#Phytochemical 3,3'-#Diindolylmethane Processes Anti-Malignant Mesothelioma Activity, Scientists Show

The bioactive compound 3,3'-Diindolylmethane or DIM may process strong and positive effects against the risk and progression of mesothelioma, according to studies.
Mesothelioma is a medical condition characterized by irregular cell growth in the thin layer of tissue that covers the internal organs, due to cell DNA alternation.

However, according to epidemiological studies, the most common area affected is the lining of the lungs and chest wall.

Most cases of mesothelioma start in the cell on the surface of the layered tissue. At the later stage, the cells of the larger size of the tumor can cause secondary tumors to develop in nearby organs and distant locations through the circulation of blood and fluids, leading to secondary metastasis.

The most common symptoms of mesothelioma are persistent dry cough or wheezing, shortness of breath, pain in the chest or abdomen, fatigue, and weakness.

In severe cases, patients may also experience symptoms of respiratory complications such as pleural effusion (fluid around the lungs).

Dr. Ombretta Melaiu, the lead scientist in the investigation of the genetic susceptibility in the development of malignant pleural mesothelioma (MPM), wrote, "Among asbestos-exposed workers, several studies reported familial cases of MPM, suggesting that heredity could be important in the tumor development,.... the risk of developing MPM is increased of a limited extent among people exposed to asbestos with a positive history of familial cancers".

And, "In families and in animal models, mutations within BAP1 are strongly predisposing to develop MPM".


The results clearly suggested that if you carry the mutation gene BAP1, you are at a higher risk of MPM and other types of cancer.


3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

Scientists concerned of malignant mesothelioma (MM), a very aggressive, lethal cancer, and its incidence is increasing worldwide examined the dietary phytochemicals that may process the potential for the treatment of MM with no side effects.

The study included several bioactive ingredients such as curcumin, epigallocatechin gallate, quercetin, genistein, pterostilbene, resveratrol, capsaicin, ellagic acid, benzyl isothiocyanate, phenethyl isothiocyanate, sulforaphane, indole-3-carbinol, 3,3'-diindolylmethane (DIM), diallyl disulfide, betulinic acid, and oleanolic acid.

DIM was found to process the anti-MM activity by boosting the function of microRNA in the regulation of the activities of the genes.


MicroRNAs (miRNAs) are a class of small noncoding RNAs of ∼22nt in length involved in the regulation of gene expression against the incidence of cancer. Increased expression of microRNAs is associated with numerous types of cancer.

Overexpression of miR-132 in vivo was associated with the severity of cancer patients.

Injection of DIM exerted anti mesothelioma potentials, through the expression of microRNA regulatory activities.was attributed to the suppression of macrophage infiltration and pro-inflammatory cytokines.

In other words, DIM modulated the immune response against the onset and progression of mesothelioma caused by the production of oxidative stress due to the overexpression of inflammatory cytokines in the initiation of the disease.

Dr. Sayeed MA the lead scientist wrote in the final report, "In addition to microRNA regulatory activities, curcumin, epigallocatechin gallate, quercetin, genistein, resveratrol, phenethyl isothiocyanate, and sulforaphane have anti-mesothelioma potentials, and pterostilbene, capsaicin, ellagic acid, benzyl isothiocyanate, indole-3-carbinol, 3,3'-diindolylmethane, diallyl disulfide, betulinic acid, and oleanolic acid have potentials to inhibit cancer by regulating the expression of various genes which are also known to be aberrant in MM".

Taken together, DIM may be considered a supplement for the prevention and combined with primary therapy for the treatment of MM, pending the confirmation of a larger sample size and multicenter human study.

#Broccoli Improves Post-Ischemic Ventricular Function and Reduces Myocardial Infarction and Cardiomyocyte Apoptosis, Researchers Say

Acute brain infarction is a severe medical emergency characterized by the obstruction of the blood supply to the brain, causing the local death of the neurons in the brain. Prompt treatment can minimize brain damage and potential complications.

The occurrence and extent of brain infarction are associated with three basic factors, including the site of arterial occlusion, the rapidity of arterial occlusion, and the presence or absence of collateral circulation.

