Phytochemical Quercetin Isolated from Onion Inhibits Parameters Involved in Oral cancer Developing

By Kyle J. Norton

Oral cancer is a medical condition of malignant cell growth in the tissue located in the oral cavity, including the tissue of the lips or the tongue, cheek lining, floor of the mouth, gums (gingiva) and the roof of the mouth (palate).

Most common types of oral cancer are
* Squamous cell carcinoma
Squamous cell carcinoma is one of the most common types of oral cancer accounting over 90% of all cases and arisen from the squamous cell layer in the lining of the oral cavity and oropharynx.

* Verrucous carcinoma
The low grade and rarely spread verrucous carcinoma is accounted for only approximately 5% of all cases. The cancer can spread deeply into the surrounding tissue.

* Minor salivary gland cancers
The cancer is originated from the salivary gland accounted for less than 1% of all cases.

According to the statistics, approximately, 53,000 Americans will be diagnosed with oral or oropharyngeal cancer in 2019.

The disease also causes the death of over 9,750 patients, roughly 1 person per hour, 24 hours per day.

The grades of oral cancer are depending on the tendency of spreading.  Low-grade cancers usually grow more slowly and are less likely to spread while high-grade cancer indicates otherwise.

The 5-year survival rate of the localized stage of oral cancer is 84%. However, If cancer has spread to surrounding tissues or organs and/or the regional lymph nodes, the overall 5-year survival rate is 65%.

The survival rate is even lower (39%) If cancer has spread to a distant part of the body,

The onion is a plant in the genus Allium, belongings to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

In the urgency to find a potential compound for the treatment of oral cancer, researchers investigated the potential effects of quercetin, a principal flavonoid compound in onions anti-oral cancer activities.

On SAS human oral cancer cells, quercetin significantly reduced wound closure of SAS cells in culture plates after 12- and 24-h treatments.

Quercetin inhibited the expression and activity of matrix metalloproteinase (MMP)-2 that promotes cancer cell growth and proliferation and (MMP)-9 involved in cancer cells angiogenesis and metastasis.

Additionally, quercetin also inhibited the proteins associated with the promotion of forming new blood vessel in SAS human oral cancer cells.

Furthermore, quercetin also blocked the expression of enzymes associated with the pathogenesis of many solid tumors through its inflammatory activation.
Moreover, proteins involved in cellular interactions with the extracellular matrix that cause tumor initiation, progression, and metastasis were also inhibited by the injection of quercetin.

More importantly, quercetin also exhibited the gene expression of tumor suppression such as P53 and apoptosis.

In other words, quercetin protects the oral cells by blocking the proteins involved in the signaling expression of tumor initiation, progression, and metastasis such as Ras homolog gene family, member A (RhoA) and protein kinase C (PKC).

Taken altogether, onion may be considered a remedy for the prevention and treatment of oral cancer, pending to the confirmation of the larger sample size and multicenter human study.

Phytochemical Betanin, the Anti-Inflammatory Natural Compound

By Kyle J. Norton

Acute inflammation is a condition that lasts in the short term caused by an immune system response to the injury.

Symptoms of pain, redness, immobility (loss of function), swelling and heat may occur within minutes or hours, depending on the size and penetration of the wound.

Overproduction of proinflammatory cytokines induced by the immune response may cause the damage or death of the nearby healthy cell, leading to the formation of scars.

The symptoms may disappear sooner in people with a stronger immune system compared to the elderly and the wound is covered quickly in a few days.

However, if the wound is quite large, then some stitches may be needed and the wound recovery will go 4 overlapped stages, including
* The stage of hemostasis is the process of the wound being closed by clotting.
* The second stage involved in the production of inflammatory cytokines in the prevention of infection.
* The third stage is the proliferation in which the wound is rebuilt with new tissues made up of collagen and extracellular matrix.
and
* The masturbating stage in which collagens are converted, the recovery is completed and the wound fully closed.

Any interruption of the overlapped stage may induce the formation of scar similar to those of overproduction of inflammatory cytokines.

Unfortunately, if the immune system can not kill off all invaders over a short period of time normally between 3 to 8 weeks, it will compromise, leading to chronic inflammation in facilitating several chronic diseases such as cardiovascular disease and diabetes.

Betanin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

With an aim to discover a potential ingredient for the treatment of diseases associated with inflammation, researchers investigated the effects of natural pigment betanin on oxidative stress and inflammation in the kidney of paraquat-treated rat.

