#Phytochemical 3,3'-Diindolylmethane Inhibits Expressions Associated with Renal Fibrosis, According to Studies

Renal fibrosis is a progressive condition caused by excessive accumulation of extracellular matrix including collagen, enzymes, and glycoproteins which provided structural and biochemical support of surrounding the renal cells during aging and chronic kidney disease (CKD).

According to the statistics, CKD and renal fibrosis affect approximately half of the adults age 70 and over and 10% of the world's population.

In other words, renal fibrosis is a direct consequence caused by the inability of the kidney to regenerate after injury, leading to the formation of renal scars that results in a progressive loss of renal function.

Most cases of renal fibrosis ultimately lead to end-stage renal failure and a need for dialysis or kidney transplantation.

At the early stage of renal fibrosis, patients may experience symptoms of dull pain in the abdomen or back, pain on one side between your upper abdomen and back, leg pain. discoloration swelling in one or both legs.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

In the urgency to discover the potential compound for the treatment of renal fibrosis, researchers examined the effects of 3,3'-Diindolylmethane (DIM) a natural acid condensation extracted from cruciferous plants, on renal fibrosis.

DIM has been found to process anti-hepatic and cardiac fibrosis models in a number of studies.

The study included the established unilateral ureteral obstruction (UUO) and transforming growth factor (TGF)-β1-stimulated normal rat kidney (NRK)-49F fibroblast cell mouse models.


Injection of DIM reduced kidney injury and production of interstitial collagens fibrosis.

In the UUO model, DIM inhibited the expression of fibronectin, and collagen-1 associated with the formation of scars but retained E-cadherin involved in the formation of adherens junctions to bind cells with each other.

In other words, DIM protects the renal cells against the induction of renal fibrosis.

In vivo and in vitro, the administration of DIM is also found to suppress local fibroblast activation observed by the levels of myofibroblast markers α-SMA and vimentin.

Most importantly, DIM significantly inhibited the levels of pro-inflammatory cytokine-induced abnormal proliferation of NRK49F cells, in a time- and dose-dependent manner.

Furthermore, DIM also inhibited the expression of Smad2 phosphorylation in the formation of renal scar tissue.

Based on the results, researchers concluded that DIM protects renal cell integrity by inhibiting TGF-β/Smad2/3 signaling against the formation of renal fibrosis.


Taken together, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of renal fibrosis, pending the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytic acid (Inositol hexaphosphate) may have a potential effect in reducing the risk and treatment of cancers, some scientists suggested.

Cancer is a class of chronic diseases associated with the growth of irregular cells that start in the cells of the inner lining of a tissue or organ caused by DNA alternation.

At the advanced stage, cells of the invasive tumor may travel a distance away from the primary site to infect other healthy tissue of the organs, through lymph and blood, leading to secondary metastasis.

Some cancers can be slow-growth such as prostate and colon cancer.

Primary cancer at stage 0 and 1 do not induce any symptoms. However, at the later stage, a larger size tumor can suppress the nearby tissue and nerve cells leading to localized and severe symptoms and pain, depending on the type of cancer.

According to the statistics, in the US, the number of new cases of cancer(cancer incidence) is 439.2 per 100,000 men and women per year (based on 2011–2015 cases). The number of cancer deaths (cancer mortality) is 163.5 per 100,000 men and women per year (based on 2011–2015 deaths).

Cancers who are diagnosed in the early stage have a 5 years survival rate close to 100%. Therefore, the annual examination may be necessary for people who are classified as having a higher risk of cancer development.


The causes of cancer are unknown. researchers cannot explain why people with the same healthy condition, and family history in the same family, are susceptible to the onset of the disease while others do not.

Either they can explain why people who carry some of the risk factors associated with the development of cancer never have cancer, while others having no risk factors are diagnosed with cancer.

Conventionally, the treatment of cancers completely depends on the types and grades of cancer. Some people with cancer at an early stage may only need one treatment. However most people with cancer in the later stage have a combination of treatments, including surgery with chemotherapy and/or radiation therapy.

Phytic acids (Inositol hexaphosphate) are phytochemicals of the organic acid found abundantly in nuts, sesame seeds, soybeans, wheat, pumpkin, beans, almonds, etc.

With an aim to find a natural ingredient for the prevention and treatment of cancers without inducing side effects, researchers examine the anticancer effects of phytic acid (IP6).

