Green tea is found to have a potential effect in the promotion of oral health against bacterial-induced tooth decay or cavities, a renowned institute suggested.

Scientists may have found a natural compound for the treatment of Parkinson's disease (PD) with no side effects, according to studies.
Parkinson's disease (PD) is a disabling, progressive condition caused by the gradual loss of neuron cells.

In other words, PD is a cognitive deficit associated with the interruption of frontal-subcortical loops that facilitate cognition and parallel the motor loop and the loss of substantia nigra pars compacta (SNc) dopamine (DA) neurons.

According to the statistics, more than 10 million people worldwide are living with PD. In the US, every year, 60,000 adults are diagnosed with PD. Approximately, 95% of people diagnosed with PD are over 50 years of age.

PD can be classified into 5 stages, depending on the severity
* Stage 1

In stage 1, symptoms of PD do not interfere with daily activities.

* Stage 2
In stage 2, the symptoms start to get worse over time. Making living alone to carry daily tasks now may be difficult.

* Stage 3

The symptoms are now interfering with daily activity, including the impairment of dressing and eating.

* Stage 4
The symptoms are severe with limited functioning, Patients in this stage require the help of others to carry out daily activities.

The exact causes of PD are unidentified. However, the epidemiological study suggested that age and sex, genetic factors, head trauma, exposure to chemicals, and long-term use of certain medications and drugs are some of the prevalent risks of Parkinson's disease (PD).

Some researchers suggested lifelong cigarette smoking may have a strong impact on the risk of PD.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers concerned about Parkinson's disease (PD) associated with the progressive degeneration of dopaminergic neurons in the nigrostriatal system and dopamine (DA) depletion in the striatum, examined the Zingerone and eugenol, essential oils extracted from ginger and cloves, free radical scavenging and antioxidant effects.


Rodents used in the study were induced PD by injected with 6-hydroxydopamine (6-OHDA) intracerebroventricularly.

According to the assays, daily oral administration of eugenol/zingerone and injection of L-DOPA intraperitoneally for 4 weeks following a single 6-OHDA injection did not improve abnormal behaviors induced by L-DOPA treatment.

Where 3-(3,4-Dihydroxyphenyl)-L-alanine (L-DOPA) is the most popular therapeutic medicine for treating PD with adverse effects.

However, injection of zingerone and eugenol alone enhanced the reduction of striatal DA and its metabolites and zingerone decreased the oxidative stress and increased the levels of antioxidants such as glutathione peroxidase and the oxidized L-ascorbate in the striatum.

In other words, compared to the effects of zingerone against PD through its antioxidant activity, pre-treatment with zingerone or eugenol does not prevent 6-OHDA-induced DA depression by preventing lipid peroxidation.


Taken altogether, ginger may be considered a functional spice for the prevention and treatment of PD, pending the confirmation of a larger sample size and multicenter human study.

According to a joint study led by the Xinyang Normal University, Epigallocatechin-3-gallate (EGCG), a bioactive compound in green tea inhibited the risk of breast cancer through many mechanisms involving primarily cell death and survival.



The study also insisted on the efficacy of Epigallocatechin-3-gallate (EGCG) in reducing the risk of breast cancer probably due to the main function in induced cancer cell cycle arrest in interference of cell division.

In the MDA-MB-231 breast cancer cell line, Epigallocatechin gallate isolated from green tea induced apoptosis and inhibited tumorigenesis through the expression of multiple signaling pathways in breast cancer cells.


Also, EGCG decreased MDA-MB-231 cell viability and significantly downregulated the expression of β-catenin in regulating the coordination of cell-cell adhesion and gene transcription, according to Western blot analysis as patients with breast cancer are found to be associated with high levels of β-catenin in breast cancer tissue than in normal tissue.

Dr. Hong OY, the lead author said, "EGCG inhibits the growth of MDA-MB-231 cells through the inactivation of the β-catenin signaling pathway. Based on these promising results, EGCG may be a potential treatment for triple-negative breast cancer patients.".


In polychlorinated biphenyls (PCBs) accumulated in human breast cancer tissue induced the growth of estrogen-sensitive tumors, EGCG inhibited the early onset of breast cancer by modifying estrogenic activity against cell proliferation in a dose-dependent manner.


