Herbal Ashwagandha, the Potent Anti Cancer Activity in Vitro and Vivo

Cancer is a class of diseases associated with abnormal cell growth in a specific tissue of an organ, due to the alternation of DNA.

A primary tumor or original tumor is a tumor that is at the original site where it first arose where tumor progression began and proceeded to yield a cancerous mass.

Cancer cells that grow to its tumor size, can invade and kill their neighbor cells by producing abnormal or excessive signaling chemicals (hormones or cytokines) which upset the normal chemical balance of the body and alter the metabolic pathways, leading to disordered and violent cellular behavior.

Most cases of cancer begin in the cells on the surface of the inner lining of tissue. At the early stage, due to the small size of the tumor, most patients are asymptomatic.

However, at the advanced stage, the oversize tumor can press on the nearby nerve cells and blood vessel, leading to bleeding and localized pain and symptoms accompanied by the general symptoms shared by other types of cancer, such as unintended weight loss.

Furthermore, at this stage, cancer cells can also travel a distance away from the originated site to infect other healthy cells and tissue, leading to secondary metastasis.

Localized symptoms are totally depending on the types of cancer.

In cell cycle division perspective, under normal circumstance, cell growth is a natural process. Every day millions of cell grow to replace millions of dead cells and keep our body in balance and healthy.

Sometimes, acceleration of cells growth is necessary to prevent further damage to the body organ, such as cut, injure, etc., but after fulfilling their purposes, the growth of cells are back to a normal rate again.

However, if the DNA of a cell is altered, it can multiply indefinitely comapred to limit cell cycle division in a normal cell.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

With an aim to discover a natural compound for the treatment of cancer, researchers examined the anticancer activity in the water extract of Ashwagandha leaves (ASH-WEX) in vitro and in vivo.

According to the results from the tested analysis, ASH-WEX not only demonstrated a cytotoxic effect on cancer cells selectively, and also induced tumor suppression in vivo.

 Triethylene glycol (TEG), an active compound found in the extract processed a strongest anti-cancer property compared to other chemical constituents.

Additionally, ASH-WEX activated the genes associated with tumor suppression in cancer cells.

Furthermore, the extract induced the increase of protein that regulates mitosis and decrease of proteins associated with cell cycle division without causing harm to normal cells.

Moreover, ASH-WEX also inhibited the matrixes involved in cancer metastasis, without affecting the normal cells.

Based on the findings, researchers wrote in the final report, "ASH-WEX and TEG have selective cancer cell growth arrest activity and hence may offer natural and economic resources for anticancer medicine".

Taken altogether, Ashwagandha may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of cancer, pending to larger example size and multicenter human study.

Lycopene Inhibits Parameters Induced Post Myocardial Infarction Complication

Acute brain infarction is the severe and medical emergency characterized by the obstruction of the blood supply to the brain, causing local death of the neurons in the brain. Prompt treatment can minimize brain damage and potential complications.

The occurrence and extent of brain infarction are asscoaited with three basic factors, including the site of arterial occlusion, the rapidity of arterial occlusion, and the presence or absence of collateral circulation.

In other words, brain infarction can be classified into myocardial infarction due to plaque rupture and myocardial oxygen supply-demand imbalance secondary to other acute illnesses.

The exact causes of a blood clot to induce acute brain infarction are unknown. However, the trouble with speaking and understanding, confusion, sudden numbness, weakness or paralysis in your face, arm or leg, headache and the trouble with walking are some of the most common symptoms found in patients with post-myocardial infarction.

Depending on the severity of the acute infarction, in serious cases, patients may also experience symptoms, paralysis of the face, arm or leg, trouble with seeing in one or both eyes.

Although, there are many factors associated with the risk of acute infarction. some researchers suggested aging may have a strong and negative implication on acute infarction.

Dr. Amelia Carro, the lead scientist wrote, "Data from 163,140 hospital admissions of Medicare beneficiaries age > or =65 years between 1994 and 1996 with AMI at U.S. hospitals were evaluated for differences in clinical characteristics and mortality across five age-based strata (in years): 65 to 69, 70 to 74, 75 to 79, 80 to 84 and > or =85".

And, "Our data indicate significant age-associated differences in clinical characteristics in elderly patients with AMI, which account for some of the age-associated differences in mortality".

Lycopene is a phytochemical found in tomato in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas.

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.

On finding a natural compound for the treatment in the post-heart attack, researchers examined supplementation on cardiac remodeling after myocardial infarction (MI).

