation is a natural response of the immune system to protect our body against foreign induction of infection to the site of injury or damage.

However, overexpression of proinflammatory cytokines is also associated to induce local cell death and the formation of local scars.

Carrot, a root vegetable with orange color is a subspecies of Daucus carota, belongings to the family Apiaceae, native to Asia and Europe.

In the investigation of the function of adipose tissue in the induction of various metabolic diseases including insulin resistance, type 2 diabetes, and non-alcoholic fatty liver disease (NAFLD) involved initiation of inflammatory development, researchers at the National Institute of Nutrition, Jamai Osmania launched a study to examine the effect of carrot intake and inflammatory status in male weanling Wistar rats.

The tested group of rats were divided into four groups and fed either a control or high fructose diet of AIN-93G composition with or without carrot juice ingestion for an 8-week period.

AIN-93G was the formulation published by the American Institute of Nutrition (AIN) committee in 1993 improving the laboratory standard, with an aim to accommodate the increased nutritional demands of rat or mouse growth.

At the end of the experiment, researchers indicated that
* Carrot juice displays an insignificant effect in adiposity and cell size of the visceral fat depot as well as the retroperitoneal white adipose tissue (RPWAT).

The role of white adipose tissue (WAT) is to store excess fat for future use as sources of energy.

* In a high fructose diet, the application of carrot juice exerts a strong activity in inhibited elevation of plasma free fatty acid (FFA), macrophage chemoattractant protein 1 (MCP1) (P ≤ 0.01), and high sensitive C-reactive protein (hs-CRP) (P ≤ 0.05) levels.

Macrophages chemoattractant protein-1 (MCP-1/CCL2) is a chemokine with function in regulated migration and infiltration of macrophages, the mobile white blood cells.

Levels of highly sensitive C-reactive protein (hsCRP) are associated with the inflammation of blood vessel walls.

Further analysis suggested that carrot juice decreased overexpression of free cholesterol and immune response through its antioxidant in inhibited the elevation of ROS in the production of pro-inflammatory cytokines.

In other words, carrot juice activated the production of anti-inflammatory factors in reduced overexpression of inflammatory cytokines in facilitated onset and progression of inflammatory-related diseases, including insulin resistance, type 2 diabetes, and non-alcoholic fatty liver disease (NAFLD).

Moreover, in the additional differentiation of the effect of root vegetables in Immune-inflammatory signaling and metabolic effects, researchers at the joint study led by Agriculture and Agri-Food Canada conducted a study to assess the bioaccessibility and bioavailability of purple carrot and potato-derived anthocyanins and the molecular mechanisms to ameliorate cellular inflammation in mono- and co-culture cell models.

In vitro gastrointestinal model, the application of bioaccessibility of 44.62 and 71.8% for anthocyanins of purple carrot and potato demonstrated strong activity in the inhibition of cellular inflammation in Caco-2 cells.

According to the assay, the effectiveness of anthocyanins injection was found to transport across a transmembrane cell model.

The inhibition of anthocyanins during the transportation also reduced the expression of pro-inflammatory factors of IL-8 and TNF-α secretion, and expression of pro-inflammatory cytokines by blocking NF-κB, and MAPK, the pro-inflammatory proteins in mediated inflammatory cellular signaling cascades.

Interleukin 8 is a chemokine produced by macrophages and other cell types in initiated production of inflammatory expression.

Tumor necrosis factor is a cell-signaling protein (cytokine) involved in systemic inflammation in acute phase infection of tissue injury and damage.

Dr. Zhang H, the lead scientist after taking into account other co and confounders said, "Anthocyanins from purple carrots and potatoes possess a promising anti-inflammatory effect in the model gut system" and "They (Anthocyanins) can be absorbed and act differently but are in general beneficial for inflammation-mediated diseases".

Taken together, carrot juice containing a high amount of anthocyanins may be considered as a functional food that prevented the onset of inflammatory-related diseases.

