Phytochemical Alliin, Processes Anti-Angiogenesis in Cancer

Angiogenesis is the process that promotes the formation of new blood vessels involving many factors including the migration, growth, and differentiation of endothelial cells.

Believe it or not, all active tissues in the body is on a distance less than a hundred micrometers from a blood capillary formed by the process of angiogenesis.

Angiogenesis is important for the cells for the diffusion exchange of nutrients and metabolites through the small blood vessels.

In normal and healthy condition, stimulating angiogenesis has been found to have a therapeutic effect for the treatment of ischemic heart disease, peripheral arterial disease, and wound healing.

However, in cancers, stimulating angiogenesis in tumor cells has been associated with tumors proliferation and metastasis.

In other words, angiogenesis process a function of a double sword, on one hand, the process provides an ideal condition for diseases and wound healing. On the other hand, it promotes invasive and overgrowth tumor.

The Genentech in the article "Tumor Angiogenesis and the VEGF Pathway" wrote, "Angiogenesis is a necessary part of the process in the progression of cancer from small, localized neoplasms to larger, growing, and potentially metastatic tumors. To grow beyond 1 to 2 mm in diameter, a tumor needs an independent blood supply, which is acquired by the expression of growth factors that recruit new vasculature from existing blood vessels".

"This process continues even as the tumor matures. Thus, upregulation of angiogenesis is a key step in sustained tumor growth and may also be critical for tumor metastasis".

In other words, by the inhibiting the process of blood vessels formation of the tumor, cancer may be curable.

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

In the urgency to find a potential ingredient for the treatment of cancer with no side effect, scientists examined the phytochemical alliin, a compound derived from garlic anti-angiogenesis activity.

In the chick chorioallantoic membrane, injection of alliin exerted a significant effect on the inhibition of fibroblast growth factor-2 (FGF2) associated with the angiogenesis.

In the CAM model, alliin also inhibited the vascular endothelial growth factor (VEGF)-induced angiogenesis.

Combined with antioxidant vitamins C and E significantly (P < 0.001) enhanced the function of alliin inhibitory efficacy on FGF2-induced EC tube formation and angiogenesis, particularly in the colon and fibrosarcoma tumor growth.

More precisely, alliin significantly inhibited both FGF2 and VEGF secretion from human fibrosarcoma cells compared to other types of tumors in a concentration-dependent manner

However, the anti-angiogenesis of alliin was reversed if the NO synthesis inhibitor nitro-L-arginine methyl ester (L-NAME) were injected.

The efficacy of alliin was attributed to the activation of the expression of a protein involved in immune tumor suppression.

In other words, alliin worked synergistically with other antioxidants to promote anti-angiogenesis and anticancer efficacy.

Dr. Mousa AS, the lead scientist wrote, "These data also suggest the implication of cellular NO and p53 as mediators of anti-angiogenesis and anticancer effects of alliin".


Taken altogether, alliin may be considered supplements for the inhibition of angiogenesis in the promotion of tumor growth, pending on the confirmation of the larger sample size and multicenter human study.

Colitis is a condition associated with the inflammation of the inner lining of the colon caused by infection, inflammatory bowel disease, ischemic colitis, and microscopic colitis.

Some cases of colitis are also induced by the overactive immune system in the response of unharmful allergens.

Most common symptoms of colitis depending on the cause of the disease, including abdominal pain, cramping, diarrhea, with or without blood in the stool.

Some patients in the early phase of colitis may also experience symptoms of fever, chills, fatigue, and skin inflammation.

Further analysis of the anti-inflammatory properties researchers found that MIC inhibits the miR-132 activity against the stages of chronic colitis without suppressing the production of inflammatory cytokines TNF-α, IL-1β, and IL-6 during the course of the disease.

In other words, there must be other mechanisms associated with the MIC anti chronic colitis efficacy other than those of the traditional differentiation.

Dr. Alzahrani AM, the lead scientist said, "miR-132 as a promising therapeutic candidate to control autoimmune inflammation and tumorigenesis in CAC patients".

