Carrots, the Veggies with Anti Cancer Potential, Researchers Find

Carrot, the root veggie may have a potential and positive effect in blocking the proliferation of cancer cells, some scientists suggested.

Cancer is a class of chronic and medical diseases characterized by irregular and disordered cell growth in one or some tissue and organs in the body.

Most cancers start from the inner lining of the tissue of the organs then slowly spread to the deeper parts of the infected organs. At the advanced stage, most malignant cancer cells can travel a distance away from the original site to infect other healthy tissue and organs.

Most cancer diagnosed in the early have a 100%  5 years survival rate.

According to the statistic provided by the Canadian public health service, during their lifetime, nearly 1 in 2 Canadians will be diagnosed with cancer, and 1 in 4 will die from the disease.

Lung, colorectal, breast and prostate cancers account for half of all cancers diagnosed in Canada and over 25% of all cancer deaths in Canada are due to lung cancer.

The causes of cancers are unknown, however, scientists in the world agreed that there are several risk factors such as gene mutations inherited from the parents and acquired gene mutations caused by smoking, radiation, viruses, chemicals carcinogens, obesity, hormones, chronic inflammation, and physical inactivity are associated to the early onset of cancer.

Carrot, a root vegetable with an orange color is a subspecies of Daucus carota, belonging to the family Apiaceae, native to Asian and Europe.

In the investigation of the claim of Lebanese folk medicine that Daucus carota, also known as wild carrot has a potency for treatment of various types of cancer, researchers at the Lebanese American University, conducted an experience to identify the major compound responsible for the anti-cancer activity of DCOE.

β-2-himachalen-6-ol, a novel sesquiterpene unique to β-2-Himachalen-6-ol isolated from the Lebanese wild carrot demonstrated potent anti-cancer activity against B16F-10, Caco-2, MB-MDA-231, A549, and SF-268 cancer cells (IC50 13-4µg/ml; 58-18µM), and SF-268 cells.

Furthermore, the sesquiterpene was shown to induce cell death through apoptosis (flow cytometry), decrease 2D cell motility (wound healing assay) and 3D invasion, as well as increase cell adhesion in SF-268 cells.

Additionally, β-2-himachalen-6-ol showed very low toxicity.

These results suggested that wild carrot β-2-himachalen-6-ol may have a potential multi-mechanistic chemotherapeutic drug with high potency and safety.

Moreover, in the concerns of the mortality rate caused by gastric cancer, researchers at the Shahid Sadoughi University of Medical Sciences conducted a review by searching the multiple databases including PubMed, Cochrane Library, Scopus, ScienceDirect, and Persian databases like Scientific Information Database (SID) and IranMedx to determine the relationship between carrot consumption and gastric cancer.

Out of 81 articles found in the databases, only 5 studies satisfied the medical guidelines and criteria were included in the review. after taking into account co and confounders, researchers found that
* Intake of carrot regularly reduced the risk of gastric cancer by 26%.

* There is an inverse relationship between the consumption of carrots and the risk of gastric cancer.

More interestingly, regarding the inconsistent results. associated with carrot intake and prostate cancer risk, researchers at the Zhejiang University also launched a review by searching the databases of PubMed, EMBASE, Scopus, Web of Science, the Cochrane register, and the Chinese National Knowledge Infrastructure to determine whether intake of carrot may reduce the risk of prostate cancer.

At the final report, researchers found that
* Intake of carrot is associated with a significantly decreased risk of prostate cancer with the odds ratio of 0.82 which is consistency with the Shahid Sadoughi University of Medical Sciences study.

* Forr each serving per week or 10 g per day increment of carrot intake, the odds ratio of the risk of prostate cancer was 0.95, according to the dose-response meta-analysis.

The findings indicated that intake of carrot may have a potential effect in reducing the risk of the onset of cancers. 

However, due to the limited number of cohort studies and substantial heterogeneity observed between studies in this meta-analysis, further well-designed prospective studies are warranted to confirm the findings from the studies.

Green Tea and Its Bioactive Polyphenols Inhibit Alpha1-Antitrypsin (AAT) Deficiency Against the Onset of Fibromyalgia

Green tea may have a therapeutic and positive effect in reducing onset. progression and treatment of Alpha1-Antitrypsin (AAT) deficiency in the induced fibromyalgia, some scientists suggested.

