#Protein Stem Bromelain (SBM) Inhibits Genes and Proteins Associated with the Onset of Obesity and #ObeseComplications, Researchers Find

Obesity is a chronic medical condition associated with excessive fat accumulation over time.
According to the BMI index, a person is considered overweight, if he/she has a BMI between 25 to 29.9 kg/m2. A BMI of 30 kg/m2 or more is an indication of obesity.

In other words, obesity is a progressive chronic disease characterized by abnormal fat accumulation in the body tissues that may impair health.


Similar to other chronic medical conditions including diabetes, cardiovascular disease, or high blood pressure, obesity has recently become a serious health problem in the US as nearly 35% of Americans are obese accompanied by a similar percentage of overweight adults.


Epidemiologically, obesity does not only have serious effects on physical and psychological health but also has a strongly negative impact on the patient's quality of life.

Today, the widespread obesity in the U.S. population has caused a significant economic impact on the U.S. healthcare system, including direct and indirect costs.


The Milken Institute wrote, "The burden of obesity, and the chronic diseases for which it is a contributing factor, has reached record economic heights. In 2016, chronic diseases driven by the risk factor of obesity and overweight accounted for $480.7 billion in direct healthcare costs in the U.S., with an additional $1.24 trillion in indirect costs due to lost economic productivity. The total cost of chronic diseases due to obesity and overweight was $1.72 trillion — equivalent to 9.3 percent of the U.S. gross domestic product (GDP). Obesity as a risk factor is by far the greatest contributor to the burden of chronic diseases in the U.S., accounting for 47.1 percent of the total cost of chronic diseases nationwide".


Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

In the concern of the rise of obesity incidence in the world, researchers examined the phytotherapeutic protein stem bromelain (SBM) anti-obesity activity.

At the cellular levels, SBM irreversibly inhibited 3T3-L1 adipocyte differentiation in the expression of the obese gene product (leptin) in white adipose and induced apoptosis and lipolysis in mature adipocytes.

At the molecular level, SBM suppressed adipogenesis by inhibiting gene expression of PPARγ in developing obesity.

Furthermore, SBM also reduced the secretion of adipocyte fatty acid-binding protein (ap2) associated with obese glucose and lipid metabolism, and fatty acid synthase (FAS) involved in food intake and body weight.

Lipoprotein lipase (LPL) which leads to increased lipid partitioning to other tissues, insulin resistance, and obesity, and gene CD36 CD36 associated with a number of metabolic pathways with relevance to obesity and enzyme acetyl-CoA carboxylase (ACC) involved in the biosynthesis and metabolism of fatty acids also inhibited by the injection of SBM.

Moreover, SBM was found to augment the expression of TNFα(tumor necrosis factor-alpha) -induced lipolysis and apoptosis in mature adipocytes, another indication of anti-obesity properties.

Where TNFα(tumor necrosis factor-alpha) has been proposed as a link between obesity and insulin resistance.

The results strongly supported the use of SBM for the prevention of obesity in an animal model.


Additionally, according to a study led by the University of British Columbia, bromelain from papaya and pineapple may be used to control obesity and obesity complications such as hypertension, and diabetes.


Taken together, bromelain used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of neurodegeneration, pending the confirmation of the larger sample size and multicenter human study.


Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

This process has been found to involve several stages after the bodily injury caused by physical impact or internal damage.

In the acute phase of infection, the white blood cells of the first line of immune defense stimulate the production of blood coagulation to cover the wound and activate the protein associated with the production of inflammatory cytokines to kill off the invaders before they can enter the body.

In RAW 264.7 cells, based on the tested analysis, administration of TGN enhanced the HO-1 expression associated with reducing inflammatory stress, oxidative stress, and ER stress and nuclear factor-E2-related factor 2 (Nrf2), associated with the antioxidant defense in a time- and dose-dependent manner.

Furthermore, TGN also suppressed the proteins associated with the production of proinflammatory cytokines such as nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) by reducing the mRNA expression of proinflammatory cytokines.


In vivo, TGN also reduced iNOS and COX-2 expression in a mouse model of skin inflammation induced by the injection of 12-O-tetradecanoylphorbol-13-acetate (TPA).

Based on the findings, researchers said, "These findings suggest an important role for TGN-induced HO-1 activation in regulating inflammatory responses.".

Taken together, coltsfoot may be considered a remedy for the improvement of skin conditions caused by inflammation, pending the confirmation of the larger sample size and multicenter human study.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

In finding a natural ingredient for the treatment of Crohn's disease (CD), researchers at the Grand Valley State University compared the effects of Remicade to curcumin for the treatment of the condition.