In other words, brain infarction can be classified into myocardial infarction due to plaque rupture and myocardial oxygen supply-demand imbalance secondary to other acute illnesses.

The exact causes of a blood clot that induces acute brain infarction are unknown. The carbohydrate pattern showed a weak relationship with AMI. We observed a U-shaped association between frequencies of fat and protein pattern and AMI risk. Excessive fat intake increased the AMI risk.".

With an aim to find a natural food for the prevention and treatment of heart disease, researchers examined the effect of sulforaphane found in broccoli against myocardium from ischaemic reperfusion injury by comparing fresh broccoli and cooked broccoli cardioprotection.

Broccoli is a mustard/cabbage plant, belonging to the family Brassicaceae. The veggie has large flower heads, usually green in color and the mass of flower heads surrounded by leaves and evolved from a wild cabbage plant on the continent of Europe.

The study compared the two groups of rats that were fed with fresh (steamed) broccoli or cooked broccoli for 30 d, while a third group was given a vehicle only for the same period of time.

Based on the differentiation of isolated working hearts that were subjected to 30 min ischemia followed by 2 h of reperfusion. 30 min ischemia followed by 2 h of reperfusion, both cooked and steamed broccolis displayed significantly improved post-ischemic ventricular function and reduced myocardial infarction and cardiomyocyte apoptosis compared to control.

Additionally, both cooked and steamed broccolis demonstrated significantly enhanced induction of the survival signaling proteins and inhibition of death signaling proteins associated with heart tissue damage.

Furthermore, both cooked and steamed broccoli also protect the heart cells against the overexpression of free radicals by enhancing the levels of antioxidants.

However, steamed broccoli was found to have more cardioprotective abilities compared to those of cooked broccoli.

Moreover, levels of Trx2 and Trx reductase associated with redox activity were only enhanced in the steamed broccoli group.

Based on the findings, researchers said, " The results of the present study documented superior cardioprotective properties of the steamed broccoli overcooked broccoli because of the ability of fresh broccoli to perform redox signaling of Trx".

Taken together, broccoli processed abundantly SFN may be considered supplements as a cardioprotective remedy, pending the confirmation of the larger sample size and multicenter human study.
Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Metabolic syndrome is a cluster of conditions that increase the risk of heart disease, stroke, and diabetes, including too much fat around the waist, high blood pressure, triglycerides, and blood sugar, and low HDL cholesterol.
People who have 3 or more aforementioned conditions are at a higher risk of heart disease, stroke, and diabetes. However, most people with metabolic syndrome have never developed cardiovascular disease (CVD) or diabetes.

Most common symptoms of metabolic symptoms are depending totally on the clustered conditions, including thirst and urination, fatigue, and blurred vision due to high blood glucose. However, in patients with no condition of hyperglycemia, metabolic syndrome is asymptomatic.

Based on the analysis, compared to the baseline, at 4 weeks, Black seed and Turmeric alone showed improvement in the parameters associated with the risk of MetS including the body-mass-index (BMI), waist circumference (WC), and body-fat-percent (BF%).


Furthermore, also at week 4, the combination improved all parameters involved in the risk of MetS, except HDL-cholesterol with lower fasting blood glucose (FBG) and LDL-cholesterol, compared to placebo.

Moreover, at 8 weeks, Black seeds reduced lipids and FBG, whereas turmeric reduced LDL-cholesterol and CRP, compared to placebo.

More importantly, the combination group with a 60% dose of the individual herbs showed an improvement in all parameters from baseline, compared to placebo.

In other words, the combination showed complete inhibition of all parameters associated with the onset of metabolic syndrome compared to the selected inhibition of individual herbal medicine. Black seeds + Turmeric showed much better efficacy than the use of Black seed and Turmeric alone.


Based on the results, researchers said, " The combination of Black seeds and Turmeric can, therefore, be recommended with lifestyle modification as a starting point for patients with MetS to halt its future complications and progression".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be used alone or combination with Black seed for the prevention and treatment of metabolic syndrome, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Scientists may have found a bioactive ingredient that can normalize the blood pressure without inducing any side effects, according to studies.

