Legune Fibers, the Best Source in the Prevention and Treatment of Hypercholesterolaemia

By Kyle J. Norton

A legume is a plant in the family Fabaceae cultivated all over the world for commercial profits.
Fiber is a type of indigestible carbohydrate.

In other words, fiber cannot be broken down into sugar molecules by the stomach and intestines.

Fiber has been found to regulate the body's use of sugars, keep hunger and blood sugar in check, thus reducing the risk of carbohydrate-related diseases such as diabetes.

According to the Mayo Clinic, daily intake of fiber may normalize bowel movements, softens stool, increase the weight and size of your stool, promote bowel health, lower levels of cholesterol, control glucose levels, etc.

On finding a potent food in the promotion of digestive health, researchers investigated the potential health benefits of legumes as a good source of dietary fiber.

The study included examing the 6 to 10 local legume including cowpeas, mung beans, pole sitao, chickpeas, green peas, groundnuts, pigeon peas, kidney beans, lima beans, and soybeans.

* In the mineral assay,  lima beans (9.5 (sem 0.1)) showed the highest Fe availability and kidney beans (49.3 (sem 4.5)) and pigeon peas (75.1 (sem 7.1)) showed a highest Zn and Ca, the highest availability.

*In cholesterol assays, kidney beans showed significant reductions of in total (6 %) and LDL-cholesterol (9 %), compared to other legumes.

Based on the findings, researchers said, "mineral availability from legumes differs and may be attributed to their mineral content, mineral-mineral interaction and from their phytic and tannic acid content; legumes are considered low-GI foods and have shown potential hypocholesterolemic effects".

Taken altogether, legume fibers may be considered functional foods for the prevention and treatment of hypercholesterolemia,  pending to the confirmation of the larger sample size and multicenter human study.

3,3'-Diindolylmethane May Induce immune-Toxicity if Overdosed

By Kyle J. Norton

The immune system is the set of cells and their activity against antigens or infectious agents that comprises the body's defense system against diseases. The immune system does a great job of keeping people healthy and preventing infections.

It is one of the body's defense system against diseases.

Immune dysfunction is a chronic condition, depending on the parts of the immune system affected.

There are 2 types of immune dysfunction
* The underactive immune system is also known as the weakened immune system. People with the underactive immune system are susceptible to infection, recurrent infections.

The most common symptoms of the underactive immune system are infection including frequent and recurrent pneumonia, bronchitis, sinus infections, ear infections, meningitis or skin infections and Inflammation and infection of internal organs.

However, blood disorders, such as low platelet counts or anemia and digestive dysfunction are also some of the common symptoms found in patients with the underactive immune system.

* The overactive immune system is also known as autoimmune diseases. In this case, patients suffer diseases associated with the immune system that attacks the body tissues.

Immunomodulation is an action with a profound and positive effect in regulated immune response and function through activation of antibody formation or the inhibition of white blood cell activity.

On finding a potential plant that enhances immune functioning, researchers compared the effects of 3,3'-Diindolylmethane (DIM), a major product of indole-3-Carbinol isolated from genus Brassicaon various immune-modulatory activity in animal models and in vitro studies.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

According to the tested analysis, the DIM treatment showed multiple immunotoxic effects in decreasing in various immune cells (F4/80(+), CD11c(+), CD19(+), and CD3(+) cells) in the spleen.

Furthermore, the DIM treatment also induced the expression of splenic white pulp atrophy, an increase in immune cell apoptosis, and decreased expression of various toll-like receptors (TLRs), a class of proteins that play a key role in the innate immune system.

The action of 3,3'-Diindolylmethane that modulates the immune functioning was attributed to the promotion of the caspase-3 activity and ratio of proteins associated with cell deaths and survivals in regulating programmed cell death or apoptosis.

Based on the findings, researchers said, "This new information about the immunotoxic roles of DIM in a newborn mouse model may provide valuable clues for the development of a safe supplement, especially one designed for human infants".

Taken altogether, DIM supplements may process immune toxic activity if overdosed.

Herbal Coltfoots (Tussilago farfara) in the Prevention of Liver Cancer

By Kyle J. Norton

Cancer is a class of chronic and medical conditions caused by the alternation of cell DNA.