The study included male Wistar rats assigned randomly into four groups: the sham group (animals that underwent simulated surgery) that received a standard chow (S; n=18), the infarcted group that received a standard chow (MI; n=13), the infarcted group supplemented with lycopene (1 mg of lycopene/kg body weight/day) (MIL; n=16) and the infarcted group supplemented with tomato (MIT; n=16).

At the end of 3 months of the experiment, based on the assay analysis, researchers found that
* The groups MIL and MIT showed decreased interstitial fibrosis induced by infarction.

* Tomato treated group attenuated the hypertrophy induced by MI.

* Lycopene reduced the expression of proteins and pathway associated with pro-inflammatory cytokines such as TGF-β1, and NF-κB and TNF-α respectively, collagen I, collagen III associated with the initiation of fibrous tissues.

* Most importantly, lycopene also ameliorated the level of ventricular remodeling post-MI.

Based on the findings, researchers said, "Lycopene may inhibit the NF-κB signaling pathway thereby reducing the inflammatory response and cardiomyocyte apoptosis post-MI, which could be a key mechanism of lycopene in attenuating ventricular remodeling".

Taken altogether, lycopene found in tomato may be considered supplements for the and treatment of in cardio-remodeling in post-myocardial infarction, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Red Cabbage Improves Sleep Quality

Most of us experience sleep disturbance sometimes. However, chronic sleep disorders are the changes in the way that you sleep.

Sleep disorders can be acute or chronic. Acute sleep disorder caused by specific or nonspecific event can resolve quickly in short times after the event is addressed.

Ignoring sleep disorders have been found to induce poor health, poor mood, and lethargy, and sometimes have a negative impact on motivation, relationships, and job performance in daily life.

Some people who use over the counter medicine or herbal medicine without consulting the experts may experience side effects and in some cases, worsening sleep problems.

Most people with chronic sleep disorder experience symptoms of excessive daytime sleepiness, irregular breathing or increased movement during sleep, and difficulty falling asleep.

Sleep quality can be measured by examing the.way that you sleep, including the time period that you need to fall to sleep and how many times you have awakened and drifted back to sleep during night sleep.

The National Sleep Foundation’s inaugural Sleep Health Index showed, 45 % of Americans have poor or insufficient sleep that affects their daily activities at least once in the past seven days.

Depending on the types of sleep disorder, the most common causes of chronic sleep disorder are caffeine consumption, lack of regular exercise, poor stress management and the increase in aging.

Dr. Keisuke Suzuki, the lead scientist at the Dokkyo Medical University in the investigation of sleep disorders in the elderly wrote, "Compared with younger people, elderly people show age‐related sleep changes, including an advanced sleep phase and decreased slow‐wave sleep, which results in fragmented sleep and early awakening".

And, "Multiple etiologies contribute to insomnia in the elderly, consistent with the observation that elderly people are likely to have comorbid conditions and medications".


Red cabbage is a species of Brassica Oleracea, belongings to the family Brassicaceae with unknown origin. Today, the plant is cultivated commercially for its thick, slightly bitter edible leaves.

 On finding the potent compound for the improvement of sleep quality, researchers investigated the effect of hydroalcoholic extract of red cabbage and its fractions on sleeping behavior in mice.

According to the tested analysis,  the extract increased sleep duration at doses of 50–200mg/kg (P < 0.001),  comparable to that of diazepam (3mg/kg) and those of control group.

The extract decreased sleep latency was attributed to the ethyl acetate fraction LD50 value in the red cabbage extract of 2.4g/kg. 

Most importantly, no toxic effect on the viability of cultured neuronal cells (PC12) was found in the tested hydroalcoholic extract.

Collectively, researchers said, "red cabbage potentiates pentobarbital hypnosis without any toxic effect. The main component(s) responsible for this effect is most likely to be an intermediate polar agent(s) such as flavonoids, which are found in ethyl acetate fraction of this plant".

Taken altogether, red cabbage may be considered a functional food for the promotion of sleep quality, pending to the confirmation of the larger sample size and multicenter human study.

Cranberry, the Potent Free Radical Scavenger

Free radicals are highly reactive atoms associated with an unpaired electron in the outermost ring produced internally by our body cell metabolism and externally by the environmental factors, including the air which we breathe and the food we eat.

In other words, any chemicals compounds or molecules which process oxygen atom can be free radicals after chemical interaction.

Believe it or not, free radical in moderate amount is important for our body functions such as NO cardio-vessel relaxation. However, in high levels, free radicals have been found epidemiologically to damage the lipid, and protein and induce the alternation of healthy cell DNA.