The reproductive system is a system of sex organs with a function to produce offspring.

In males, the reproductive system consists of the testes, where sperm is produced, and the penis.

In males, most cases of infection are found on the penis, testicles, urethra or the vas deferens.

Some cases of reproductive disease such as cancer have been found to affect the reproductive functioning, leading to infertility.

Fertility is a natural capability to produce offspring. Infertility is a condition in which the couple fails to conceive after 12 months of unprotected sexual intercourse or the female cannot carry the pregnancy to full term.

According to the statistics, over 5 million couple alone in the U. S. and many times more in the world, because of unawareness of treatments, only 10% seeks help from a professional specialist.

Turmeric is a perennial plant in the genus Curcuma, belongings to the family Zingiberaceae, native to tropical South Asia.

Furthermore, C. longa and curcumin exerted a protective effect on the activity of the reproductive organs through their antioxidant effects in normal cells.

Moreover, C. longa and curcumin also exhibited different reproductive effects depending on the doses and the type of cells.

In order to discover more information of CUR reproductive activity, researchers examined the effect of on testis and testosterone, follicle stimulating hormone (FSH) and luteinizing hormone (LH) in rats with establishing chronic variable stress (CVS).

A total of 21 adult male Sprague-Dawley rats included in the study were divided into three groups: 1) control, 2) CVS and 3) CVS+ CUR (100 mg/kg/day dissolved in 0.5 mL of olive oil). All of the animals in control, CVS, and CVS+CUR groups were sacrificed after 15 days.

Injection CUR showed a significant effect in protecting the reproductive functioning by inhibiting the change of normal range testosterone, follicle stimulating hormone (FSH) and luteinizing hormone (LH) serum levels and seminiferous tubule apoptotic cells in the CVS group compared to the control.

Based on the results, Dr. the lead scientist said, "Our findings propose that curcumin might have curative potential on the reproductive system function and its impairment. It's regulated by stress and reproductive-related hormones".

Whole Food Tomato Inhibits Gene and Factors Associated with Obesity Complications in Vivo

Obesity is a medical condition characterized by fat accumulated in the body over time.

In other words, obesity is a progressive disease similar to those of diabetes and high blood pressure and lipid, leading to the impairment of health in many aspects, including heart disease, diabetes, and high blood pressure.

According to the Body Mass Index(BMI), a BMI between 25 to 29.9 is considered overweight, while a BMI of over 30 is an indication of obesity. 

According to the statistic, 68% of the American population is either overweight or obese.

The causes of obesity are the results overconsumption of food that produces energy, compared to energy expenditure (physical inactivity) over the period of time.

Most common physical symptoms of obesity are breathlessness. increased sweating, snoring, tiredness, back, and joint pains accompanied by psychological symptoms of low confidence and self-esteem. 

Some researchers in the evaluation of the risk factors that cause widespread obesity in the US population suggested that the incidence may correlate to the promotion of the Western diet over the past few decades.

Dr. Farah Naja, the lead scientist wrote, "The Western pattern was characterized by high consumption of red meat, eggs, and fast-food sandwiches".

And, "After adjustment, subjects belonging to the 3rd tertile of the Western pattern scores had significantly higher odds of overweight compared with those belonging to the 1st tertile (OR 2·3; 95 % CI 1·12, 4·73)".

Tomato is red, edible fruit, genus Solanum, belonging to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find a potential compound for the treatment of obesity, scientists evaluated and compared the effect of lycopene and tomato powder on obesity-associated disorders.

Selected male C57BL/J6 mice included in the study were assigned into four groups to receive: control diet (CD), high-fat diet (HFD), high-fat diet supplemented with lycopene or with tomato powder (TP) for 12 weeks.

According to the tested assays, both lycopene and TP supplementation significantly reduced adiposity index, organ, and relative organ weights, serum triglycerides, free fatty acids, 8-iso-prostaglandin GF2α induced by inflammation and improved glucose homeostasis, but did not affect total body weight.