The liver organ is one of the organs with a lack of nerve tissue, therefore liver cancer does not induce pain until the later stage.

Liver cancer is a medical condition characterized by cell growth irregularly in the liver tissue.

The causes of liver cancer are associated with the alternation of cells DNA which normally begins in the cells on the surface of the inner lining.

Liver cancer at the early stage no not cause any symptoms. However, at the advanced stage, liver cancer cells can cause a wide range of symptoms including jaundice, abdominal swelling and pain, and dark urine and white stool accompanied by the general symptoms of cancers such as unintended weight loss, loss of appetite, chronic fatigue and tiredness and gastrointestinal discomforts.

The most common cause of liver cancer is infected by the hepatitis virus. However, in North America, due to overexpression of overweight and obese of the population, liver cancer may be slowly progressed from nonalcoholic fatty liver disease through cirrhosis to liver cancer.

According to the statistics provided by the Canadian Cancer Society, every year, approximately, 1,900 men and 580 women are diagnosed with liver cancer, causing the death of 950 men and 270 women.

In 2017, 2,500 Canadians were diagnosed with liver cancer and 1,200 Canadians died from liver cancer.

Compared to the early stage of other cancer, the 5 years survival rate in the early stage diagnosis is only 31% and 43% of people with liver cancer diagnosed at stage 0 and 1.

On finding a natural ingredient for the protection of the liver functioning, researchers examined the effect of herbal on liver cancer, particularly in the induction of apoptosis through activation of the TRAIL pathway.

Coltsfoot(Tussilago farfara) is a perennial herbaceous plant, genus Tussilago, belonging to the family Asteraceae, native to Europe and Asia. The herb has been used over thousands of years to treat asthma, various coughs, bronchial congestion, respiratory disorders, headaches and obstruction in the nasal passage, etc.

Where TRAIL is a cytokine that is produced and secreted by most normal tissue cells, with a function to cause cell death.

According to the results of the analysis, the administration of Tussilago farfara L. (TF) exerted the effects of novel Indazole‐based MKK7–TIPRL inhibitor against liver cancer as TRAIL sensitizers.

Furthermore, co-treatment of human liver cells (Huh7 cells) with TF and TRAILdemonstrated a strong activity in the induction of apoptosis via inhibition of the MKK7-TIPRL interaction, a signal transduction pathway associated with cell death.

Based on the experimental results, researchers said, "TF as a novel TRAIL sensitizer, unveiling a potentially novel therapeutic strategy in cancer therapy".

Taken altogether, coltsfoot may be considered a remedy for the prevention and treatment of liver cancer, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Angelica Exerts Neuro-Protective Activity Against Cerebral Ischemia Injury

By Kyle J. Norton

Brain ischemia is a condition caused by insufficient blood flow to the brain to meet metabolic demand, leading to irreversible brain damage in severe cases.

In other words, brain ischemia-induced the death of neurons of the affected tissues.

A neuron or nerve cell made of a cell body (also called soma), dendrites and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord and peripheral nerves that play an essential role to process and transmit information.

The function of neurons can be classified into 3 types
* The sensory neurons found within the nervous system that convert external stimuli from the organism's environment into internal electrical impulses. In other words, sensory neurons are activated by sensory input from the environment, including physical input of sound, touch, heat, or light.

*The motor neurons transmit impulses from the spinal cord to skeletal and smooth muscles,. In other words, motor neurons directly control all of the muscle movements.

* The interneurons connect the motor and sensory neurons. In other words, the interneurons transfer signals between sensory and motor neurons.

Brain ischemia can be classified into
* Focal ischemia is a condition caused by damage to a specific region of the brain; and

* Global ischemia is a condition of damage that affects the wide areas of brain tissue, leading to symptoms of impairments in vision, body movement, and speaking.

Dr. Taoufik E, the lead scientist, in the study of the neuron death following a stroke, may help to develop effectively neuroprotective strategies, said, "The two principal models for human stroke are induced in rodents either by global or focal ischemia. In both cases, blood flow disruptions limit the delivery of oxygen and glucose to neurons causing ATP reduction and energy depletion, initiating excitotoxic mechanisms that are deleterious for neurons".

