#Fennel Seed Extract and 5-Methoxypsoralen (5-MoP) Found in Fennel Inhibit Toxins Caused by Drug Interaction, Scientists Show

Fennel seeds may have the potential to act as liver antitoxin enzymes in removing xenobiotics from our body, some scientists suggested.

Liver cytochrome P450 3A4 (abbreviated CYP3A4) is an important enzyme, mainly found in the liver with the function in stimulating the removal of toxins from our body through the oxidation of small foreign organic molecules (xenobiotics).

Acute drug toxicity is a condition characterized by having too much of a drug in the body at once caused by overdoses of a certain medicine.

Otherwise, drug toxicity is a condition of toxin accumulation caused by long-term intake of medicine which has a strong and negative impact on the liver or kidney enzymes that are responsible for toxin removals.

The signs and symptoms of toxicity differ depending on the medication. However, the most common symptoms may include diarrhea, dizziness, nausea, stomach pains, vomiting, and weakness.

Untreated drug toxicity may induce complications of ataxia, muscle twitches, slurred speech, nystagmus, seizures, coma,....... and, in rare cases, heart problems and death.

Dr. ALASDAIR BRECKENRIDGE, in the article " A clinical pharmacologist's view of drug toxicity," wrote, "Our ability to understand the basis of adverse drug reactions has improved progressively, and with it, the ability to prevent their occurrence; one aim of the clinical pharmacologist is to identify patients at special risk" and " With a fair degree of certainty one can also predict that advances in bioinformatics, genomics, and combinatorial chemistry will progressively contribute to clinical toxicology in the future"(6).

Among these compounds, 5-methoxypsoralen (5-MoP) showed the strongest inhibition of toxins induced by drug interaction with an IC50 value of 18.3 μm and with a mixed type of inhibition(1).

According to the Brescia University Hospital, 5-methoxypsoralen (5-MOP) may be considered an alternative to 8-methoxypsoralen (8-MOP) for photochemotherapy of psoriasis, proven by the study in compared the clinical efficacy and tolerability of 5-MOP (1.2 mg/kg)-UVA versus 8-MOP (0.6 mg/kg)-UVA therapy in 25 patients of skin type III and IV(5).

The University of Toyama conducted an experiment to examine whether methanolic extract of fennel showed a characteristic of mechanism-based inactivation on erythromycin N-demethylation mediated by human liver microsomal cytochrome P450 3A4 (CYP3A4), suggested, 5-methoxypsoralen (5-MOP) showed the strongest inhibition with an IC50 value of 18.3 microM and a mixed type of inhibition, similar to those of above study.

With the preincubation time of 20 min only, 5-MOP showed preincubation time dependency with the IC50 value decreased from 18.3 microM with a preincubation time of 0 min to 4.6 microM with a preincubation time of 20 min(2).

These results suggested that 5-MOP acts as a human Liver Cytochrome P450 3A4 in the inhibition of toxins caused by drug interaction in a time-dependent manner.

Additionally, in the examination of the antitoxic effect of 30 sample plants of Indonesia, caused by the injection of xenobiotics, researchers showed that methanol extracts isolated from fennel seed exerted time-dependent inhibition toxins oxidized similarly to those of cytochrome P450 3A4 (CYP3A4) and CYP2D6.

In comparing the experimental injection to another plant with the same volume, fennel seed extract should contain mechanism-based inhibitors on CYP3A4.

Furthermore, SFN also inhibited NSCLC cells to form spherical colonies, to induce proliferation.

Moreover, SFN also suppressed the activation of signaling pathways to induce cancer cell adhesion.

Based on the findings, researchers said, "These results provide new insight into mechanisms underlying the chemopreventive ability of SFN and suggest a potential benefit of SFN to interfere with tumor metastasis".

Taken altogether, Cruciferous vegetables may be considered functional foods for the prevention and treatment of non-small cell lung cancer, pending to the confirmation of the larger sample size and multicenter human study.