Selected rats were injected by Paraquat intraperitoneally to induce renal damage, then randomly divided into four groups: a control group, a paraquat group, and two paraquat groups that were treated with betanin at 25 and 100 mg/kg/d three days before and two days after paraquat administration.

Betanin treated group showed a significant effect in alleviating the paraquat-incurred acute kidney injury, evidenced by histological improvement, reducing serum and urine markers for kidney injury compared to the untreated group.

The bioactive compound also attenuated the inflammation induced by the injection of Paraquat observed by the reduced expression of inflammatory proteins.

Furthermore, betanin also inhibited the kidney oxidative stress through its antioxidant activity.

Based on the finding, researchers said, "betanin may have a protective effect against paraquat-induced acute kidney damage. The mechanisms of the protection appear to be the inhibition of oxidative stress and inflammation".

Taken altogether, betanin may be considered supplements for the prevention and treatment of diseases associated with inflammation, pending to the confirmation of larger sample size and multicenter human study.

Intake of betanin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Cucumber Protects the Blood Vessels Against the Risk and Onset of Vascular Disease

By Kyle J. Norton

Vascular disease is a disease associated with an abnormal condition of the blood vessels, affecting the blood flow to the body.

There are many types of vascular disease, including
* Stroke caused by the blockage of blood flow to the brain, leading to the death of the neurons.

* Peripheral artery disease (PAD), a disease is caused by reduced blood flow to the limbs

* Abdominal aortic aneurysm (AAA), is a condition of a localized enlargement of the abdominal aorta.

*Carotid artery disease (CAD) is a condition caused by plaque builds up inside the carotid arteries. of the neck.

* Arteriovenous malformation (AVM), is a condition associated with an abnormal tangle of blood vessels connecting arteries and veins.

*Pulmonary embolism (blood clots) is a condition caused by the sudden blockage of an artery in the lung,

* Deep vein thrombosis (DVT) is caused by a blood clot formed in a vein deep.

Symptoms of vascular disease are totally depending on the types of conditions.

According to the statistics, vascular diseases are becoming an epidemic in the US. Approximately 8.5 million people in the US are living with Peripheral artery disease (PAD), one of the most common types of vascular disease.

Some researchers suggested that the rise of aging patients with vascular disease may be correlated to the promotion of high-fat diet over the past few decades in inducing a large population with obesity and Type II diabetes in the US.

Dr. Morteza Mowlaie, the lead scientist wrote, " Significant positive association between left ventricular hypertrophy and plaque score and also a significant positive association between left ventricular hypertrophy with the presence of chest pain was found (p<0.05)"

And, "Association of diabetes mellitus with the presence of chest pain was positive. A positive correlation between hypertension with plaque score was demonstrated too (p<0.05). Also, an inverse association of plaque score with left ventricular ejection fraction was detected too (p<0.05)".

In other words, vascular disease is a condition associated with patients with obesity and diabetes.

The cucumber plant is a species of Cucumis Sativus, belongings to the family Cucurbitaceae and native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

With an aim to find a potential ingredient for the treatment of impaired cells of the endothelium in the onset of many chronic and acute vascular diseases, researches examined the effect of cucumbers water/ethanol extract of Cucumis sativus L. (CSE) fruit on the damaging effect of pro-inflammatory lipopolysaccharide (LPS).

In vitro, a model of endothelial cells induced overexpression of pro-inflammatory cytokines with lipopolysaccharide (LPS) are incubated with a water/ethanol extract of Cucumis sativus L. (CSE) fruit.

CSE protected the integrity and viability of endothelial cells against LPS-induced cytotoxicity and decreased the cellular detachment, restoring the expression of tight junction ZO-1 that interacts with proteins associated with adherens junctions.

The increase of TLR4 expression associated with pathogen recognition and activation of innate immunity caused by LPS was also inhibited by the injection of CSE.

Futhermore, CSE also protected the endothelial cells by increasing the function of gene Heme oxygenase (HO)-1 involved in inhibition of oxidative stress.

Moreover, CSE not only inhibited the levels of pro-inflammatory cytokines produced by the administration of LPS but also exhibited the expression of anti-inflammatory cytokines and its detoxed enzymes.

Additonally, also in vitro, the angiogenesis induced by inflammatory LPS was prevented by the presence of Cucumis sativus L. extract (CSE), at any doses tested.

After taking into account other factors, researchers suggested that Cucumis sativus L. extract has attenuated lipopolysaccharide-induced inflammatory response in endothelial cells, thus reducing the onset of many chronic and acute vascular diseases.