In vitro and in vivo phytic acid (IP6) exerted a strong anticancer activity by inhibiting the tumor growth and cell proliferation in a series of cancer cell lines, including colon, nipple, breast, prostate, cervix, liver, pancreas, melanoma, and glioblastoma.

The efficacy of IP6 was probably associated with the promoted antioxidant production, improved signal transduction, enhanced functions of the NK-cells, and exhibited cell cycle arrest and activated proteins associated with cell death programming.

Furthermore, in the findings, the proper role for the treatment of cancer researchers looked into the concentration of IP6 and its lower phosphorylated forms (IP(1-5)),

At the 10 microM to 1 mM concentrations, IP6 and its lower phosphorylated forms (IP(1-5)) showed a potential activity in regulating vital cellular functions such as signal transduction, cell proliferation, and differentiation, similar to those of the previous study.

In vivo and in vitro, exogenously administered IP6 and internalized, IP6 and IP(1-5) inhibited cell growth.

Moreover, researchers found that the application of IP6 not only decreased cellular proliferation but also caused the differentiation of malignant cells often resulting in a reversion to a normal phenotype.

The results strongly indicated that IP6 inhibits the cancer cell lines through a number of mechanisms, including cell cycle regulatory genes, differentiation genes, and tumor suppressor genes.

Taken together, phytic acid (IP6) may be considered an anti-cancer bioactive compound for the prevention and treatment of cancer with no side effects, pending the confirmation of a large sample size and multicenter human study.

The function of neurons is to process and transmit information that controls the body function.

Amyloid beta is the main component of the amyloid plaques found in the brains of Alzheimer patients.

Alzheimer's disease is a type of dementia correlated with major reductions of neurons to the respective target areas, named after German physician Alois Alzheimer.

In other words, Alzheimer's disease is caused by the gradual loss of neuron functioning that usually starts with the early symptom of the difficulty of remembering recent events and worsens over time.

Dementia is a group of neurodegenerative diseases caused by a decline in mental ability in the older population that is severe enough to interfere with daily life, such as memory loss.

Compared to dementia, neurological disorders are a class of brain diseases associated with reduced function of the brain, depending on the region of the neurons that were affected.

Alzheimer's destroys brain cells, causing cognitive modalities severe enough to affect language communication, memory, lifelong hobbies, or social life. Alzheimer's gets worse over time, and it is fatal.

The disease is most prevalent in the elderly and causes over 60% of all cases of dementia.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On findings a potential compound for the prevention of neuronal cell death in Alzheimer's disease, researchers compared the effects of curcuminoids, curcumin, demethoxycurcumin, and bisdemethoxycurcumin and alpha-Tocopherol on beta-Amyloid (betaA) induced oxidative stress in brain cells.

According to the results from the tested analysis, all phytochemicals isolated from turmeric blocked the insults from the beta (1-42) in PC12 rat pheochromocytoma and normal human umbilical vein endothelial (HUVEC) cells.

Furthermore, the ED(50) values of curcumin, demethoxycurcumin, and bisdemethoxycurcumin toward PC12 and HUVEC cells were 7.1+/-0.3, 4.7+/-0.1, 3.5+/-0.2 microg/ml and 6.8+/-0.4, 4.2+/-0.3, and 3.0+/-0.3 microg/ml, respectively.

Moreover, according to the DPPH radical trapping experiment assay, all 4 compounds exerted a strong antioxidant activity compared to that of alpha-tocopherol.
Additionally, the injection of alpha-tocopherol did not protect the cells from betaA(1-42) insult even at>50 microg/ml concentration.
Based on the results, researchers wrote, " The results suggest that these compounds may be protecting the cells from betaA(1-42) insult through antioxidant pathway".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of Alzheimer's disease, pending the confirmation of the larger sample size and multicenter human study.

Scientists may have found a bioactive compound for the prevention and treatment of lung diseases with no side effects, according to studies.
Lung diseases are medical conditions, affecting the upper respiratory tract, trachea, bronchi, bronchioles, alveoli, pleura and pleural cavity, and the nerves and muscles of breathing.

People with lung diseases have breathing problems, preventing them to get enough oxygen for the body's needs.

According to the statistics from the World Health Organization, respiratory diseases are the leading causes of death and disability in the world. chronic obstructive pulmonary disease (COPD) and asthma are 2 of the most common respiratory disease in the world, affecting 65 million and 334 million people, respectively.

3 million people die from chronic obstructive pulmonary disease (COPD) each year, making it the third leading cause of death worldwide. And 14% of 334 million people suffer from asthma are children.