Fibromyalgia (FM) is a chronic condition characterized by muscle and soft tissue pain, affecting over 10 million (3.6% of the population) people the in the US alone.

Serum of alpha1-Antitrypsin (AAT) impregnated in most parts of body circulates in our body in high concentrations with function in the inhibition of the enzymes of inflammatory cells.

Alpha1-Antitrypsin (AAT) deficiency is a genetic disorder inherited from the parent characterized by of the inability of the body to produce a protein called alpha-1 anti-trypsin (AAT), leading to inflammation of fatty tissue under the skin (subcutaneous), and at other parts of the body, including the joints, the major cause of fibromyalgia.

Additionally, deficiency of AAT enynes also can cause the liver and lung damage, leading to poor function of the organ and increasing the risk of cirrhosis, COPD, liver and lung cancer

Although the deficiency is presented since birth, most patients with mutated gene do not experience any symptoms until the age of 50 or 60.

According to the statistic, AAT deficiency is one of the most common inherited disorders among Caucasian compared to other race groups. 

Approximately over 1% of patients diagnosed with chronic obstructive pulmonary disease (COPD)  processes the  alpha1-antitrypsin deficiency gene

In the study of the effect of serum of alpha1-Antitrypsin (AAT) and risk of fibromyalgia (FM), researchers at the Hospital Valle del Nalón found that deficiency of AAT has been found in numbers of patients with fibromyalgia (FM) with severe hereditary.

Functional magnetic resonance imaging of these patients revealed a significantly greater activity in pain-sensitive areas of the brain in response to cutaneous stimuli, which may be another evidence for a physiological explanation for FM pain.

Furthermore, patients with FM also were found to consist of abnormal profiles of inflammation markers in biopsies analysis.

The results suggested that these patients may have an imbalance level of pro-inflammatory and anti-inflammatory substances, in the soft body tissues caused by alpha1-Antitrypsin (AAT) deficiency.

Due to surprising findings, Dr. Blanco LE, the lead author, after taking into account of other confounders, said, "identification of genetic, familial, and environmental risk factors suspected to participate in the FM syndrome development (may be necessary) to determine the gene frequency of AAT deficiency alleles in FM patients".

Green tea, bioactive polyphenols EGCG, according to the Dr Helieh S in animal model induced by alpha1-Antitrypsin (AAT) (α1-antitrypsin) deficiency exerted a significant anti-inflammatory effect in inhibited the PI3K/Akt signalling pathway with function in activated proinflammatory factors through the innate immune response involved in pathogen killing, antigen processing and immune cell survival.

Moreover, green tea EGCG application also enhanced antioxidant activity not only through its antioxidant profiles but also stimulated the production of the body natural antioxidants in reduced inflammatory response through nitric oxide (NO) and inducible nitric oxide synthase (iNOS) expression, observed by Griess reagent and western blotting.

The evidence indicated that green tea EGCG may have a profound effect in reducing serum alpha1-Antitrypsin (AAT) deficiency in initiated the onset of fibromyalgia

The above differentiations were supported by the review of Chronic Inflammatory Diseases and Green Tea Polyphenol, by Dr. Helieh S. Oz at the University of Kentucky Medical Center.

According to the authors, green tea polyphenols are potent antioxidants with important roles in regulating vital signaling pathways in activated inflammatory responses by attenuating nuclear factor (NF)-kappaB IKK, NF-κB in facilitated innate- and adaptive immune responses, cell death and inflammation and cytokines like TNFα, inflammatory markers and cyclooxygenase-2 in elevated symptoms of inflammation and pain.

Dr. Helieh S. in the summary said, "green tea polyphenols (GrTPs) are potent antioxidants with important roles in regulating vital signaling pathways" and "these comprise transcription nuclear factor-kappa B mediated I kappa B kinase complex pathways, programmed cell death pathways like caspases and B-cell lymphoma-2 (through regulating the cytochrome c release from the mitochondria) and intervention with the surge of inflammatory markers like cytokines and production of cyclooxygenase-2"".

Taken together, green tea and its bioactive polyphenols may be considered as a functional food in ameliorated serum alpha1-Antitrypsin (AAT) deficiency effect in initiated the onset of fibromyalgia. However, intake of green tea supplement should be taken with extreme care due to recent medical reports of acute liver toxicity.

