#Luteolin and Caffeoylquinic acid derivatives and #Quercetin Diclycosides Found in Herbal #Dandelion Inhibits Dengue Virus Serotype 2 Replication, According to Studies

Kyle J. Norton

Infection is a condition that causes the invasion of stimuli and micropathogens, including viral infection.
Similar to bacterial infection, both infections share common characteristics by spreading through coughing and sneezing and contact with infected people, especially through kissing and sex.

However, unlike bacteria, a virus replicates only inside the living cells of an organism. Viruses can infect all types of life forms, including humans and microorganisms, such as bacteria and archaea.

Anti-viral effects are the process that which certain substances are used to cause the destruction of a virus.

In the acute phase of viral infection, depending on the types of viruses, the viruses after entering the body start to attack the specific host cells. The cold and flu virus will attack the cells in the respiratory system and the human immunodeficiency virus (HIV), which causes AIDS, will attack the white blood cells of the immune system,

The viral infection has been known to carry over some different stage, after successfully attaching to the host cell, the virus to release its mRNA into the host cell, leading to the recruitment of the host cell's enzymes to accelerate the viral multiplying, then breaking free from the host cell to start the cycle all over again.

The white blood cells in the systematic immune response in the acute phase of infection stimulate the production of the proinflammatory cytokines to the site of infection with an aim to protect our body by killing off the invaders.

The battle between the immune white blood cell and viral invaders in most cases is in the flavor of the side of the immune system, the invaders are killed and the infection is healed.

However, if the immune system can not kill off the invaders within 3 - 8 weeks, it then will adapt to the change, leading to chronic inflammatory and infectious diseases.

Dengue virus (DENV) belongs to the Flavivirus genus and is transmitted by Aedes aegypti and Ae. Albopictus mosquitoes, found in tropical and subtropical regions of the world. In severe cases, dengue virus serotype 2 has been found to induce symptoms of severe plasma leakage, severe hemorrhage, and/or severe organ impairment.

Dandelion is a herbaceous perennial plant, genus Taraxacum, belonging to the family Asteraceae, native to temperate regions of the world. It has been used in traditional and herbal medicine to treat gout, eczema, acne, gall bladder, kidney, liver, and urinary disorders, hypoglycemia, dyspepsia with constipation, edema, blood pressure, and heart weakness, chronic joint, skin diseases, etc.

On finding a natural ingredient for the prevention and treatment of infectious diseases, scientists examined the cytotoxic concentration and the inhibitory effect of extracts or fractions from Urtica dioica, Taraxacum officinale, Calea integrifolia and Caesalpinia pulcherrima on the dengue virus serotype 2 (DENV2 replication on the BHK-21 cell line.
According to the tested differentiation,
* The methanolic extracts at 60 °C of T. officinale and U. dioica showed higher inhibitory effects on DENV2 replication.

* The chemical analysis suggested that the higher activity fraction is found for U. dioica and T. officinale, at the IC50 values of 165.7 ± 3.85 and 126.1 ± 2.80 μg/ml, respectively.

The compounds which exhibited the efficacy of T. officinale were luteolin and caffeoylquinic acid derivatives and quercetin diclycosides compared to the compounds in the active fraction chlorogenic acid, quercetin derivatives and flavonol glycosides (quercetin and kaempferol) in the U. dioica.

Based on the results, researchers suggest, "Two fractions from U. dioica and T. officinale methanolic extracts with anti-dengue activity were found. The compounds present in both fractions were identified, and several recognized molecules have demonstrated activity against other viral species".

Dandelion may be considered an antioxidant remedy for the treatment of dengue virus serotype 2, pending confirmation of the larger sample size and multicenter human study.

#Onionin A (ONA) Found in Onion Promotes Healthy Gut Microbiota and Intestinal Histomorphology, Researchers Suggest

Kyle J. Norton

Gut microbiota is a community of microbes living in the gastrointestinal tract, including at least 1000 different species of known bacteria with more than 3 million genes.
In a healthy individual, the ratio of good and bad microbes in the gut is balanced. However, people who have a psychological condition such as anxiety, and depression, and are under constant stress and long-term use of antibiotics, particularly your diet may have a negative impact on the ratio.

