Ursolic Acid, An Adjunct Therapy for the Treatment of Liver Cancer

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing them to other parts of the body.

In the US, approximately 42,030 new cases including 29,480 in men and 12,550 in women will be diagnosed with liver cancer. The liver cancer also will cause of 31,780 American, including 21,600 men and 10,180 women in 2019.

Hepatocellular carcinoma (HCC) is a common form of primary liver cancer.

The most common symptoms of liver cancer are abdominal pain, tenderness, easy bruising or bleeding, enlarged abdomen, jaundice, severe pain, particularly in the later stage and drowsiness and confusion accompanied by the general symptoms shared by other types of cancer.

Most cases of contact dermatitis are nonlife-threatening, however, the condition can induce certain discomforts, particularly in people with an allergic reaction to other conditions.

Contact dermatitis can be healed within a week or 2. The condition can be preventable if the cause is identified.

Besides the common symptoms of a red, itchy rash on the skin, some people may experience more serious symptoms including crusting swelling, burning or tenderness skin.

Atopic dermatitis or eczema is an inflammatory condition associated with red and itchy skin.

* Histamine and motivation
Brain histamine may affect a variety of different behavioral and physiological functions; however, its role in promoting wakefulness has overshadowed its other important functions.

In the study to investigate the effect of histamine-3 (H(3)) receptors, expressed in the tuberomammillary nucleus (TMN) of the hypothalamus and in the prefrontal cortex (PFC), on histamine neurotransmission in the rat brain, showed that systemic administration of the selective H(3)-agonist, immepip, decreases, and the reverse H(3) /H(4)-agonist, thioperamide, increases the firing activity of histamine neurons in the TMN and the release of histamine in TMN and PFC.

* The FS-3 fraction, obtained from the dichloromethane extract, comprised triterpenes β-sitosterol (1), α-amyrin (2), moronic acid (3), and ursolic acid (4), and all the compounds showed significant activity in comparison with that of indomethacin (41.5 ± 0.6%) at 0.1 mg/mouse ear.

* Moronic acid displayed the highest inhibitory effect (68.1 ± 1.3%).

* Moronic acid was the most active (73.3 ± 1.1%, indomethacin 33.8 ± 0.8%) on histaminic activity.

Finally, moronic acid was found to be a principal anti-inflammatory and antihistaminic compound present in B. cuneata. 

Furthermore, in RAW 264.7 cell stimulated with LPS, moronic acid at 30 and 15 mg/mL a significant reduction of ON was observed (36% and 28% respectively) but had no significant effect on TNFα production.

Based on the findings, researchers said, "the organic extracts and isolated compounds from the aerial parts of B. cuneata had topical anti-inflammatory and antihistaminic activities in vivo, but in vitro only modified the production of ON in RAW cells. The results of this study validated the use of B. cuneata in folk medicine for the treatment of inflammatory diseases".

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of bacterial infectious diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Carotenoids Inhibit the Risk and Progression of Chronic Low-Grade Systemic Inflammation

Inflammation is a natural defense of the immune system that protects the body against the insult from the foreign invasion.

Acute inflammation occurs quickly or within the hours and lasts a few days or weeks and the damaged tissues are healed themselves after all pathogens are killed off.

However, if the immune system can not completely eradicate all invaders in the acute phase of infection, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

Chronic inflammation is a low-grade inflammation caused by the inability of the immune system to eliminate the invasive pathogens that occurred in the acute phase of infection.

Any inflammation that lasts more than 8 weeks is considered chronic inflammation.

Believe it or not, worldwide, 3 of 5 death are results from chronic inflammatory diseases like stroke, chronic respiratory diseases, heart disorders, cancer, obesity, and diabetes.

In the US, chronic inflammatory diseases affect more than 50 million Americans each year.

Diseases associated with low-grade inflammation include diabetes and cardiovascular obesity, depression, and chronic pain.

Conventionally, Nonsteroidal anti-inflammatory drugs (NSAIDs) are the most common medical drugs such as aspirin. ibuprofen in reducing inflammation associated with pain-relieving.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of chronic diseases, researchers examined the relationship between dietary patterns, circulating carotenoid and tocopherol concentrations, and biomarkers of chronic low-grade systemic inflammation in a 10-year longitudinal study of Scottish postmenopausal women.

According to the assessment of FFQ during 1997-2000 (n 3237, mean age 54·8 (SD 2·2) years). Participants (n 2130, mean age 66·0 (SD 2·2) years) returned during 2007-11 for follow-up, 
* The past 'prudent' dietary pattern showed a decrease of serum concentrations of hs-CRP and IL-6 (which decreased across the quintiles of the dietary pattern; P= 0·002 and P= 0·001, respectively; ANCOVA), 

* The Inflammatory biomarker concentrations were decreased markedly across the quintiles of carotenoids component score (P< 0·001 for hs-CRP and IL-6, and P= 0·016 for E-selectin, compared to the increase of inflammatory biomarkers in contemporary dietary patterns.