Taken altogether, cucumber may be used for the treatment of chronic and acute vascular diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of CuB in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Bilberry Protects the Neurons Against Memory Impairment in Vivo

By Kyle J. Norton

Memory impairment is a condition of partial or complete loss of memory, depending on the quality of the neuron that has been affected.

Memory impairment other than the physical impact is a progressive disease that can lead to permanent loss of memory due to the gradual loss of neurons in the brain.

The most common risk factors associated with memory loss in the general population are long-term use of certain medication, excessive alcohol drinking, smoking, drug abuse, depression, nutrient deficiency, stroke, and daily stress.

In elderly or aging-induced memory loss are associated with the deterioration of a region of the brain involved in the formation and retrieval of memories, the hippocampus, and the reduced hormones and protein that protect and repair brain cells and stimulate neural growth.

However, some researchers suggested the rise of memory impairment in the elderly probably is correlated to the promotion of the Western diet over the past few decades.

Dr. Scott E. Kanoski, the lead scientist in the review the risk cognitive impairment in the Western diet consumption wrote, "Intake of saturated fats and simple carbohydrates, two of the primary components of a modern Western diet, is linked with the development of obesity and Alzheimer's Disease".

And, "Western diet intake is associated with cognitive impairment, with a specific emphasis on learning and memory functions that are dependent on the integrity of the hippocampus".

The results clearly stated if you are Western diet follower, you are at a higher risk of memory impairment compared to healthy diet individuals.

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

With an aim to find a potential ingredient for the treatment of memory impairment researchers investigated the effects of Vaccinium uliginosum L. (bilberry) on the learning and memory impairments induced by amyloid-β protein (AβP) 1-42.

The evaluation included selected ICR Swiss mice divided randomly into 4 groups: the control (Aβ40-1A), control with 5% bilberry group (Aβ40-1B), amyloid β protein 1-42 treated group (Aβ1-42A), and Aβ1-42 with 5% bilberry group (Aβ1-42B).

According to the tested assays, bilberry added groups showed to decrease the finding time of hidden platform, compared to those of escape latencies of AβP to that of Aβ40-1.

Furthermore, bilberry added Aβ1-42B group also showed a significant (P<0.05) increase of probe crossing frequency compared to Aβ1-42A.

In the passive avoidance test, bilberry significantly (P<0.05) increased the time of staying in the lighted area compared to AD control.

These results suggested that bilberry improved memory and learning capability in chemically induced Alzheimer's disease in experimental animal models.

In the incidence of degenerative disorders including Alzheimer's disease (AD), rats consumed approximately 3.2 mg kg(-1)day (oral), of the anthocyanins significantly enhanced short-term memory, but not long-term memory in the inhibitory avoidance task.

Moreover, injection of approximately 3.2 mg kg(-1)day (oral), of the anthocyanins also induced an increase in the number of crossings in the first exposure to the open field.

However, in the comparison of the radial maze and elevated plus-maze, lyophilized berries (blueberry, bilberry) improved working memory in the radial maze, with significant differences observed during sessions 1-2 and 4, compared to no effects in the elevated plus-maze task.

The findings strongly indicated the positive effects of administration of lyophilized Vaccinium berries (blueberry, bilberry) and anthocyanin in the prevention of memory deficits related to AD.

Taken altogether, bilberry processed abundantly bioactive compound anthocyanin may be considered a functional remedy for the prevention and treatment of memory impairment, pending to the validation of larger sample size and multicenter human study.

Whole Food Tomato, the Best Anti-Inflammatory Remedy with No Side Effects

By Kyle J. Norton

Inflammation is a natural defense of the immune system that protects the body against the invasion of  the foreign pathogens.

Acute inflammation occurs quickly or within the hours and lasts a few days or weeks.

In the acute phase of inflammation, damage tissues are healed itself after all pathogens are killed off.

However, if the immune system can not completely destroy all invaders in the acute phase of infection within 3 to 8weeks, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

In most cases, depending on the types of injury, overproduction of pro-inflammatory cytokines in the acute phase of infection can cause damage to the nearby healthy tissues, leading to the formation of scars.

Anti-inflammatory is a process that inhibits the inflammatory symptoms without affecting the pro-inflammatory function of killing microorganisms.

Conventionally, the most common use of anti-inflammatory medicine for the treatment of symptoms induced by pro-inflammatory cytokines are Nonsteroidal anti-inflammatory drugs (NSAIDs) focusing on pain relief, inclduing aspirin, ibuprofen (Advil, Motrin, Midol) and naproxen (Aleve, Naprosyn).

