Elecampane (Inula helenium) Exerts Radio-Protective Activity in Vivo

By Kyle J. Norton

Cancer is the class of diseases associated with the growth of the cells disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Compared to all cancers, lung, breast, prostate, and colorectal cancers are the most common cancer in the US.

Some cases of cancer are treated by radiotherapy which uses high doses of radiation to kill cancer cells and shrink tumors.

Radiotherapy has been found to induce certain adverse effects in cancer treatment.
For example, osteoradionecrosis is a condition associated with bone death due to exposure to radiation or radiation treatment.

The death of bone cells is a result of radiation induce-damage of the blood vessel that supplies the nutrients to the bone.

Osteoradionecrosis is a rare side effect occurred sometime after radiation therapy has ended. The complication usually occurs in the lower jaw or mandible.

Radioprotection refers to the protection of people from the harmful effects of exposure to ionizing radiation.

On finding a natural ingredient for the protection against radiation-induced adverse effects, scientists examined the radio-modulatory efficacy of aqueous root extract of I. racemosa.

Elecampane, also known as Horse-heal (Inula helenium) is a perennial composite plant, genus nulla, belonging to Asteraceae, native to Southern Europe, and Asia.

The plant has been used in traditional and herbal medicine in treating bronchitis or emphysema, night sweats, pulmonary tuberculosis, irritating cough, catarrhal discharges, etc.

According to the tested analysis of normal kidney epithelium cells that were treated with the extract (50-200 μg/ml) and exposed to 3 Gy of γ-radiation, while C57BL/6 mice were administered with extract (300-600 mg/kg BW) intraperitoneally prior to exposure to 7.5 Gy of γ-radiation,
* The administration of the extract (30 min and 1 h) prior to radiation exposure improved the NKE cell survival.

* The proteins associated with the production of antioxidant enzymes were significantly induced by the administration of the extract observed by the increase of Nrf2 protein levels at 7 h and HO-1 and NOQ1 protein at 24 h.

* The 30 days whole body survival study also suggested that extract pre-treatment increases survival or delays the onset of radiation-induced mortality as against 70% mortality of 7.5 Gy of γ-radiation.

Based on the results, researchers suggest, "The aqueous extract of roots of I. racemosa enhanced the survival of irradiated NKE cells and rescued C57BL/6 mice against WBI-induced mortality.".

Taken altogether, Dandelion may be considered a remedy for the protection of radio-side effects, pending to the confirmation of the larger sample size and multicenter human study.

Omega-3 Fatty Acids Induce Apoptosis in Mammary Cancer Cells in Vitro

By Kyle J. Norton

Cancer is a class of chronic diseases associated with irregular cell growth in the tissues of the affected organ.

The mammary gland located in the breasts of females is responsible for lactation.

Both males and females have glandular tissue within the breasts; however, in females, the glandular tissue begins to develop after puberty in response to estrogen release.

A mammary tumor is a medical and chronic condition originated from the milk-producing gland.

The mammary gland located in the breasts of females is responsible for lactation.

Breast cancer (malignant breast neoplasm) is cancer that starts in the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.

There are also rare cases that breast cancer starts in other areas of the breast.

In 2010, over 250,000 new cases of breast cancer were expected to be diagnosed in women in the U.S. alone and the risk of getting invasive breast cancer during the lifetime of a woman is 1/8.

Most cases of the mammary cancer are first found in the cell on the surface of the inner lining of the milk-producing gland, before penetrating deeper into the layers.

At the advanced stage, the malignant cancer cells can also travel a distance away from the original site to infect other healthy tissues and organs, leading to secondary metastasis.

Epidemiologically, mammary cancer has been found to spread to the colon and the liver.

Most common risk factors of mammary cancer are similar to those of breast cancer, including, aging, physical inactivity, being obese or overweight, family and personal history, having a dense breast, radiation therapy, pregnancy history, genetic proposition, particularly in women who had used drug diethylstilbestrol (DES).

Conventionally, treatment of mammary gland cancer is totally dependent on the stage and grade of cancer, including surgery, radiation therapy, chemotherapy, or a combination.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of breast cancer, researchers examined the effects of omega 3 on the signaling pathway impacted by syndecan-1 (SDC-1) induced apoptosis in vitro and Vivo.

According to the tested analysis,
* In MCF-7 cells and SK-BR-3 cells, DHA and an SDC-1 ectodomain impaired signaling of the p44/42 mitogen-activated protein kinase (MAPK) pathway associated with mitogens, and growth factors and pro-apoptotic protein to induce apoptosis.

