Angelica Sinensis Induces Bladder Cancer Cell Death

By Kyle J. Norton

The bladder is a hollow elastic organ in the center of the lower abdomen that collects urine from the kidneys and excreted them through the urethra.

Bladder cancer is the most common type of cancer that affects men twice as frequently as it affects women.

Usually, cancer starts from the lining of the bladder caused by several types of malignant growths of the urinary bladder cells. However, after growing large enough, the cancerous call can spread to other regions in the body through the circulation of fluids and blood.

According to the statistic. 90% of bladder cancer were diagnosed in people who are older than 55. and the average age of people are diagnosed with bladder cancer is 73.

Every year, the disease causes over 17,000 deaths and bladder cancer is the eighth most common cause of cancer death in men.

According to the statistic, men are 4 times more likely than women to be diagnosed with the disease.

Out of the many causes of bladder cancer, researchers found that lifelong smoking, aging, and chronic bladder inflammation are some of the most common prevalent risk factors of bladder cancer cases found in the Western world.

Bladder cancer can be classified into 2 types,
* Superficial types of bladder cancer
This type of bladder cancers has a characteristic of often recurrence but usually non-invasive. It can be controlled or removed by regular cystoscopy.

*Invasive types of bladder cancer.
Invasive types of bladder cancer tend to spread after beginning in cells in the lining of the bladder. It often requires surgery by removing part or the whole bladder.

On finding a natural ingredient for the prevention and treatment of cancer, scientists examined the anti bladder cancer activity of N-butylidenephthalide (BP) isolated from Radix Angelica Sinensis (Danggui) both in vivo and in vitro.

Angelica or Dang Gui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to tested analysis,
* The administration of BP caused bladder cancer cell death in a time- and dose-dependent manner and induced apoptosis via the activation of pro-apoptotic proteins caspase-9 and caspase-3.

* Furthermore, BP also suppressed the migration of bladder cancer cells, revealed by the trans-well and wound healing assays.

* In vitro, BP up-regulated the tumor suppressor E-cadherin and down-regulation of N-cadherin involved in cancer metastasis and increase tumor cell proliferation.

* Moreover, BP also suppressed BFTC xenograft tumor growth and caused a 44.2% reduction of tumor volume after treatment for 26 days.

* Most importantly, in combination with BP, the sensitivity of these bladder cancer cells to cisplatin increased significantly.

Based on the results, researchers suggest, "The incidence of bladder cancer decreased in patients who were exposed to Angelica sinensis, suggesting that BP could serve as a potential adjuvant in bladder cancer therapy regimen".

Taken altogether, Angelica may be considered a remedy for the treatment of bladder cancer, pending to the confirmation of the larger sample size and multicenter human study.

Devil's Claw (Harpagophytum procumbens) Processes Strong Antioxidant Properties

By Kyle J. Norton

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid, and cell damage.

Antioxidants can either produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

However, in people with a weakened immune system, intake dietary sources of antioxidants may be necessary.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily to donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.

Oxidative stress is a result of an imbalance of the ratio of free radicals and antioxidants in the body.

On finding a natural ingredient for the prevention and treatment of chronic inflammatory diseases, scientists examined the antioxidative reparations from the dried tubers of Harpagophytum procumbens (Burch.) DC ex Meisn, commonly known as devil's claw on joint pain and inflammation in patients suffering from rheumatic and arthritic disorders.

Devil's claw also known as the wood spider, is a plant of genus Harpagophytum in the family of Pedaliaceae, native to South Africa.

The plant has been used as an herb in traditional and herbal medicine to treat fever, rheumatoid arthritis, skin, gallbladder, pancreas, stomach and kidneys conditions, etc.

According to the chemical analysis of the content of specialized metabolites in the three samples,
* Harpagoside, the main compound, followed by the two isomers acteoside and isoacteoside exerted a significant antioxidant capacity at the IC 50 concentration compared to others.