The review study includes a search in electronic databases (ProQuest Medical Library, CINAHL Complete, and PUBMED) to locate and identify articles to meet a predetermined set of inclusion criteria, including articles published after 2007 in human participants 18 years or older.


Compared to Remicade in reducing TNF-a, the protein associated with inflammation in adults with CD, curcumin induced a 55 points mean reduction in the Crohn's Disease Activity Index, reduced the expression of IL-1 and levels of C-reactive protein (CRP), associated with elevated levels of proinflammatory cytokines in the colon, particularly in the reduced levels of TNF-a and PPMTase involved in the improvement of colorectal cancer outcomes.

Based on the findings, researchers said, "...Preliminary data would suggest using both would reduce LOR. Curcumin, even by itself, was found to be a cheap and safe way to reduce CD symptoms and inflammatory markers"


Furthermore, according to the study conducted by the Medical School of Marília in the search for articles associating curcuminoids and CD and UC on the database of MEDLINE-PubMed, curcumin demonstrated an intestinal protective effect on reducing oxidative stress and inhibiting the migration of neutrophils and inducible nitric oxide synthase in the intestine.

#Phytochemical #Sinigrin Inhibits Proteins Associated with Inflammation and #ImmuneResponse, Researchers Say

Sinigrin may be used as a dietary supplement for the improvement of the antioxidant profile in patients with overexpression of reactive oxygen species (ROS), according to studies.

ROS, a phrase used to describe a number of reactive molecules. Free radicals are unstable molecules with unpaired electrons on the outer ring. In order for them to become stable, free radicals are ready to donate or intercept an electron from other stable atoms, leading to a chain reaction that can not be stopped until all electrons are paired or inhibited by antioxidants.

Reactive Oxygen Species (ROS) have long been known to be a component that impairs the function of the response of immune cells to microbial invasion and induces cytotoxicity in healthy cells, leading to the damage of lipids, proteins, and alternation of cell DNA.

In other words, overexpression of ROS can cause tissue and DNA damage, causing chronic diseases such as cardiovascular and inflammatory disease, cataracts, and cancer.


Free radicals can be produced by either cell metabolism or contact with outside sources, including the food and air we breathe.

In healthy individuals, free radicals are neutralized by the antioxidant enzymes produced by the body's defense system. People with low levels of antioxidant enzymes can acquire antioxidants from dietary sources found in fruits and vegetables.

In fact, free radicals play a dual role as both toxic and beneficial compounds, since they can be either harmful or helpful to the body. Free radicals can exert beneficial effects on cellular responses and immune function at low levels. However, at high concentrations, free radicals generate oxidative stress, a deleterious process that can damage all cell structures.


Antioxidants are stable molecules that inhibit free radicals.

Sinigrin is a phytochemical glucosinolate, that belongs to the family of glucosides found abundantly in Brussels sprouts, broccoli, the seeds of black mustard, etc.

Scientists on finding a natural compound that processes a strong antioxidant activity against oxidative stress examined by the effect of sinigrin anti-free radicals activity.

Sinigrin enhanced the immune macrophage function, by enhancing the activity of Nod-like receptor protein 3 (NLRP3) against cell stress.

The chemical compound also inhibited the expression of free radicals on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in a concentration-dependent manner.

Sinigrin also exhibited anti-inflammatory activity against the expression of COX-2 and prostaglandin E2 (PGE2) in RAW 264.7 cells and proinflammatory cytokines such as tumor necrosis factor (TNF)-α and interleukin (IL)-6.

Treatment with sinigrin decreased pro-inflammatory expression (IL-1β and IL-18) and concurrently suppressed NLRP3, the protein associated with innate immune response and inflammation.

In other words, by blocking the expression of NLRP3, sinigrin inhibited the pro-inflammatory expression (IL-1β and IL-18).


Furthermore, the efficacy of sinigrin immunomodulatory activity was attributed to the decreasing activation of the signaling in the activation of pro-inflammatory pathways such as the NF-κB/MAPK pathways or NLRP3 inflammasome.

The findings strongly suggested that sinigrin has immunomodulatory effects by suppressing the production of inflammatory mediators.

Taken altogether, sinigrin may be considered an antioxidant supplement for modulatory activity against oxidative stress in the induction of inflammation.

However, the intake of sinigrin should be taken with extreme care to prevent acute liver toxicity.

Garlic Powder Tablets are More Effects in Treating Hypertension Compared to Regular Garlic Supplements, Researchers Say

Scientists may have found a bioactive ingredient that can normalize blood pressure without inducing any side effects, according to studies.
Allicin is a phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in onion and garlic.

Blood pressure is the force of blood pushing against the walls of the arteries as the heart pumps out blood.

