Cancer is a class of diseases characterized by abnormal cell growth that begins in the cells on the surface of the inner lining of the tissue of an organ caused by the alternation of cell DNA with unknown nature.
Under normal circumstances, cell growth is a natural process that happens daily to replace millions of dead cells. In some cases, acceleration of cell growth is necessary to prevent further damage to the body organ, such as cut, injure, etc., but after fulfilling their purposes, the growth of cells are back to a normal rate again regulated growth inhibitors.
In other words, healthy cell division is limited to several cycles. Comapred to normal cells, cells with an alternation of DNA or cancer cells can grow indefinitely.
Unfortunately, skin cells sometimes overgrow while healing, leading to scar to the skin.
At the early, most people with cancer are asymptomatic, however, later, due to the size and the tumor, patients may experience general cancer symptoms such as unintended weight loss, fatigue, gastrointestinal discomfort, and loss of appetite and localized symptoms such as severe pain and bleeding.
Cancer can be classified by the types of cells that have become malignant.
* Carcinoma
Carcinoma is a malignant tumor found in anywhere of epithelial tissues lining the cavities, surfaces of structures and glands.
* Sarcoma
Sarcoma is a type of cancer that arises from malignantly connective tissue cells that originated from embryonic germ cells or bone, cartilage, and fat tissues and cells.
* Lymphoma and leukemia
Hematopoietic cells are multipotent stem cells that give rise to all types of blood cells, including red blood cells, white blood cells, and platelets become cancerous.
* Germ cell tumor
Cancer is derived from pluripotent cells. In adults, these are most often found in the testicle and ovary but are more common in babies and young children.
* Blastoma
Blastoma is cancer derived from immature "precursor" or embryonic tissue, mostly in children.
The causes of cancer are not identified. However, most prevalent factors associated with the cancer onset include age, family history, long-term exposure to certain cancer-induced chemicals, use of tobacco and excessive alcohol drinking,..... the list goes on and on, depending on the types of cancer.
Some researchers suggested that the most common risk factor that causes cancer in the Western world may be associated with widespread obesity.
Dr. Karen Basen-Engquist, in the evaluation of the obesity risk of cancer, said, "In the United States, approximately 85,000 new cancer cases per year are related to obesity. Recent research has found that as the body mass index increases by 5 kg/m2, cancer mortality increases by 10%".And, "High priorities for future research involve additional work on the underlying mechanisms, and trials to examine the effect of lifestyle behavior change and weight loss interventions on cancer and intermediate biomarkers".
The findings strongly indicated the risk of cancer is increased substantially in obese.
Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.
In finding a potential compound for the treatment of invasive tumors, researchers examined the potential effects of cranberry on cancer prevention and treatment.
According to the review of the medical literature by searching the database of PubMed, cranberry and/or its components may act as chemopreventive agents, that diminish the parameters associated with the onset of cancer, including the inhibition of cells oxidation and inflammatory-related processes, in vitro.
Cell proliferation angiogenesis and metastasis involved in cancer cell invasion were also attenuated by the administration of cranberry's apoptotic activity.
Dr. Mantzorou M, the lead scientist, after taking other factors into account wrote, " Cranberry could be considered as a conglomeration of potential effective anticancer drug-like compounds".
Additionally, in chemical constituents analysis, cranberry showed inhibitory action against cancers of the esophagus, stomach, colon, bladder, prostate, glioblastoma, and lymphoma.
The efficacy of anti-cancer activity was attributed to the chemical compounds that work synergistically to result in cellular death induction via apoptosis, necrosis, and autophagy, reduction of cellular proliferation, the elevation of reactive oxygen species, and modification of cytokine and signal transduction pathways, similar to those of aforementioned study.
Taken together, cranberry may be considered a remedy for the prevention and treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.