Allicin is phytochemical containing sulfur in the class of organosulfur compound, found abundantly in onion and garlic.

Nine patients with rather severe hypertension (diastolic blood pressure > or = 115 mm Hg) were included in the study.

Application of the preparation reduced the sitting blood pressure by 7/16 (+/- 3/2 SD) mm Hg at peak effect approximately 5 hours after the dose, with a significant decrease in diastolic blood pressure (p < 0.05) from 5-14 hours after the dose.
No significant side effects were reported. The findings strongly indicate that this garlic preparation can reduce blood pressure.

Futhermore, in order to compare the antihypertensive effect of the different fractions of garlic, researchers investigated the hypotensive action of time-released garlic powder tablets (Allicor) vs regular garlic pills (Kwai).

84 men with mild or moderate arterial hypertension have participated in the study.

After an 8-week placebo treatment run-in phase, patients were randomized either to 600 mg Allicor (n=30) or to placebo (n=20) daily for 8 weeks. In addition, in the open-label branch, patients received either 2400 mg Allicor daily (n=18) or 900 mg Kwai daily (n=16).

Allicor treatment (600 mg daily) resulted in a reduction of both systolic and diastolic blood pressures by 7.0 mm Hg (95% confidence interval (95% CI): 5.3-8.7) and 3.8 mm Hg (95% CI: 2.7-4.8), respectively. Increasing the Allicor dosage to 2400 mg daily did not provide any additional benefit.
Treatment with Kwai resulted in the same decrease in systolic blood pressure (5.4 mm Hg, 95% CI: 1.9-8.8) as that seen with Allicor, but no decrease in diastolic blood pressure.

In the depth analysis, researchers found that different effects of Allicor and Kwai on diastolic blood pressure may be associated with the prolonged action of Allicor.

Dr. Sobenin IA, wrote in the final report, "The results of this study show that time-released garlic powder tablets are more effective for the treatment of mild and moderate arterial hypertension than are regular garlic supplements".

Taken together,. allicin, a bioactive compound of garlic may be used for the prevention and treatment of high blood pressure with no side effects

Green tea may have a potential effect on the regulation of Lewy bodies against the onset of Lewy body disease, some scientists suggested.

Lewy body disease is one of the common forms of neurodegenerative diseases occurring in the elderly characterized by mutation of the Lewy bodies as a result of the abnormal structure of Lewy bodies, building up in areas of the brain.

Lewy bodies, mainly composed of α-synuclein (αS), are pathological hallmarks of Parkinson's disease and dementia with Lewy bodies.

People with building Lewy bodies in most cases also have Alzheimer's disease, causing reduced mental ability, including symptoms of visual hallucinations, and changes in alertness and attention.

Some patients with the condition are also experiencing symptoms of rigid muscles, slow movement, and tremors, the hallmarks of Parkinson's disease.

Researchers do not know why the elderly with the same health conditions and family history, are susceptible to the early onset of the disease while others do not. However, they do know that certain risk factors such as old age, family history, and male gender are associated with the onset of Lewy body disease.

Some researchers also believe due to the severity of brain damage, patients with the condition also express certain mental instability including aggressive behavior, depression, and worsening of Parkinsonian-like symptoms.

A genetic alteration such as SNCA or SNCB gene mutations is also found in patients with the disease.

In the study of N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)- and 6-hydroxydopamine-induced neurodegeneration in rodents and nonhuman primates with the increased presence of iron and alpha-synuclein in the SNpc, researchers found that application of green tea catechin polyphenol (-)-epigallocatechin-3-gallate exerts a significant effect as an iron chelator in protecting the brain against neurotoxins by inhibiting the increased presence of iron and alpha-synuclein in the SNpc.

Pretreatment of green tea on N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)- and 6-hydroxydopamine-induced neurodegeneration in rodents was also found to demonstrate high levels of antioxidant activity in preventing the accumulation of iron and alpha-synuclein in SNpc.

