#Herbal #Chastetreeberry Induces Cytotoxicity of Cancer, According to Studies

The study conducted by respectable institutions suggested that Chaste tree berries may have a potential effect in reducing the risk and treatment of gastric/stomach cancer.

The efficacy of anti-gastric cancer extract is a result of antioxidant activity in reduced progression of oxidative stress which can be detected through using 2',7'-dichlorofluorescin-diacetate.

Dr.Ohyama K, the lead author said, " Our results demonstrate that intracellular oxidative stress and mitochondrial membrane damage is responsible for Vitex-induced apoptosis, which may be mediated by a diminution of reduced type glutathione within the cell".

Furthermore, in support of the above differentiation, another study conducted by the same institution, suggested that Vitex extracted expressed a similar effect against human gastric signet ring carcinoma cell line, KATO-III, through separating or breaking of DNA in a time- and dose-dependent manner.

Additionally, the study also indicated, Vitex-induced apoptosis by elevated the intracellular oxidative stress and mitochondrial membrane damage of gastric cancer cell lines through the mediation of the presence of reduced type glutathione within the cell.


Other; investigations of the cytotoxicity of the extract against several human cancer cell lines, including gastric signet ring carcinoma (KATO-III), and colon carcinoma (COLO 201), showed that injection of the extract exhibited a significant effect in interference of the cell cycle division, reduced DNA abnormal expression through antioxidative reagent pyrrolidine dithiocarbamate or N-acetyl-L-cysteine (NAC).

Finally, according to Dr. Ohyama K, the lead author, the cytotoxic activity of Vitex extract may be attributed to the effect on cell growth in the expression of apoptotic cell death by increased intracellular oxidation.

Taken together, chaste tree berries exert an anti-cancer effect through various phytochemicals and may be used as a functional food for ameliorated risk and treatment of gastric cancer, but intake of a large amount should be taken with care.

#Ginger's 6 #Shogaol Inhibits Aging Processes, According to Scientists

Aging is a natural process of becoming older. Antiaging is a term that relates to delayed aging. In other words, anti-aging is an action that makes your appearance look younger than your biological age.
The cause of aging has many theories, including the theory of the accumulation of damage due to oxidative stress in the initiation of DNA oxidation and the programmed aging concept due to DNA methylation.


However, regardless of the theories, the decrease in hormone production gradually found in the human body may be one of the major causes of aging progression.

In fact, reduced production of hormones affects the amount of hormone released into the bloodstream, leading to decreased functioning of the target organs and systems.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine to treat dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers on finding a natural ingredient that processes anti-aging properties examined the ameliorative effects of two antioxidant supplements, ginger and alpha lipoic acid (ALA), on hepatic ultrastructural alterations in old rats by evaluating the livers of young (4 months) and old (24 months) Wistar rats.

Additionally, in order to get more information about ginger's antiaging effects, researchers examined the anti-aging natural products, the active constituent 6-shogaol isolated from ginger.

Application of the tested compound showed the lifespan-extending effect in a dose-dependent manner in Caenorhabditis elegans.


The anti-aging activity effects were attributed to 6 gingerols in increasing oxidative stress tolerances via antioxidant enzyme induction.

In the depth differentiation, researchers surprisingly found that 6-shogaol inhibits the aging process independent from other aging-related factors of C. elegans.

Where Nuclear factor (erythroid-derived 2)-like 2, (Nrf2), is a pro- antioxidant protein that plays a critical role in protecting our body against oxidative damage triggered by injury and inflammation.

CYP2E1 is a protein involved in the metabolism of xenobiotics in the body.

The application of sulforaphane (SFN) treatment increased the levels of antioxidants by activating the levels of the Nrf2 and lowering oxidant stress observed by the decline in lipid peroxidation and increase in glutathione (GSH) levels.

SFN also decreased liver levels of triglycerides and cholesterol elevated by the injection of ethanol.

The results strongly suggested that SFN inhibited liver disease by increasing the expression of antioxidant enzymes and lowering the expression of free radicals as well as reducing the liver fat deposits.

Moreover, in the investigation to determine whether glucoraphanin and its breakdown product sulforaphane, are potent modulators of various phase I and phase II enzymes involved in carcinogen-metabolizing enzyme systems in vitro, researchers showed that injection of glucoraphanin at higher concentrations (25 μM) decreased dealkylation of methoxyresorufin, a marker for cytochrome P4501 activity

Where cytochrome P4501 is an enzyme involved in the metabolism of endogenous substrates and drugs, as well as the activation of certain toxins and environmental pollutants.