#Phytochemical #Allicin Inhibits Liver Oxidativestress by Preventing Lipid Peroxidation of Liver Homogenate, Researchers Say

Phytochemicals may be the next potential source of new medications for the treatment of diseases with little or no side effects.
Scientists may have found a dietary and bioactive ingredient for the prevention and treatment of diseases associated with oxidative stress.
Allicin is a phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in onion and garlic.
Oxidative stress is a condition characterized by the overexpression of free radicals in the host body.
Most cases of oxidative stress are caused by the inability of the host liver to stimulate the production of antioxidant enzymes which play a critical role in inhibiting the free radical expression, causing oxidative stress.
In a healthy individual, the ratio of free radicals and antioxidant enzymes is always balanced.
However, an individual whose antioxidant enzymes were suppressed due to damage to the antioxidant enzymes' productive system or other sources may have a greater risk of the onset of oxidative stress.
Dietary antioxidant intake such as fruits and vegetables may be helpful to keep the free radicals in check.
Oxidative stress has been found to cause damage to cell lipids, proteins, and DNA, leading to many chronic diseases, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.
The oxidative stress that causes chronic ailment is a result of a chain reaction started by free radical donating or stealing an electron from another atom, leading to a domino effect that can not be stopped until all outer ring electron of free radicals is paired or inhibited by antioxidants.
A free radical is any atom or molecule that has a single unpaired electron in an outer shell and is highly reactive to react with another cell.
Most free radicals and other ROS are derived internally either from normal essential metabolic processes in the human body or from external sources such as exposure to X-rays, ozone, cigarette smoking, air pollutants, and industrial chemicals.
Antioxidants are biomolecules that inhibit oxidation.
On finding a natural ingredient and whole food for the prevention and treatment of diseases associated with oxidative stress, researchers investigated the effect of allicin (the active ingredient of garlic) contained in the commercial preparation Garlicin against hydroxyl radicals (.OH) using high-pressure liquid chromatographic (HPLC) method.
Allicin equivalent in Garlicin (1.8, 3.6, 7.2, 14.4, 21.6, 28.8, and 36 micrograms) produced concentration-dependent decreases in the formation of 2,3-DHBA and 2,5-DHBA, the OH adduct products.
The inhibition of the formation of 2,3-DHBA and 2,5-DHBA with 1.8 micrograms/ml was 32.36% and 43.2% respectively while with 36.0 micrograms/ml the inhibition was approximately 94.0% and 90.0% respectively.
These results suggested that allicin is highly active against the OH adduct products through its free radical scavenging activity.
Furthermore, Allicin also exerted a significant antioxidant activity by preventing the lipid peroxidation of liver homogenate in a concentration-dependent manner.
Additionally, by examining the antioxidant effects of four main chemical classes, alliin, allyl cysteine, allyl disulfide, and allicin, prepared by chemical synthesis or purification, researchers discovered
* Alliin scavenged superoxide, while allyl cysteine and allyl disulfide did not react with superoxide.
* Allicin suppressed the formation of superoxide by the xanthine/xanthine oxidase system, probably via a thiol exchange mechanism.
* Allicin did not exhibit hydroxyl radical scavenging activity in this study, probably due to some unknown mechanisms.
The finding suggested that allyl disulfide, alliin, allicin, and allyl cysteine exhibit different patterns of antioxidant activities as protective compounds against free radical damage.
Taken altogether, allicin-processed high levels of free radicals scavenging activity may be used for the prevention of the overexpression of ROS in the initiation of oxidative stress.

