Omega 3 Fatty Acids in the Prevention, Treatment and Management of Obesity

Obesity is a medical condition caused by excess body fat accumulated to some extent that impacts personal health. The hallmark of obesity is attenuated metabolism and structural cell changes in the obese fat observed by reducing metabolic activity.

According to the BMI index, a person is considered overweight, if he/she has a BMI between 25 to 29.9 kg/m2. A BMI of 30 kg/m2 or more is an indication of obesity.

In other words, obesity is a progressive chronic disease characterized by abnormal fat accumulation in the body tissues that may impair health.

According to the statistics provided by the National Center for Health Statistics (2015-2016), in the US, approximately 39.8% of adults aged 20 and over were obese accompanied by another 31.8% overweight.

Visceral fat obesity is associated with 90% of patients with ischemic heart disease. In obese, accumulation of visceral fat is predominant in the intra-abdominal cavity.

Visceral fat is a layer of fat that surrounds the organs. Overexpression of visceral fat has been found to promote insulin resistance, diabetes, heart disease, and inflammation.

Similar to other chronic medical conditions including diabetes, cardiovascular disease or high blood pressure, obesity has recently become a serious health problem.

Epidemiologically, obesity does not only have serious effects on physical and psychological health but also has a strongly negative impact on the patient's quality of life.

Today, the widespread obesity in the US population have caused a significant economic impact on the U.S. health care system, including direct and indirect costs.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of diseases associated with obesity, researchers examined the Omega 3 fatty acid activity in the prevalence of overweight and obesity.

According to the results of the study,
* Omega-6 and omega-3 fatty acids elicit divergent effects on body fat gain through mechanisms associated with the risk of weight gain, including adipogenesis, browning of adipose tissue, lipid homeostasis, brain-gut-adipose tissue axis.

* Omega 3 fatty acids also modulated the immune response in the stimulation of the production of pro-inflammatory cytokines.

* Furthermore, the risk of obesity is increased if the level of omega-6 fatty acids and the omega-6/omega-3 ratio was increased in red blood cell (RBC) membrane phospholipids.

* However, the high omega-3 RBC membrane phospholipids were associated with the decreased risk of obesity.

Moreover, researchers said, " in addition to absolute amounts of omega-6 and omega-3 fatty acid intake, the omega-6/omega-3 ratio plays an important role in increasing the development of obesity via both AA eicosanoid metabolites and hyperactivity of the cannabinoid system, which can be reversed with increased intake of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). A balanced omega-6/omega-3 ratio is important for health and in the prevention and management of obesity".


Taken altogether, Omega-3 fatty acids may be considered a functional food for the prevention and treatment of obesity, pending to the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Brassica Vegetables Inhibit the Onset of Hepatic Cancer Cells in Vitro

The liver is the largest internal organ found in the human which plays an essential role in filtering the blood from the digestive system before passing them to other parts of the body.

The liver also produces cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Liver disease is a class of medical conditions characterized by the reducing partly or completely the function of the liver, including liver failure.

Liver cancer is a medical condition characterized by cell growth irregularly in the liver tissue.

The causes of liver cancer are associated with the alternation of cells DNA which normally begins in the cells on the surface of the inner lining.

Liver cancer at the early stage no not cause any symptoms. However, at the advanced stage, liver cancer cells can cause a wide range of symptoms including jaundice, abdominal swelling and pain, and dark urine and white stool accompanied by the general symptoms of cancers such as unintended weight loss, loss of appetite, chronic fatigue and tiredness and gastrointestinal discomforts.

The most common cause of liver cancer is infected by the hepatitis virus. However, in North America, due to overexpression of overweight and obese of the population, liver cancer may be slowly progressed from nonalcoholic fatty liver disease through cirrhosis to liver cancer.

In 2017, 2,500 Canadians were diagnosed with liver cancer and 1,200 Canadians died from liver cancer.

Compared to the early stage of other cancer, the 5 years survival rate in the early stage diagnosis is only 31% and 43% of people with liver cancer diagnosed at stage 0 and 1.

Cabbage is a species of Brassica oleracea belongings to the family Brassicaceae (or Cruciferae), native to the Mediterranean region along the seacoast. The veggie has a short stem of which is crowded a mass of leaves, usually green but in some varieties, it may be red or purplish.

With an aim to find a natural food that inhibits cancer, researchers examined the bioactivity of synthetic and plant-derived 1-cyano-2,3-epithiopropane (CETP) - the predominant epithionitrile in Brassica vegetables in three human hepatocellular carcinomas (HCC) cell lines.