If you are planning to take them in a long-term, please make sure that you know all the side effects.

Tomato is red, edible fruit, genus Solanum, belonging to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

On finding healthy food for the treatment of diseases associated with inflammation, researchers examined the tomato effects on the inflammation that causes intestinal dysbiosis associated with the onset of inflammatory bowel diseases (IBD).

The study included Winnie mice, a model of spontaneous ulcerative colitis received chows enriched with 1% Bronze tomato.

According to the tested assays, at week 2, the Bronze-enriched chow administered showed no macroscopic effect on the IBD symptoms. However, at the molecular level, the Bronze-enriched chow demonstrated significant activity in the activation of anti-inflammatory genes.

According to the intracellular staining of T cells, proteins associated with the production of pro-inflammatory cytokines ( IL17A, IFNγ) were significantly inhibited by the injection of Bronez tomato.

Based on the analysis of the microbial composition, researchers also found that two weeks of Bronze enriched diet was sufficient to alter positively the microbial composition of the tested subjects.

In other words, polyphenols found in the Bronze tomato has created unfavorable conditions for bad bacterial species in the gastrointestinal tract to stimulate the progression of IBD.

Dr. Liso M, the lead scientist wrote, after taking other factors into account, "dietary regimes enriched in polyphenols may efficiently support IBD remission affecting the intestinal dysbiosis:.

Taken altogether, tomato may be considered a dietary supplements for the prevention and treatment of IBD, pending to large sample size and multicenter human study.

Herbal Turmeric Protects the Kidney Against the Onset of Lupus Nephritis

By Kyle J. Norton

Lupus nephritis is a condition of kidneys an inflammation caused by systemic lupus erythematosus (SLE), an autoimmune disease.

It is a type of glomerulonephritis in which the glomeruli become inflamed.

Kidneys are a pair of bean-shaped organs, located just below the rib cage, one on each side of your spine with a function to filter out blood toxins through urinary secretion.

Believe it or not, every day, the blood passes the kidneys many times. Some people with only 10% of kidney function showed no symptoms of toxin accumulation.

However, if the kidney fails to cleanse the blood due to kidney failure, patients may experience symptoms of decreased urine output, fluid retention in your legs, ankles or feet.

Depending on the severity of toxins accumulation, some patients may also experience symptoms of shortness of breath and confusion.
Lupus nephritis reduced the kidney function may cause symptoms of sudden and unexplained swelling feet, ankles, legs, fingers, arms or the eyes, foamy appearance in urine and frequent urination.

In severe cases, patients may also have symptoms of blood in urine and elevated blood pressure.

Out of many risk factors associated with the risk of lupus nephritis, some researchers suggested genetic preposition may be a strong impact that leads to early onset of the disease.

Dr. the lead scientist wrote, " Although many SLE susceptibility genes have been identified, the genetic basis of LN is not as well understood".

And, "some of the established general SLE susceptibility genes are associated with LN, recent discoveries highlight a number of genes with renal functions that are specifically associated with LN. Some of these genes associated with LN help to explain the disparity in the prevalence of nephritis between individuals with SLE, and also partially explain differences in LN between ancestral backgrounds".

Turmeric is a perennial plant in the genus Curcuma, belongings to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a potential ingredient for the treatment of lupus nephritis, researchers investigated the effects of curcumin on lupus nephritis (LN) induced animal model.

LN established New Zealand Black/White (NZB/W) F1 female mice, at 18 weeks of age were fed a standard diet or a diet containing 1 % curcumin until the end of the study.

During the study, infection of curcumin decreased the proteinuria level and serum levels of IgG1, IgG2a which were elevated in LN mice.

Where proteinuria is the ratio of a protein/creatinine greater than 45 mg/mmol and high levels of IgG1, IgG2a are an indication of immune defect.

Additonally, curcumin also protected the kidneys against inflammation by reducing the anti-dsDNA IgG antibodies which were also elevated in NZB/W F1 female mice.

Where elevated levels of anti-dsDNA IgG antibodies are associated with ongoing inflammation and damage to the kidneys.

Furthermore, IgG immune complex deposition in the glomeruli was also reduced in curcumin-treated mice.

In other words, curcumin suppressed the pro-inflammatory expression in the kidney glomeruli.

Moreover, according to the kidney tested assays, kidney inflammation was also decreased after curcumin treatment.