#Enzyme #Bromelain Shows A Significant Effect on #SkinHealing, Researchers Say

By Kyle J. Norton

Skin healing is a natural process to reconstruct the skin back to its originated stage after skin damage.
Skin is a large organ in the human body which only protects our body against foreign invaders but also serves as a third detoxed system after the liver and kidney.

Skin healing is depending on the size of the wound, the small size can be healed quickly without using any medication. However, the large size wound in most cases is required stitches.

Similar to wound healing, at the acute phase of infection, the macrophages of the immune first line of defense stimulate the blood clot to cover the wound or cut and production of proinflammatory cytokines to destroy foreign invaders.

In this phase, patients may experience symptoms of skin redness, tenderness, and swelling around the wound or cut.

Depending on the size of a skin wound, the next overlap stage is broken blood vessels in the wound is repaired and new tissue is grown followed by the creation of collagen, the fibers that form the foundation for new tissue.

This process continues until the skin is stronger or back to the original form.

In the acute phase of infection, overproduction of pro-inflammatory cytokines induced by immune response may cause damage to healthy cells, leading to scar forming.

Additionally, any interruption of these processes may induce the formation of scars.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

In finding a potential compound for speeding up wound healing, researchers compared the structural integrity of bromelain (debridement enzyme) in the healing process.

The study included a combinational multiphasic delivery system that delivers biomolecules of bromelain through the fabrication of coaxial electrospinning of different biocompatible polymers.

In vitro, the fabricated scaffold delivery system sustained the release of debriding protease and its bioactive components in a timely fashion showing a significant effect on wound healing.

Furthermore, the fabricated scaffold used in debridement enzyme also promoted the wound healing process by its angiogenic potential through in vitro migration of endothelial cells and increased new capillaries from the existing blood vessel in response to wound healing.

Moreover, in vivo, studies also confirm the efficacy of the fabricated scaffold used in the delivery of bromelain.

Based on the findings, researchers said, "Our results, therefore, open up a new avenue for designing a bioactive combinational multiphasic delivery system to enhance wound healing".

In order to assess the role and clinical advantages of enzymatic debridement by a mixture of proteolytic enzymes enriched in Bromelain (Nexobrid®) on wound healing, researchers examined the mutual experience of applying enzymatic debridement in more than 500 adult and pediatric patients by the consensus panelists.

The statement included the answer to questions related to pain management and anesthesia, the timing of application, technique of application, after-intervention care, skin grafting after enzymatic debridement, blood loss, training strategies and learning curve and areas of future research needs.

According to the statements returned by the participant, 68 consensus statements, enzymatic debridement used showed a higher degree of consensus with a unanimous consensus in 88.2% of statements, and the lowest degree of consensus of 70% in only 3 statements.

Taken altogether, bromelain used alone or combined with the multiphasic delivery system may be considered supplements for speedup wound healing, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthyfood #Carrot's Bioactive Compounds Exert Cytotoxic Activities That Inhibit #BreastCancer Cell Motility and Metastasis

By Kyle J. Norton

Carrots may have a profound and positive effect on reduced risk, progression, and treatment of breast cancer, some scientists suggested.
Breast cancer is a condition characterized by cell growth disorderly and uncontrollably in breast tissue. At the later stage, the cancerous cells may infect other healthy tissue and organ a distance away from the original site.

Carrot, a root vegetable with orange color is a subspecies of Daucus carota, belongings to the family Apiaceae, native to Asia and Europe.

In the review of carrot fractions' anticancer activities in vitro and in vivo, researchers found that pentane/diethyl ether (50:50) isolated from carrots express a significant anti-cancer activity against various tumors, including lung, skin, breast, and glioblastoma cancer cells.

Further analysis and application of the ether demonstrated a pronounced inhibition of mobility of the above 4 cancer cell lines through a decrease in cancer cell invasion and increased cell adhesion.