Furthermore, both lycopene and TP supplementation prevented HFD-induced hepatosteatosis and hypertrophy of adipocytes associated with induced obesity.

Moreover, both lycopene and TP decreased HFD-induced proinflammatory cytokine mRNA expression in the liver and in the epididymal adipose tissue.

In other words, lycopene and TP inhibited the expression of hepatosteatosis involved in liver disease and hypertrophy of adipocytes in obese.

Lycopene and TP protected the adipocytes dysfunction in the onset of obese complications by inhibiting the overexpression of pro-inflammatory cytokines in the tissues.

Based on the results, researchers said, " lycopene or TP supplementation display similar beneficial health effects that could be particularly relevant in the context of nutritional approaches to fight obesity-associated pathologies".

In order to reveal more information about tomato effects on obese complications, researchers examined the Mediterranean tomato-based sofrito sauce against obese fibroblast growth factor 21 (FGF21)-resistant state.

In obese Zucker rats (OZR) fed with a sofrito-supplemented diet or control diet, researchers found that the sofrito-supplemented diet improved insulin and FGF21 sensitivity in OZR, by exhibiting the effect on glucose metabolism.

The sofrito-fed rats also showed a strong effect on the protection of the UCP1 gene associated with reduced obesity and improved insulin sensitivity.


Taken altogether, tomatoes processed abundantly bioactive compound lycopene may be considered functional food for the prevention of obese complications, pending to the confirmation of the larger sample size and multicenter human study.


Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Honey Protects the neurons Against Neuro-inflammation with No Side Effects

Neuroinflammation is a medical condition caused by inflammation of the nervous tissue affecting the behavior, noradrenaline (NA) and other functions in the CNS.

It may be initiated by infection, traumatic brain injury, toxic metabolites, or autoimmunity.

Inflammation is a natural response of the first line defense of macrophages in the immune system after tissues damage by stimulating the production of blood palettes to cover the wound and pro-inflammatory cytokines to kill off the invading stimuli or pathogens in the acute phase of infection.

A nerve cell is a brain cell specialized in transmitting nerve impulses.

Chronic inflammation is a medical condition caused by the adaptation of the immune system after failing to kill off the invaders with a period of 3-8 weeks. Chronic inflammation has been found to initiate several diseases and conditions, including some cancers and rheumatoid arthritis.

Most cases of neuroinflammation are associated with the exposure to neurotoxic stimuli, ischemic injury, protein accumulation or age-related homeostatic perturbations caused by the microglia respond by upregulating inflammatory signals.

Similar to those of chronic inflammation in other parts of the body if the acute brain inflammation cannot be resolved within a set period of time, the inflammation may be chronic.

In other words, neuroinflammation is a complex response to brain injury involving the activation of inflammatory cytokines and chemokines, and generation of reactive oxygen and nitrogen species.

Overnutrition in the Western world may be one of the main culprit associated with the widespread neuroinflammation in the aging population.

Dr. Dongsheng Cai in the differentiation of neuroinflammation in overnutrition-induced diseases, said, "overnutrition-induced inflammation occurs in the brain and thus plays abroad and leadership role in overnutrition-induced diseases".

With an aim to find a natural therapy for the treatment of neuroinflammation against the progressive death of neurons, researchers evaluated the bioactive compound chrysin contained in honey anti-neuroinflammatory effects.

In LPS-induced BV2, primary microglial cells in mice, honey' chrysin protected the neuron integrity against the proteins associated with apoptosis and cycle arrest by stimulating the protein A 20 involved in the negative feedback regulation of NF-κB ( cell apoptosis) signaling.

In other words, chrysin exerts an anti-neuroinflammatory effect via NF-κB ( cell apoptosis) signaling and the exhibition of A20 expression.

Furthermore, in order to reveal more information about the protective effect of Tualang honey (TH) on neuroinflammation, researchers analyzed the cell death protein signaling in rat cerebral cortex, cerebellum, and brainstem after kainic acid- (KA-) induced status epilepticus. 