More precisely, under normal conditions, gut microbiota plays a critical role in overall health,
* Gut microbiota improves the absorption of dietary nutrients in the digestive system

* Gut microbiota protects the body against xenobiotics and drug toxicity by facilitating the excretion.

* Gut microbiota also maintains the structural integrity of the gut mucosal barrier by enhancing the epithelial cells that protect the intestine against invasive pathogens and ensure the function of intestinal in nutrient adsorption.

* Gut microbiota interacts with the immune system against inflammatory immune disorders and inhibits the risk of gastrointestinal diseases and non-gastrointestinal diseases caused by overexpression of bad bacteria.

However, imbalanced the ratio of gut microbiota may have a negative impact on human health and diseases.
Epidemiological studies suggested imbalanced gut microbiota dysbiosis found in overweight and obese has been found to promote diet-induced obesity and metabolic complications.

Dr. Karina Al-Ass wrote, "Obesity is a worldwide epidemiologic syndrome characterized by fat mass accumulation, mainly visceral fat. Current thinking considers the potential role of gut microbiota on the development of obesity and its related comorbidities"

And " Gut microbiota can influence energy extraction from food, lipid metabolism, immune response, and endocrine functions and its profile has shown to differ between obese and lean subjects".
In a recent joint study led by the Tianjin Medical University, General Hospital, an onion supplement may be potent to improve gut microflora and intestinal histomorphology on an animal model.

The onion is a plant in the genus Allium, belonging to the family Alliaceae, a closely related of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

The study included a total of 300 12-day-old broiler chicks (Ross) that were randomly divided into four groups as A served as the control without any supplementation, and B, C, and D received supplemented with onion powder at the rate of 1.5, 2 and 2.5g/kg of feed respectively.

Based on the tested results,* Supplementation of onion exerted a significant (P<0.05) increased weight gain and feed consumption compared to the control.

* Furthermore, supplementation of onion significantly inhibited the population of E. coli and increased significantly Lactobacillus and Streptococcus species.

In other words, onion supplement promotes colon health by improving the ratio of gut microbiota.

* More importantly, injection of onion supplement also significantly increases the morphometry of length, width, crypt depth, and surface area of the villus in the duodenum, jejunum, and ileum of the small intestine associated with colon health optimization.

Based on the findings, researchers said, "Onion supplementation in broiler produced a positive effect on performance, gut microflora, and intestinal histomorphology".

Taken altogether, onion processed with a higher amount of onionin A (ONA) may be considered a remedy for the promotion of colon health, pending the confirmation of the larger sample size and multicenter human study.

#Healthyfood #Cucumber Fruit #Extract Processes a Strong Free Radical Scavenging Activity, Researchers Suggest

Kyle J. Norton

Free radical scavengers are antioxidants that inhibit free radicals before they can induce a chain of domino reactions that have been found to cause protein, lipid, and cell damage, leading to cell injury and cell death.
Most common free radical scavengers can be either produced by the host or from dietary sources, including catalases, glutathione peroxidase, superoxide dismutase (SOD), and vitamin A, C, and E.

Oxidative stress is the imbalance of the ratio of antioxidants and free radicals. In other words, overexpression of free radicals and low levels of antioxidants in the body can lead to oxidative stress, a major cause of chronic diseases such as cardiovascular disease, diabetes, and cancer.

In a healthy individual, dietary, antioxidant, and antioxidant enzymes produced by the liver are more than enough to counter free radicals produced from all sources.

However, for people with a weakened immune system, intake of antioxidant supplements may be necessary, and in chronic illness patients, antioxidant supplementation at an early stage of illness has been found to improve therapies.

Due to the complexity of treating chronic and critical illness patients, some researchers suggested that
professionals need to understand the mechanism of damage caused by free radicals and free radical scavengers protective activity for the treatment of varied pathological conditions.