Also, pentane/diethyl ether (50:50) decreased activation of the Rho family of GTPases, a family of small GTP-binding proteins with a function in initiated cancer mobility involved in cell migration, transcription, and proliferation and hyperactivation of Cdc42 with function in stimulating the development, progression, and metastasis of breast cancer.

Overexpression of levels of Cdc42 is found in patients with breast cancer tissue compared to healthy women.

Dr. Zgheib P, after taking into account another confounder, said, " the treatment (of pentane/diethyl ether ) induced decrease in cell motility" and "(These results) demonstrates a crucial effect of the DC pentane/diethyl ether fraction on cancer cell motility and metastasis".

Additionally, in human breast adenocarcinoma cell lines MDA-MB-231 and MCF-7, administration of Daucus carota oil extract (DCOE) fractions, namely, the pentane fraction (F1) and 1:1 pentane: diethyl ether fraction (F2) exerted potential anti both cancer lines through induction of cytotoxicity.

According to the Flow cytometric assay, both factions of carrot induced cell cycle arrest in the sub-G1 phase and increased apoptotic cell death.

In Western blot, F1 and F2 also promoted the increase in apoptosis through an increase in BAX in suppressed the tumor proliferation and decrease in Bcl-2 levels in promoted anti-breast cancer apoptosis and caspase-3 and poly ADP ribose polymerase in activated death protease through cell cycle arrest.

Also, both factions also increased chromatin condensation in facilitated cell apoptosis observed by chromatin activity of cancer cells undergoing of phase change from a heterogeneous, genetically active network to an inert, highly condensed form.

Interestingly, injection of F1 and F2 in MDA-MB-231 cells did not significantly reduce the level of phosphorylated Erk and phosphorylated Akt protein in the induction of cancer cellular processes involved in cancer cell proliferation, differentiation, and survival.

In other words, F1 and F2 inhibited the expression and proliferation of MDA-MB-231 cells by mediating a variety of biological responses including cell growth, proliferation, and survival through the phosphorylated Erk pathway without alternated expression of phosphorylated Akt proteins.

Moreover, observation of combined treatment with wortmannin, a potent PI3K inhibitor, and F1 or F2 fraction revealed that the two drugs work on different pathways in the synergistically inhibitory effect on cell survival.

The above differentiation was supported by the National Institute of Environmental Health Sciences in estimating nutrients intake in nine questions on food and supplement in a population-based case-control study of breast cancer risk conducted in Maine, Massachusetts, New Hampshire, and Wisconsin in 1988-1991 with data for 3543 cases and 9406 controls.

According to the results, eating carrots or spinach more than twice weekly was associated with a relative odds ratio of 0.56 compared with no intake.

However, these data did not distinguish among several potential explanations for the protective association observed between the intake of carrots and spinach and the risk of breast cancer but consistent with a diet rich in these foods having a modest protective effect.

The result findings strongly suggested that long-term intake of carrots in moderate amounts may have substantial implications in the prevention and treatment of breast cancer. But Intake of carrot supplements should be taken with care to prevent acute liver toxicity.

#Herbal #BilberryExtracts in the Inhibition and Treatment of #DryEye, Scientists Discover

By Kyle J. Norton

In seeking natural therapy with no side effects, scientists may have found a herbal fruit for the cure of dry eye, according to studies.

A dry eye is a condition characterized by a lack of quality tears to lubricate and nourish the eye for clear vision.

In other words, dry eye is a condition caused by an imbalance of tear production and drainage or low-quality tear production. The condition is very common in the elderly.

There are certain risk factors that may affect the levels of quality of tears, such as aging (most seniors age 65 and over have experience with dry eye), gender (the risk of dry eye is higher in women due to the change of hormone as a result of pregnancy and menopause), medications and long term of use of contact lens.

According to the statistics, in the United States, dry eye affects over 4.88 million Americans age 50 and older. The syndrome is most common in the population of middle-aged or older.