Green tea may have a therapeutic and positive effect in reducing the symptoms, risk, progression, and treatment of polymyalgia rheumatica (PMR), some scientists suggested.


Polymyalgia rheumatica an inflammatory disease with muscle pain and stiffness, especially in the shoulders, affecting mostly older adults beyond the age of 65.

The exact causes of polymyalgia rheumatica are unknown. However, researchers do know that certain prevalent risk factors are associated with the onset of diseases, such as age, family history, gender, ethnicity and exposure to environmental weather.

Genetic mutations inherited from the parents are also the major risk factor for the development of polymyalgia arthritis such as ICAM-1 (intercellular adhesion molecule 1), RANTES (regulated upon activation, normal T cell expressed, and presumably secreted). 

It is important to know that Giant cell arteritis (GCA) (temporal arteritis) and polymyalgia rheumatica (PMR) share the common identity, frequently related conditions in people generally over 50 years of age, according to the Dr. Miguel A Gonzalez-Gay in the study of " Genetic epidemiology: Giant cell arteritis and polymyalgia rheumatica".

Symptoms of PMR are depending on the severity of the condition, including aches or pain shoulders,  neck, upper arms, buttocks, hips or thighs, and stiffness in affected areas.

People with PMR also experience pain in the morning or after being inactive for a time,  limited range of motion in the affected areas.

The bioactive compound also inhibited reactive oxygen species (ROS) in stimulating the production of inflammatory cytokines through stopping the free radicals chain reactions that facilitate inflammation and the precipitate skeletal muscle dysfunction and degradation by infiltrating immune cells to induce direct injury to muscle tissue.

In fact, overproduction of ROS increased expression of mitochondria, NAD(P)H oxidases, xanthine oxidoreductases, and nitric oxide synthases can cause an imbalance of the free radicals and antioxidant ratio, in orchestrated oxidative stress to muscle cells.

The further evaluation suggested that green tea EGCG also reduced expression of interleukin 6 (IL-6), with function in response to infections and tissue injuries.

Long-term overproduction of IL 6 due to the duo factors as a pro and anti-inflammatory protein may increase high-level concentrations of serum amyloid A (SAA), which has been found to induce serious complications of several chronic inflammatory diseases through the generated and deposited amyloid A amyloidosis into the extracellular space in an abnormal, insoluble, fibrillar form,

Moreover, EGCG also attenuated levels of nuclear factor kappa B (NF-κB) in the regulation of innate and adaptive immune functions and served as a pivotal mediator of inflammatory responses through activation of tumor necrosis factor alpha (TNF-α) in promoting muscle wasting, leading to muscle atrophy in the induction of pain and stiffness and swelling.

Importantly, application of green tea EGCG enhanced the natural antioxidant enzymes in the host such as glutathione, an antioxidant enzyme produced naturally found in every cell and tissue of the body.

These actions together with antioxidant initiated by injection of green tea EGCG stimulated cell regeneration, preserved the muscle cells and tissues and prevented muscle tension to precipitate polymyalgia arthritis.

Deficiency of glutathione levels was found to associate to the early onset of polymyalgia rheumatica, inducing symptoms of pain, stiffness, achiness, fatigue, and fever.

Differentiation of the above was supported by conventional medicine corticosteroids, a powerful anti-inflammatory drugs with the function to target pro-inflammation cytokines in the relieved stiffness and pain in patients with polymyalgia arthritis.

Taken all together, green tea and its bioactive polyphenols EGCG may be considered a functional food for alleviating symptoms, risk, progression and treatment of polymyalgia rheumatica(PMR).

However, ingestion green tea supplements should be taken with extreme care as acute liver toxicity has been reported in several medical literature.

Green may have processed a drug interaction property that inhibits the effect of conventional medicine, some scientists suggested.

Drug interactions are the greatest concern of some scientists, particularly in the pharmaceutical community as these effects can potentially reduce the desired effects for the treatment of certain diseases.

Intake of some drugs together, can reduce the absorption or increase the metabolism or elimination of other drugs treatment potential.

Some drugs interaction can cause an allergic reaction from skin rash and itching to a life-threatening anaphylactic reaction.

