Omega-3 Fatty Acids Kills Myeloid Leukemia Cells in Vivo

By Kyle J. Norton

Cancer is a class of diseases in which a group of cells growing and multiplying disorderly and uncontrollably in our body.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of the organ or gland. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Leukemia is a condition of an abnormal increase of white blood cells produced by the bone marrow and/or the lymphatic system. Depending on the malignant granulocytes or lymphocytes, leukemia is classified into myelogenous or lymphoblastic leukemia.

Bone marrow is the soft tissue inside the hollow center of the major bone. including spine, pelvis, underarm, leg. etc.

Myeloid Leukemia can be classified into acute and chronic
* Acute myeloid leukemia
Acute myeloid leukemia (AML) is a cancer of myelogenous lining cause of the rapid growth of abnormal granulocytes accumulated in the bone marrow, interfering with the normal blood cells. In AML, granulocytes appear immature in the bloodstream.

* Chronic myeloid leukemia (CML) is a medical condition associated with the overgrowth of immature cells started in the blood-forming cells of the bone marrow.

In other words, compared to normal cells that will die after a limited cell division, chronic myeloid leukemia cells continue to divide with no meaning of dying or stopping, leading to proliferation that affects other healthy cells in the bone marrow.

According to the statistics provided by the American Cancer Society, in 2019, approximately, 8,990 new cases will be diagnosed with CML (5,250 in men and 3,740 in women. and CML is expected to cause the death of 1,140 people including 660 men and 480 women.

More precisely, CML accounts for 15% of all cases of leukemia diagnosed each year. In other words, the lifetime risk of CML in the US is 1 person in 526.

The 5-year survival rate of patients treated with chemo drug imatinib (Gleevec®) including all stages of CML is 90%.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of cancer, researchers investigated the effects of eicosapentaenoic acid (EPA, C20:5), a polyunsaturated omega-3 fatty acid on myeloid leukemia.

According to the tested analysis in a murine model of chronic myelogenous leukemia (CML) induced by bone marrow transplantation of BCR-ABL-expressing hematopoietic stem cells,
* EPA showed a decrease in the leukemia stem cells (LSC) population and reduced the abnormal enlargement of the spleen and above the normal range in the blood associated with chronic myelogenous leukemia progression, compared to mice treated with oleic acid diet.

* EPA exerted a similar effect on CML mice carrying the T315I mutation (in BCR-ABL).

* Δ(12)-PGJ3, a derivative of EPA also rescued indomethacin-treated BCR-ABL mice and decreased LSCs.

Based on the findings, researchers said, "EPA supplementation was effective in a murine model of acute myeloid leukemia. EPA-supplemented mice exhibited a decrease in leukemia burden and a decrease in the LSC colony-forming unit (LSC-CFU)".

And, "he results support a chemopreventive role for EPA in myeloid leukemia, which is dependent on the ability to efficiently convert EPA to....Δ(12)-PGJ3".

Taken altogether, Omega-3 fatty acids processed abundantly eicosapentaenoic acid may be considered a functional food for the treatment of myeloid leukemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol, A Promising Natural Hormone Estrogen Therapy Alternative

By Kyle J. Norton

Estrogen is a primary female sex hormone, responsible for the development and regulation of the female reproductive system and secondary sex characteristics.

The estrogenic effect is an action that stimulates the production of the hormone estrogen in case of deficiency.

On the other hand, the anti-estrogenic effect is an action of binding the estrogen receptor(ERα ) and/ERβ in inhibiting the excessive levels of estrogen circulating in the body.

Menopause is a condition in which a woman is in the transition stage of permanent cessation of the ovaries functions in egg production because of less production of estrogen and progesterone, signaling the end of the reproductive phase a woman's life.

In menopause, menstruation has become irregular and slowly stop overtime, but in some women, menstrual flow comes to a sudden halt.

Some researchers suggested that you officially enter the stage of menopause if you have gone 12 months without a menstrual period.

Menopause symptoms are a set of symptoms caused by the gradual decline of levels of estrogen and progesterone due to aging loss menstrual periods in women.