Free radicals are unstable atoms with an unpaired electron in the outermost ring. Therefore, in order for them to become stable, free radicals must steal or donate an electron from or to other atoms.

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, chard, etc.

On finding a natural pigment for the prevention and treatment of diseases associated with oxidative stress, researchers examined the antioxidant effects of beetroot (Beta vulgaris L.) juice (BRJ), a good source of betalain (betacyanins and betaxanthin) pigments.

The study included rats with isoproterenol (ISP85 mg/kg injected at 24 h intervals)-induced myocardial ischemia followed by oral administration of BRJ for 28 days at doses of 150 and 300 mg/kg.


According to the tested results* Pretreatment with BRJ (150 and 300 mg/kg) restored the cardiac endogenous antioxidants and reduced levels of oxidative/nitrosative stress, in rats treated by ISP.
* Overexpression of cardiac inflammatory cytokines such as TNF-α, IL-6, and IL-10 caused by ISP was inhibited by the pretreatment of BRJ compared to the untreated group.


* Furthermore, hyperlipidemia level-induced ISP was significantly reduced by the BRJ pretreatment.


* Moreover, BRJ also protected against the extent of histological damage, myonecrosis, and edema, and maintained the architecture of cardiomyocytes by exhibiting the effects of anti-apoptotic proteins such as Bcl-2 and inhibiting the effects of apoptotic proteins.

These results strongly suggested that betacyanins and betaxanthin found in BRJ mitigate cardiac dysfunction and structural damage by decreasing oxidative stress, inflammation, and apoptosis in cardiac tissues.

Based on the findings, researchers said, "These results further support the use of BRJ in traditional medicine against cardiovascular diseases".


Taken together, betacyanins found abundantly in beetroot may be considered a remedy for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.

Intake of betacyanins in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Herbal #Cinnamon Inhibits Syndromes and Complications in #Diabetes, Researchers Say

Cinnamon, a kitchen spicy in many cultures, may have gained many believers, including medical professionals because of its anti-diabetic effect, through regulating the blood glucose levels in healthy and diabetic subjects. According to herbalists, intake of 1/4 tsp. in the morning and evening may reduce blood sugar significantly with little or no adverse side effects.

The herb has been used in herbal and traditional medicine as an anti-fungal and bacteria levels to improve the reproductive organ, prevent flatulence and intestinal cramping, and treat indigestion, diarrhea, bad breath, headache, migraine, etc.(1).

According to the 2011 statistic from National Diabetes InformationClearinghouse (NDIC), among U.S. residents ages 65 years and older, 10.9 million, or 26.9 percent, had diabetes in 2010, about 215,000 people younger than 20 years had diabetes—type 1 or type 2—in the United States in 2010 and approximately, 1.9 million people ages 20 years or older were newly diagnosed with diabetes in 2010 in the United States.

In a total of 66 patients with type 2 diabetes study conducted by Xuhui District Central Hospital, showed a reduction significant of blood triglyceride levels even in the low-dose group, after 3 months of oral administration(2)

Some researchers suggested that cinnamon extract improved fasting blood glucose levels and glucose tolerance without altering insulin secretion, through lower triglyceride and increased liver glycogen content, and improved insulin action in liver tissues(3).

Dr. Kim SH and Dr. Choung SY. told PubMed that "the effect of cinnamon significantly increases insulin sensitivity, reduces serum, and hepatic lipids, and improves hyperglycemia and hyperlipidemia possibly by regulating the PPAR-medicated glucose and lipid metabolism"(4)

In support of the above, chromium(Cr) and polyphenols found in cinnamon (Cinnamomum cassia) indicated an improvement of glucose, insulin, cholesterol, and HbA1c in patients with type 2 diabetes following Cr supplementation, in a double-blind placebo-controlled study(7).

Unfortunately, according to the Western University of Health Sciences, consumption of cinnamon is associated with a statistically significant decrease in levels of fasting plasma glucose but insisted that a high degree of heterogeneity may limit the ability to apply these results to patient care, due to the unclear preferred dose and duration of therapy(5)

A randomized clinical trial in which 70 patients with type II diabetes were assigned randomly to two groups (35 in cinnamon and 35 in the placebo group the test the effect of cinnamon on the glucose level in the blood, indicated an insignificant effect of using cinnamon on glucose level of diabetic patients and the herb should not be recommended to patients with type II diabetes depending on further studies(6).