* Green tea extract and its main catechin, (-)-epigallocatechin-3-gallate, to varying degrees, inhibit the activity of the cytochrome P450 superfamily of enzymes of CYP1A1, CYP1A2, CYP2B6, CYP2C8, CYP2C9, CYP2D6, and CYP3A4. UGT1A1 and UGT1A4 but not the major cytochrome P450 enzyme, in vitro

These results suggested that patients should be cautious that consumption of significant amounts of green tea beverages or green tea extract may interact with the substrates of organic anion transporting polypeptide as mentioned above.

Further to investigate the effects of green tea extract (GTE) and (-)-epigallocatechin-3-gallate (EGCG) on the pharmacokinetics of a non-selective hydrophilic β-blocker nadolol, which is reported to be a substrate for several drug transporters and is not metabolized by cytochrome P450 enzymes launched a study o the. male Sprague-Dawley rats.

The tested animals were given GTE (400 mg/kg), EGCG (150 mg/kg) or saline (control) by oral gavage, 30 min before a single intragastric administration of 10 mg/kg nadolol.

At the end of the experiment, researchers showed that

* Pretreatment with GTE resulted in marked reductions in the maximum concentration (Cmax) and area under the time-plasma concentration curve (AUC) of nadolol by 85% and 74%, respectively, compared with control.

* EGCG alone significantly reduced Cmax and AUC of nadolol.

* The amounts of nadolol excreted into the urine were decreased by pretreatments with GTE and EGCG, while the terminal half-life of nadolol was not different among groups.

These results suggest that the coadministration with green tea catechins, particularly EGCG, causes a significant alteration in the pharmacokinetics of nadolol, possibly through the inhibition of its intestinal absorption mediated by uptake transporters.

Taken all together, green tea, green tea extract (GTE) and (-)-epigallocatechin-3-gallate (EGCG) can interact with the drug-metabolizing enzymes and drug transporters including nadolol.

Green Tea And Its Bioactive Polyphenols Which inhibits Fibromyalgia Caused by Sleep Disturbance

Green tea may have a therapeutic and positive effect in reduced onset, progression and treatment of fibromyalgia caused by sleep disturbance, some scientists suggested.


Fibromyalgia (FM) is a chronic condition characterized by muscle and soft tissue pain affecting over 10 million (3.6% of the population) people in the US alone.

Sleep disturbances are the symptoms of physical, mental, and spiritual problems. The major causes of sleeplessness and insomnia.

Insomnia, a  short-term sleep disturbance affects about 30% of adults and chronic insomnia by 10%, according to the statistic provided by the American Sleep Association.

Some researchers suggested that long-term untreated sleep disturbance is a major cause of stroke and heart disease. Dr. Dae Lim Koo, the lead author in the explanation of some risk factor of stroke, wrote, "Any causes of sleep curtailment or fragmentation such as sleep restriction, sleep apnea, insomnia, periodic limb movements during sleep, and shift work, not only impair cardiovascular restoration but also impose a stress on the cardiovascular system" and  "Sleep disturbances have been reported to play a role in the development of stroke and other cardiovascular disorders" 

Green tea, a precious drink processes numbers of health benefit known to almost everyone in Asia and the Western world.

In the study of sleep disturbance that causes fibromyalgia, Dr. Choy EH suggested that patients with fibromyalgia demonstrated a reduced short-wave sleep and abnormal α-rhythms, suggestive of wakefulness during non-REM (rapid eye movement) sleep compared to the healthy individuals according to the observation of polysomnography.

Furthermore, in healthy participants, the study indicated that sleep deprivation can also cause fibromyalgia-like symptoms such as myalgia, tenderness, and fatigue and suggested that sleep dysfunction might be not only a consequence of pain.

These results showed a strongly correlated condition of sleep disturbance and initiation of chronic fibromyalgia if sleep disorders are left untreated for a prolonged period of time.

Moreover, according to the presentation, long time effect of sleep disturbance may influence alter the pathophysiology of the central nervous system in abnormal pain processing and central sensitization, according to the observation of the neuroimaging of patients with fibromyalgia neural activation compared to a lower pressure-pain threshold in the healthy age-matched and gender-matched individuals.

After taking into account confounders, researchers said, " Improving sleep quality can reduce pain and fatigue, further supporting the hypothesis that sleep dysfunction is a pathogenic stimulus of fibromyalgia".