The reproductive tract, the urinary tract, the heart and blood vessels, bones, breasts, skin, hair, mucous membranes, pelvic muscles normally regulated by hormone estrogen also are affected by the decline levels of the hormone.

In the concern of hormone therapy used for the treatment of menopause symptoms, herbal medicine with estrogenic effect have been under intensive investigation, with an aim to find a natural ingredient with the similar effects as of those used in hormone therapy with no side effects.

The aim of estrogenic therapy is to relieve symptoms of sleep interruption, according to the study of health-related quality of life, fatigue, and hormone-related symptoms in breast cancer survivors, and menopausal symptoms, primarily hot flashes, affects approximately two-thirds of menopausal and postmenopausal women, botanicals and dietary supplement (BDS).

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of menopausal symptoms, researchers elucidated the effects of β-sitosterol and stigmasterol in the hydroalcoholic extract of Bambusa bambos as an alternative Hormone Replacement Therapy.

According to the results of the experiment
* The extract containing β-sitosterol and stigmasterol showed an estrogenic effect on the growth of MCF-7 cells in vitro.

* The tested extract showed no cytotoxicity to the to MCF-7 cells in any of the dilutions, but induced cell proliferation and all the dilutions except <500μg/ml have significantly increased cell multiplication.

* Furthermore, the doses of 15.6, 31.2, 62.5 and 125μg/ml of HEBB have shown influence on the proliferation rates similar to the standard 17β-estradiol.

Based on the findings, researchers said, "The results suggest that HEBB might be used as a safe alternative to estrogen replacement therapies".

Beta-Sitosterol may be considered a natural estrogenic remedy for the prevention and treatment of menopausal symptoms, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Devil's claw (Harpagophytum procumbens) Induce Liver Toxicity in Treatment of Cancer in Vitro

By Kyle J. Norton

The liver not only plays a critical role to filter the blood coming from the intestine before passing them to other parts of the body but also protects our body against toxin by detoxifying chemicals and metabolizes drugs.

Furthermore, the liver produces bile that not only aids the digestive function in food absorption but also builds strong cell membranes, produces vitamin D and steroid hormone.

Additionally, the liver also inhibits oxidative stress by producing the antioxidant enzyme that counters the expression of free radicals in the body.

Liver cancer is a medical condition characterized by cell growth irregularly in the liver tissue.

The liver organ is one of the organs with a lack of nerve tissue, therefore liver cancer does not induce pain until the later stage.

At the advanced stage of liver cancer, the cancer cell may travel a distance away to infect other tissues and organs, leading to secondary metastasis.

According to the statistics provided by the Canadian Cancer Society, every year, approximately, 1,900 men and 580 women are diagnosed with liver cancer, causing the death of 950 men and 270 women.

In 2017, 2,500 Canadians were diagnosed with liver cancer and 1,200 Canadians died from liver cancer.

Compared to the early stage of other cancer, the 5 years survival rate in the early stage diagnosis is only 31% and 43% of people with liver cancer diagnosed at stage 0 and 1.

Toxic liver disease or liver toxicity is a condition that damages the liver.

On finding a natural ingredient for the prevention and treatment of liver diseases, scientists examined the anti toxicity of harpagoside (HA), the main compound from Harpagophytum procumbens (Hp) in a hepatic cell line (HepG2/C3A).

Devil's claw also known as the wood spider, is a plant of genus Harpagophytum in the family of Pedaliaceae, native to South Africa.

The plant has been used as an herb in traditional and herbal medicine to treat fever, rheumatoid arthritis, skin, gallbladder, pancreas, stomach and kidneys conditions, etc.

According to tested analysis,
* The administration of HA was found to decrease the number of cells in the S phase, increase the number of cells in the G2/M phase followed by inducing apoptosis.

* Neither Hp nor HA was genotoxic.

* Hp inhibited the expression of CDK6 and CTP3A4 in removing liver toxins or drugs.

* The cells upon exposure to Hp and HA exhibited decreased proliferation.

More precisely, HA and Hp induced cytotoxicity through mechanisms that involved in the inhibition of metabolism instead of involving transcriptional regulation of the cyclins and CDKs associated with cell cycle arrest.