The cucumber plant is a species of Cucumis Sativus, belonging to the family Cucurbitaceae and native to Western Asia. It is a creeping vine with roots in the ground and grows with the support of frames.

With an aim to find a natural compound that processes anti-free radical overexpression, researchers investigated the aqueous fruit extract of Cucumis sativus L. anti-free radical scavenging activities.

In vitro, aqueous fruit extract at 250 and 500 μg/ml showed a significant effect in the inhibition of free radicals through scavenging activity.

Compared to other chemical compounds, The C. sativus fruit extract showed maximum antioxidant and at 500 μg/ml and 500 mg/kg,

The efficacy of the extracts was attributed to the presence of flavonoids and tannins evidenced by preliminary phytochemical screening.

Additionally, in the comparison of carrot, cucumber, and broccoli seed flours extracted with 50% acetone for their free radical scavenging properties researcher found that
* Cucumber showed a strong free radical scavenging property against DPPH radical scavenging capacity, oxygen radical absorbing capacity, and hydroxyl radical (HO•) scavenging capacity secondary to the broccoli seed flour extract.

* in ABTS•+ scavenging capacity of 250 μmol TE/g, carrot showed a better effect compared to cucumber.

Based on the findings, Dr. Choe U, the lead scientist wrote in the final report, "The results might be used to promote the value-added utilization of these vegetable seed flours in improving human health".

Taken together, cucumber may be used for the treatment of disease associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

#Herbal #Ashwagandha and Its Phytochemical Withanolide A Inhibit the Parameters Associated with Management of #Epilepsy, Researchers Say

Kyle J. Norton

Epilepsy is a neurological disorder associated with sudden recurrent episodes of sensory disturbance, loss of consciousness, or convulsions, caused by abnormal electrical activity in the brain.
Approximately, 1.2 percent of U.S. people have active epilepsy. More than 65 million people have epilepsy globally.

Additionally, about 1 in 26 people will develop epilepsy at some point during their lifetime.

The most common causes of epilepsy are low oxygen during birth, head injuries that occur during birth or from accidents during youth or adulthood, brain tumors, genetic mutation, and infections such as meningitis or encephalitis.

The most common symptoms of epilepsy are temporary confusion, uncontrollable jerking movements of the arms and legs, loss of consciousness or awareness, and psychic symptoms such as fear, anxiety, or deja vu.

Experimentally, melatonin, is a hormone that regulates the sleep-wake cycle. has been found to attenuate seizures, and it is most effective in the treatment of juvenile intractable epilepsy.

Melatonin also processed additional benefits include improved physical, emotional, cognitive, and social functions.

On the other hand, melatonin has been shown to induce electroencephalographic abnormalities in patients with temporal lobe epilepsy and increase seizure activity in neurologically disabled children.

The hormone showed very low toxicity in clinical practice, according to the study by the Department of Pathophysiology, Medical University, PL 20-090 Lublin, Jaczewskiego 8, Poland(22).

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine for over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered an anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

In the urgency to discover a natural ingredient for the management of the neurological disorder, researchers investigated the effect of Withania somnifera (WS) root extract and withanolide A (WA) in the management of temporal lobe epilepsy.

According to the tested analysis, in the epileptic test group of rats, levels of antioxidants are found to substantially decrease compared to the increase of the levels of peroxide.

Oxidative stress caused by an imbalance of the ratio of antioxidants and free radicals in the tested rats, induced significant decrease levels of proteins associated with systemic inflammation, compared to the increase of pro-apoptotic proteins.

Moreover, the epileptic condition of the tested rats also showed to increase the muscarinic receptor binding, which leads to reduced function of transduction of cholinergic signals and increases mRNA expression involved of the inhibition of the cell growth in the cerebral cortex.

However, the administration of the WS reversed the altered muscarinic receptor expression, oxidative stress, and resultant derailment in cell signaling in experimental rats.

Based on the findings, researchers said, "Withania somnifera and withanolide A play important roles in central muscarinic receptor functional balance and activation of the antioxidant system in the cerebral cortex in temporal lobe epilepsy".