High blood pressure is a condition of abnormally high blood pressure, leading to rising pressure in your heart.

If it stays high over time it can cause hardening and stiffening arterial walls,, a major risk factor for heart diseases.

The ideal blood pressure is more than 120 over 80 and less than 140 over 90 (120/80-140/90), according to the consensus.

Abnormal high blood pressure, depending on how high the numbers, patients may experience symptoms of a headache to severe headache., fatigue, and vision problems. However, in severe cases, high blood pressure can also induce chest pain, difficulty breathing, irregular heartbeat, and confusion.

Untreated high blood pressure, epidemiologically can cause complications of retinopathy, lead to damage to the retina or bleeding in the eye, blurred vision, and complete loss of vision.


Blood pressure medications have undoubtedly prevented many deaths from heart disease in the past 30 years, but they have many side effects such as damaging the kidneys.

If you can control your diet, such as making changes to your lifestyle, maintaining an ideal weight, and dieting with low salt, you could do just fine without the drugs.

The hypertension clinical guideline from the National Institute for Health and Clinical Excellence (NICE) usefully recommends regular aerobic exercise and reduction of salt, alcohol, and smoking, and advocates ‘healthy, low-calorie diets' for ‘overweight individuals with raised blood pressure'".

This suggestion once again, highlighted that high blood pressure can be prevented by a change to a healthy lifestyle.

In finding a natural ingredient for the prevention and treatment of high blood pressure with no side effects, scientists at the Clinical Research Center looked into the antihypertensive effect of a popular garlic preparation containing 1.3% allicin at a large dose (2400 mg).

Nine patients with rather severe hypertension (diastolic blood pressure > or = 115 mm Hg) were included in the study.

Application of the preparation reduced the sitting blood pressure by 7/16 (+/- 3/2 SD) mm Hg at peak effect approximately 5 hours after the dose, with a significant decrease in diastolic blood pressure (p < 0.05) from 5-14 hours after the dose.
No significant side effects were reported. The findings strongly indicate that this garlic preparation can reduce blood pressure.

Furthermore, in order to compare the antihypertensive effect of the different fractions of garlic, researchers investigated the hypotensive action of time-released garlic powder tablets (Allicor) vs. regular garlic pills (Kwai).


84 men with mild or moderate arterial hypertension participated in the study.

After an 8-week placebo treatment run-in phase, patients were randomized either to 600 mg Allicor (n=30) or to placebo (n=20) daily for 8 weeks. In addition, in the open-label branch, patients received either 2400 mg Allicor daily (n=18) or 900 mg Kwai daily (n=16).


Allicor treatment (600 mg daily) resulted in a reduction of both systolic and diastolic blood pressures by 7.0 mm Hg (95% confidence interval (95% CI): 5.3-8.7) and 3.8 mm Hg (95% CI: 2.7-4.8), respectively. Increasing the Allicor dosage to 2400 mg daily did not provide any additional benefit.
Treatment with Kwai resulted in the same decrease in systolic blood pressure (5.4 mm Hg, 95% CI: 1.9-8.8) as that seen with Allicor, but no decrease in diastolic blood pressure.

In the depth analysis, researchers found that different effects of Allicor and Kwai on diastolic blood pressure may be associated with the prolonged action of Allicor.


Dr. Sobenin IA, wrote in the final report, "The results of this study show that time-released garlic powder tablets are more effective for the treatment of mild and moderate arterial hypertension than are regular garlic supplements".

Taken together,. allicin, a bioactive compound of garlic may be used for the prevention and treatment of high blood pressure with no side effects

#Coffee Caffeine Intake During Pregnancy Does Not Associate with Poor Child Cognition and Behavior, Researchers Suggest

Epidemiological studies do not agree that maternal Caffeine Intake during pregnancy may have a negative impact on child cognition and behavior, a new study suggested.

Coffee, second to green tea is a popular and social beverage all over the world, particularly in the West, made from roast beans from the Coffea plant, native to tropical Africa and Madagascar.

Cognition and Behavior is a group of neuron functions involving sensation and perception, learning and memory, attention, mental imagery, conceptual presentation, language expression, emotion response, motor control,.....

In the study of examining maternal serum paraxanthine at <20 and ≥26 weeks gestation with the child's intelligence quotient (IQ) and problem behaviors at ages 4 and 7 years among 2,197 mother-child pairs, researchers showed that paraxanthine, a structurally related to caffeine.and caffeine's primary metabolite concentration has a significantly inverted effect at ≥26 weeks gestation with child's IQ at age 7 years.

At <20 weeks, paraxanthine also expressed a linearly associated with internalizing behavior at age 4 years.