Furthermore, at a dose of 25 μM, EGCG also exerted antiproliferative and pro-apoptotic action in the activation of the ERβ receptor.


With all the information collected above, green tea, containing rich sources of anti-malignant and nonmalignant tumors phytochemicals, may be considered as a functional food in reduced risk and treatment of breast cancer.

In finding a potential compound for the treatment of gastrointestinal tumors, researchers examined the effect of a purified cranberry-derived proanthocyanidin extract (C-PAC) on esophageal adenocarcinoma (EAC) cell lines, including acid-sensitive JHAD1 and OE33 cells, and esophageal tumor xenografts in athymic NU/NU mice.

According to the tested assays, C-PAC shared a strong activity in inducing the expression of a protein associated with cell death programming mainly in JHAD1 and OE33 cells.

Autophagy that promotes cancer cell growth and anti-apoptosis of JHAD1 and OE33 cells were also inhibited by the injection of C-PAC.

In vitro, C-PAC-induced cell death was attributed to the deactivation of the phosphatidylinositol-3-kinase (PI3K)/Akt and the mammalian target of rapamycin (mTOR) signaling pathways expression in the promotion of cancer cellular processes, including cell survival, growth, and differentiation.

Additionally, C-PAC not only inhibited the levels of anti-apoptotic proteins such as BCL-xL, Cytochrome C, PARP), pathways involved in the occurrence and progression of cancer but also exhibited the expression of proteins that induce cell cycle arrest.


Similarly, in vivo, oral delivery of C-PAC significantly inhibited OE19 tumor xenograft growth as the aforementioned activities.


In other words, in vitro and vivo purified cranberry-derived proanthocyanidin extract (C-PAC) eradicates the esophageal adenocarcinoma (EAC) cells through several mechanisms involved in pro-apoptosis, anti-anti-apoptosis, and blocking cellular division.

Based on the finding, researchers after taking other factors into account wrote, " C-PAC is a potent inducer of EAC cell death and is efficacious in vivo at non-toxic behaviorally achievable concentrations, holding promise for preventive or therapeutic interventions in cohorts at increased risk for EAC, a rapidly rising and extremely deadly malignancy".

Taken altogether, cranberry extract may be considered a remedy for the prevention and treatment of esophageal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Scientists may have found a bioactive compound for the prevention and treatment of pancreatic cancer, according to studies.
Cancer of the pancreas or pancreatic cancer is a condition of a malignant neoplasm of the pancreas.

Most cases of primary pancreatic cancer start in the cells on the surface of the inner lining of the pancreas as a result of irregular cell growth caused by the alternation of DNA.

The pancreatic cancers start in the ducts (an exocrine component of the pancreas) carried pancreatic juices with cancer of an epithelium that originates in glandular tissue and accounts for over 75 - 90% of all cases of exocrine cancers.

According to the statistics, pancreatic cancer is the fourth leading cause of cancer death in both men and women in the US. Cancer has the poorest prognosis compared to other major types of cancer, with a 5-year survival rate of approximately 5%.

More precisely, in the US, the age-adjusted incidence of pancreatic cancer is higher in men compared to women regardless of ethnicity.

In human pancreatic cancer cells (Panc-1), injection of 13C inhibited the expression of miR-21 positively correlated in a time- and dose-dependent manner.

13C and gemcitabine combination therapy increased cytotoxicity in Panc-1 cells.


The efficacy of 13C was attributed to its activity in promoting cancer cell apoptosis by attenuating miR-21 transfection.

The finding suggested that I3C would be effective in enhancing the sensitivity of pancreatic cancer cells to gemcitabine via the inhibition of the overexpression of miR-21.

An additional experiment demonstrating the ability of a dietary agent, indole-3-carbinol (I3C), to lower the LD(50) of gemcitabine (Gemzar) in decreasing growth of both male (MiaPaca2) and female (SU86.86) pancreatic cancer cells, researchers found that* Without an injection of 13C, female pancreatic cancer cells were more resistant to gemcitabine.
* In male MiaPaca2 cells application of 1, 10, or 100 μM of I3C induced apoptosis by reactivating the tumor suppressor gene in pancreatic cancer cells.


* 13C also protects the healthy pancreas cells by enhancing the activity of the tumor suppressor gene.