Compared to glucoraphanin, sulforaphane is superior to its precursor in modulating carcinogen-metabolizing enzyme systems in vitro.


The University of Rhode Island, Kingston study also suggested that SF activates Nrf2 activation in inhibited lipid accumulation in white adipose tissue, suppressed adipogenesis, insulin resistance, and glucose intolerance, and hepatic steatosis in Lep(ob/ob) mice.

Inflammation is a natural defense of the immune system that protects the body against the invasion of harmful stimuli and pathogens.
Anti-inflammatory activity is the process of reducing inflammation or swelling without affecting the function of the immune system.

In the acute phase of infection, the immune first line of defense, the white blood cells, macrophages stimulates the production of blood platelets to cover the wound or cut and proinflammatory cytokines to destroy all invaders.

Most cases of acute infection are stopped at this phase and the wound is healed quickly.

However, if the wound is too large, then the recovery may take a longer period and through 4 overlap recovery stages. Any interruption in the overlapped recover stage may cause the formation of scars.

* iNOS (71.9%) and COX-2 (65.9%) enzyme expressions involved in the inflammatory processes that recruit the leukocytes are also inhibited by the bioactive compound.

* Moreover, enzymes associated with the induction of free radicals chain reaction were also curbed by the indicaxanthin at a time- and dose-dependently.

Pharmacokinetically, the most efficacy in inhibition of inflammation was found to have a maximum 0.22 ± 0.02 μmol/L (n = 15) plasma concentration of indicaxanthin, with a half-life of 1.15 ± 0.11 h.


Based on the findings, researchers said, "This dietary pigment has the potential to improve health and prevent inflammation-based disorders".

Taken together, indicaxanthin may be considered a remedy for the prevention and treatment of inflammation, pending the confirmation of the larger sample size and multicenter human study.

A metabolic profile is a tool to diagnose metabolic disease.
A metabolic disease is a group of medical conditions that disrupt normal cell metabolism in converting food to energy.
The most common causes of metabolic disease are genetic prepositions. However, people with the mutated gene remain healthy for years.

Interestingly, cellular stress and family history have been found to be a major cause of the initiation of the symptoms of metabolic disease, epidemiologically.

As of today, there is no effective treatment for metabolic disease. Some children with metabolic disease diagnosed at an early age may respond very well. However, many do not respond well to therapy and cause severe problems, despite the early diagnosis.

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of hypercholesterolemia, researchers examined the effect of cranberry extract (CE) on the improvement of lipid dysfunction in HFD-induced obesity in an animal model.

The study included postnatal day 120 (PN120), male Wistar rats assigned into two groups: (1) SD (n = 36) fed with a standard diet and (2) HFD (n = 36), fed with a diet containing 44.5% (35.2% from lard) energy from fat.

According to the results from rats 12 animals from SD and HFD groups were killed at at At PN150 and others subdivided into four groups (n = 12/group): animals that received 200 mg/kg cranberry extract (SD CE, HFD CE) gavage/daily/30 days or water (SD, HFD) killed at PN180, HFD group showed higher body mass and visceral fat, cholesterol and triglyceride contents.

Furthermore, the HFD group demonstrated a significantly higher lipid peroxidation in plasma and tissues and higher protein carbonylation in the liver and adipose tissue compared to the SD group.
Application of CE not only inhibited the aforementioned parameters induced by a high-fat diet but also protected the liver tissues against by lowering lipid peroxidation and protein carbonylation and accumulation of liver fat without completely reversing the adiposity.

Based on the findings, researchers aid, "cranberry extract improved the metabolic profile and reduced oxidative damage and steatosis in HFD-fed rats, which suggests that it can help manage obesity-related disorders".

Taken together, cranberry may be considered a remedy for the improvement of the metabolic profile of obesity, pending the confirmation of the larger sample size and multicenter human study.

Betanin/isIsobetanin Inhibits Breast Cell Toxicity Induced Breast Cancer Associated with Oxidative Stress Overtime, Researchers Say

The breast-formed part of the women's reproductive system is the tissue overlying the chest muscles with a primary function to produce milk for nutrition to the infant and baby.
The size of a woman's breast is totally depending on the amount of fat accumulation.