And, "Both in vitro and in vivo studies, targeting a single death mechanism either by the inhibition of death-inducing molecules or the overexpression of antiapoptotic components in neurons, have shown tremendous neuroprotective potential".

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China. The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

On finding the potential and natural remedy for the treatment of post cerebral ischemia injury,
evaluated the effects of Angelica Sinensis extract [Dang Gui (DG)] administered before 60min of middle cerebral artery occlusion followed by 3 days of reperfusion.

According to the results of DG intraperitoneally administered at a dose of 0.25 g/kg, 0.5 g/kg (DG-0.5g), or 1 g/kg (DG-1g) 30 min before the onset of cerebral ischemia
* DG-0.5g and DG-1g pretreatment effectively attenuated cerebral infarct and improved neurological deficits.

* DG-0.5g and DG-1g pretreatment significantly reduced proteins associated with the expression inflammatory response after injury, depredation, and apoptosis of neurons and pathways involved in the reduction of oxidative stress.

Furthermore, DG-0.5g and DG-1g pretreatment also protected the neurons against additional damage induced by cerebral ischemia injury by inhibiting the production of pro-inflammatory cytokines and
cell apoptosis due to oxygen deficiency and improving the kinases in neurotransmission.

More precisely, DG-0.5g and DG-1g pretreatment provided neuroprotective effects against astrocyte-mediated cerebral infarction by activating angiogenic and anti-apoptotic signaling.

Based on the findings, Angelica Sinensis in a dose depend-manner may be used as a functional remedy for the protection of further neuron damage in patients with cerebral ischemia injury, pending to the confirmation of the larger sample size and multicenter human study.

Cranberry Inhibited Oxidation Induced Liver Mitochondrial Damage

By Kyle J. Norton

The liver is the largest internal organ located in the upper right portion of the abdomen, beneath your diaphragm and above your stomach.

The liver not only plays a critical role to filter the blood coming from the intestine before passing them to other parts of the body but also protects our body against toxin by detoxifying chemicals and metabolizes drugs.

Additionally, the liver also inhibits oxidative stress by producing the antioxidant enzyme that counters the expression of free radicals in the body.

Mitochondria are power generators of the cell found abundantly in the liver and play a critical role in converting oxygen and nutrients to power the cell's metabolisms.

In other words, mitochondria convert nutrients from the cell into energy for cell functioning.

Mitochondrial permeability transition (MPT) and the mitochondrial generation of oxidative stress have been found to initiate hepatic mitochondrial dysfunction caused by hepatocellular necrosis and apoptosis.

Liver mitochondrial damage has been found to orchestrate the symptoms, jaundice, vomiting, weakness, low blood sugar levels and sleepiness and an increased amount of lactic acid in the blood.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of diseases associated with oxidative stress, researchers investigated the cranberry flavonoids prevent toxic rat liver mitochondrial damage.

The study included rat induced mitochondrial damage by chronic carbon tetrachloride-induced intoxication.

According to the tested analysis, the treatment of cranberry flavonoids (7 mg/kg) protected the liver mitochondrial damage by inhibiting the fragmentation, rupture and local loss of the outer mitochondrial membrane.

Moreover, cranberry flavonoids also acted as free radical scavengers that inhibit oxidative stress in inducing liver mitochondrial damage by effectively scavenged nitric oxide (IC50  = 4.4 ± 0.4 µg/ml), superoxide anion radicals (IC50  = 2.8 ± 0.3 µg/ml) and hydroxyl radicals (IC50  = 53 ± 4 µg/ml).

In liposomal membranes and glutathione oxidation in liver erythrocytes, flavonoids isolated from cranberry exerted some protective activity against lipid peroxidation induced by UV irradiation or organic hydroperoxides.

Based on the findings, researchers wrote, "The hepatoprotective potential of cranberry flavonoids could be due to specific prevention of rat liver mitochondrial damage. The mitochondria-addressed effects of flavonoids might be related both to radical-scavenging properties and modulation of various mitochondrial events".

