In the urgency to find a potential compound for the treatment of leukemia, researchers examined the antiproliferative effects of betanin, a principle betacyanin pigment, isolated from the fruits of Opuntia ficus-indica on chronic myeloid leukemia.

In the human chronic myeloid leukemia cell line (K562), the application of betanin decreased the proliferation of K562 cells at a dose and time-dependent with an IC(50) of 40 microM.

According to the tested analysis, the antiproliferative activity of betanin was associated with chromatin condensation, cell shrinkage, and membrane blebbing.


In other words, cells treated with betanin showed a fragmentation pattern typical for apoptotic cells.
Particularly, at 40 microM betanin, the phytochemical compound induced 28.4% cell cycle arrest in the sub-G0/G1 phase.

Moreover, treatment with betanin not only activated the proteins such as PARP cleavage associated with pro-apoptosis but also inhibited the proteins such as Bcl-2 involved in anti-apoptosis.

Based on the findings, Dr. Sreekanth D, the lead scientist wrote, "Betanin induces apoptosis in K562 cells through the intrinsic pathway and is mediated by the release of cytochrome c from mitochondria into the cytosol, and PARP cleavage".

Taken together, betacyanins processed abundantly bioactive compound betanin may be considered a remedy for the prevention and treatment of chronic myeloid leukemia, pending to the confirmation of the larger sample size and multicenter human study.

The antibacterial activity of onion extracts may be attributed to the presence of a high concentration of phenolic compounds in the onion.

In other words, the antibacterial property of onion is totally dependent on the antioxidant content which may be altered due to the different methods of growth, including conventional and organic cultivation.

Based on the findings, researchers said, "This study has provided an opportunity to establish valuable baseline information on the antibiotic potentiating activity of onion and... which can be further exploited for the treatment and/or management of infectious diseases".

Breast cancer is a chronic medical condition characterized by cell growth irregularly in the breast tissues due to DNA alternation.
Most cases of breast cancer begin in the cells on the surface of the inner lining of the breast tissues before penetrating into the deeper layers.

According to the statistics, in the US the lifetime of breast cancer in women is 1/8. In other words, if you are a woman, your risk of breast cancer development during your lifetime is 12.5%.

According to the prediction using historical data, in 2019, approximately 268,600 new cases of invasive breast cancer are expected to be diagnosed in women in the U.S., along with 62,930 new cases of non-invasive (in situ) breast cancer.

The exact causes of breast cancer are not identified. However, according to epidemiological studies, certain conditions may increase the risk of the disease such as being overweight, diet, inactivity, excessive alcohol consumption, smoking, and exposure to xenoestrogen.


Some researchers suggested that the discovery of oral contraceptive pills over the past few decades may correlate to the rise of breast cancer.

In other words, if you currently use an oral contraceptive, you are at a higher risk of the disease onset.

The study involved the analysis of the cytotoxicity of applying betanin-enriched extract then characterized on cancer and normal cells.

Betanin/Isobetanin concentrate significantly decreased cancer cell proliferation and viability of MCF-7-treated cells, by inducing the production of proteins associated with proapoptosis and genes associated with tumor suppression.

Observation of the increased alternation of the mitochondrial membrane potential suggested that the extracts cause cancer cell death by both intrinsic and extrinsic apoptotic pathways.

The death of MCF-7-treated cells is also attributed to betanin/isobetanin-induced autophagy.

Interestingly, the injection of betanin-enriched extract showed no obvious effect on normal cell lines.

Dr. Nowacki L, the lead scientist said, "The betanin/isobetanin mix as a therapeutic anticancer compound, alone or in combination with classical chemotherapeutic drugs, especially in functional p53 tumors".

In other words, betanin/isobetanin mix extracts processed anti-breast cancer cell proliferation by inducing cell death without affecting the integrity and viability of the normal breast cells.


In the analysis of the FDA-approved red food color E162, used in suppressing the development of multi-organ tumors in experimental animals, researchers compared the cytotoxic effect of the red beetroot extract with an anticancer drug, doxorubicin (Adriamycin) in the estrogen receptor-positive human breast cancer cells (MCF-7).

Beetroot extract exerted a significantly lower cancer cell inhibition compared to doxorubicin.

The findings suggested that betanin, the major betacyanin constituent, may play an important role in the cytotoxicity exhibited by the red beetroot extract, according to the study.

Taken together, betanin used alone or combined with other phytochemicals may be considered supplements for the prevention and treatment of breast cancer, pending the confirmation of the larger sample size and multicenter human study.

Bioactive dithiolthiones found in cruciferous vegetables may be the next-generation ingredient for the prevention and treatment of cardiovascular disease, some scientists suggested.
Dithiolthiones are phytochemicals in the class of Organosulfides, found abundantly in cruciferous vegetables, garden sorrel, horseradish, etc.