Based on the results, researchers suggest, "treatment with either Hp or HA can be cytotoxic, and this should be taken into consideration when balancing the risks and benefits of treatments for rheumatic disease".

Taken altogether, Devil's claw injection with a large dose may induce toxicity, pending to the confirmation of the larger sample size and multicenter human study.

Healthy Foods Broccoli May Process Anti-Influenza Virus Properties

By Kyle J. Norton

Influenza also known as "the flu", is an infectious disease caused by an influenza virus.

In other words, flu (influenza) is an infectious disease caused by RNA viruses of the family Orthomyxoviridae (the influenza viruses) that can lead to symptoms of chills, fever, sore throat, muscle pains, severe headache, coughing, etc.

There are 3 types of flu influenza: A, B, and C. the influenza virus A and B are considered seasonal flu that causes sniffling, aching, coughing, and running and stuffy nose and fever to over 20% of the general population.

Influenza virus types C tends to cause mild upper respiratory infections, similar to those of cold.

Healthy individuals who have influenza are usually recovered within one week.

However, people with immature and weakened immune system such as young children under age 5, adults older than age 65 residents of nursing homes and other long-term care facilities, chronic illnesses, such as asthma, heart disease, kidney disease, liver disease, and diabetes are not only associated with the risk of influenza but also increase the risk of influenza deadly complication.

Pregnant women and women up to two weeks postpartum due to change of the immune system and other organs have been found to make a woman more prone to influenza and influenza complications.

Most common H and N subtype are influenza A that has been mutated to affect the combination of human, swine, and bird flu viruses, including
* H1N1 - Swine Flu
H1N1 normally is only transmitted among pigs. However, as of today, H1N1 became the first flu pandemic in the world that causes respiratory illness.

* H5N1 - Bird Flu
H5N1 is the strain of influenza known as a bird or avian flu. Before, it is transmitted between birds, but today, it now can be passed from bird to human. The strain is dangerous to humans with high death rates.

Broccoli is a mustard/cabbage plant, belongings to the family Brassicaceae. The veggie has large flower heads, usually green in color with a mass of flower heads surrounded by leaves and evolved from a wild cabbage plant from the continent of Europe.

With an aim to find a natural food for the prevention and treatment of viral diseases, researchers examined the effects of broccoli sprout homogenates (BSH) on nasal cytokines, virus replication, and Nrf2-dependent enzyme expression in smokers and nonsmokers.

The randomized, double-blind, placebo-controlled trial included healthy young adult smokers and nonsmokers ingested BSH or placebo (alfalfa sprout homogenate) for 4 days, designated Days -1, 0, 1, 2. and on day 0 they received standard vaccine dose of live attenuated influenza virus (LAIV) intranasally.
According to the tested analysis,
* In the area under curve analyses, post-LAIV IL-6 responses (P = 0.03) and influenza sequences (P = 0.01) were significantly reduced in NLF from BSH-treated smokers.

* However, In nonsmokers, a similar trend for reduction in virus quantity with BSH did not reach statistical significance.

In other words, in smokers, short term ingestion of broccoli sprout homogenates appears to significantly reduce some virus-induced markers of inflammation, and virus quantity compared to less significant in non-smokers.

Based on the results, researchers wrote, " Nutritional antioxidant interventions have promise as a safe, low-cost strategy for reducing influenza risk among smokers and other at-risk populations".
And "However, because variability exists in the timing and methods of detection for SFN conjugate levels, it is difficult to compare systemic SFN levels across studies".

In other words, SFN protected the integrity of the neurons against neuron damage through it's anti-neuroinflammatory and neuroprotective activities.

Taken altogether, broccoli sprouts may be considered supplements for the prevention of influenza, pending to the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Angelica Sinensis, A Potential Anti Weight Gain Remedy

By Kyle J. Norton

Obesity is a medical condition caused by excess body fat accumulated to some extent that impacts personal health. The hallmark of obesity is attenuated metabolism and structural cell changes in the obese fat observed by reducing metabolic activity.

According to the Body Mass Index(BMI), people who have a BMI between 25 to 29.9 are considered overweight and a BMI of over 30 is an indication of obesity.

