In further confirmation of carrot effect in inhibited leukaemia, five fractions from carrot juice extract (CJE) were tested on human lymphoid leukaemia cell lines, including 3 polyacetylenes (falcarinol, falcarindiol and falcarindiol-3-acetate) and 2 carotenoids (beta-carotene and lutein).

According to the Annexin V/PI and Caspase 3 activity assays and Cell Titer Glo assay and cell cycle analysis,
* Injection of carrot extracts which contained polyacetylenes and carotenoids was significantly more cytotoxic in all three lymphoid leukaemia cell linesthan the 4 other fractions.

* Treatments with purified polyacetylenes also induced apoptosis in a dose and time responsive manner.

* Falcarinol and falcarindiol-3-acetate isolated from Daucus carota L were more cytotoxic in compared to falcarindiol

* Interestingly, the carotenoids treatment showed no significant effect on either apoptosis or proliferation in any of the tested cell lines.

These results suggested that carrot processed anti leukemia through induction of cell apoptosis via cytotocity and cell cycle arrest was attributed to the contents of polyacetylenes rather than beta-carotene or lutein.

Additionally, in the study of carrot juice extract effect against leukemia cells, researchers at the Sheffield Hallam University found that on myeloid and lymphoid leukemia cell lines together with normal hematopoietic stem cells, injection of carrot juice extracts to 72 hours induces apoptosis and inhibited progression through the cell cycle.

In classification of types of cell line, also indicated that lymphoid cell lines are affected to a greater extent in compared to myeloid cell lines. Application of the extract showed no effect on normal hematopoietic stem cells.

The experiment strongly suggested that carrot juice extract significantly induces apoptosis and causes cell cycle arrest in leukemia cell lines without harming to the healthy nearby cells.

Taken together, carrot and its extract may be considered as potent and therapeutic therapies for prevention of leukemia.

However, intake of carrot supplement should be taken with extreme care in prevention of acute liver toxicity.

Intake of nitrate-rich beetroot juice displayed a significant increase in the FMD response of 1.1% in compared to placebo group of decrease of 0.3%

Nitrate-rich beetroot juice group also demonstrated a small improvement of stiffness measured by Aortic Pulse Wave Velocity and showed a trend of continuous improvement in compared to placebo.

Aortic Pulse Wave Velocity is a test to measure stiffness of the largest artery in the body, Increased stiffness will result in reduced mobility in related regions.


Result of aortic pulse wave velocity measurement is said to have an ability to identify people in their 20’s who are at increased risk for development of heart disease, stroke and dementia.


Administration of nitrate-rich beetroot juice showed a strong reduction of 7.6% of platelet-monocyte aggregates in compared to increase of 10.1% in placebo. This finding indicated that nitrate-rich beetroot juice intake had a substantial effect in decreased risk of acute myocardial infarction.

Platelet–monocyte aggregates is a novel index with platelet activation and a maker of early onset of acute myocardial infarction.


Furthermore, intake of nitrate-rich beetroot juice group expressed an exceptional improvement of vascular function in reduced P-selectin expression in activated platelet aggression

P-selectin is a cell adhesion molecule (CAM) with function in mediated the interaction of activated endothelial cells or platelets.


Additionally, application of the juice also improved the nonpathogenic, commensal bacteria present in the healthy human salivary glands measured by salivary microbiome in compared to placebo group.


Interestingly, intake of the nitrate-rich beetroot juice also responded to change of 78 bacterial taxa out of thousands harbored in human gut.


The proportions of Rothia mucilaginosa trending to increase in nitrate-rich beetroot juice group, in compared to Neisseria flavescens (P < 0.01) increased in the placebo group.


Rothia mucilaginosa, is considered part of the normal microflora of the human mouth and the upper respiratory tract.

Neisseria flavescens is an uncommon pathogen of human infection, pneumonia and empyema


There were no adverse effects of dietary nitrate were detected through out the experiment.

The finding evidences suggested that intake of nitrate-rich beetroot juice daily may have a sustainable effect in improve vascular function, thus reducing risk of heart disease.

Monoaminergic systems and glutamate and γ-aminobutyric acid (GABA) homeostasis are vital for iron metabolism.

Investigation of the green tea (−)-epigallocatechin-3-gallate (EGCG) effect in risk and progression of PD has been associated to many mechanisms involved numbers of aspects.


Application of metal chelation was found to ameliorate the expression of ROS in induction of oxidative stress through a chain of reaction and precipitation of aggregation of alpha-synuclein and beta-amyloid peptides deposit in the brain, a main cause on the onset of PD.


In iron abnormality PD subjects, oxidopamine, also known as 6-hydroxydopamine or 2,4,5-trihydroxyphenethylamine(6-OHD), a neurotoxic synthetic organic compound was found to disrupt iron metabolism, leading to over expression of the iron in induction of dyshomeostasis cause of iron toxicity and oxidative damage in dopaminergic neurons.


Green tea major polyphenol, (−)-epigallocatechin-3-gallate used in metal chelation, ameliorated iron dependent free radicals located in the subcellular compartments and cellular membranes, through the effect in inhibited iron input and exhibited iron output in brain areas where it preferentially accumulates in neurodegenerative diseases, particularly Parkinsons' disease.


Further analysis of the neurotoxin 6-OHDA causes of iron unstable equilibrium, green extract GECG administration improved the function of hepcidin, a liver-derived hormone in regulated iron dyshomeostasis and gene expression of the iron metabolism through expressions of iron regulatory proteins (IRP1 and IRP2) in regulated the RNA level, between the transcription and the translation of the gene in expression of the iron homeostasis.

Further study of the mechanism on disruption of iron metabolism by 6-OHDA in PD cell line N27, discovered that application of the iron chelator EGCG also disrupts the iron dyshomeostasis in N27 cells (dopaminergic neural cell line) induced by 6-OHDA through regulating iron transporters and regulators expression, such as DMT1 with function in mediation of the entry of dietary iron into these mucosal cells and TfR, a carrier protein for transferrin with function in import iron into the cell and regulated response to intracellular iron concentration and hepcidin, a central regulator in maintain iron homeostasis.

Dysregulation of hepcidin production results in a variety of iron disorders.


Additionally, in 6-OHDA induced neurotoxicity to cause PD, administration of green tea EGCG also reduced the intracellular iron retention by decreasing the iron importers and increasing iron exporters, through regulation of the function of iron-regulated transporters, such as ferroportin.


Interestingly, there was a report suggested that intervention of 3 cups of green tea daily for 3 months improved the antioxidant status and reduced oxidative damage in early PD patients without affecting their iron status.

The findings revealed that green tea bioactive polyphenol, (−)-epigallocatechin-3-gallate may be considered as iron chelator in regulation of PD progression.