Moreover, green tea bioactive polyphenols (-)-Epigallocatechin gallate (EGCG) in the evaluation of the chick brain under an acute stressful condition-induced sleep disturbance, researchers found that EGCG attenuates stress behavior with special reference to gamma-aminobutyric acid (GABA), a neurotransmitter with function in sending chemical messages through the brain and the nervous system, and involving in regulating communication between brain cells in stress environment.

Deficiency of is associated with many nervous conditions, including depression, anxiety, and sleep disorders.

Application of EGCG of doses of 50, 100 and 200 microg, intracerebroventricularly exerted a profound effect in suppressing the vocalization which normally occurs during social separation-stress, one of the risk factors of sleep disturbance.

This illustration truly suggested that EGCG reduced the time spent in active wakefulness and induced sleep-like behavior in a dose-dependent manner.

Taken all together, the findings suggested that green tea and its bioactive polyphenols EGCG and L-theanine may be considered a secondary therapy in reducing risk, progression, and treatment of fibromyalgia caused by a sleep disorder.

However, intake of green tea supplement should be taken with extreme care due to recent reports of acute liver toxicity.

Fennel may have a potential and positive effect in protecting healthy cells against mutagenesis, some scientists suggested.

Mutagenesis is an action that causes the alteration of DNA transcripts in organisms caused either by adaptation to the new environment or mutagens. In other words, it is the change in cells genetic information for survival in a new environment or by the invasion of mutagens

In human, the change of genetic information can be classified in different ways. In healthy cells, the change of DNA transcript can cause cells to become cancerous.

Dt. Basu AK, at the University of Connecticut, in the study of "DNA Damage, Mutagenesis and Cancer" said, "Generation of DNA damage (also known as DNA adducts or lesions) induced by these agents is an important first step in the process of carcinogenesis".
In cancers cells, the change of DNA may prevent proliferation via induction of cancer death.

Dr. Apostolos G. The mice bone marrow chromosomal aberration (CA), micronucleus, and sperm abnormality assays were employed to measure genotoxicity and cytotoxicity in the entire experiment.

Mice used in the testing were administered two different doses of FEO (1 and 2 mL/kg) continuously for 3 days at intervals of 24 hours by the oral route before tissue sampling.

At the and of the study, researchers found that before administration of fennel essential oil, cyclophosphamide (CP) causes a significant increase in the average percentage of aberrant metaphases and induces cytotoxicity in mouse bone marrow cells, and abnormal sperms in the male germ line.

The application of CP also decreased the levels of antioxidants produced by the mice tissue such as
superoxide dismutase (SOD), glutathione (GSH), catalase (CAT) and increased levels of malondialdehyde (MDA).

These result suggested that CP induces a significant oxidative stress by causing an imbalance of the ratio of antioxidants and free radicals in the body of tested subjects.

Mice group pretreatment with FEO exerts a significantly inhibited against the induction of CP, including the frequencies of aberrant metaphases, CAs, micronuclei formation, and cytotoxicity in mouse bone marrow cells.

Furthermore, application of fennel essential oils also reduced the production of abnormal sperm and improved the antioxidants enzymes SOD, CAT, and GSH activities and inhibited oxidative stress observed by decreasing MDA content in the liver.

Dr. Tripathi P, the lead author, after taking into account co and confounders said, "FEO inhibits genotoxicity and oxidative stress induced by CP.

Additionally, in the comparison to examine the effect of plant peptides of thionine Ns-W2 extracted from seeds of fennel flower (Nigella sativa) and β-purothionine from wheat germs (Triticum kiharae), as well as a synthetic antimutagen (crown-compound), on the expression of several genes involved in the.control of cellular homeostasis, processes of carcinogenesis, and radiation response in human rhabdomyosarcoma cells (RD cells), researchers found that all of these agents acted as antimutagens-anticarcinogens, reducing the expression of genes involved in carcinogenesis through some complex pathways involving cellular proliferation, anti-apoptosis, cellular survival in tumor cells.

In compared to other agents, thionine Ns-W2 exerts the most pronounced reduction in the mRNA level of these genes.

Taken all together, fennel may be considered a chemotherapeutic functional food in the protection of our body against mutagenesis, the process that can lead to the onset of cancer.
