Herbal Arnica, The Functional Herb Which Stops the Onset of Chronic Diseases Associated with Oxidative Stress, Researchers Found

Arnica may have a profound and therapeutic effect in reducing the risk of diseases associated with overexpression oxidative stress, some scientist suggested.

The expression of oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

Dr. Ercal N, the lead author in the study of the relation of toxic metals and oxidative stress, wrote, "antioxidants may play an important role in abating some hazards of heavy metals. In order to prove the importance of using antioxidants in heavy metal poisoning, pertinent biochemical mechanisms for metal-induced oxidative stress".

There is no single cause of oxidative stress. Some researchers suggested that numbers of factors can facilitate the onset of oxidative stress, including cigarette smoke, oxidative burst from activated macrophages, particularly, the free radicals chain reaction.

Sadly, a prolonged period of oxidative stress presented in the human body can cause oxidative damage to biomolecules, (lipids, proteins, DNA) and death of healthy cells through cytotoxicity.

Certain chronic diseases are found to be associated with oxidative stress, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

By changing diet and lifestyle, oxidative stress can be managed or avoided, Dr. Seungyoun Jung, the lead author in the study of Healthy Dietary Patterns and Oxidative Stress suggested, "Healthy diets may lower oxidative stress and risk of chronic diseases" and "Among specific food groups that contribute to these diet scores, significantly positive associations were observed with legumes and vegetables for FlOP_360, vegetables and fruits for FlOP_320, and legumes and alcohol for FlOP_400".

Arnica is a herbaceous species, genus perennial, belonging to the family Asteraceae, native to Europe and Asia.

The Herbal medicine used in herbal medicine for centuries to treat strains, sprains, and bruises.

The ingredients
Helenalin, chamissonolid, Sesquiterpene lactone, lignans of the furofuran, dibenzylbutyrolactone, dibenzylbutyrolactol types, pinoresinol, epipinoresinol, phillygenin, matairesinol, nortrachelogenin, and nortracheloside, six dibenzylbutyrolactol derivatives.

In the analysis of the effect of Arnica Arnica on mitochondrial oxidative stress induced by Ca(2+) plus inorganic phosphate and/or Fe(2+)-citrate-mediated lipid peroxidation through changes in oxygen consumption rates in adult male Wistar rat, researchers found that application of Arnica at dose of 6cH, 12cH, 30cH or succussed 30% ethanol (control) for 21 days, exert a strong activity in reduced expression of lipid peroxidation of mitochondrial membranes through inhibiting the antimycin-A, a toxic chemical with function in initiated production of free radicals.

Under oxidative stress conditions caused by injection of Ca(2+) and inorganic phosphate, tested group with a dose of Arnica 30cH demonstrated the effect in decreasing mitochondrial O(2), an indication of lesser expression of free radicals presented in the mitochondrion, through the herbal antioxidant activity.

In liver tissues, administration of Anise at dose 30cH in the tested group also protected the hepatic mitochondrial membrane permeabilization by inhibited ROS on the induction of lipid peroxidation and precipitated fragmentation of proteins.

Further evaluation of the study showed that these results of free radical inhibitory effect were attributed to levels of antioxidant from the herb and the herb in the stimulated production of antioxidants of the tested host.

Additionally, in the investigate the therapeutic effects and mechanism of action of A. Montana flower methanol extract (AMME) against both inflammation and oxidative stress in a collagen-induced arthritis (CIA) rat model, scientists suggested that oral administration of AMME reduces clinical signs and improves the histological and radiological status of the hind limb joints, through reducing overexpression of levels of nitric oxide, tumor necrosis factor-α, interleukins (IL-1β, IL-6 and IL-12) and titer of anti-type II collagen antibody activated by high levels of oxidative stress.

This inhibitory effect of AMME against overproduction inflammatory factors not only reverses the disturbed antioxidant levels and peroxidative damage but also inhibits the overproduction of inflammatory cytokines in facilitated cell damage and healthy cell death at the site of infection.

Moreover, according to the gas chromatography/mass spectrometry (GC/MS) analysis, the function of Arnica in the alleviation of arthritis by ameliorating ROS expression was attributed to the combined action of phenolic and flavonoid compounds.

Dr. Sharma S, the lead author said, " The study also shed some light on mechanisms involved in the diminution of inflammatory mediators and free radical-generating toxicants and enhancement of the antioxidant armory, thereby preventing further tissue damage, injury and synovial hyperproliferation in arthritis".