* In Fat-1 mice that is a genetic model able to convert n-6 to n-3 PUFA to result in higher SDC-1 levels in Fat-1 mammary tissue compared with that of wild-type (wt) mice,  which showed a lower level of apoptosis compared to elevated signaling of the p44/42 mitogen-activated protein kinase (MAPK) in MCF-7 cells and SK-BR-3 cells.

* Moreover, SDC-1(-/-) mice that demonstrated markedly higher levels of mitogens, and growth factors and pro-apoptotic protein in mammary gland tissue compared with those of SDC(+/+) mice.

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the prevention of breast cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol Demonstrates Anti-Hyperuricemic Effects in Vivo

By Kyle J. Norton

Anti-hyperuricemic activity is the process including the use of anti-Xanthine oxidase inhibitors to reduce the levels of uric acid levels in the body.

Uric acid, the form of ions and salts is a by-product of chemical after the body breaks down purines, the substance is found in many foods, including Asparagus, Bacon, Beef, Bluefish, Bouillon, Calf tongue, Carp, Cauliflower, Chicken, Chicken soup, Codfish, Crab, Duck, Goose, etc..

High levels of uric acid in blood serum can be harmful, leading to gout and forming of kidney stones.

Gout mostly affected one joint, is an acute and recurrent condition of arthritis as a result of uric acid building up in the blood, inducing joint inflammation.

According to a cross-sectional survey of a total of 6584 adults (3394 women, 3190 men) conducted by Second Military Medical University, on the prevalence of musculoskeletal symptoms, rheumatoid arthritis (RA), ankylosing spondylitis (AS), and gout showed that symptoms occurred more frequently in the following sites:
* Knee 7.0%
* Lower back 5.6%(95% CI 5.0-6.2%),
* Shoulder 4.7%
* Neck 2.4%
Women complained of rheumatic symptoms more frequently than men. In fact, symptoms are quite noticeable, you may feel well when you go to bed but wake up during the night with intense pain in one or few joints and some times with fever. The symptoms may go away in a few days but can return from time to time. Chronic gout can cause lumps below the skin around joints.

Kidney stones are composed of mineral salts formed in the kidneys. Men account for 80% of those with kidney stones and are at risk of the formings between 30 and 40 years of age. About 75% of kidney stones are calcium stones.

Uric acid stones occur in 10% of all kidney stones and are the second-most-common cause of urinary stones after calcium oxalate and calcium phosphate calculi(

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of diseases associated with uric acid, researchers elucidated the L. trichocarpha ethanolic extract (LTE), ethyl acetate fraction from ethanolic extract (LTA) and isolated compounds urate-lowering activity and liver xanthine oxidase (XOD) inhibition on oxonate-induced hyperuricemic mice.

According to the tested differentiation,
* Crude ethanolic extract and its ethyl acetate fraction showed significant urate-lowering effects.

* LTE also significantly inhibited liver xanthine oxidase (XOD) activity in vivo at the dose of 250mg/kg.

* Luteolin, apigenin, lupeol, lychnopholide, and eremantholide C isolated compounds showed anti-hyperuricemic activities among tested compounds.

* Furthermore, LTE, LTA, lupeol, β-sitosterol, lychnopholide, eremantholide, luteolin, and apigenin were also found to inhibit monosodium urate crystals-induced paw edema in mice.

Based on the findings, researchers said, "Ethanolic extract of Lychnophora trichocarpha and some of its bioactive compounds may be promising agents for the treatment of gouty arthritis since they possess both anti-hyperuricemic and anti-inflammatory properties".

Taken altogether, beta-Sitosterol found abundantly in Ethanolic extract of Lychnophora trichocarpha may be considered a supplement for the prevention and treatment of hyperuricemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Tocopherols inhibits the Onset of Esophageal Cancer in Vivo

By Kyle J. Norton

The esophagus or gullet, an organ in vertebrates, is the tube that leads foods from the pharynx to the stomach.

Esophageal cancer is not very uncommon, caused by malignant of the esophagus due to abnormal cell growth as a result of the DNA alternation of the cells that line the upper part of the esophagus or glandular cells that are present at the lower part of the esophagus that connected with the stomach.

Esophageal cancer tends to spread if it left untreated and starts from the lining of the esophagus, then later penetrates the wall of the esophagus and spread to the lymph node around the bottom of the esophagus, stomach and the chest, then to the distant parts of the body.

Most common types of esophageal cancer
* Squamous cancer
In the upper part of the esophagus caused by the squamous cell of which has become malignant caused by mutation of DNA in cells replication of division uncontrollably.

* Adenocarcinomas
In the lower part of the esophagus caused by the glandular cells of which have become malignant caused by mutation of DNA in cells replication of division uncontrollably.