*More importantly, other chemical differentiation on antioxidant capacities such as 2,2'-di-phenyl-1-picrylhydrazyl (DPPH) and 2.2'-azin-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods also resulted in very similar IC50 values.

* Most importantly, researchers also found that a higher ferric in the extract showed to reduce antioxidant ability.

In other words, Devil's claw processes a number of antioxidants that play a critical role in decreasing the symptoms of patients suffering from rheumatic and arthritic disorders.

Based on the results, researchers suggest, " the two preparations could generally maintain good stability and biological activity at least for the four years from the production to the expiration date.".

Taken altogether, Dandelion may be considered a remedy for the treatment of diseases associated with oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Elder (Sambucus), a Natural Antioxidant Remedy

By Kyle J. Norton

Antioxidants are stable molecules that inhibit oxidation induced by free radicals.

Some foods are antioxidants producers such as fruits and vegetables and whole-grain, while others such as processed meats, alcohol are radicals producers. Particularly, fats and oils can easily be oxidized if they are cooked in high heat.

A free radical is a highly reactive atom with a single electron in the outer ring.

Free radicals produced in our body are a natural process derived either from essential metabolism through the processes of food intake or from external sources through directly contacting the skin or the air we breathe including exposure to X-rays, ozone, cigarette smoking, air pollutants, and industrial chemicals.

In order to counter the overexpression of free radicals which add to the body's burden. However,  our body produces enzymes antioxidants and metal carrier proteins to neutralize them before they can cause damage.

Depending on the degrees of free radical invasion, antioxidant enzymes can be categorized into first, second, third and even fourth line of defense.

Oxidative stress is a result of the imbalance of free radicals and antioxidants in the body. The syndrome is either caused by overexpression of free radicals or depressed levels of antioxidants or both.

Oxidative stress has been found to induce widespread damage due to a set of a chain reaction auto-catalytically by free radicals that attacks the lipid bilayers of the cell walls, induces lipid, protein damage and alternation of cell DNA, leading to the onset of chronic diseases, including cancers.

On finding a natural ingredient for the prevention and treatment of diseases of oxidative stress, scientists compared the chemical profile that induces the antityrosinase and antioxidant activities of ethanol (EtOH) and methanol (MeOH) extracts of Sambucus nigra L. (Adoxaceae) flowers and leaves

Elder or Common Elder, is a shrub or small tree of the genus Sambucus of 30 species, belonging to the family Adoxaceae, native to the temperate-to-subtropical regions of both the Northern Hemisphere and the Southern Hemisphere.

The herb has been used in traditional and herbal medicine to treat constipation, colic, diarrhea, colds, flu, bronchial and upper-respiratory, irritation of the skin, rheumatism, etc.

According to the results of a novel lower-melting-point ethylene-chlorotrifluoroethylene (LMP ECTFE) nanofiltration (NF) membrane was employed to produce the concentrated fractions of S. nigra., the NF retentate fractions of floral ethanol extracts exerted the highest tyrosinase inhibitory activity with an IC50 of 53.9 µg/mL and the highest ABTS radical scavenging activity (IC50 of 46.4 µg/mL).

The major chemical compounds that exert the aforementioned activity, according to the performance liquid chromatography (HPLC) profile were rutin, quercetin, protocatechuic acid, 3,5-dicaffeoylquinic acid, and neochlorogenic acid.

Based on the results, researchers suggest, " ...the present investigation revealed the potential benefits of NF application in S. nigra extracts processing, suggesting the use of retentate fractions as a promising source for antioxidant and tyrosinase inhibitory compounds which could pave the way for future applications".

Taken altogether, Dandelion may be considered an antioxidative remedy for the treatment of diseases induced by oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Elecampane (Inula helenium) May Be Used for the Inhibition of Oxidative Stress-Related Human Diseases

By Kyle J. Norton

Oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.

In other words, antioxidants inhibited the onset of free radical before they can cause a chain reaction in facilitating the protein, lipid and cell damage.