Taken together, Ashwagandha may be considered supplements for the management of epilepsy, pending to the confirmation of the larger sample size and multicenter human study.

#Ginger's 6-Shogaol (6-SHO) and Extract Reduce Survival and Induce Apoptosis of #ProstateCancer Cells, Scientists Reveal

Kyle J. Norton

Scientists may have found a pungent spice that processes the potential for the prevention and treatment of prostate cancer, some studies suggested.
Prostate cancer is a chronic condition caused by the cell growth irregularly and disorderly in the prostate tissue due to the alternation of DNA.

Prostate cancer is a slow-growth of cancer and can be detected during an annual rectum exam.

At the early stage, prostate cancer cell which has grown on the surface of the inner lining of the prostate does not cause any symptoms due to the very small size of the tumor.

However, at the later stage, the large tumor size may suppress the nearby blood vessels and nerve cells, leading to internal bleeding and severe pain. Also, cancer cells may also travel a distance away from the stomach to infect other healthy tissue and organs, leading to a secondary metastasis.

Common sites metastases of prostate cancer are bone, lymph nodes, lung, and liver.

Researchers do not know the exact causes of prostate cancer. However, they do know the increase in age, family and personal history, ethnicity, genetic preposition, a poor diet, and being overweight or obese are some of the prevalent risk factors of prostate cancer.

According to the Canadian Cancer Society, in 2017, 21,300 men were diagnosed with prostate cancer, represented 21% of all new cancer cases and 4,100 men died from prostate cancer, and 10% of all cancer deaths.

The 5 years survival rate of localized prostate cancer is 100% and 98% of survivors are alive after 10 years compared to the advanced stage of only 31%.

Some researchers concerned the widespread prostate cancer in North America, northwestern Europe, Australia, and on Caribbean islands compared to Asia, Africa, Central America, and South America, said, "One of the most striking characteristics of prostate cancer is the degree of geographic variation in its patterns of occurrence and progression; this variation is apparent at local, national, and international levels".

And "the known factors influencing the biology and epidemiology of prostate cancer and some of the ways in which findings to date have benefited from geography".

The results strongly indicated there must be something happened in the Western world that does not found in the East and Africa, Central America, and South America.

Some researchers suggested it may be the Western diet.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine to treat dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

In the concern of prostate cancer risk in men in the Western world and its related mortality and morbidity, researchers in the cancer team at the The University of Texas at Austin examined the anti-prostate cancer effect of 6-Shogaol (6-SHO), a potent bioactive compound in ginger (Zingiber officinale Roscoe).

Injection of the bioactive compound effectively reduced survival and induced apoptosis of cultured human (LNCaP, DU145, and PC3) and mouse (HMVP2) prostate cancer cells.

6-SHO reduced the expression of pro-inflammatory cytokines in the activation of the proteins involved in cell proliferation and the capacity for metastasis.

Furthermore, 6-SHO also inhibited the expression of cytokines in the stimulation of proteins and genes associated with anti-apoptotic activity and tumor cell survival.

6-SHO was more effective than two other compounds found in ginger, 6-gingerol, and 6-paradol at reducing survival of prostate cancer cells.

Moreover, 6-SHO also showed significant tumor growth inhibitory activity in an allograft model in HMVP2 cells.

In other words, 6-SHO inhibited prostate cancer through several mechanisms, including chemokine, cytokine, cell cycle, apoptosis, and genes.

Dr. Saha A and colleagues wrote in the final report, "6-SHO may have potential as a chemopreventive and/or therapeutic agent for prostate cancer".

Additionally, in order to reveal more information about ginger as a promising chemopreventive agent, researchers examined ginger extract (GE) growth-inhibitory and death-inductive effects in a spectrum of prostate cancer cells.

Injection of the extract showed a significant effect on perturbed cell-cycle progression, impaired reproductive capacity, modulated cell-cycle, and apoptosis in human prostate cancer cells.

Daily oral feeding of 100 mg/kg body weight of GE inhibited growth and progression of PC-3 xenografts by approximately 56 % in nude mice, observed by the tumor tissue from GE-treated mice compared to controls.