According to the statistics provided by the American Cancer Society, in 2019, approximately 17,650 new esophageal cancer cases will be diagnosed, including 13,750 in men and 3,900 in women.

Also, in 2019, esophageal cancer will also cause the death of 16,080 Americans, including 13,020 in men and 3,060 in women.

The 5-year survival rate of people diagnosed in the local stage of esophageal cancer is 45%, much less than other cancer, including 100% of breast cancer.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of chronic diseases, researchers examined the effect of dietary tocopherols on esophageal carcinogenesis.

According to the analysis,
* In murine models, dietary tocopherols significantly prevent esophageal cancer by inhibiting the activation of NF-κB, associated with blocking the cancer cell apoptosis and the subsequent interaction of chemokine CXCL9/10/11 with their receptor CXCR3 to prevent ESCC proliferation, angiogenesis, and metastasis.

* Furthermore, dietary supplementation with 0.15% α-tocopherol (α-T), δ-tocopherol (δ-T), or γ-tocopherol rich mixture (γ-TmT) markedly suppressed the production of pro-inflammatory cytokines, as well as the induction of CXCR3+ effector T cells (CD4+ Th1 and CD8+ CTLs) infiltration, at the early stage of carcinogenesis.

In other words, dietary tocopherols significantly that prevent the onset of esophageal cancer In experiments in vivo and in vitro, were tightly correlated with the blockade of NF-κB activation

After talking about other factors into account, researchers said, "tocopherols decrease carcinogenesis through inhibiting NF-κB and CXCR3 signaling, as well as related inflammation in early premalignant lesions. This pathway may offer a novel target for chemoprevention of esophageal cancer".

Taken altogether, tocopherols may be considered a remedy for the prevention of the onset of esophageal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Angelica Prevents Oxidative Injury in Vivo

By Kyle J. Norton

Oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

Free radical is an unstable atom with a single electron on the outermost ring. Given its nature, free radical readily donate or intercept an electron from another atom, leading a chain of the free radical cascade that cannot be stopped until all single electron is paired or inhibited by antioxidants.

The antioxidant is a stable atom with a function to inhibit oxidation, come either from dietary sources or produced by the body.

Contrast to general belief, free radical is also needed in moderated amount for the function of the heart muscle to pump blood such as NO for vasodilation and increasing blood flow.

There is no single cause of oxidative stress. Some researchers suggested that a number of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Some researchers suggested that oxidative stress may also arise due to the chain reaction of free radicals in which free radicals donate or intercept an electron from other stable molecules.

Sadly, a prolonged period of oxidative stress presented in the human body can cause oxidative injury to biomolecules, (lipids, proteins, DNA) and the death of healthy cells through cytotoxicity.

Certain chronic diseases are found to be associated with oxidative stress, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

On finding a natural ingredient for the prevention and treatment of chronic diseases, scientists examined the ultra-filtration extract from Danggui Buxue Decoction (EDBD) was able to protect cardiomyocytes from oxidative injury in an animal model.
Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to the results of myocardial cells from 1- to 2-day-old neonatal rats cultured in Dulbecco's modified Eagle's medium low-glucose and Ham's F12 medium (1:1), and the cellular injury induced by H(2)O(2), treatment of ultra-filtration extract mixture from Angelica sinensis and Hedysarum Polyporus at three doses of 3.75, 7.5, and 15 mg/mL.
* Protect the cardiomyocytes from H(2)O(2) injury in a dose-dependent manner (3.75, 7.50, and 15.00 mg/mL).

* Could significantly decrease LDH and CK leakages and intracellular MDA and MPO contents, increase SOD activity, up-regulate hsp70 expression, and down-regulate caspase-3 expression.

Based on the results, researchers suggest, "he EDBD has protection on cardiomyocytes injured by H(2)O(2) through improving cell antioxidant ability, up-regulating hsp70 expression, and inhibiting caspase-3 activity".

Taken altogether, herbal angelica may be considered an antioxidant remedy for the prevention of oxidative injury, pending to the confirmation of the larger sample size and multicenter human study.

Devil's club (Oplopanax horridus) Gobbles Up Human Colorectal Cancer Cells in Vitro

By Kyle J. Norton

The colon-rectum formed a part of the digestive system is involved in the absorption of water from the small bowel contents and broken down of certain materials in the feces into substances of which some of them to be reabsorbed and reused by the body.

Colorectal cancer is a medical condition caused by cell growth irregularly in the tissue of the colon or rectum due to the alternation of cells DNA.

Colon cancer is relatively very common and slowly growing and progressing in a predictable way.