Although it is rare, untreated dry eye or serious dry eye syndrome has been associated with the risk of infection, leading to scarring, thinning, and even perforation of the cornea.

People with dry eyes may experience symptoms of burning or scratchy sensation, stringy mucus in the eyes, eye redness, and sensitivity to light.

Some people with severe conditions may have difficulty with nighttime driving.

If you have experienced some of the aforementioned symptoms, please check with your doctor to rule out the possibility of preventive measures.

Bilberry is a species of low-growing shrub in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, psoriasis, diabetes, etc.

Researchers on finding a natural ingredient for the treatment of dry eye, a chronic disease of lachrymal fluid and cornea-conjunctival epithelium examined the bioavailability of a standardized bilberry extract in an animal model.

The study included 22 otherwise healthy subjects suffering from dry eye symptoms who were enrolled and randomly assigned to receive the more bioavailable bilberry extract or placebo.

Out of 22 participants,11 subjects in the bilberry extract (Mirtoselect®) group and 10 subjects in the placebo group completed the clinical study.

According to the results of the assay analysis, bilberry extract showed a significant improvement in the volume of tear secretion compared to no change in the placebo group.

Mirtoselect® also more effective in subjects with a higher tendency of dry eye.

The efficacy of bilberry in the treatment of dry eye was attributed to the stimulation of the production of antioxidant enzymes in the host and suppression of the overexpression of free radicals found in the dry eye participants.

Dr. Riva A the lead scientist suggested that bilberry extract can improve tear secretion and plasmatic antioxidant potential in subjects suffering from DED symptoms.

Taken altogether, bilberry-processed abundantly bioactive compounds may be considered a natural remedy for the treatment of dry eye with no side effects, pending the validation of a larger sample size and multicenter human study.

#Antioxidant #Sulforaphane (SFN) Found in #CollardGreens Builds-up #BoneDensity Against #Osteoporosis, Researchers Find

By Kyle J. Norton

Bone density is also known as bone mineral density associated with the amount of bone minerals in bone tissue. The concept of bone density has been used to measure the mass of minerals per volume of bone to identify people in risk and onset of osteoporosis.

Osteoarthritis (OA) is a common chronic inflammatory joint disease characterized by the breaking down of joint cartilage and the underlying bone, mostly in women in menopause and older adult, leading to symptoms of joint pain, tenderness, and lack of flexibility and stiffness.

Some osteoporotic patients may also experience symptoms of grating sensation and bone spurs.

Aging, genetic preposition, overweight, and obesity, injury, and overuse of the joints are the most common risk factor associated with the onset of the disease. However, occupation with the repetitive movements of the joint has been found to affect people in the industrial world.

Interestingly, osteoarthritis symptoms can come and go throughout the day and vary from person to person. Weather change has been one of the most complaints that trigger the symptoms of OA in the elderly.

According to the statistics, approximately, more than 200 million people are suffering from osteoporosis, including 44 million Americans with either osteoporosis or low bone mass, and 1 in 3 women over the age of 50 years and 1 in 5 men will experience osteoporotic fractures in their lifetime.

Dr. Jakob F, the lead scientist in the study of the risk of osteoporosis in the aging population wrote, "Age is a high-risk factor for osteoporosis. Vitamin D insufficiency and reduced calcium absorption are common in the elderly. Loss of bone and muscle development in a vicious circle of immobilization caused by underlying diseases. In addition, deficits in cognition and coordination promote falls and fragility fractures".

And, "Specific medication is indicated if the 10-year fracture risk exceeds 30 %, wherein women > 75 and in men > 85 years of age bone density measurement can be relinquished".

Collard Green is a species of Brassica Oleracea, belongings to the family Brassicaceae with unknown origin. Today, the plant is cultivated commercially for its thick, slightly bitter edible leaves.

With an aim to find a natural compound for the treatment of osteoporosis, researchers examined the effects of sulforaphane (SFN) on age-related shifts in anabolic and catabolic responses.