Most importantly, GE is safe as toxicity is not detected in normal, rapidly dividing tissues such as gut and bone marrow.

Taken together, ginger-processed 6-Shogaol (6-SHO) may be considered a functioning remedy for the prevention and combined with the primary medicine for the treatment of prostate cancer with no side effects, pending the confirmation of a larger sample size and multicenter human study.

#Green Tea #Polyphenols (GrTPs) Inhibit #Oxidativestress Associated with Impair Fertilization Capacity in Females, Scientists Find

Kyle J. Norton

Scientists may have found a beverage bioactive compound in improves the chance of fertility in females without inducing any side effects, some studies found,
Infertility is a condition characterized by the inability to get pregnant after 12 months of frequent and unprotected sex of a couple. Women who can carry the term of pregnancy are also considered infertile.

According to the statistics, approximately 8 to 12 percent of couples worldwide experience fertility problems. Between 45 and 50 percent of cases of infertility are attributed to male reproductive problems.

The most common causes of infertility in female in the US are aging, medical condition of endometriosis, polycystic ovarian syndrome (PCOS) which have a strong interruption of the female menstrual cycle, sexually transmitted infections (STIs) and obesity.

Other prevalent risk factors including physical and medical conditions, such as blocked fallopian tubes due to pelvic inflammatory disease, physical problems with the uterus diabetes, long-term intakes of a certain medicine, such as anti-inflammatory drugs and hormone imbalances that affect the menstrual cycle

Genetic preposition such as The HOXA13 gene. Only one single gene disorder has been shown to affect uterine development in infertile females.

Age, smoking, use of illicit drugs, excessive alcohol drinking, and long-term exposure to chemicals are the common causes of female infertility.

In terms of diet, researchers at the Harvard T.H. Chan School of Public Health in the study "Diet and female fertility: doctor, what should I eat?" wrote, "According to existing data, women trying to achieve pregnancy are encouraged to increase consumption of whole grains, omega-3 fatty acids, fish, and soy and to reduce consumption of trans fats and red meat. In addition, a daily multivitamin that contains folic acid before and during pregnancy may not only prevent birth defects but also improve the chance of achieving and maintaining a pregnancy".

These results suggested that women who follow a diet high in saturated and trans fat, intake of a large amount of red meat and processed food, and less in fruits, nuts, and vegetables are at a higher risk of infertility or of carrying the pregnancy to full term.

In the concerns of oxygen species (ROS), which are produced during cellular oxidation may reduce the chance of conception, researchers at the joint study led by the Uniwersytet Medyczny w Łodzi. launched an investigation to examine whether increased production of reactive oxygen species is an important factor in the etiopathogenesis of pregnancy and affects female reproduction.

Before the application of green tea s (GrTPs), researchers found that oxidative stress caused by overexpression of free radicals may damage the oocytes and may impair their fertilization capacity.

The increased ROS levels probably are the results of antioxidant enzymes produced by the tested subjects being suppressed due to some unknown mechanisms.

Oxidative stress may also lead to embryo fragmentation and formation of numerous developmental abnormalities and is regarded to be one of the important reasons for spontaneous and recurrent miscarriage
Oral administered green tea polyphenols (GrTPs) inhibit the overexpression of ROS which is shown to restore the embryo fragmentation and formation of numerous developmental abnormalities and increase the quality of female gametes, mostly via the capability of catechins to reduce ROS production.

Further analysis also suggested that GrTPs processed antioxidant properties that only restore the health of oocytes caused by overexpression of ROS but also improve age-related decline in female fertility rate and gamete quality.

These unique properties of green tea catechins in improving female reproductive health suggested green tea polyphenols (GrTPs) may be considered a functional food in protecting against damage to the oocytes and their fertilization capacity and the integrity of the embryo.

Dr, Rahman SU, the lead scientist in the final report, said, "the therapeutic effects of GrTPs against infertility, their possible mechanisms of action, and recommended supportive therapy for improving fertility in humans and in animals".