Most cases of primary colon cancer start in the cell on the surface of the inner lining of the colon. At the early stage. most patients with colon cancer do not experience any symptoms.

Rectal cancer is the slowest growing cancer. Therefore, most cases of colorectal cancer are diagnosed at the advanced stage in the older population.

According to the statistics provided by the American Cancer Society, in the US, colorectal cancer is the third most common cancer diagnosed in both men and women compared to other types of cancer.

The lifetime risk of colorectal cancer is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

Approximately, 140,000 cases of colorectal cancer were diagnosed in the US in 2018. Cancer also causes the death of over 50, 000 people, representing 8.3% of total cancer death.

Colorectal cancer is relatively very common and slowly growing and progressing cancer.

The disease is also the third most commonly diagnosed cancer in developed countries, including the US and Canada.

Being overweight or obese. smoking, physical inactivity, excessive alcohol drinking, the increase in age, family and personal history, genetic preposition are some of the prevalent risks of the disease.

Devil's Club is a large shrub, genus Oplopanax, belonging to the family Aralioideae, native to the cool moist forests of western North America.

The herb has been used in traditional and herbal medicine as Adaptogen, Antipyretic, Pain Reliever, Antitussive, Antibacterial agent and to treat Arthritis, Diabetes, Colds, Cough, Sore throat, Chest congestion, Stomach pain, Constipation, Gall stones, Influenza, Cancer, etc.

On finding a potential compound for the treatment of cancer, researchers examined the low-toxic dichloromethane fraction of Oplopanax elatus, (OED) from the genus Oplopanax anticancer effects on colorectal cancer.

According to the tested analysis,
* The 5 μg/ml of OED combined with 40 μM of irinotecan possessed significant synergetic inhibition on SW-480 cells with a combination index (CI) of 0.56.

* The percentage of apoptotic cells was significantly increased from 69.57% (40 μM of irinotecan) or 72.7% (5 μg/ml of OED) to 95.6% after treatment of OED combined with irinotecan (OCI).

In other words, OED and irinotecan exerted a strong synergistic apoptotic effect against the tested cells by activating the protein associated with pro-apoptosis.

Based on the evidence, researchers said, "OED could enhance antiproliferative effects of irinotecan on colorectal cancer cells, which was related to the induction of apoptosis and regulations of activity of caspase-3".

Taken altogether, Devil's club may be considered a remedy for the treatment of colorectal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Brassica Vegetables Reduce the Risk of Prostate Cancer

By Kyle J. Norton

The prostate warped around a tube (the urethra) is a gland of the male reproductive system found in front of the rectum and just below the bladder and responsible for making some of the semen that carries sperm.

The prostate gland is considered a reproductive organ in men.

Prostate cancer is a medical and chronic condition in which the cells of the prostate have become cancerous due to the alternation of the cell's DNA.

Most prostate cancers are slow-growing and enlarged prostate and prostate cancer may be detected during the Physical (rectum) exams.

Primary prostate cancer normally starts from the cell on the surface of the inner lining of the prostate tissue and gradually spreads to the deeper part of the prostate.

At the early stage, patients may not experience any symptoms, However, at the advanced stage, prostate cancer can induce symptoms of blood in the urine, frequent urination and urinary difficulty.

The cancer cells at this stage can also travel a distance away from the prostate to invade other healthy tissue and organ, leading secondary metastasis tumors.

According to the statistic, the 5-year survival rate for local and regional (stage 0 and 1) stages of prostate cancer is nearly 100%.

Believe it or not, more than 80% of prostate cancers are diagnosed in men who are 65 or older.

Therefore, men who are at higher risk of prostate cancer due to family history and increase in age and inherited gene changes should have their prostate gland checked annually.

On finding a potential plant that processes anti-cancer activity researchers examined the effects of Brassica vegetables, including mustard greens on prostate cancer risk.

According to the tested results
* Brassica vegetables exerted a strong effect on the metabolic breakdown products associated with the modulation of xenobiotic-metabolizing enzymes that protect DNA from damage.

According to the 12 published studies that analyze the relationship of Brassica vegetable intake and prostate cancer risk
* Three reported statistically significant reduced risks (P < 0.05) and one reported a borderline significantly reduced risk (P = 0.06) with high Brassica vegetable consumption.

* 6 showed a clear explanation of the prostate cancer risk involved in the consumption of Brassica vegetables.

Collectively, researchers said, " The epidemiological literature provides modest support for the hypothesis that high intakes of Brassica vegetables reduce prostate cancer risk".

Taken altogether, mustard greens may be considered a functional food for reducing the risk of prostate cancer, pending to the confirmation of the larger sample size and multicenter human study.