According to the tested differentiation, in both normal and ovariectomized mice that were treated with SFN for 5 weeks, sulforaphane (SFN), a naturally occurring in collard greens promotes osteoblast differentiation by epigenetic mechanisms associated with enhancing the production of bone density.

Furthermore, SFN enhanced active DNA demethylation via Tet1 and Tet2 involved in the production of bone cells against the pathogenesis of osteoporosis and promoted pre-osteoblast differentiation by enhancing extracellular matrix mineralization and the expression of osteoblastic markers in the process of making new bone cells.

Where Tet loss has been associated with abnormal osteoclast function.

Moreover, SFN inhibited the expression of the osteoclast activator RANKL in osteocytes associated with bone reabsorption and induced apoptosis in pre-osteoclastic cells through the several mechanisms involved in bone degenerative pathology.

The results suggested that these mechanistic effects not only lower the rate of bone reabsorption but also accelerate the production of higher bone volume.

More importantly, SFN protected the bone density against the shifts in mineral density distribution observed by quantitative back-scattered electron imaging (qBEI).

Based on the findings, researchers concluded, "Our data indicate that the food-derived compound SFN epigenetically stimulates osteoblast activity and diminishes osteoclast bone resorption shifting the balance of bone homeostasis and favoring bone acquisition and/or mitigation of bone resorption in vivo".

Taken altogether, collard green processed a higher amount of sulforaphane (SFN) may be considered supplements for the prevention and treatment of osteoporosis, pending the confirmation of the larger sample size and multicenter human study.

Intake of sulforaphane (SFN) in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

# RetinoicAcidAmide Found in #Healthyfood #Carrots In Attenuating the Risk of #LungCancer in Vitros, Researchers Suggest

By Kyle J. Norton

Carrots may have a profound and positive effect on reduced risk, progression, and treatment of lung cancer, some scientists suggested.

Lung cancer is a condition characterized by cell growth disorderly and uncontrollably in lung tissue. At the later stage, the cancerous cells may infect other healthy tissue and organ a distance away from the original site.

Carrot, a root vegetable with orange color is a subspecies of Daucus carota, belongings to the family Apiaceae, native to Asia and Europe.

In the evaluation of a total of 417 lung cancer cases and 849 controls of a hospital-based case-control with 'cases' of lung cancer patients diagnosed during the period 1979/80 at seven hospitals in the Lombardy region by interviews on their life-long tobacco usage and their current intake of four food items rich in retinol or carotene, the researcher found that risk of lung cancer increased 3-4 fold in current smokers who did not consume carrots in compared with those who ate them more than once a week.

The odd ratio of carrot consumption reduced significantly and linearly to the risk of lung cancer.
However, among ex-smokers or non-smokers, the intake of carrots did not offer a decrease in lung cancer risk.

Dr. Pisani P the lead author, after taking into account another cofounder such as vegetables said, "The effect of carrots is independent of histological type of lung cancer while the effect of green vegetables was confined to epidermoid carcinomas".

Further analysis of the antiproliferative effect of retinoic acid amide found abundantly in carrots suggested that retinoic acid amide at a dose of 30 μM expressed a significant activity in the inhibition of lung cancer cell lines and clonogenic growth.

Application of retinoic acid amide also induced cancer cell apoptosis in the G2/M-phase cell cycle arrest through inhibition of JAK2 in cancer cell line in signaling cell functions by transmitting messages between cells in initiated growth and proliferation; inhibited STAT3, and STAT5 in cancer cell in expression of inflammatory cytokine to fuel the cancer development and expansion and level of p21WAF1, a key mediator of p53-dependent in cell cycle arrest with function as a tumor suppressor in cancer

In the meantime, the application of retinoic acid amide stimulated the decreased cyclin A, and cyclin B1 in the induction of cell mitosis, and Bcl-XL in the expression of anti-apoptotic activity in lung cancer cell line by preventing the release of mitochondrial contents such as cytochrome c.