The above differentiation was supported by the National Institute of Research and Development for Biological Sciences study of A. montana and A. absinthium ethanolic extracts antioxidant activity and protective effect against H2O2-induced oxidative stress in a mouse fibroblast-like NCTC cell line.

According to the final assessment, researchers suggested that both A. absinthium and A. montana extract showed a strong antioxidant capacity but lesser in A. montana, compared to A. absinthium.

At the doses of the range of 10-100 mg/L and A. montana and 10-500 mg/L A. absinthium, both extracts exhibited a significant oxygen radical absorbance capacity and 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging activity and protected fibroblast cells against hydrogen peroxide-induced oxidative damage, depending on the levels of flavonoids and phenolic acids contents presented in the extracts.

According to the neutral red and lactate dehydrogenase assays, pretreatment with 10 mg/L A. montana and 10-300 mg/L A. absinthium was considered the best protection against the early onset of ROS production in H2O2-induced oxidative stress in a mouse fibroblast-like NCTC cell line.

Importantly, pre-treatment with A. montana and A. absinthium extracts also restored the proportion of cells in each phase of the cell cycle through inhibiting oxidative stress in the induction of cytotoxicity.

Taken together, Arnica and its bioactive chemical may be considered a functional food in reduced inflammatory-related diseases caused by overexpression of oxidative stress. However, intake of Arnica supplement should be taken with extreme care for the prevention of acute liver toxicity.

Yoga Therapy In the Prevention and Treatment of Chronic Inflammatory Diseases

Yoga therapy may have a strong impact in reducing the risk of inflammation by significantly decreased expression of pro-inflammatory cytokines, a US university study suggested.

Inflammation is a natural response of the immune system in the stimulation of the production of inflammatory cytokines with an aim to protect our body against infectious agents in the acute phase of bodily injury and tissue damage.

However, in many instances, overexpression of pro-inflammatory cytokines can cause cells death and tissue damage, leading to irreversible organs damage with large scars formation.

For example, overexpression of pro-inflammatory cytokines in the healing of hepatitis virus infection can lead to liver scars.

Furthermore, inflammation that lasts more than 3 months may be considered chronic inflammation, a leading cause of chronic diseases.

Chronic inflammation occurs when the immune system no longer has the ability to kill off all the infectious agents, during the onset of acute infection.

Moreover, chronic inflammation also happens in people with the autoimmune disorder. In this case, the immune system attacks the bodily normal healthy tissue, by mistaking them for foreign pathogens that cause disease.

Taking all together, Yoga may be considered as an integrated form of exercise in reduced risk and treatment of inflammation-inducing poor behavioral and physical activities.

Green Tea and Its' Polyphenols (GTPs) That Protect Our Bodily Skin Against the Onset of Cancer, Scientists Say

Green tea may have a strong and positive effect in reduced risk and treatment of skin cancer, some scientists suggested.

Skin cancer is a medical condition characterized by irregular cell growth in the issues of skin.

At the later stage, the cancerous cell may travel a distance away to invade other healthy tissues and organs.

According to statistic, approximately, 87,110 new cases of invasive melanoma are diagnosed in the U.S every year.

The exact causes of skin cancer are unknown. Researchers believed that there are a set of factors that can have the substantial effect to initiate the onset of skin cancer.

Truly, according to the studies, people who have fair skin, regardless of skin color, a history of sunburns, excessive sun exposure, including occupations which are required to work outside during Summar months, moles, family history of skin cancer. and previous and personal history of skin cancer is at the increased risk of skin cancer or recurrence of skin cancer.

Particularly, (-)-Epigallocatechin-3-gallate (EGCG), the major and most photoprotective polyphenolic component of green tea also exhibited a protective activity against UV in induction of skin cancer, through induction of immunoregulatory cytokine interleukin (IL) 12 in differentiation of naive T cells into Th1 cells and acted as T cell-stimulating factor in stimulate the growth and function of T cells.

Moreover, interleukin (IL) 12 also demonstrated a significant effect as an antagonist against immunosuppression induced by solar/ultraviolet (UV) radiation by restoring immune responses and also stimulating DNA repair.

Further analysis also indicated that green tea (-)-Epigallocatechin-3-gallate (EGCG) also inhibited UV in the induction of skin tumors; the angiogenetic effect in the stimulated production of new blood vessels to
provide nutrients and oxygen for cancer cells proliferation and survivals and initiated cytotoxic T lymphocyte cells in induced cancer and infectious and damage cells apoptosis.