Moreover, by examining the DNA methylation researchers suggested suppressor tumor gene (p16INK4a) inactivation in the promotion of hypermethylation is considered an early event in pancreatic carcinogenesis.

DNA methylation is a process by which methyl groups are added to the DNA molecule.

These results clearly indicated, that I3C, a possible non-toxic hypomethylating agent, combined with the anticancer agent, gemcitabine, is a powerful strategy for treating pancreatic cancer.


Taken altogether, the bioactive compound indole-3-carbinol (I3C) may be used for the prevention and treatment of pancreas cancer, pending the results of the larger sample size and multicenter human study.

#Carotenoids Inhibits the Risk of #ColorectalCancer in Patients with DICER1 rs3742330 Genotypeancer, Researchers Say

Cancer is a class of diseases associated with irregular cell growth in the tissue of organs.
The colon-rectum formed as a part of the digestive system is involved in the absorption of water from the small bowel contents and broken down of certain materials in the feces into substances which some of them to be reabsorbed and reused by the body.

Colorectal cancer is cancer located in the colon or rectum caused alternation of cell DNA. Most cases of colorectal cancer begin in the cells on the surface of inner ling tissue in either the colon or rectum, before multiplying to form a tumor.

Colorectal cancer is relatively very common and slowly growing and progressing cancer.
The disease is also the third most commonly diagnosed cancer in developed countries, including the US and Canada.

According to the statistics provided by the American Cancer Society, in the US,
101,420 new cases of colon cancer and 44,180 new cases of rectal cancer will be diagnosed in 2019.

Additionally, colorectal cancer will also cause the death of 51,020 Americans in 2019.

Overall, the lifetime risk of colorectal cancer developing is 1 in 22 (4.49%) in men and 1 in 24 (4.15%) in women.

Besides adenocarcinomas, the common type of colorectal cancer originates in epithelium tissues and squamous cells that make up the bowel lining together with the gland cells, other types of colorectal tumors include

* Carcinoid
Carcinoid is an unusual type of slow growth neuroendocrine tumor in the hormone-producing tissue in the digestive system. The condition may look benign but can spread to other parts of the body as they grow

* Leiomyosarcomas
Leiomyosarcomas are the rare and resistant type of colorectal cancer and generally not very responsive to chemotherapy or radiation. They are a malignant abnormal mass of tissue of smooth muscles of the colon, comprising between 5–10% of soft tissue sarcomas.

* Lymphomas
Bowel lymphomas are rare and more likely to start in the rectum rather in the colon. Many patients with lymphomas are diagnosed in the late stage and already spread to other organs.

* Melanomas
Melanomas are a rare malignant tumor of cells that produce the dark pigment. This type of bowel cancer usually originates from somewhere else and then spread to the bowel.

Carotenoids are a class of mainly yellow, orange, or red fat-soluble pigments, including lycopene and carotene, found abundantly in ripe tomato, pumpkins, carrots, corn, and daffodils.

On finding a potential compound for the treatment of colorectal cancer, researchers investigated whether dietary lutein/zeaxanthin, the major compounds found in carotenoid intake affect colorectal cancer risk in patients with a DICER1 rs3742330 polymorphism.

The experiment included 923 colorectal cancer patients and 1,846 controls based on eligibility criteria, that answered the semiquantitative food frequency questionnaire and the DICER1 rs3742330 genotype.

Based on the returned questionnaire differentiation, the highest quartile of lutein/zeaxanthin consumption was inversely associated with colorectal cancer risk.

In genotype analysis, patients who carry the G allele (AG + GG) showed a significantly reduced colorectal cancer incidence compared to AA carriers.
Furthermore, patients with the G allele (AG + GG) along with high lutein/zeaxanthin consumption were markedly associated with a decreased colorectal cancer risk, particularly in rectal cancer compared to that of AA carriers with low lutein/zeaxanthin intakes.

The results strongly suggested that carotenoids major compounds lutein/zeaxanthin has a strong implication on the colorectal cancer risk in patients with DICER1 rs3742330 genotype.


Dr. Kim J, the lead scientist after taking into account co and confounders wrote in the final report, "Colorectal cancer risk was related to an interactive effect between dietary lutein/zeaxanthin intake and the DICER1 rs3742330 polymorphism".

Taken together, carotenoids may be considered a remedy for the prevention and treatment of colorectal cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of carotenoids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