Antioxidants can either produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

There is no single cause of oxidative stress. Some researchers suggested that a number of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Sadly, a prolonged period of oxidative stress presented in the human body can cause oxidative damage to biomolecules, (lipids, proteins, DNA) and the death of healthy cells through cytotoxicity.

Certain chronic diseases are found to be associated with oxidative stress, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

On finding a natural ingredient for the prevention and treatment of diseases involved in oxidative stress, scientists examined the effects of secondary metabolites from Inula species on the oxidative stress-related diseases.

Elecampane, also known as Horse-heal (Inula helenium) is a perennial composite plant, genus nulla, belonging to Asteraceae, native to Southern Europe, and Asia.

The plant has been used in traditional and herbal medicine in treating bronchitis or emphysema, night sweats, pulmonary tuberculosis, irritating cough, catarrhal discharges, etc.

According to the search of the database of medical literature from Scopus, PubMed, and Web of Science, that identified more than 120 articles are reviewed, highlighting the most active compounds with special emphasis on the elucidation of their antioxidative-stress mechanism of action,
* Alantolactone found in Elecampane was the most polyvalent compound, reporting interesting EC50 values for several bioactivities that showed an effect on on the oxidative stress-related diseases.

* 1-O-acetylbritannilactone also found in Elecampane was found to be a promising lead compound for the development of analogs with interesting properties.

Based on the results, researchers suggest, "The Inula genus is a good bet as a source of structurally diverse compounds with antioxidant activity that can act via different mechanisms to fight several oxidative stress-related human diseases, being useful for the development of new drugs".

Taken altogether, Elecampane processed abundantly Alantolactone and 1-O-acetylbritannilactone and may be considered a remedy for the treatment of diseases associated with oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Angelica Improves Impairment of Testosterone Synthesis Induced Infertility

By Kyle J. Norton

Testosterone is a male hormone. The hormone besides promoting libido it has other important functions such as maintaining muscle mass and forming bone and regulating heart muscle and cholesterol.

The Hormone also helps to improve the oxygen levels throughout the body as well as controlling blood glucose and strengthening the immune system.

However, as men age, the pituitary gland produces less free testosterone while more free testosterone is being converted to estrogen which results in many health problems.

Testosterone is synthesized primarily in Leydig cells stimulated by the pituitary gland to release FSH and LH.

In other words, when the levels of testosterone are low, testosterone is synthesized induced by gonadotropin-releasing hormone (GnRH) that is released by the hypothalamus.

Symptoms associated with  testosterone deficiency are
* Decreased sexual function
As we mentioned in the last article, starting at age 40 levels of testosterone start to diminish. The levels of the by-product prolactin of testosterone of men increases, stimulating the production of the enzyme 5-alpha reductase that causes the conversion of testosterone to dihydro-testosterones DHT thus triggering low levels of testosterone resulting in sexual dysfunction.

* Loss of bone density
The brain and bone are the important issues that have the primary effect of testosterone are by way of aromatization to 17² estradiol in the bones. 17²-estradiol accelerates the maturation of cartilage into bone, leading to the closure of the epiphyses (a rounded end of a long bone) and a conclusion of growth.

* Loss of muscle mass
Testosterone effects can be classified as anabolic effects that include growth of muscle mass and strength, increased bone density and strength, and stimulation of height growth and bone maturation. Testosterone effects can also be classified by the age of usual occurrence.

* Memory loss
Low levels of testosterone allow beta-amyloid, a toxic peptide to accumulate in certain regions of the brain causing memory loss.

* Abdominal fat
Fat cells create aromatase enzymes that contribute to fat build up and low levels of testosterone which allows the forming of abdominal fat that produces more aromatase enzymes resulting in more formation of estrogen. It also causes insulin resistance by increasing fat around the stomach/waist area and fat mass.

Impairment of testosterone synthesis is a condition associated with the testis impaired by the cryptorchid state,

On finding a natural ingredient for the prevention and treatment of cancers, scientists examined the Chinese angelica polysaccharide (CAP), the main effective ingredient of angelica sinensis anti-inflammatory and anti-apoptotic effects.