The study included male Sprague-Dawley rats were pretreated orally with TH (1.0 g/kg body weight) five times at 12 h intervals and KA (15 mg/kg body weight) injected subcutaneously 30 min after last oral treatment.

Administration of KA-induced epileptic seizures and caused significant increase levels of proinflammatory cytokines and protein associated with neuron apoptosis in the rat cerebral cortex, cerebellum, and brainstem at multiple time points.

Pretreatment with TH significantly reduced the elevation pro-inflammatory level in those brain regions at multiple time points and attenuated the increased caspase-3 activity, a protein associated with cell death in the cerebral cortex.

Taken altogether, Tualang honey (TH), containing chrysin may be considered a  functional remedy for the prevention and treatment of neuroinflammation with no side effects, pending to the confirmation of large sample size and multicenter human study.

Herbal Bilberry, a Natural Treatment of Steatohepatitis with No Side Effects

Steatohepatitis is a fatty liver disease caused by inflammation of the liver tissue accompanied by fat accumulation.

A most common form of steatohepatitis is non-alcoholic steatohepatitis (NASH), an advanced form of non-alcoholic fatty liver disease (NAFLD) that is not caused by excessive alcohol drinking.

People with steatohepatitis have been associated with the increased risk of liver failure and liver cancer, due to excessive formation of liver scar tissues, epidemiologically.

According to the statistics, fatty liver affects approximately 25% of the general population worldwide. Non-alcoholic steatohepatitis (NASH), is very common in overweight persons.

If you experience chronic symptoms of swollen belly enlarged blood vessels underneath your skin, larger-than-normal breasts in men, particularly chronic red palms, skin and eyes, and jaundice, you may have steatohepatitis.

Please make sure that you check with your doctor to rule out the possibility.

Researchers do not know the exact cause of steatohepatitis. However, they do know certain medical conditions such as diabetes, being overweight or obese and people with metabolic syndrome, including high blood cholesterol are some most prevalent risks of the disease.

Some researchers suggested that people who are viral hepatitis carriers are also at risk of steatohepatitis.

Dr. Anish Patel, the lead scientist in the evaluation of hepatitis C virus infection and nonalcoholic steatohepatitis (NASH) wrote, "Hepatitis C virus infection is another common cause of liver disease worldwide. Up to 70% of patients with chronic hepatitis C (CHC) will have concomitant steatosis" and "Worldwide, chronic hepatitis C (CHC) affects up to 200 million individuals—3% of the world's population".

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

With an aim to discover a potential ingredient for the treatment of steatohepatitis, an inflammatory type of non-alcoholic fatty liver disease, researchers examined the effects of anthocyanins found abundantly in bilberry on disorders related to NASH induced by a methionine-choline-deficient (MCD) diet in mice.

Treatment of anthocyanins not only showed a significantly alleviated inflammation, oxidative stress, steatosis but also inhibited the formation of fibrosis.

Anthocyanins improved depletion of mitochondrial content and damage of mitochondrial biogenesis and electron transfer chain developed concomitantly caused by overexpression of reactive oxygen species in the liver of mice fed the MCD diet.

In proteins perspectively, anthocyanins treatment promoted activation of AMP-activated protein kinase (AMPK) in controlling energy balance and lipid metabolism against oxidation and expression of peroxisome proliferator-activated receptor gamma coactivator-1α (PGC-1α) against inflammation.

Researchers, at the end of the study, wrote, "anthocyanins protected the liver against the development of steatohepatitis by improving the antioxidants in the host defense against overexpression of free radicals by modulating the expression of the genes associated with oxidation and inflammation".

In ApoE(∗)3Leiden mice fed a Western-type cholesterol-containing diet without (HC) or with 0.1% (w/w) Mirtoselect (HCM) for 20weeks diet-induced non-alcoholic steatohepatitis (NASH), researchers found that Mirtoselect attenuated HC-induced hepatic steatosis, by decreasing the macro- and microvesicular hepatocellular lipid accumulation and hepatic cholesteryl ester content associated with lipid metabolism.