Cardiovascular disease is a class of conditions associated with the heart and blood vessels. In other words, CVD can be a result of anything that impedes the function of the heart and blood vessels.

According to the CVD statistics, heart disease kills more than 2,000 Americans every day. Approximately 60 million Americans have heart disease.
There are many causes of cardiovascular disease. Most CVD is caused by high blood pressure which contributes to the hardening of the arteries.

Plaques building up inside the arteries as a result of high blood cholesterol and other substances such as calcium over time can harden and narrow the arteries, leading to a blood clot, a major cause of CVD.

Other risk factors associated with the onset of cardiovascular disease include unhealthy diet, lack of exercise, being overweight, and smoking.

With an aim to find a natural ingredient or whole food with a cardioprotective effect, researchers examined whether sulforaphane protects the heart against I/R injury.

Sprague-Dawley rats selected for the study were given sulforaphane (500microg/kg/day) or vehicle intraperitoneally for 3 days and global ischemia was performed using isolated perfused Langendorff hearts.

Treatment with sulforaphane inhibited an increase in the post-ischemic left ventricular end-diastolic pressure (LVEDP) and improved the post-ischemic left ventricular developed pressure (LVDP), +/-dP/dt, and coronary flow as compared with the untreated control hearts.

Sulforaphane markedly decreased the infarcted size and attenuated the increased lactate dehydrogenase level in the effluent during reperfusion, similar to that of 5-hydroxydecanoic acid (5-HD).

Application of sulforaphane also attenuated the post-ischemia increased the concentration of atrial natriuretic peptide in coronary effluent and decreased levels of free radicals produced during and after injury.

Particularly, sulforaphane also protects the heart cell against damage and apoptosis by promoting heart cell cycle division.

The finding clearly indicated the cardioprotective effect of the sulforaphane against acute I/R injury.

In order to understand the therapeutic effect of sulforaphane protecting the myocardium from ischaemic reperfusion injury, researchers compared the cardio effects among cooked and fresh broccoli.

The study included two groups of rats fed with fresh (steamed) broccoli or cooked broccoli for 30 d, while a third group was given a vehicle only for the same period of time.

At the end of the study, by examining the 30 min ischemia followed by 2 h of reperfusion, researchers surprisingly found that both cooked and steamed broccoli display significantly improved post-ischemic ventricular function and reduced myocardial infarction and cardiomyocyte apoptosis compared to control, but steamed broccoli showed superior cardioprotective abilities compared with the cooked broccoli.

These results were attributed to the broccoli effect in enhancing the levels of proteins associated with cell survival, increasing the levels of antioxidants, and decreasing the levels of proteins associated with cell damage and apoptosis.


More importantly, the expressions of redox proteins (the thioredoxin (Trx)) associated with the reduction of oxidative stress were enhanced only in the steamed broccoli group.

Dr. Mukherjee S, the lead scientist said, "The present study documented superior cardioprotective properties of the steamed broccoli overcooked broccoli because of the ability of fresh broccoli to perform redox signaling of Trx"

Taken together, Isothiocyanates or sulforaphane may have a potential effect on the prevention and treatment of cardiovascular disease.

Myeloperoxidase (MPO) is a peroxidase enzyme found most abundantly in white blood cells neutrophil granulocytes.
Myeloperoxidase (MPO) plays a critical role that protecting our body against the invasion of microorganisms.

At cellular levels, myeloperoxidase (MPO) is demonstrated to be a local mediator of tissue damage by stimulating the production of proinflammatory cytokines with an aim to protect the cell against the invasion of pathogens that may enter the body to induce the acute phase of infection.

In other words, MPO deficiency that induces the exaggeration of the inflammatory response and affects neutrophil functions including cytokine production has been found to reduce the immune function in the acute phase of infection that leads to chronic low-grade inflammatory disease.

MPO deficiency has been found in patients with diabetes mellitus, pregnancy, iron deficiency, renal transplantation, thrombotic diseases, lead poisoning, obstructive jaundice, disseminated cancers, hematologic disorders, and neoplasms.


However, genetic predisposition may be the primary condition that causes the onset of PMO deficiency.


Dr. Roma Pahwa wrote, "Primary MPO deficiency is inherited as an autosomal recessive disorder and is generally present with varying degrees of severity and clinical features. A number of point germline mutations resulting in MPO deficiency have been detected in primary form. Some of these mutations are associated with the defective posttranslational processing of the MPO precursor protein and others with pre-translational defects caused by mutations in the regulatory portion of the MPO gene".