The formula of the BMI indexBMI= weight (kg)/ height (m2)

According to the statistic, 68% of the American population is either overweight or obese. However,
in 2016, more than 1.9 billion adults, 18 years and older, were overweight, including over 650 million were obese worldwide.

Visceral fat obesity is associated with 90% of patients with ischemic heart disease. In obese, accumulation of visceral fat is predominant in the intra-abdominal cavity.

Visceral fat is a layer of fat that surrounds the organs. Overexpression of visceral fat has been found to promote insulin resistance, diabetes, heart disease, and inflammation.

However, in non-obese subjects, visceral fat accumulation is correlated with glucose intolerance, hyperlipidemia, and hypertension.

Obesity rates have increased for all age groups in the US over the last several decades, coincidently with the periods that promoted the Western diet.

Weight gain is an increase in body weight associated with some parameters, including increased muscle mass, fat deposits, excess fluids such as water or other factors.

However, weight gain has also been found to be induced by serious medical conditions.

On finding a natural ingredient for the prevention and treatment of obesity, scientists examined the radix Angelica sinensis (RAS) suppressed body weight by altering the expression of the fat mass and obesity-associated (FTO) gene in an animal model.
Angelica or Dang Gui is a herb of Genus Angelica from the family Apiaceae, indigenous to China.

The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

According to tested analysis in mice fed with high-fat diet (HFD)-induced obesity,
* 36 differentially-expressed genes (DEGs) associated with the increased onset of obesity were found in adipose tissues from the RAS supplementation group (DH) and control group (HC).

* Six out of the 36 DEGs identified to be involved in lipid metabolism, with the APOA2 gene a potential anti-obesogenic influence were inhibited by the injection of RAS compared to control.

In other words, RAS supplementation in HFD-induced obese mice was associated with an anti-obesogenic global transcriptomic response.

Based on the results, researchers suggest, "This study provides insight into potential applications of RAS in obesity therapy".

Taken altogether, Dandelion may be considered a remedy for the treatment of weight gain in obese, pending to the confirmation of the larger sample size and multicenter human study.

Ficus religiosa L. (Moraceae) ot Sacred Fig Protects the Kidney Against Acute Renal Failure in Vivo

By Kyle J. Norton

Kidneys are to bean-shaped organs located below the rib cage, one on each side of your spine with a function to filter out blood and remove waste through urinary secretion.

The kidney also plays a critical in maintaining the levels of fluids in the body, by removing extra fluids to make urine.

Furthermore, the kidney also regulates electrolytes, blood pressure, maintains acid-base balance, produces hormone calcitriol, renin, and erythropoietin.

Kidney diseases are a class of medical conditions associated with reduced function of the kidney, in cases, complete loss of kidney function such as kidney failure.

Kidney dysfunction reduces the body's ability to clean and filter extra water from your blood, leading to fluid retention and high blood pressure in the early stage. Believe it or not, people with a kidney function of 10% do not show signs of kidney dysfunction and symptoms.

In other words, kidney disease often has no symptoms until at a very advanced stage.
In the US, the leading causes of kidney failure and the end stage of chronic kidney diseases are diabetes and high blood pressure.

Kidney failure or renal failure or end-stage kidney disease is a medical condition in which the kidneys are functioning at less than 15% of normal.

Acute kidney failure, which develops rapidly and may resolve quickly, is a result of insufficient blood flow to the kidneys such as renal hypertension. These types of kidneys dysfunction can be cured if the causes are found.

Most cases of acute kidney dysfunction may be associated with toxin accumulated in the kidney organs, leading to the condition of nephrotoxicity.

Ficus religiosa or sacred fig is a species of fig belongs to Moraceae, the fig or mulberry family, native to the Indian subcontinent and Indochina.

On finding a potential phytochemical for the prevention and treatment of kidney diseases, researchers elucidated the effects of nephroprotective and curative effects of F. religiosa latex methanol extract against cisplatin-induced acute renal failure.