On finding a potential compound for the treatment of male infertility, researchers examined the protective effect of a mixture of 2 herbal extracts, KH-465, which consisted of Epimedium koreanum Nakai and Angelica Gigas Nakai, on spermatogenesis in a luteinizing hormone-releasing hormone (LHRH) agonist-induced rat model of male infertility.

The study included 75 12-week-old male Sprague-Dawley rats that were randomly divided into 5 groups, containing 15 rats each: a normal control group that received no treatment and 4 experimental groups (I, II, III, and IV) in which an LHRH agonist was administered for 4 weeks to induce spermatogenic failure. Group I received distilled water, and groups II, III, and IV received 200 mg/kg/day of KH-465, 400 mg/kg/day KH-465, and depo-testosterone for 4 weeks, respectively.

According to the results of differentiation,
* There are no significant changes in Body, testis, and epididymis weight among the control and experimental groups.
* Treatment with KH-465 increased sperm count and motility.

* Serum hormone levels of T, free T, and FSH were not significantly different in the experimental groups, while the LH level was higher than in the LHRH agonist-induced control group, but not to a significant extent.

* Levels of SOD were higher and 8-OHdG were lower in the groups that received KH-465 than in the LHRH agonist-induced control group.

Based on the results, researchers suggest, "Our results suggest that KH-465 increased sperm production via reducing oxidative stress and had a positive effect in a male infertility model".

Taken altogether, herbal Angelica alone or in combination with another herbal remedy may be used for the treatment of male infertility, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Devil's Club Processes Anti-Inflammatory Property

By Kyle J. Norton

Anti-inflammation is a process that inhibits the pro-inflammatory cytokines without affecting the immune system activity against pathogens.

Acute inflammation is a natural response of the immune system to protect the human body against harmful stimuli and pathogens.

Acute information is the first phase of injection. At this phase, after sensing the body injury by the central nervous system, the immune first line of defense stimulates the production of blood platelets to the cover the site of damage and pro-inflammatory to kill off pathogens that try to enter the body through the wound or damaged tissue.

Acute inflammation usually receded after a few days or weeks, depending on the size and damage. However, if all pathogens were not eradicated after 3 to 8 weeks, the immune system will compromise, leading to chronic low-grade inflammation, a leading cause of chronic illness.

The most common symptoms of acute inflammation occurred with a short period of time such as hours is pain, redness, immobility, swelling, and heat.

Conventionally, depending on the types of acute inflammation, non-steroidal anti-inflammatory drugs (NSAIDs) may be prescribed to alleviate the pain by counteracting an enzyme that contributes to inflammation.

Chronic inflammation is a low-grade inflammation caused by the inability of the immune system to eliminate the invasive pathogens that occurred in the acute phase of infection.

Any inflammation that lasts more than 8 weeks is considered chronic inflammation.

Believe it or not, worldwide, 3 of 5 death are results from chronic inflammatory diseases like stroke, chronic respiratory diseases, heart disorders, cancer, obesity, and diabetes.

In the US, chronic inflammatory diseases affect more than 50 million Americans each year.

Devil's Club is a large shrub, genus Oplopanax, belonging to the family Aralioideae, native to the cool moist forests of western North America.

The herb has been used in traditional and herbal medicine as Adaptogen, Antipyretic, Pain Reliever, Antitussive, Antibacterial agent and to treat Arthritis, Diabetes, Colds, Cough, Sore throat, Chest congestion, Stomach pain, Constipation, Gall stones, Influenza, Cancer, etc.

On finding a potential compound for the treatment of chronic inflammatory diseases, scientists examined the antioxidative stress activity of the ethanolic extracts of the devil's club (Oplopanax horridus) leaves, stems, and roots.

Based on the chemical differentiation,
* The leaf extract had higher total phenolic and flavonoid contents and anti-oxidative activity (radical scavenging, reducing power, and inhibition of lipid oxidation) compared to the root and stem extracts.