More importantly, researchers discovered that treatment with anti-CD25 antibody injection that causes Treg depletion also affects the therapeutic effects of curcumin.

Where the anti-CD25 antibody is a conventional medicine used for enhancing the immune response.

More precisely, curcumin cannot be used in combination with anti-CD25 antibody daclizumab that
targets the T cells for the treatment of lupus nephritis.

Based on the findings, researchers said, "Curcumin exerted a protective effect against LN in NZB/W F1 mice. We speculate that the protective effects of curcumin in LN may involve, at least in part, its interaction with Treg cells".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of lupus nephritis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical Lycopene Inhibits Hyperproliferation In Gastric Epithelial Cells

By Kyle J. Norton

Hyperproliferation is a progression of highly invasive tumor associated with an abnormally high rate of cancer cells division, leading to rapid overgrowth of the tumor.

Gastric cancer or stomach cancer is a medical condition caused by irregular cell growth of the inner lining tissues of the stomach.

Most cancer of gastric start in the cells on the inner lining of the stomach before penetrating into deeper layers.

At the advanced stage, cancerous gastric cells can also travel a distance away to an alternate DNA of healthy cells, leading a secondary metastasis.

According to the statistics provided by the Canadian Cancer Society, In 2017, approximately  3,500 Canadians were diagnosed with stomach cancer, and gastric cancer also caused the death of 2,100 including 1,300 women and 790 men.

There are major types of gastric cancer
*Adenocarcinoma, accounting to over 90% of gastric cancer diagnosed each year is a type of cancer developed in the innermost lining of the stomach (the mucosa).

* Lymphoma is a type of cancer associated with the immune system found in the wall of the stomach.

Most common symptoms of gastric cancer are heartburn or indigestion, abdominal pain or vague discomfort in the abdomen, usually above the navel and a sense of fullness in the upper abdomen after eating a small meal accompanied by the general symptoms shared by other types of cancer.

In severe cases, if the oversize tumor has pressed on the nearby blood vessel, patients may experience the symptom of vomiting blood.

Out of many prevalent factors involved in the cancer onset, some researchers suggested the promotion of the Western diet may have a strong and negative implication on the cancer occurrence in North America.

Dr. Bertuccio P, the lead scientist wrote, "An unfavorable role on GC emerged for the 'Western/unhealthy' dietary pattern, with an OR of 1.51 (95% CI: 1.21-1.89). This association was weaker for the distal/NOS (not otherwise specified) category (OR = 1.36) compared with the cardia GC (OR = 2.05). Among the a priori scores, the ORs ranged from 0.2 to 0.7 for the favorable and from 1.8 to 6.9 for the unfavorable ones".

And, "There is a ~2-fold difference in GC risk between a 'Prudent/healthy' diet-rich in fruits and vegetables, and a 'Western/unhealthy' diet-rich in starchy foods, meat, and fats".

Lycopene is a phytochemical in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

With an aim to find a potential compound for the treatment of invasive gastric cancer, researchers examined the lycopene effect on Helicobacter pylori (H pylori) colonized human stomach in the risk of gastric cancer.

According to the tested differentiation, injection lycopene reduced ROS levels by inhibiting Jak1/Stat3 activation associated with increased levels of proinflammatory cytokines, alteration of Wnt/β-catenin multiprotein signal in induced cells proliferation, and differentiation.

Furthermore, the levels of ROS also are decreased observed by the inhibition of c-Myc, a protein which plays a critical role in cellular growth regulation and cellular metabolism and cyclin E in cell duplication and cell proliferation in H pylori-infected AGS cells, derived from an adenocarcinoma of the stomach.

In other words, lycopene protects the gastric cell by constraining the expression of ROS through several aforementioned mechanisms.

Based on the findings, researchers said, " Lycopene might be a potential and promising nutrient for preventing H pylori-associated gastric diseases including gastric cancer".

Furthermore, according to the results of HGC-27 cells which were treated with varying concentration lycopene for 24, 48, 72,
* Lycopene displayed antiproliferative activity in HGC-27 cell lines.

* Lycopene inhibited HGC-27 by enhancing the protein LC3 which is essential for cancer cell autophagy and p-ERK proteins expression that impaired regeneration of intracellular antioxidants.

Furthermore, in gastric cancer nude mice model, lycopene treatment also significantly decreased tumor weight or tumor proliferation.

Taken altogether, lycopene may be considered supplements for the prevention and treatment of gastric cancer, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