With an aim to find a natural compound for the treatment of lung diseases researchers examined the effects of sulforaphane (SF) in the protein Nuclear factor-like 2 (Nrf2) in mice from arsenic-induced toxicity.
Where Nuclear factor-like 2 has been found to protect the body against a broad spectrum of diseases induced by environmental exposure to harmful substances through its detoxed activity.

Selected mice were exposed to 2 weeks of arsenic-containing dust. At the end of 2 weeks, tested mice showed pathological alterations, oxidative DNA damage, and mild apoptotic cell death in the lung.


Pretreatment of sulforaphane activated the expression of Nrf2 only blocking the alternation caused by arsenic-containing dust but also restoring all the damages induced by the substances.

Furthermore, SF-mediated activation of Nrf2 also alleviated inflammatory responses by modulating cytokine production, thus reducing the oxidative stress in the tested mice.


These results clearly suggested that dietary intervention which targets the Nrf2 activation may have a significant effect on the reduction of adverse health effects associated with arsenic exposure.

In the mouse model, activation of NRF2 protein was also found to regulate the rhythmic expression of antioxidant genes involved in glutathione redox homeostasis in the mouse lung.

Moreover, in vivo bleomycin-induced lung fibrosis model, the pretreatment of SF restored the lung function by blocking the phenotype caused by bleomycin.

According to the observation of the lungs of the arrhythmic Clock(Δ19) mice, SF increased the levels of NRF2 and glutathione, the antioxidant enzymes produced by the host mice.

The results suggested that NRF2 activated by SF not only inhibited the oxidative damage but also protect the tested mice lungs against fibrositis.


Dr. Pekovic-Vaughan V, the lead scientist, after taking into account co and confounders, said, "Our findings reveal a pivotal role for the circadian control of the NRF2/glutathione pathway in combating oxidative/fibrotic lung damage, which might prompt new chronotherapeutic strategies for the treatment of human lung diseases, including idiopathic pulmonary fibrosis".


However, in wild-type neonatal mice exposed to hyperoxia. SF was also found to activate Nrf2 activation through the induced expression of anti-oxidant genes but did not attenuate alveolar growth arrest caused by exposure to hyperoxia.

This discovery strongly suggested that activation of activated Nrf2 may have a certain limit in protecting the lung and lung tissues against alveolar growth exposed to an excess supply of oxygen (O2) or higher than the normal partial pressure of oxygen.

Taken together, depending on the types of lung diseases, bioactive compound sulforaphane may be used for the prevention and combined with the primary therapy for the treatment of lung diseases with no side effects.

#Turmeric Reduces Cholesterol Accumulation Associated with the Onset of #Atherosclerosis, Researchers Suggest

Cardiovascular diseases (CVD) are a class of conditions associated with the dysfunction of the arteries and heart, affecting the heart functioning.
According to the statistics provided by the CDC, heart disease is the leading cause of death for both men and women. Men are associated with 1/2 of the deaths due to heart disease in 2015 were in men, compared to women.
Believe it or not, more than 63),000 Americans die from heart disease each year or 1 in every 4 deaths are caused by heart disease.

Atherosclerosis is a condition caused by long-term plaques built up on the wall of the arteries. Over time, continued plaque accumulation can lead to cardiovascular disease.

In other words, narrowing the arteries due to plaque accumulation is a major cause of heart disease and stroke.

Plaque is made up of fat, cholesterol, calcium, and other substances found in the blood.

The most common symptoms found in patients with antiatherosclerosis involve reduced blood flow to the body, including chest pain or angina, pain in the leg, and arm, shortness of breath fatigue, and confusion.

According to Dr. Roma Pahwa and colleagues, atherosclerosis causes approximately 50% of all deaths in Westernized society associated with the inflammation in the arteries involved in the accumulation of cholesterol in the bloodstream.

Epidemiologically, the most common risk factors of atherosclerosis are unhealthy blood cholesterol levels associated with high levels of bad and low levels of good cholesterol, high blood pressure, smoking, insulin resistance, diabetes, overweight or obesity, and physical inactivity.

Some researchers suggested that the promotion of a high-fat diet that induces widespread obesity in the Western world may be one of the major culprits that cause atherosclerosis.

Dr. Lovren F and colleagues wrote, "Obesity is a multifactorial chronic disease characterized by an accumulation of visceral and subcutaneous fat, which leads to a predisposition toward cardiometabolic diseases".