Out of all damages caused by overexpression of free radicals, some researchers suggested that lipid peroxidation induced by free radical has a strong implication in the pathogenesis of various disorders and diseases, including free radical-mediated chain reaction, enzyme-mediated oxidation, and nonradical, non-enzymatic oxidation. 

These chain of reaction can not be stopped until the electrons on the outermost ring of free radical are paired or inhibited by antioxidants.

Free radical scavengers are antioxidants that process a specific function to prevent the forming of free radicals by removing them before they can damage vital components of the cell.

Dr. Slemmer JE, the lead scientist in the examination of free radical scavengers for the treatment of stroke wrote, "The overproduction of reactive oxygen species (ROS) and reactive nitrogen species (RNS) is a common underlying mechanism of many neuropathologies, as they have been shown to damage various cellular components, including proteins, lipids, and DNA".

And "several acute and chronic pharmacological therapies that have been extensively studied in order to reduce ROS/RNS loads in cells and the subsequent oxidative stress, so-called "free-radical scavengers".

These results strongly suggested that antioxidants with free radical scavenging activity protect the body against the damage of various cellular components, including proteins, lipids, and DNA alternation in the initiation of diseases.


Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of diseases associated with oxidative stress, researchers examined the cranberry antioxidative and protective effects of cranberry flavonoids in free radical-generating systems on mitochondrial ultrastructure.

In radical-generating systems, cranberry flavonoids effectively inhibit the generation of free radical nitric oxide (IC50  = 4.4 ± 0.4 µg/ml), superoxide anion radicals (IC50  = 2.8 ± 0.3 µg/ml) and hydroxyl radicals (IC50  = 53 ± 4 µg/ml). 

1,1-diphenyl-2-picrylhydrazyl radicals (DPPH) was scavenged at the concentration of 2.2 ± 0.3 µg/ml with the IC50.

Furthermore, cranberry flavonoids exerted the antioxidant and free radical scavenging activity against lipid peroxidation in liposomal membranes and glutathione oxidation in erythrocytes induced by UV irradiation or organic hydroperoxides.

Moreover, the phytochemical compound also decreased the rigidity of the outer leaflet of the liposomal membranes. 

In the liver, cranberry flavonoids also displayed hepatoprotective potential by preventing rat liver mitochondrial damage.

Collectively, researchers said, "The mitochondria-addressed effects of flavonoids might be related both to radical-scavenging properties and modulation of various mitochondrial events".

Taken altogether, cranberry may be considered free radical scavenging remedy, pending to the confirmation of the larger sample size and multicenter human study.

Whole Food Tomato Apoptoses Liver Cancer Better Than of Doxorubicin and Camptothecin in Vitro

Cancer is a class of diseases caused by cells growth disorderly and uncontrollably in our body.

Liver cancer is a chronic and medical condition associated with irregular cell growth in the liver tissues.

The liver is the largest internal organ that only filters out blood from the intestine before passing them to other parts of the body, but also produces bile to aid digestion, build strong cell walls and vitamin D and steroid hormones.

The liver jointed with kidney and skin is considered 3 detoxed organs in the human.

Most common liver cancers are
* Hepatocellular carcinoma, is a most common liver cancer accounting, accounting for 75 percent of all cases caused by the DNA alternation of hepatocytes, predominant liver cells.

Epidemiologically, primarily hepatocellular carcinoma has been found to metastasize to pancreas, intestines, and stomach.

* Cholangiocarcinoma, also known as bile duct cancer, is caused by abnormal cells growth in the small, tube-like bile ducts in the liver, accounting to 10 to 20% of all liver cancer cases.

Out of many prevalent factors that cause the onset of liver cancer, some researchers suggested that the promotion of a high-fat diet over the past few decades may have a strong impact to increase the risk of liver cancer.

Dr. Qiu-Ye Lan, the lead scientist wrote, "Healthy dietary patterns may prevent many chronic diseases and is emphasized by 2015 US dietary guideline, but it remains unclear which dietary patterns may be a benefit to prevention of primary liver cancer (PLC)"

And, "Our findings suggest that the UPDP (an urban prudent dietary pattern (UPDP)) and TCDP (traditional Cantonese dietary pattern (TCDP) )were associated with lower, whereas the MPFP with the higher, risk of PLC".

Tomato is red, edible fruit, genus Solanum, belongings to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

In the urgency to discover healthy foods for the treatment of liver cancer, researchers examined the effect of the tomato antiproliferative activities against human colon (HT29) and liver (HepG2) cancer cells.