* The leaf extract also had anti-inflammatory effects against the activation of proteins associated with the production of pro-inflammatory cytokines, such as lipopolysaccharide (LPS)-induced nitric oxide (NO; 71.0% at 50 μg/mL), tumor necrosis factor (TNF)-α (87.6% at 100 μg/mL), and interleukin (IL)-6 (36.2% at 100 μg/mL) in the murine RAW 264.7 macrophages.

* Furthermore, LPS-induced inducible nitric oxide synthase (iNOS) expression was also decreased by the leaf extract at the concentration of IC50=24.4 μg/mL.

* The major compounds that exert the aforementioned inhibition was attributed to the expression of gallic acid, protocatechuic acid, chlorogenic acid, and maltol.

Collectively, researchers wrote, " leaf extract could be utilized as a functional food material because of its anti-oxidative and anti-inflammatory activities".

Taken altogether, coltsfoot may be considered a remedy for the protection of inflammatory diseases induced by oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Omega-3 Fatty Acids Has NO Effects on Chronic Kidney Diseases

By Kyle J. Norton

Kidneys formed part of the secretory system, are to bean-shaped organs located below the rib cage, one on each side of your spine with a function to filter out blood and remove waste through urinary secretion.

The kidney also plays a critical in maintaining the levels of fluids in the body, by removing extra fluids to make urine.

Furthermore, the kidney also regulates electrolytes, blood pressure, maintains acid-base balance, produces hormone calcitriol, renin, and erythropoietin.

Kidney diseases are a class of medical conditions associated with reduced function of the kidney, in cases, complete loss of kidney function such as kidney failure.

Chronic kidney disease, also known as chronic kidney failure, is a condition of a gradual loss of kidney function.

According to the observation of patients data associated with the diseases, diabetes and high blood pressure are responsible for up to two-thirds of the cases.

According to the statistic, approximately, 30 million American adults have chronic kidney disease, and a large percentage of these people aren't aware of it. 1 in 3 American adults is at risk for kidney disease.

In other words, approximately 14 percent of the US population suffer some forms of chronic kidney disease (CKD), including 661,000 cases of kidney failure.

Believe it or not, almost half of the individuals with CKD also have diabetes and/or self-reported cardiovascular disease (CVD).

Diabetes is a medical condition caused by long-term abnormally high blood glucose, leading to the damage of other tissue and organs, including the kidneys.

Another cause of chronic kidney disease is kidney disease. Early diagnosis of chronic kidney disease can often keep chronic kidney disease from getting worse.

Late diagnosis of chronic kidney disease may eventually lead to kidney failure, leading to dialysis or a kidney transplant to maintain life.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of kidney, researchers evaluated the effects of the anti-inflammatory effect of omega-3 fatty acids on patients with chronic kidney disease(CKD).

By searching the medical literature on the database from inception to August 2016, including randomized controlled trials (RCTs) derived from multiple databases, and the effect of omega-3 fatty acids supplementation compared to control cohorts, researchers identified a total of 12 RCTs involving 487 patients were included in the meta-analysis.

Among 254 patients received omega-3 fatty acids and 233 patients served as controls who received placebo, there is
* There is no statistical significance in serum levels of C-reactive protein (CRP) (SMD, -0.20; 95% CI, -0.44 to 0.05; P = 0.11),

* The levels of pro-inflammatory cytokine IL-6 associated with the CKD showed insignificant change in both omega 3 and control groups.

* Additionally, chronic kidney disease (CKD) in the constitution of chronic inflammation through the expression of elevated levels of tumor necrosis factor-alpha (TNF-α) also showed insignificant changes in both groups.

Based on the findings, researchers suggested, "there is insufficient evidence to conclude the benefit of omega-3 fatty acids oral supplementation in reducing serum levels of CRP, IL-6, and TNF-α in patients with CKD".

In other words, omega-3 fatty acids may not be used for reducing complications of patients with CKD, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