According to the formazan dye assays, the concentrations of 380-1500 nM CETP isolated from Brassica vegetables inhibited the mitochondrial dehydrogenase activity-induced cytotoxicity of human HCC cells without impairment of cell growth.

Furthermore, at 100-fold higher CETP concentrations, the bioactive induced cell death.

The presence of a plant matrix associated with oxidative stress increased CETP-based toxicity without exhibiting the epithionitriles in human health.

Taken altogether, Brassica vegetables may be considered probiotics for the promotion of digestive functioning, pending to the confirmation of the larger sample size and multicenter human study.

Green-Plant Membrane inhibits the Onset of Hypertriglyceridemia

Triglycerides are accumulated in the blood for future use by converting any unused calories.

In other words, triglycerides are an ester derived from glycerol and three fatty acids (from tri- and glyceride).

Overexpression of hyperglyceridemia is found to patients with atherosclerosis even without the presence of high cholesterol levels, and predispose.

Hypertriglyceridemia is a condition characterized by high levels of triglycerides in the blood.

Hypertriglyceridemia can also be accumulated to the arterial wall or combined with other substances to form plaque that narrows the arteries in transporting rich oxygenated blood to the heart and other parts of the body, leading to the onset of atherosclerosis.

Primary hypertriglyceridemia is found in 5% of patients associated with genetic preposition compared to 95% of secondary hypertriglyceridemia found in obese metabolic syndrome, diabetes,... patients.

Treating hypertriglyceridemia is difficult, as the condition is frequent coexistence with other conditions that affect cardiovascular disease risks, such as high levels of low-density lipoprotein, obesity, metabolic syndrome, and type 2 diabetes.

In severe cases of hypertriglyceridemia, patients may also experience the complication of acute pancreatitis, independent of the risk of CVD.

On finding a potential wholefood for the treatment of diabetes, researchers examined the effects of the once-daily intake of the green-plant membrane with dietary oil on postprandial hypertriglyceridemia in vitro and in vivo.

In vitro, green-plant membrane-bound hydrophobic bile acids but did not inhibit pancreatic lipase activity.

The green-plant membrane in rats, oral fat tolerance tests, increased serum triglycerides levels were significantly reduced.

Moreover, fecal total lipid and bile acid volumes were significantly increased in rats that administered 200 mg/mL green-plant membrane.

Based on the evidence, researchers said, "...green-plant membrane with dietary oil inhibits dietary fat absorption via promotion of bile acid excretion in feces and the effectiveness of eating green-plant membrane, such as green leafy vegetables, with meals".

Taken altogether, the green-plant membrane may be considered a functional food for the prevention of hypertriglyceridemia, pending to the confirmation of the larger sample size and multicenter human study.

The digestive system included the gastrointestinal tract, the liver, pancreas, and gallbladder play a critical role to maintain the body normal function by providing nutrients and fluids to the body and remove waste through the secretary system.

Digestion is a process of the digestive system by breaking out down food after food intake through mechanical and enzymatic action.

Dr. Catalioto RM, the lead scientist in examing the intestinal epithelial barrier dysfunction wrote, "Epithelial cells are linked together at the apical junctional complex by tight junctions that reduce the extracellular space and the passage of charge entities while forming a physical barrier to lipophilic molecules".

In vitro mild irritants, proinflammatory cytokines, toxins and pathogens, and adverse environmental conditions open tight junctions and increase paracellular permeability, an effect often accompanied by immune activation of the enterocytes".

In other words, intestinal epithelium dysfunction induces the risk of various intestinal and non-intestinal disorders including inflammatory bowel disease, celiac disease, and diarrhoeal infection.

Coltsfoot(Tussilago farfara) is a perennial herbaceous plant, genus Tussilago, belonging to the family Asteraceae, native to Europe and Asia. The herb has been used over thousands of years to treat asthma, various coughs, bronchial congestion, respiratory disorders, headaches and obstruction in the nasal passage, etc.

On finding a potential compound for the improvement of the digestive functioning, researchers examined the protective effect of Tussilagone (TSL) on mall intestinal epithelium against foreign pathogens.

According to the DNA comet assay, in C57Bl/6 mice, the chemical constituent of polysaccharides from Tussilago farfara L. reduced the intensity of polychemotherapy-induced apoptosis in the intestinal epithelial cells.