Breast cancer is a type of cancer that starts in the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.

However, at the advanced stage, invasive cancer cells can travel a distance away from the breast to infect other healthy tissues and organs via lymph and blood, leading to secondary metastasis.

According to the joint study led by the Unité Génie Enzymatique et Cellulaire, phytochemical isobetanin may process chemopreventive activity against breast cancer.

In MCF-7-treated cells, betanin/isobetanin concentrate exerted anti-cancer activity by significantly decreasingand  cancer cell proliferation and viability.

Betanin/isobetanin concentrates increased the levels of proteins associated with the promotion of apoptosis such as Bad, and p53, and alternated the cancer cell's mitochondrial membrane potential that processed energy storage during oxidative phosphorylation.


Betanin/isobetanin treatment also induced autophagic cell death in the MCF-7 cancer cell line.

According to the statistics, the lifetime risk of breast cancer in a woman is 12%. In the US, approximately 268,600 new cases of invasive breast cancer will be diagnosed in women along with 62,930 new cases of non-invasive (in situ) breast cancer, compared to 2,670 new cases of invasive breast cancer in men in 2019.

The most common symptoms at the early stage of breast cancer are a lump (mass) in the breast, in the armpit (lymph nodes), nipple discharge (clear or bloody), inverted or retracted nipple, scaly or pitted skin on nipple persistent tenderness of the breast and unusual breast pain or discomfort.


However, at the advanced stage, patients with breast cancer may also experience additional symptoms of bone pain (secondary tumors in the bone), shortness of breath (secondary tumors in the lung), unintentional weight loss, and drop in appetite (secondary tumors in the liver) and headaches, neurological pain or weakness (secondary tumors in the nervous system).

The causes of breast cancer are unknown. Out of many risk factors associated with the risk of breast cancer, aging may be a major factor as most breast cancers are diagnosed after age 50.

Dr. Christopher C. Benz wrote, "Early-onset breast cancers largely represent inherited or early life-transforming effects on the immature mammary epithelium, late-onset breast cancers likely follow extended exposures to promoting stimuli of susceptible epithelium that has failed to age normally"


And " late-onset breast cancers grow more slowly and are biologically less aggressive than early-onset breast cancers, even when controlled for hormone receptor (e.g. estrogen receptor, ER) and growth factor receptor (e.g. HER2) expression, supporting the conclusion that the biology of breast cancer is age-dependent".

More precisely, betanin/isobetanin inhibits breast cell toxicity-induced breast cancer caused by accumulated breast tissue damage due to oxidative stress over time.

Based on the finding, researchers said, "Our data bring new insight to consider the betanin/isobetanin mix as a therapeutic anticancer compound, alone or in combination with classical chemotherapeutic drugs, especially in functional p53 tumors".

Taken together, isobetanin combined with betanin/may be considered a remedy for the prevention and treatment of breast cancer, pending the confirmation of the larger sample size and multicenter human study.

#Coffee Inhibits Risk Factors Such as Obedity and Depression of Cardiovascular Disease, According to Studies

Coffee consumers may have one less risk to worry about. A recent study by a well-known institute suggested that long-term and moderate coffee consumption is inversely associated with CVD risk.
Coffee, becoming a popular and social beverage all over the world, particularly in the West, is a drink made from roast beans from the Coffea plant, native to tropical Africa and Madagascar.

Cardiovascular disease is a medical condition characterized by a heart attack, chest pain (angina), or stroke caused by narrow blood vessels induced block blood.

However, the effect of caffeine in lipid files may be associated with endothelial function in increased CVD risk, depending on how the coffee is prepared.

More importantly, epidemiological studies also suggest that risks of mortality in both CV and all causeson are substantially attenuated in regular coffee drinkers.

Furthermore, other researchers insisted that the health benefits of coffee consumption regarding cardiovascular system risk, mostly depend on its antioxidant compounds involved many factors, such as the variety of coffee species, roasting degree, type of brewing method, and serving size.

Since most of the data on coffee's health effects are based on observational data, the risk of high caffeine adverse effects such as anxiety, insomnia, tremulousness, and palpitations have rarely been mentioned, therefore, more randomized, controlled studies are necessary, some researchers suggested.