Interestingly, additional study of green tea polyphenols (GTPs) in prevention of UVB irradiated (80 mJ/cm(2)) 3 times each week for 24 wk in induced skin cancer in mice divided in to group consumed water and group fed with water containing 2 g/L GTPs, showed that group treated with GTPs displayed a significantly reduced UVB-induced tumor incidence by 35%, tumor multiplicity by 63%, and tumor growth by 55% in compared to non treatment group.

Group treated with GTPs+UVB also demonstrated a reduced expression of the matrix metalloproteinases (MMP)-2 and MMP-9, which are activator associated with progression from benign to advanced cancer and enhanced expression of specific endogenous tissue inhibitors of metalloproteinases (TIMPs), tissues inhibitor of MMP in skin tumors in compared to mice treated with UVB alone.

Furthermore, GTPs+UVB group also reduced expressions of CD31 and vascular endothelial growth factor, inactivated development of new blood vessel to nourish skin tumor cells proliferation and site expansion and inhibited expression of proliferating cell nuclear antigen, a key factor in DNA replication and cell cycle regulation in skin cancer in compared to UVB group.

More importantly, mice treated with GTPs+UVB group also showed more cytotoxic CD8(+) T cells in the tumors in compared to UVB group by exhibited greater activation of caspase-3 expression in induced apoptosis of the tumor cells.

The information findings suggested a potential and therapeutic effect of green tea and its bioactive polyphenols (GTPs) in attenuated risk, progression, and treatment of melanoma and non-melanoma skin cancers.

Furthermore, application of LPS also induced production of inflammatory mediators such as matrix metalloproteinase 9 (MMP-9), tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) all systematic inflammatory cytokines in response to cellular damage and tissue injury.

Moreover, LPS also exhibited overproduction of nitric oxide (NO) observed in bronchoalveolar lavage fluid in facilitated cellular cytotoxicity.

In the compared nontreatment group, injection anethole group demonstrated a strong anti-inflammatory effect through decrease activities of proinflammatory factors without affecting the immune activity in protects against infection.

Additional analysis also found that treatment group reduced acute lung injury caused by injection LPS through its antioxidants and induced production of host natural antioxidant in the body tissue.


The inhibited LPS results according to were attributed to anethole effect in suppressed the activation of NF-κB in signaling production of inflammatory cytokines by blocking the initiation of IκB-α degradation by inhibited TRAF2 in the induction of inflammation through TNF receptor superfamily members and NIK, a key signaling molecule of the noncanonical NF-κB.

Interestingly, further investigation of anethole as an inhibitor of TNF-induced NF-kB activation (an early response) as monitored by electrophoretic mobility shift assay, by suppression of IkBa phosphorylation and NF-kB reporter gene, researchers also indicated that
* Anethole also suppressed TNF-induced activation of the transcription factor AP-1 in regulated gene expression in response to a variety of stimuli, including LPS application.

*c-Jun N-terminal kinases (JNKs) with function in response to stress stimuli, such as cytokines,
And

* MAPKkinase in signal inflammatory response through transmitting information from a receptor on the surface of the cell to the DNA in the nucleus of the cell.

* Application of anethole also suppressed TNF-induced both lipid peroxidation and ROS generation, through the stimulated production of natural antioxidant in the host and itself, resulting in balancing the ratio of antioxidant and ROS expression.

There were no differences between the two groups in pain relief and improvement of hand stiffness.

However, compared to 2 groups, researchers found that adverse effects were reported by six patients (6.1%) on ibuprofen and by five patients (4.8%) on arnica.

These result suggested that Arnica topical preparation is compatible with those of ibuprofen for the treatment of patients with osteoarthritis, particularly in the patients with hand's osteoarthritis.

Further analysis of the safety and efficacy of an Arnica montana fresh plant gel, applied twice daily, in 26 men and 53 women with mild to moderate osteoarthritis (OA) of the knee, after 3 and weeks of application, researchers indicated that
* All groups (treatment with 3 and 6 weeks) display a significant improvement of scores on the pain, stiffness, and function subscales.

* The topical application also shows significant reductions at these time points.

* Total scores on the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) are decreased substantially in all patients.

* According to the returned questionnaire, 69 patients (87%) rate the tolerability of the topical gel as "good" or "fairly good,"

However, the overall local adverse-event rate is 7.6% included only one allergic reaction.