Mirtoselect (HCM) anti-NASH, accompanied by local anti-inflammatory effects in the liver was attributed to the levels of anthocyanin in reducing the inflammatory cell clusters and reduced neutrophil infiltration.

Genes associated with pro-fibrotic development such as Acta2 and Col1a1 were also inhibited by the injection of HCM.

Dr. Morrison MC, the lead scientist, wrote in the final report, "Mirtoselect significantly reduced accumulation and crystallization of intrahepatic free cholesterol, providing a possible mechanism for the observed hepatoprotective effects".

In other words, HCM protected the liver against fat accumulated caused by inflammation via its promotion of antioxidant production.

Taken altogether, bilberry processed abundantly bioactive compound anthocyanin may be considered a functioning remedy for the treatment of steatohepatitis, pending to the validation of larger sample size and multicenter human study.

Phytochemical 3,3'-Diindolylmethane kills Gastrointestinal Cancer Cells In Vivo and Vitro

Scientists may have found a bioactive ingredient for the treatment of gastrointestinal cancer with no side effects, according to studies.

Gastrointestinal (GI) cancer is a group of cancers, affecting the digestive system, including esophagus, gallbladder & biliary tract, liver, pancreas, stomach, small intestine, bowel, and anus

The digestive system consists of the gastrointestinal tract and other parts of the accessory organs of digestion including the tongue, salivary glands, pancreas, liver, and gallbladder), involved in the function of breaking down food into smaller and smaller components then absorbed before passing them as nutrients and fluids to the body.

Most cases of gastrointestinal cancer start in the cells on the surface of the inner lining tissues, depending on the location of the affected organs.

At the early stage, most patients are asymptomatic due to the small size of the tumor. However, at the later stage, after penetrating into deeper layers of the organ, cancer cells can travel through blood and fluids circulation to infect other healthy tissues or organs, leading to secondary metastasis.

At this stage, besides sharing the general symptoms of other types of cancer, gastrointestinal cancer also cause symptoms of severe pain and bleeding due to the overgrowth tumor size have suppressed the nearby blood vessels and nerve cells.

The exact causes of gastrointestinal cancer are not identified. However, genetic preposition, unhealthy lifestyle and medical conditions are some of the prevalent risks of the disease.

Some researchers suggested that a poor diet may have a strong implication on the incidence of gastrointestinal cancer.

Dr. Reilly T. Enos, the leads scientist said, "High-fat-diet (HFD) consumption is associated with colon cancer risk".

And, "HFDs with the highest SF content had the greatest concentration of Muc2. Our data suggest that high dietary SF is protective in the AOM/DSS model of colon cancer, which may be due, at least in part, to the ability of SF to maintain intestinal barrier integrity through increased colonic Muc2".


With an aim to find a natural compound for the treatment of gastrointestinal cancers, researchers examined the 3,3'-Diindolylmethane (DIM) activity against effective gastrointestinal cancers.

In vivo and vitro, injection of the DIM acted upon several cellular and molecular processes in gastrointestinal cancer cells.

In cancer cells apoptosis, DIM  initiated production reactive oxygen species (ROS) in generating stress, triggering DNA damage by increasing the cytotoxicity.

In angiogenesis, DIM inhibited the protein associated with the generation of a new blood vessel for cells survival and proliferation.

In cell cycle regulation, DIM deactivated the expression of protein kinases which play the critical roles in the cancer cells division.

Furthermore, DIM triggers cellular endoplasmic reticulum (ER) stress and apoptosis within the cancer cells.

In autophagy, DIM inhibited autophagy that leads to cell death.

In metastasis, DIM  inhibited the growth of human cancer cells by interfering with multiple signaling pathways, involved restraining invasion, migration, and promoting apoptosis.

Furthermore, DIM used combined with conventional chemo drugs or therapeutic chemicals increased the efficacy of other drugs for the treatment of gastrointestinal cancer.

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of gastrointestinal cancers, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