The study included rats that were divided into five groups.
* Group 1 was administered acacia (2% w/v) of 5 ml/kg throughout the experiment;

* Group 2 was treated with a single dose of cisplatin (5 mg/kg i.p.) on the 1st day;

* Group 3 (200 mg/kg p.o.) of extract control for the 1st to 10th day,

* Group 4 (200 mg/kg p.o.) of extract from the 1st to 10th day and a single dose of cisplatin (5 mg/kg, i.p.) on 11th day while

* Group 5 received the same dose of cisplatin on day 1 and extract (200 mg/kg p.o.) from the 7th to 16th day.

According to the results of the experiment
* The cisplatin-treated group 2 showed significantly reduced renal functions, the changes of biochemical parameters and histopathology in the kidney including increased lipid peroxidation, kidney oxidative stress, and inflammatory markers and reduced levels of antioxidant enzymes.

* However, all parameters were restored back to the near-normal with the 200 mg/kg of extract in the curative and protective groups.

In other words, F. religiosa latex methanol extract showed a strong kidney protective activity against toxicity induced by the cisplatin treatment.

Based on the findings, researchers said, "These findings demonstrated that F. religiosa latex and constituents have excellent nephroprotective and curative activities and thus have great potential as a source for natural health products".
Taken altogether, beta-Sitosterol may be considered a supplement for the prevention of acute kidney toxicity, pending to the confirmation of the larger sample size and multicenter human study.

Healthy Food Cabbage Causes Cytotoxicity to Endometrial Cancer Cells in Vivo

By Kyle J. Norton

The endometrium is the inner lining of the mammalian uterus and very susceptible hormone change, particularly in the menstrual cycle.

Endometrial cancer is a chronic condition characterized by the irregular cell in the tissue of the endometrium.

It's the fourth most common cancer among women overall, after breast cancer, lung cancer, and bowel cancer.

Most cases of endometrial cancer start on the cells on the surface of the inner lining of the endometrium. At the advanced stage, after penetrating into the deeper layers, malignant cancer cells may travel a distance away to infect other healthy tissue and organs.

Endometrial cancer has been found to metastasize to the rectum, bladder, pelvic and para-aortic lymph nodes, vagina.

The exact causes of endometrial cancer are unknown. Epidemiological studies suggested that the increase in age, more years of menstruation, never been pregnant, obesity and the use of hormone therapy are some of the major prevalent risk factors associated with endometrial cancer.

Some researchers indicated that hormone changes in women's body such as higher androgen and estrogen levels and lower levels of progesterone due to polycystic.

In the US, endometrial cancer is the most common cancer of the female reproductive organs. Every year, approximately About 63,000 new cases of uterus cancer are diagnosed, causing the death of 11,000 women.

The 5-year survival rates for white and black women with the disease are 84% and 62% respectively.
At the invasive stage, the survival rate is 16%.

Cabbage is a species of Brassica oleracea belongings to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem with a crowded mass of leaves, usually green but in some varieties, it may be red or purplish.

On findings a natural wholefood for the treatment of cancer, researchers examined the effects of 3,3'-Diindolylmethane (DIM), a major in vivo product of indole-3-carbinol (I3C), is a promising anticancer agent derived from vegetables of the Brassica genus including cabbage on the endometrial cancer cell line.

According to the results of the tested analysis,
* DIM induced cytotoxic activity by increasing the level of TGF-alpha transcripts by approximately 4-fold within 24 h of indole treatment in the tested cancer cells.

* DIM also induced a 4-fold increase in the activity of the estrogen response marker, alkaline phosphatase (AP), the cancer markers.

In other words, DIM increased the maximum stimulatory effect of estrogen on TGF-alpha expression to induce cancer cell death.

* DIM alone treatment stimulated levels of secreted TGF-alpha protein by >10-fold. More precisely, the elevated addition of TGF-alpha inhibited the growth of Ishikawa cells, whereas incubation with a TGF-alpha antibody partially reversed the growth inhibitory effects of DIM.

Based on the results, researchers wrote, " ligand-independent estrogen receptor agonist activity of DIM, and uncover an essential role for the stimulation in TGF-alpha expression and the TGF-alpha activated signal transduction pathway in the potent cytostatic effects of DIM in endometrial cancer cells".
Taken altogether, cabbage may be considered a functional food for the treatment of endometrial cancer pending to the confirmation of the larger sample size and multicenter human study.