And, " Most of the mutations associated with the hereditary form are R569W (the commonest), Y173C, M251T, G501S, and R499C and a deletion of 14 bases (D14) within exon 9. It is noteworthy that eosinophils are never involved in MPO deficiency since eosinophil peroxidase is encoded by another gene than MPO".

With an aim to find a potent compound for the prevention and treatment of bacterial infection, researchers examined the effects of phytochemical indicaxanthin on the enzyme myeloperoxidase.

The kinetic study conducted by the University of Palermo was to examine the interaction between the betalains, betanin, and indicaxanthin as redox intermediates of compound I and compound II of MPO.

Based on the tested differentiation, both compound I and compound II exhibited significant antioxidant activity was found to be associated with the concentration of betanin and indicaxanthin.

The presence of betalains and indicaxanthin exhibited the function of MPO in scavenging hypochlorous acid at pH 7.0 and 25 degrees C.


At neutral pH and depending on their concentration, both betalains and indicaxanthin.can exhibit a stimulating and inhibitory effect on the chlorination activity of MPO.

However, at pH 5.0 both betalains and can only exhibit a stimulating and inhibitory effect on the chlorination activity of MPO in the micromolar concentrations.

Taken altogether, indicaxanthin may be considered a remedy for the promotion of the antibacterial activity of myeloperoxidase, pending the confirmation of the larger sample size and multicenter human study.

Intake of Isobetanin and betalains in the form of supplements should be taken with extreme care to prevent liver toxicity.

#Sulforaphane Found in #BroccoliSprouts Inhibits Proteins Associated with the Onset of Neuro-degenerative Disease, According to Studies

Age-related neurological disorders or neurodegenerative diseases are the conditions associated with the progressive death of neurons due to aging, leading to problems with movement (called ataxias), or mental functioning including Alzheimer's disease (AD) and dementia.

Dementia is a general term for neurodegenerative disease that causes a decline in memory and thinking skills that are severe enough to reduce a person's ability to perform everyday activities and quality of life.

About 5-8% of all people over the age of 65 have some form of dementia, and this number doubles every five years above that age.

The most common type of dementia is Alzheimer's disease, the condition that destroys brain cells, causing problems with memory, thinking, and behavior severe enough to affect language communication, memory, lifelong hobbies, or social life. Alzheimer's gets worse over time, and it is fatal.

Over 1 million people in the US alone are currently afflicted by Alzheimer's disease because of the degeneration of the hippocampus and cerebral cortex(3) of the brain where memory, language, and cognition are located.

Parkinson's disease (PD) is a disabling, progressive condition that causes cognitive deficit due to the interruption of frontal-subcortical loops that facilitate cognition and parallel the motor loop, and the loss of substantia nigra pars compacta (SNc) dopamine (DA) neurons.

Besides aging, geographical, hereditary viral, and toxicological exposure may also correlate to the development of diseases in the elderly.

Dr. Hung CW in the examination of the neurodegenerative disease in the aging population wrote, "Ageing, which all creatures must encounter, is a challenge to every living organism. In the human body, it is estimated that cell division and metabolism occur exuberantly until about 25 years of age. Beyond this age, subsidiary products of metabolism and cell damage accumulate, and the phenotypes of aging appear, causing disease formation".
And, "Among these age-related diseases, neurodegenerative diseases have drawn a lot of attention due to their irreversibility, lack of effective treatment, and accompanied social and economic burdens".


With an aim to find a natural food for the prevention and treatment of neurodegenerative disease, researchers examined the effect of sulforaphane, an active compound produced after conversion of glucoraphanin by the myrosinase enzyme in broccoli (Brassica oleracea var) sprouts inflammatory conditions in aged models involving microglial activation.

According to the results of pretreatment of pulsed electric fields that trigger the biological role of normal broccoli against lipopolysaccharide-activated microglia, sulforaphane-enriched broccoli sprouts showed an excellent potency against neuroinflammation conditions, particularly in both 6 and 24 h of microglial activation.

Sulforaphane in broccoli sprouts also inhibited an inflammatory cascade by decreasing the function of proteins associated with the production of pro-inflammatory cytokines.

Furthermore, proteins associated with the production of antioxidant enzymes and inhibition of oxidative stress (Nrf2 and HO-1) were also increased by SFN followed by the lowered neuronal apoptosis induced by activated microglia.

Based on the results, researchers wrote, " sulforaphane-enriched broccoli sprouts improved the scopolamine-induced memory impairment in mice through Nrf2 activation, inhibiting neuronal apoptosis, particularly through inhibition of caspase-3 activation which could lead to the neuroprotection against neurodegenerative disorders".

In other words, SFN protects the integrity of the neurons against neuron damage through its anti-neuroinflammatory and neuroprotective activities.

Taken altogether, broccoli sprouts processed abundantly SFN may be considered supplements for the prevention of neurodegenerative disease, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