Taken together, cinnamon in a low dose may be effective in controlling the levels of blood glucose in patients with diabetes without altering the insulin level. according to DR. Georgakopoulou EA(7), Oral intake of Cinnamon may cause Stomatitis and a burning sensation in the mouth with a large amount. Traditional Chinese medicine also insists that an Overdose of cinnamon can be toxic and damage to the liver(1).

Cancer is a deadly class of chronic disease caused by irregular cell growth in the tissue or specific organs in the body due to the alternation of cell DNA.
The ovaries are oval-shaped ovum-producing reproductive organs that form part of women's reproductive system that connect to the fallopian tubes and attach to the outer layer of the uterus via the ovarian ligament. Usually, the left and right ovaries take a turn to release an egg every month.

Ovarian cancer is medical a condition characterized by abnormal cells growth of ovarian cells that start on the surface of the inner lining of the ovary. Over time, the oversized tumor can travel a distance away to infect the nearby organs and tissue via fluid and blood circulation.

According to the statistics provided by the American Cancer Society, ovarian cancer is one of the deadly reproductive cancer in women. Every year, the disease causes more deaths than any other cancer of the female reproductive system.


The lifetime risk of getting ovarian cancer in a woman is about 1 in 78. comapred to the lifetime of dying from ovarian cancer risk of 1 in 108.


The 5-year relative survival of all stages of ovarian cancer is 47%. However, women with ovarian cancer diagnosed at the localized stage have a 5 years survival rate of over 92%.

The exact causes of ovarian cancer are not identified. However, epidemiological studies suggested that women who have endometriosis, genetic preposition such as the BRCA1 and BRCA2 gene, and acquired genetic mutation such as the TP53 tumor suppressor gene are susceptible to the risk of the development of the disease.


The Canadian Cancer Society in the evaluation of the risk of ovarian cancer in women who are never pregnant or giving birth wrote, "Women who have never been pregnant have a higher risk of developing ovarian cancer than women who have been pregnant".

And, "Researchers think that the lower risk may be because the hormones that are present during pregnancy have a protective effect. It is possible that the higher risk in women who have never been pregnant is linked to the factors that may make it difficult for her to become pregnant".


In other words, women who have at least one pregnancy are at a lower risk of ovarian cancer compared to women who do not.

Additionally, WS also exerted the immunomodulatory effect by targeting 16 proteins and protein-protein interactions.

Chemical analysis also suggested that the efficacy of WS in immunomodulation was also attributed to
the potential of withanolide-phytosterol combination and seven novel bioactive-immune target combinations.

Based on the findings, researchers said, "The study elucidated an in silico molecular mechanism of immunomodulation of WS. It unveils the potential of the withanolide-phytosterol combination to achieve better immunomodulation. Experimental validation of the network findings would aid in understanding the rationale behind WS immunomodulation as well as aid in bioactive formulation-based drug discovery.".

Taken together, Ashwagandha may be considered a functional herb for immune modulation, pending  the result of a larger sample size and multicenter study

Furthermore, the gene hormone expression of SRD5A1 that converts testosterone dihydrotestosterone (DHT) induced elevated androgen levels seldom found in blood and urine from patients with acne were significantly reduced by the application of AE1.


* AE2 suppressed gene expression level of all aforementioned parameters, similar to those of AE1, including IL-1α, SRD5A1, TNFα.


Based on the findings, researchers wrote, "Both herbal extracts demonstrated strong antibacterial and anti-inflammatory activity in this preliminary trial. In conclusion, the topical application of these botanical extracts can be good candidates for local acne treatment".

Taken together, broccoli combined with cauliflower may be considered functional foods for the prevention and treatment of acne, pending the confirmation of the larger sample size and multicenter human study.