Dr. Mandel S, the lead author suggested that a combination of iron chelation and antioxidant therapy might be a significant approach to neuroprotection in PD and other neurodegenerative diseases, including Lewy body disease.

Furthermore, in the concerns of the primary α-synucleinopathgenic expression in patients with Parkinson's disease (PD), dementia with Lewy bodies, and multiple system atrophy, researchers at Lanzhou University, launched an investigation to examine whether green tea (-)-Epigallocatechin-3-gallate (EGCG) can inhibit the fibrillation of αS.

After taking into account co and confounders, researchers showed that
* EGCG displays a potential effect in remodeling the αS fibrils and breaking the initially ordered pattern by reducing the β-sheet content.

Inflammation is a process of the immune system that responds to harmful pathogens.
Inflammation can be acute or chronic. In the acute phase of infection, the immune system after receiving information from the neurons, macrophages, the largest white blood cells in the first line of defense stimulate the production of blood palette to cover the wound and the production of inflammatory cytokines to kill off micro-organisms, leading to symptoms of fever, redness, and swelling.

However, in some cases, the overproduction of proinflammatory cytokines may damage nearby healthy cells and tissues, causing the formation of scars.

In severe cases, a protein with duo functions in the production of pro and anti-inflammatory cytokines which produce anti-inflammatory cytokines under normal conditions also switch to produce pro-inflammatory cytokines.

Most cases of acute inflammation are resolved within a few days without causing damage to the wound tissues. However, if the immune first line of defense can not completely destroy the invaders within a period of 3 to 8 weeks, it will compromise, leading to low-grade chronic inflammation.


Diseases associated with low-grade chronic inflammation include diabetes, cardiovascular disease, and cancer.

Dr. Duprez DA, the lead scientist at the University of Minnesota.in the investigation of inflammatory makers and chronic inflammation wrote, "Numerous studies have shown that high-sensitivity C-reactive protein (hsCRP), one of the most frequently studied inflammatory markers, is associated with the chronic inflammatory-related disease, cardiovascular disease, cancer and mortality [1]. While hs-CRP is a single marker of inflammation".


In other words, if you have a chronic inflammatory disease, your levels of hsCRP are much higher compared to people who do not.

Conventionally, treatment of acute phase inflammation is to manage inflammation and prevent the risk of long-term damage, including anti-inflammatory nonsteroidal anti-inflammatory drugs (NSAIDs) which can be bought without a prescription.


Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of diseases associated with inflammation, researchers examined the effect of the effects of freeze-dried cranberry powder on anti-inflammation and lipid profiles.

The study included a total of 40 Sprague-Dawley male rats (6 weeks old) were equally divided into the following five groups: 1) normal diet group + saline (NC); 2) atherogenic diet + saline (HFC); 3) atherogenic diet + LPS (HL); 4) atherogenic diet with 5% cranberry power + LPS (C5); 5) atherogenic diet with 10% cranberry power + LPS (C10).

Based on the analysis after LPS (0.5 mg/kg) was injected into the abdominal cavities of rats 18 hours prior to sacrifice, researchers found that* C5 rats treated with an atherogenic diet with 5% cranberry power + LPS showed a mean serum high-density lipoprotein (HDL)-cholesterol level in C5 rats was significantly higher compared to those in NC and HL rats.

* The mean serum levels of CRP and IL-1β associated with pro-inflammatory markers and expression were significantly lower (P < 0.05) in the cranberry powder groups compared to HL rats.


* Furthermore, levels of anti-inflammatory cytokines expressed by the serum IL-10 and NO showed 29% and 88% higher mean values in the C5 group and 49% and 24% higher in the C10 group than those in the HL group, respectively.


In other words, cranberry powder protects the Sprague-Dawley male rats induced inflammation by elevating the levels of anti-inflammatory and lowering the levels of pro-inflammatory cytokines.

Collectively, researchers wrote, " freeze-dried cranberry powder may have beneficial effects on cardiovascular diseases by modifying serum lipids and the early inflammatory response."

Taken together, cranberry may be considered an anti-inflammatory remedy, pending the confirmation of the larger sample size and multicenter human study.