* More precisely, prudent dietary pattern and carotenoid component scores were negatively associated with serum hs-CRP concentration (unstandardised β for prudent component: -0·053, 95% CI -0·102, -0·003; carotenoid component: -0·183, 95% CI -0·233, -0·134).

Where, a prudent dietary pattern (which reflects a diet high in the intakes of fish, yogurt, pulses, rice, pasta, and wine, in addition to fruit and vegetable-rich with carotenoids diet.

In other words, the prudent diet with rich of carotenoids is associated with a reduced risk of chronic inflammatory diseases.

Taken altogether, carotenoids may be considered an adjunct therapy in the treatment of diseases associated with chronic low-grade inflammation, pending to the confirmation of the larger sample size and multicenter human study.

The study included 15 patients with chronic pancreatitis complicated by steatorrhea who consumed an average of 48 g of dietary fats a day were given 3 g of high-lipase pancreatin (lipase, 379,800 USP U/g), at each meal (the total daily dose is 9 g) for a mean duration of 28.5 days.

According to the fecal output and fecal fat neutral sterol, bile acid, and short-chain fatty acid excretion analysis,
* Pancreatin administration resulted in significant reductions (P < 0.01) in fecal output (from 243.2 to 149.1 g), excretion of fecal fat,

* Furthermore, animal sterols (from 816.3 to 604.6 mg), and short-chain fatty acids were reduced (from 12.3 to 3.9 g), and (from 52.6 to 18.5 mM), respectively.

Interestingly, the administration of pancreatin showed no marked changes in fecal excretion of beta-sitosterol (a plant sterol), bile acids, or the hydroxy fatty acid fraction.

In other words, Pancreatin improved patients with pancreatic insufficiency to absorb fat, protein, and energy.

Taken altogether, beta-Sitosterol may be considered a supplement for the prevention and treatment of obesity-related chronic inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Tocopherols, the Natural Anti-hypercholesterolemic Phytochemicals

The liver is a large internal organ of the human body that protects us by filtering the blood that comes from the digestive system before passing to other parts of the body.

In other words, if the liver was damaged due to toxicity, the blood will be toxic, affecting the function of all organs and tissues in the body.

The liver also produces cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Cholesterol found in our body is needed for our body to build cell walls, hormones, and vitamin D, and create bile salts that aid digestion. However, too much of them, in most cases due to the intake of animal protein can cause plaques built up on the arterial wall, the major cause of heart disease and stroke.

More precisely, if levels of low-density lipoprotein are high compared to high-density lipoprotein which returns the accessed cholesterol back to the liver, you have high blood cholesterol.

In other words, healthy blood cholesterol is any level with either less than or equal to 4 (low-density lipoprotein/ 1 (high-density lipoprotein).

According to the statistics, in 2015-2016, by age and gender, a total of 12.4 percent of adults aged 20 years and over had high cholesterol in the United States.

Hypercholesterolemia is a condition characterized by abnormally high levels of cholesterol in the bloodstream.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of diseases associated with high blood cholesterol, researchers examined the antioxidant defense activity of alpha-tocopherol in familial hypercholesterolemia and the effects of lipoprotein apheresis.

The study included a group of hypercholesterolemic patients (n = 14) that were treated by lipoprotein apheresis was compared to healthy adult normolipidemic controls to induce the decrease of tocopherol in serum and lipoprotein fractions with no change of the content of in membrane alpha-tocopherol.

According to the analysis of a simple isocratic reversed-phase HPLC method that has been developed and validated for the determination of alpha-tocopherol in human erythrocytes in a clinical setting, alpha-tocopherol significantly decreased lipoperoxidation as revealed by serum TBARS, representing end products of lipid peroxidation which increased from third day afterward and remained significantly higher in comparison to controls until the next LDL-apheresis.

More precisely, the phytochemical demonstrated an aggressive lipid-lowering procedure with lipoprotein apheresis associated with a favorable transient decrease of lipoperoxidation.

In other words, alpha-tocopherol inhibited the lipoperoxidation in the induction of hypercholesterolemia in the tested patients through its antioxidant activity.
 
After talking about other factors into account, researchers said, "Simultaneously the cell membrane-bound antioxidative defense mechanisms as reflected by the content of alpha-tocopherol in human erythrocyte membrane were not depressed in spite of its decreased plasma lipid carrier".

Taken altogether, tocopherols may be considered an adjunct therapy in the prevention and treatment of hypercholesterolemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