Additionally, in the concerns of the Reactive oxygen species (ROS) that may increase the risk of female infertility, researchers launched an investigation to examine whether green tea catechins protect the female gametes over the expression of ROS.

After taking into account levels of oxidative stress before and after application of green tea catechin, researchers wrote,
* Epigallocatechin-3-gallate (EGCG), a major bioactive compound in green tea inhibited the expression of ROS in facilitating the damage of female gametes through to its strong antioxidant activity.

* Green tea catechins potentially improve the oocyte maturation and may inhibit in-vitro maturation of the oocyte

Dr. Roychoudhury S, the lead author said, " (These results further confirmed) the potential roles of green tea catechins on oxidative stress in male and female reproduction and fertility".

Taken together, Green tea polyphenols (GrTPs) may be considered an adjunct therapy in the protection of the female gametes and oocyte maturation against the onset of infertility.

#Herbal #Turmeric Inhibits #OralMucositis in Cancer Patients Undergoing Chemo and/or Radiotherapy, Researchers Say

Kyle J. Norton

Oral mucositis is considered a side effect in patients who undergo cancer treatment, particularly chemotherapy and radiation.
In cancer, oral mucositis is caused by the reduced function of the immune system to protect against the infection of open sores in the mucosa.

However, poor oral or dental health, smoking or chewing tobacco and drinking alcohol, female gender, dehydration, and patients who had previous cancer treatment, have kidney disease are some of the most common risk factors that cause the onset of oral mucositis.

The most common symptoms of oral mucositis are red, shiny, or swollen mouth and gums, sores in the mouth or on the gums or tongue, pain in the mouth or throat, mild burning, or pain when eating food.

Some patients may also experience symptoms of difficulty swallowing or talking, soft, whitish patches or pus in the mouth or on the tongue, particularly the increase of mucus or thicker saliva in the mouth.

Some researchers suggested the onset of oral mucositis in most cases is associated with people with a weakened immune system, including the use of chemotherapy.

Dr. Francesco Chiappelli wrote, "Mucositis is observed in a variety of immunosuppressed patients, but is most often consequential to cancer therapy. It appears as early as the third day of the intervention, and is usually established by Day 7 of treatment".

And, "The precise immune components involved in the etiology of mucositis are unclear, but evidence-based research (EBR) data has shown that applications of granulocyte–macrophage-colony stimulating factor prevent the onset of the exacerbation of oropharyngeal mucositis".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

With an aim to find a potential compound for the treatment of oral mucositis(OM), researchers evaluated the effects of turmeric and curcumin in the management of oral mucositis in cancer patients undergoing chemo and/or radiotherapy by searching the database of Cochrane Library, LILACS, LIVIVO, PubMed, Scopus, and Web of Science.

After a two-step selection process, four randomized and one nonrandomized clinical trials were included in the analysis in which 2 studies suggested that topical application of turmeric/curcumin as a gel or mouthwash showed a reduced grade of mucositis, pain, erythema intensity, and ulcerative area.

In other words, the topical application of turmeric or curcumin may be effective in controlling signs and symptoms of oral mucositis in tested patients.

Furthermore, according to the study of 72 golden Syrian hamsters induced mucositis (OM) by 5-fluorouracil (5-FU) for 4 days randomly allocated into four groups: control, placebo, chamomilla, and a mucoadhesive formulation containing curcuminoid (MFC)
* On day 5, all groups exhibited OM

* On day 8, both MFC and chamomilla groups exhibited better wound healing compared to other groups.

* Moreover, also on day 8, the MFC group demonstrated inhibited protein associated with stimulating new blood vessels for the OM to grow and proinflammatory cytokines to facilitate symptoms of OM, compared to placebo and control groups.

Collectively, Dr. Schmidt TR, the lead scientist wrote in the final report, "These findings suggest that MFC has a therapeutic effect on OM, accelerating wound healing through re-epithelization and anti-inflammatory action as modulation of angiogenesis and TGF-β1 expression".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of OM, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