Dithiolthione, a phytochemical found abundantly in cruciferous vegetables may be the next target for the development of chemomedicine for the treatment of cancer without inducing any side effects, some studies suggested.
Cancers are a class of chronic and medical conditions characterized by cell growth disorderly and uncontrollably that starts on the inner lining of the tissue of an organ due to the alternation of cell DNA.
At the advanced stage, most cancer cells can travel a distance away from the original site to infect other healthy tissues and organs through blood and fluid circulation.
The causes of cancer are unknown. Epidemiological studies suggested that depending on the type of cancer, the most common risk factors are aging, tobacco, sun exposure, radiation exposure, chemical toxins, viruses and bacteria, change of hormones, family history, excessive alcohol drinking, poor diet, lack of physical activity and overweight or obesity.
However, most people with some of the above risk factors have never developed cancer.
A genetic preposition and acquired genetic mutation are the most risk factors that can precisely predict the percentage of prevalent risk of the onset of the disease.
Mutation of gene TP53 and PTEN. which produces a protein that suppresses the growth of tumors, genetic mutation of BRCA1 and BRCA2 genes is associated with the higher risk of cancer.
In acquiring genetic mutation, some researchers suggested that the result of errors in the cell cycle division caused by long-term exposure to carcinogenic substances such as tobacco smoke and occupations that require chemical exposure that also damages the cells' DNA are also some prevalent factors associated with the increased risk of cancers.
The US National Library of Medicine wrote "For example, certain mutations in the BRCA1 or BRCA2 genes greatly increase a person's risk of developing breast cancer and ovarian cancer. Variations in other genes, such as BARD1 and BRIP1, also increase breast cancer risk, but the contribution of these genetic changes to a person's overall risk appears to be much smaller".
Futhermore, on the other front of acquired genetic mutation, Dr. Michael R. Stratton, the lead author in the study "The cancer genome" said, "The cancer cell may have acquired, from exogenous sources, completely new DNA sequences, notably those of viruses such as human papillomavirus, Epstein Barr virus, hepatitis B virus, human T lymphotropic virus 1 and human herpesvirus 8, each of which is known to contribute to the genesis of one or more type of cancer".
Some researchers proposed that cancer may be the result of weakened immunity, Dr. Louise M.E. Janssen, the lead scientist in the study "The immune system in cancer metastasis: friend or foe?" said, " the primary tumor induces an immune response, which may not be sufficient to destroy the primary tumor, but prevents the growth of a secondary tumor or metastases. Since that time, many different immune cells have been shown to play a role in both inhibiting and promoting metastatic disease".
Dithiolthiones are phytochemicals in the class of Organosulfides, found abundantly in cruciferous vegetables, garden sorrel, horseradish, etc.
In the concerns of chemo drug side effects in the treatment of invasive cancers, researchers in the cancer team at the Shanghai Jiaotong University School of Medicine conducted an experiment to test the association between urinary levels of isothiocyanates (ITCs) and the risk of pancreatic cancer in urban Shanghai.
The case-control study was conducted in urban Shanghai from December 2006 to December 2008) through a newly established "instant case reporting" system.
Intake of cruciferous vegetable intake and ITC consumption, urinary ITC levels (median (P25, P75)) were 95.0 (66.9, 135.8) g/d, 11.0 (7.1, 16.0) µmol/d, 0.95 (0.12, 2.92) µmol/g Cr respectively in treatment cases, all lower than those of controls, separately 107.4 (80.1, 154.1) g/d, 12.3 (8.0, 18.0) µmol/d, 1.78 (0.53, 5.28) µmol/g Cr with a statistically significant differences of 3.75.
The urinary levels of ITCs all cases were found to a correlation with cruciferous vegetable consumption and dietary ITC exposure.
Further analysis suggested that there is an inverse association between urinary ITCs and the risk of pancreatic cancer after adjusting for possible confounding factors such as age, sex, history of diabetes, and pancreatitis.
Dr. the lead author said, " High levels of cruciferous vegetables or ITC consumption were associated with a reduced risk of pancreatic cancer (all P trend <0.05)".
Moreover, in the study to test the combination of Thiazolo and isothiocyanates antiproliferative activity in a panel of 8 cancer cell lines, including lung (NCI-H322 and A549), epidermal (A431), glioblastoma (T98G), pancreatic (MIAPaCa-2), prostate (PC-3), human leukemia (HL-60) and breast (T47D) cells, researchers found that
* The application compound showed the induction of apoptosis in A549 cells at 10 μM, as indicated by the increase in the sub-G1 population.
* The compounds also induced apoptosis in HL-60 cells at 20 μM. The effect of compound 4a on mitochondrial potential loss in HL-60 cells. was also studied.ct
These results suggested that Thiazolo under the influence of isothiocyanates demonstrated potency in the inhibition of all cancer cell lines in a concentration and dose-dependent manner without inducing any side effects.
More importantly, Dr. Figueiredo SM, the lead scientist at the Campus Morro do Cruzeiro in the study "The anti-oxidant properties of isothiocyanates: a review" revealed that ITCs induce Phase II enzymes related to detoxification processes of chemical carcinogens to prevent the start of carcinogenesis and exhibit antitumor activity at post-initiation phase.
Additionally, the lead researcher also suggested that ITCs inhibit the cell viability of human cervical cancer cells, human pancreatic cancer cells, human hepatocellular carcinoma cells, and human ovarian cancer cells, and have anti-inflammatory properties in the treatment of human T-cell leukemia cells probably through improving antioxidant status.
Taken altogether, Phytochemical Isothiocyanates found abundantly in cruciferous vegetables may be considered a bioactive functional ingredient for the prevention and treatment of cancers, depending on the large sample size and multi-center in the human study.