Green Pea (Pisum Sativum) Eradicates Ehrlich Ascites Carcinoma Cells

Cancer is the class of diseases associated with cell growth disorderly and uncontrollably due to the alternation of DNA.

Most cases of cancer begin in the cells on the surface of the inner lining tissues of organs or glands. At the early stage, most cancers are asymptomatic.

However, at the advanced stage, most cancers not can induce common symptoms such as gastrointestinal discomforts, unintended weight loss, loss of appetite and fatigue but also localized symptoms depending on the affected tissues and organs.

Cancers at this stage also can travel a distance away from the original site to induce secondary metastasis.

According to the statistics provided by the American cancer society, in 2018,1,735,350 new cases were diagnosed, representing more than 4,700 new cancer diagnoses each day.

The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.
Ehrlich-Lettre ascites carcinoma (EAC) known as Ehrlich cell is a type of abnormal buildup of fluid in the abdomen tumor cultured in mice.

Ehrlich Ascites Carcinoma cells induced subcutaneous tumors in 129/SvJ mice has been studied a number of researchers such as Sneha Mishra,  Ankit Kumar Tamta for its effect on several types of cancer.

Compared to all cancers, lung, breast, prostate, and colorectal cancers are the most common cancer in the US.

The causes of cancer are unknown. age, excessive alcohol drinking, exposure to cancer causing-substances, diseases associated with chronic inflammation, unhealthy diet, hormones therapy, use of immunosuppression are some of the prevalent factors involved in the onset of cancer.

 
Green peas are species of Pisum Sativum belongings to the family Fabaceae and native to western Asia from the Mediterranean Sea to the Himalaya Mountains. The pea is a green, pod-shaped vegetable and a cool-season crop grown in many parts of the world.

On finding a potent ingredient for the treatment of inflammatory bowel disease, researchers investigated the impact of green pea (GP) effects on Ehrlich ascites carcinoma cell line.

Based on the results, a lectin isolated from green pea at the concentration range of 8-120 μg/ml in RPMI 1640 medium showed an 11.7-84% inhibitory effect against Ehrlich ascites carcinoma (EAC) cells.  

Furthermore, in vivo, administered 2.8 mg/kg/day and 1.4 mg/kg/day (i.p.) respectively for five consequent days exerted a strong 63% and 44% growth inhibition against EAC cells in a mice model.

More importantly, pea lectin injection into the EAC bearing mice for 10 days significantly increased the hemoglobin and RBC with the decreased of WBC levels toward the normal and induced the cell cycle arrest at the G2/M phase of EAC cells, according to flow cytometry assay.

Pea lectin also induced cancer cell apoptosis by increasing the levels of pro-apoptotic protein and decreasing the anti-apoptosis in the tested cells, after 5 consecutive days of treatment.

Taken altogether, green peas with abundant phytochemical lectin may be considered a functional food for the prevention and treatment of cancers,  pending to the confirmation of the larger sample size and multicenter human study.

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.

On finding a potent compound for the treatment of kidney cancer, researchers examined the effects of lycopene on renal cell carcinoma (RCC) associated with frequent upper urinary tract cancer in humans, accounting for 80-85% of malignant renal tumors.

The study included  10 weeks old Eker rat, the unique animal model to study RCC induced spontaneous renal tumors with a genetic mutation of TSC2 received 0, 100 or 200mg/kg of lycopene as part of their daily diet for 18 months.

According to the tested analysis associated with tumors counted measured in individual kidneys, lycopene treated group showed a decrease of 94% of tumor counts compared to 65% in the control group.

The mean numbers of renal carcinomas in the treated group were statistically significantly decreased compared to untreated controls.

Furthermore, the decrease of tumor counts in the lycopene was linearly depending on the concentration of the phytochemical observed by the increase of the supplement from 0 to 200 mg/kg.

Moreover, compared to control rats fed an only basal diet with a greater length of tumors (23.98 mm)compared to rats fed lycopene supplement with the length of 12.90 mm and 11.07 mm at the concentration 0 to 200mg/kg respectively.

The results of this efficacy were attributed to lycopene in the promotion of antibodies against proteins mTOR associated with cell proliferation and cancer progression and phospho-S6 protein associated with predicts unfavorable clinical survival and epidermal growth factor receptor (EGFR) in the promotion of solid tumor growth.

Taken altogether, lycopene found in tomato may be considered supplements for the prevention and treatment of kidney cancer pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