Additionally, researchers also discovered that the compound injection also increases the expression of caspase-3 and caspase-9 in tumors with the function of being responsible for executing cell death following cytochrome c release.

In comparison to those of conventional medicine, Dr. Li HX, the lead author said, "Retinoic acid amide exhibits promising antiproliferative effects against human lung cancer cells in vitro and in vivo and enhances the antiproliferative effect of methotrexate".

The above differentiation was supported by reviewing the 16 studies selected for elevation between lung cancer and vitamin A in humans, conducted by Dr. Palgi A.

According to the study, a high intake of dietary vitamin A has a protective effect against the development of squamous and small cell carcinoma of the lung in smokers and people who consumed carrots daily have a lower risk of lung cancer compared to nonconsumers.

However, due to the limitation of several factors, the researcher postulated, "(A sample size study with) more complete vitamin A indices, examination of the histological type of lung cancer, a large enough pool of both male and female patients for statistical analysis, and proper adjustment for the effects of age, sex, smoking, and socioeconomic status (are necessary for the viability".

The result findings strongly suggested that long-term intake of carrots in moderate amounts may have a substantial implication in the prevention and treatment of lung cancer. But Intake of carrot supplements should be taken with care to prevent acute liver toxicity.

#Bromelain Modulates the #ImmuneSystem For the Prevention and Treatment of Allergies, According to Studies

By Kyle J. Norton

Allergy, also known as an allergic disease is a group of conditions including include hay fever, food allergies, atopic dermatitis, allergic asthma, and anaphylaxis caused by the overreaction of the immune system response to harmless substances in the environment.

The most common symptoms of allergies are sneezing and runny nose, persistent coughing, skin rash, fatigue, gastrointestinal discomforts, headache, and fever.

In serious cases, people affected by allergies may also experience symptoms of shortness of breath.

Believe it or not, food allergies affect 4-6% of children and 4% of adults epidemiologically. According to the statistics, approximately, 32 million Americans have food allergies, including 5.6 million children under age 18.

Also, every year, in the U.S., 200,000 people require emergency medical care for allergic reactions to food.

There are many risk factors associated with the onset of allergies. However, foods, medications, and insect stings are the most common triggers of anaphylaxis, a life-threatening condition.

People who have asthma, a family history of asthma and allergies, and are under 18 years of age are most at risk to develop an allergy and allergic complications.

Treatment of allergies is totally depending on the severity and types of the syndrome. Over-the-counter remedies such as antihistamines that block the effects of histamine, decongestants, and nasal steroid sprays may reduce symptoms in patients with the mild condition.

In a severe case, due to the condition have interfered with the quality of life of the patients, more options may be needed including allergy shots (immunotherapy), please check with your doctors

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural ingredient against allergic symptoms, researchers examined the effects of Bromelain (sBr), a mixture of cysteine proteases from pineapple in the incidence of atopic conditions.

The study included C57BL/6J mice sensitized with OVA/alum and challenged with 7 days of OVA aerosol followed by the sBr 6 mg/kg/0.5 ml or PBS vehicle administered throughout sensitization.

According to the tested assays, throughout the study, sBr treatment through OVA-alum sensitization significantly reduced the development of AAD (BAL eosinophils and lymphocytes).

In other words, sBr prevents the expression of allergic symptoms induced by OVA-specific IgE and OVA TET(+) cells by modulating immune functioning.

In vitro, treatment of sBr significantly reduced CD44, a key receptor involved in lymphocyte infiltration of inflamed tissue in the initiation of the development of airway inflammation in a murine model of allergic asthma.

The results strongly suggested that sBr's processes have anti-inflammatory and antiallergic properties.

Based on the findings, Dr. Secor ER Jr, the lead scientist said, "sBr was shown to reduce allergic sensitization and the generation of AAD upon antigen challenge".

Taken altogether, bromelain may be considered a supplement for the prevention and treatment of allergies, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