And, " A plethora of mechanisms, including abnormalities in lipid metabolism, insulin resistance, inflammation, endothelial dysfunction, adipokine imbalance, and inflammasome activation have been suggested to underlie the relationship between obesity and atherosclerosis.".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

In finding a potential compound for the treatment of multiple sclerosis (MS), researchers examined the efficacy of supplementation with curcumin in reducing intestinal cholesterol absorption in animal models.

The study included high-fat diet-fed apolipoprotein E knockout (ApoE-/-) mice that prevented atherosclerosis fed a high-fat diet supplemented with or without curcumin (0.1% w/w) for 16 weeks.

Based on the tested differentiation, the curcumin treatment group showed not only a reduced cholesterol accumulation in the aortas by 56% but also lowered plasma total cholesterol and low-density lipoprotein cholesterol levels compared to the non-curcumin group.

Furthermore, curcumin supplementation group also demonstrated a reduced risk of atherosclerotic lesions by 45%.

Moreover, the efficacy of the anti antiatherogenic effect of curcumin was associated with a significant decrease in intestinal cholesterol absorption by 51% by inhibiting the intestinal gene expression of Niemann-Pick C1-like 1 involved in cholesterol uptakes.

Collectively, researchers said, "These findings support the hypothesis that curcumin supplementation reduces intestinal cholesterol absorption and prevents atherosclerosis in high-fat diet-fed ApoE-/- mice".

And " Curcumin affords a potent antiatherogenic action by inhibiting intestinal cholesterol absorption in the mouse".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of atherosclerosis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Cranberry Inhibits Lung Cancer Cells Metabolism, Protein Modifications, and Cell Cycle Arrests, Researchers Show

The lung formed part of the respiratory system is a vital organ located near the backbone on either side of the heart with functions of inhaling oxygen from the air then transporting it to the bloodstream and exhaling carbon dioxide from the bloodstream.
Lung diseases are a class of conditions associated with reduced function of the lung, including lung cancer.

Lung cancer is a medical condition associated with abnormal growth of the cells in the lung's tissue that begins in the cells on the surface of the inner lining tissues of the lung due to the alternation of cell DNA.

The most common form of primary lung cancer is derived from epithelial cells.

In the US, lung cancer is the leading cause of cancer deaths. According to the statistics from the American Cancer Society, in 2019, about 228,150 new cases of lung cancer, including 116,440 in men and 111,710 in women will be diagnosed in the US.

Sadly, lung cancer also caused the death of 142,670 Americans death, including 76,650 in men and 66,020 in women in the same year.

The lifetime risk of lung cancer in a man is 6.7% compared to 5.9% in a woman.

The exact causes of lung cancer are not known. However, epidemiologically, smoking tobacco, second-hand smoke, radon, asbestos, occupational exposure to certain chemicals, personal or family history of lung cancer and lung disease and exposure to radiation are some major factors found in patients with lung cancer.

Dr. Ozlü T, the lead scientist wrote, " Tobacco use has been reported to be the main cause of 90% of male and 79% of female lung cancers. 90% of deaths from lung cancer are estimated to be due to smoking".

Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.


On finding a potential compound for the protection of lung cell integrity, researchers examined the effect of the efficacy of proanthocyanidin-rich cranberry fraction (PAC) on lung cancer.


According to the tested analysis, NCI-H460 human lung cancer cells treated with 50 µg/mL PAC or vehicle for 6 hours, PAC exerted a significant activity in the regulation of apoptosis, regulation of programmed cell death, positive regulation of cell death, positive regulation of apoptosis, and apoptotic mitochondrial changes.


Moreover, NCI-H460 human lung cancer cells treated with 50 µg/mL PAC showed a strong effect on cancer cell metabolism, protein modifications, and cell cycle by inhibiting the M phase of the mitotic cell cycle, mitosis, M phase, cell cycle checkpoint, and regulation of cell cycle process.

More precisely, PAC-induced lung cancer cell apoptosis and anti-proliferative activity through cell cycle arrest.

Furthermore, PAC treatment also enhanced the levels of glutathione S-transferases associated
with blocking cancer cell metastasis.


PAC treatment also down-regulated DNA metabolism, cell cycle, meiosis, mitosis and interestingly oncogenesis signaling.

PAC treatment also inhibited the alternation of ErbB receptors and signaling pathways and mTOR signaling pathways mutated in lung tumors, involved in cancer development and progression

Taken together, cranberry used in conjunction with other remedies may be considered a remedy for the prevention and treatment of lung cancer, pending the confirmation of the larger sample size and multicenter human study.