Application of the tetrasaccharide tomato glycoalkaloid alpha-tomatine, the trisaccharide beta(1)-tomatine, the disaccharide gamma-tomatine, the monosaccharide delta-tomatine, and their common aglycon tomatidine showed better inhibition of liver cancer cell compared to the tested colon cell line.

Additonally, alpha-tomatine isolated from tomato at a concentration of 1 microg/mL demonstrated  a greater effect against liver carcinoma cells compared to those of anticancer drugs doxorubicin and camptothecin

Furthermore, alpha-tomatine also inhibited normal human liver HeLa (Chang) cells without inducing any side effects.

Also in vitro, according to the tested assays,
* Alpha-tomatine isolated from green tomatoes induced liver cancer cells death compared to little inhibition of the cells by the three low-tomatine red tomato extracts.

* Alpha-Tomatine from tomato also was highly effective in inhibiting all of the liver cell lines, compared to had little cell inhibition of ehydrotomatine, tomatidenol, and tomatidine.

Collectively, the results not only showed the susceptibility to destruction varies with the nature of the alkaloid and plant extract but also how phytochemical in plant worked synergistically in cancer inhibition.

Dr. Friedman M, the lead scientist said, "These findings extend related observations on the anticarcinogenic potential of glycoalkaloids and suggest that consumers may benefit by eating not only high-lycopene red tomatoes but also green tomatoes containing glycoalkaloids".



Taken altogether, tomato processed a high amount of lycopene may be considered a dietary supplements for the prevention and treatment of liver cancer, pending to large sample size and multicenter human study.


Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Futhermore, in vitro, quercetin exerted multiple actions on the NO-guanylyl cyclase pathway associated with the smooth muscle tone in bronchi or vessels, endothelium-derived hyperpolarizing factor(s) and endothelin-1 involved in vasoconstriction and endothelial function.

Moreover, the phytochemical also protected endothelial cells against apoptosis.

In vivo, quercetin prevented endothelial dysfunction by reducing blood pressure, oxidative stress and end-organ damage in hypertensive animals.

In other words, regular intake of quercetin improves the endothelium functioning against the risk of VED inducing cardiovascular disease, particularly in patients with hypertension and ischemic heart disease.

Taken altogether, onion processed a higher mount of quercetin may be considered a remedy for the prevention and treatment of vascular endothelial dysfunction, pending to the confirmation of the larger sample size and multicenter human study.

Inflammation is a natural defense of the immune system that protects the body against the invasion of harmful stimuli and pathogens.

Anti-inflammatory activity is the process to reduce inflammation or swelling without affecting the function of the immune system.

In the acute phase of infection, the immune first line of defense, the white blood cells, macrophages stimulates the production of blood platelet to cover the wound or cut and proinflammatory cytokines to destroy all invaders.

Most cases of acute infection are stopped at this phase and the wound is healed itself quickly.

However, if the wound is too large, then the recovery may take a longer period and through 4 overlap recovery stages. Any interruption in the overlapped recover stage may cause the formation of scars.

 Moreover, overproduction of proinflammatory cytokines that kills harmful pathogens may cause damage and death of nearby healthy cells, leading to the formation of scars, such as scar in the liver caused by hepatitis infection.

According to the tested differentiation of rat induced by injection of 0.2 mL of λ-carrageenin in the pleural cavity pretreated by indicaxanthin at 2 μmol/kg,
* Pretreatment of the phytochemical inhibited the release of pro-inflammatory PGE(2) (91.4%), NO (67.7%), IL-1β (53.6%), and TNF-α (71.1%) induced by carrageenan-induced rats.

* Furthermore, enzymes and protein associated with the production of proinflammatory cytokines such as IL-1β (34.5%), TNF-α (81.6%), iNOS (75.2%), and COX2 (87.7%) mRNA were also inhibited by the pretreatment of indicaxanthin.

* iNOS (71.9%) and COX-2 (65.9%) enzyme expressions involved in the inflammatory processes that recruits the leukocytes also inhibited by the bioactive compound.

* Moreover, enzymes associated with the induction of free radicals chain reaction were also curbed by the indicaxanthin at a time- and dose-dependently.

Pharmacokinetically, the most efficacy in inhibition of inflammation was found to have a maximum 0.22 ± 0.02 μmol/L (n = 15) plasma concentration of indicaxanthin, with a half-life of 1.15 ± 0.11 h.

Based on the findings, researchers said, " this dietary pigment has the potential to improve health and prevent inflammation-based disorders".