The urinary levels of ITCs all cases were found to a correlation with cruciferous vegetable consumption and dietary ITC exposure.
Further analysis suggested that there is an inverse association between urinary ITCs and the risk of pancreatic cancer after adjusting for possible confounding factors such as age, sex, history of diabetes, and pancreatitis.
Dr. the lead author said, " High levels of cruciferous vegetables or ITC consumption were associated with a reduced risk of pancreatic cancer (all P trend <0.05)".

Among the groups, the GC group received orally the generally prescribed dose of 1,350 mg of GC twice daily and the CSE group received orally10 mg twice daily of CSE.
According to the assays, on days 30, 60, 90, 120, 150, and 180 compared to the baseline (Day 0), the WOMAC score in the CSE group was decreased by 22.44% and 70.29% on days 30 and 180, respectively, compared to a 14.80% and 32.81% respectively decrease in the GC group.
The pain scores also showed similar trends in flavor to the CSE group. There was no adverse effect on the course of the experiment.
In other words, CSE is more effective for the treatment of OA compared to that of GC.
Dr. the lead scientists wrote, "The use of 10 mg CSE, twice daily, was effective in reducing pain related to moderate knee OA and can be potentially used in the management of knee pain, stiffness, and physical functions related to OA".

Additionally, both cooked and steamed broccolis demonstrated significantly enhanced induction of the survival signaling proteins and inhibition of death signaling proteins associated with heart tissue damage.
Furthermore, both cooked and steamed broccoli also protect the heart cells against the overexpression of free radicals by enhancing the levels of antioxidants.
However, steamed broccoli was found to have more cardioprotective abilities compared to those of cooked broccoli.
Moreover, levels of Trx2 and Trx reductase associated with redox activity were only enhanced in the steamed broccoli group.

Based on the analysis, compared to the baseline, at 4 weeks, Black seed and Turmeric alone showed improvement in the parameters associated with the risk of MetS including the body-mass-index (BMI), waist circumference (WC), and body-fat-percent (BF%).
Furthermore, also at week 4, the combination improved all parameters involved in the risk of MetS, except HDL-cholesterol with lower fasting blood glucose (FBG) and LDL-cholesterol, compared to placebo.
Moreover, at 8 weeks, Black seeds reduced lipids and FBG, whereas turmeric reduced LDL-cholesterol and CRP, compared to placebo.
More importantly, the combination group with a 60% dose of the individual herbs showed an improvement in all parameters from baseline, compared to placebo.
In other words, the combination showed complete inhibition of all parameters associated with the onset of metabolic syndrome compared to the selected inhibition of individual herbal medicine. Black seeds + Turmeric showed much better efficacy than the use of Black seed and Turmeric alone.