Interestingly, after taking into account a range of values applicable to most pregnant women, researchers indicated that coffee caffeine in contribution to serum paraxanthine concentration showed no significant effect in association with childhood IQ or problem behaviors.

In contrast to the healthy mothers, Women's reported coffee intake per day during pregnancy was associated with lower child's Behavior Rating Scale (BRS), a complete overview of child and adolescent concerns and disorders, ratings in anemic children. Even with the inclusion of other contributing factors and after anemic children were given appropriate treatment, coffee consumption continuously showed Poor Child Cognition and Behavior.

Dr.Engle PL, the lead author said, " The effects of postnatal coffee ingestion in Guatemala were seen for sleep duration, but not for cognitive development. Prenatal coffee ingestion was negatively associated with Behavior Rating Scales".

Taking together, coffee caffeine intake during pregnancy may not have any meaningful effect in association with childhood cognitive development and measurement of Behavior Rating Scales, excluding anemic children,

#AITC-rich Mustard Seed Powder (MSP-1) Processes Anti #BladderCancer Activity, Scientists Reveal

Phytochemical Sinigrin may be used as a natural ingredient for the treatment of bladder cancer, according to studies.

Bladder cancer is a medical condition characterized by cell growth uncontrollably in the bladder tissue, due to the alternation of DNA.

Most cases of bladder cancer start in the cells of the inner lining of the bladder tissue. At the early, due to the small size and on the surface of the inner lining, most patients do not experience any symptoms.

However, at the advanced stage, the larger size tumor may press on the nearby blood vessels and nerve cells, leading to the symptoms of a burning sensation, blood in the urine, abnormal urinary activity, particularly, severe abdominal pain and back pain on one side of the body.

Compared to the RR ratio, obesity, and cigarette smoking shared an extremely close ratio of 1.09 and 1.10 respectively. In other words, if you are obese, your risk of bladder cancer is 0.01 higher compared to smokers.

Sinigrin is a phytochemical glucosinolate, that belongs to the family of glucosides found abundantly in Brussels sprouts, broccoli, the seeds of black mustard, etc.

Researchers on finding a bioactive compound for the treatment of bladder cancer examined the effects of AITC-rich mustard seed powder (MSP-1) with AITC stably stored as its glucosinolate precursor (sinigrin) in MSP-1.


In a rat model, hydrated MSP-1 caused apoptosis and G(2)/M phase arrest in bladder cancer cell lines in vitro.

In the comparison of cancer activity between hydrated MSP-1 and pure sinigrin with added myrosinase, researchers surprisingly found that the anticancer effect of MSP-1 was derived principally, if not entirely, from the AITC generated from sinigrin.


In an orthotopic rat bladder cancer model, oral MSP-1 at 71.5 mg/kg (sinigrin dose of 9 μmol/kg) inhibited bladder cancer growth by 34.5% (P < 0.05) and blocked muscle invasion by 100%.

The in-depth analysis suggested that the anti-bladder cancer efficacy of MSP-1 was attributed to significant modulation of key cancer therapeutic targets, including blocking the vascular endothelial growth factor in the generation of the new blood vessels to promote cancer growth, a cyclin-associated cancer cell division and caspase correlated with significantly increased rate of cancer recurrence and deaths.

Furthermore, the anticancer activity of AITC delivered as MSP-1 appears to be more robust compared to that of pure AITC.
The finding strongly suggested that MSP-1 exhibited the AITC in the inhibition of bladder cancer development and progression.

Moreover, in the investigation of the effect of various dose levels of AITC on the activities of the phase II detoxification enzymes quinone reductase (QR) and glutathione S-transferase (GST) in rat tissues, researchers conducted an experiment with the selected rat given high and low dose level of AITC daily for 5 days.

At low doses, AITC increased the production of antioxidant enzyme activity observed only in the urinary bladder of the rats, at a dose of AITC of only 10 micromol/kg/day.


In the activities of phase II, detoxification enzyme quinone reductase (QR), a plateau of activity of rats given the injection AITC for 21 days was reached after 15 daily doses of AITC, but GST activity continued to increase with continued exposure, and no plateau was reached after 21 doses.

Further examination of the levels of enzymes in the treated rats, researchers surprisingly found that the dose level of AITC is effective in rats approaches the level that could only be achieved through human consumption of Brassica.

The results strongly support the use of dietary approaches for the prevention and treatment of bladder cancer. Dr. the lead scientist said, "Induction of phase II enzymes by food-derived isothiocyanates could contribute to the lower incidence of bladder cancer observed in individuals who regularly consume such vegetables".

Taken altogether, sinigrin, a bioactive ingredient found in the Brassica may be considered a supplement for the prevention and treatment of bladder cancer, pending the confirmation of a larger sample size and multicenter human study.