Taken altogether, cranberry combined with other herbal medicine for the protection of liver mitochondrial damage, pending to the confirmation of the larger sample size and multicenter human study.

Retinal degeneration is a condition of retinopathy associated with the gradual loss of neurons near the inner surface of the retina of the eyes.

Most cases of retinopathy are caused by abnormal blood flow.

The most common forms of retinal degeneration are macular degeneration which is a condition when the small central portion of the retina, (the macula) deteriorates and retinitis pigmentosa (RP) which is a rare genetic disorder involved a breakdown and loss of cells in the retina.

Retinal degeneration disease has been found to have a strong impact on daily life, ranging from performing basic functions to personal independence and mental health and vision loss.

According to the statistics, the overall prevalence of age-related macular degeneration (AMD) is in the range of 1.38%-1.55%) and retinitis pigmentosa is 1 in 3,500 to 1 in 4,000 individuals.

Age-related macular degeneration is a medical condition that causes irreversible blurring of the central vision in people over age 60.

The elderly at the early stage of age-related macular degeneration is asymptomatic. However, some patients may experience symptoms of slightly blurred central vision. Most cases of the early stage of AMD were diagnosed during routine eye health examinations.

Over time, age-related macular degeneration may affect the vision including the reading as the blurred area increased in size.

The advanced age-related macular degeneration has been found to induce irreversible blindness and visual impairment, according to the epidemiological studies.

Turmeric is a perennial plant in the genus Curcuma, belongings to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

In the urgency to discover a natural ingredient for the prevention of retinal degeneration, researchers examined the beneficial effects of curcumin on multiple retinal diseases.

Herbal Ashwagandha Exerts Cytotoxicity Against Melanoma in Vitro

By Kyle J. Norton

Cytotoxicity is the process to cause cell death through toxins.

In other words, cell cytotoxicity is the process to use certain chemicals or mediator cells to destroy living cells.

In normal, cytotoxicity can cause the death of healthy cells.

Most cases of cytotoxicity of a substance are to determine the potential toxicity of the quality of the substance, including plant extracts or biologically active compounds isolated from plants.

In other words, the aim of the cytotoxic studies is to find the minimal quality that is essential for the successful development of pharmaceutical or cosmetic preparation that induces cellular toxicity for the treatment of cancer.

For example, cytotoxicity induced by oxidative stress due to the imbalanced ratio of free radicals and antioxidant enzymes produced by the body can cause protein, lipid and cellular damage that leads to chronic low-grade inflammatory diseases and cancer.

However, stimulation of ROS and antioxidants production to induce cytotoxicity to the tumors can also cause cancer cell death without damage to nearby healthy cells.

Cancer is a group of medical and chronic diseases characterized by cell growth disorderly and uncontrollably in the tissue of specific organs in the body.

Over time, the alternative cells induced tumor may become progressively worse and damage other healthy tissues, depending on the grade of the tumor.

At the later stage, the most invasive tumor can induce secondary metastasis by spreading to other organs in the body via lymph or blood.

According to the National Cancer Institute, approximately 38.4% of men and women will be diagnosed with cancer at some points during their lifetimes, according to the data from 2013–2015.
In cancers, cytotoxicity can be benefits to humans by destroying the cancer cells and prevent them from proliferation.

Melanoma is a kind of skin cancer but less common to other skin cancer as a result of malignancy of melanocytes, which is the cell produced dark pigment for your skin.

Approximate 120,000 new cases of melanoma in the US are diagnosed in a year. melanoma tends to spread along the skin in most of the cases, but also can grow down to the deeper layers of the skin.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound that processes anti-cancer activity, researchers examined the cytotoxic effect of Withania root extract on human malignant melanoma A375 cells.

The structured changes also were observed in the A375 cells treated groups compared to untreated or vehicle-treated control.

Treatment of Withania root extract also induces A375 cells apoptosis, according to the ladder of fragmented DNA in treated cells.

Based on the findings, researchers said, "might be said that the crude water extract of Withania somnifera has a potent cytotoxic effect on human malignant melanoma A375 cells".

Taken altogether, Ashwagandha may be considered a cytotoxic herb for the treatment of melanoma, pending to the result of larger sample size and multicenter study
