#Herbal Turmeric Extract Inhibits Human Papillomavirus-Positive and Negative HPV+, HPV- Cells, Through Its Cell Cycle Arrest, Researchers Find

Kyle J. Norton

Cancer is a group of chronic diseases associated with cell growth disorderly in the tissue of some specific organ. Depending on the grade and stage, most cases of cancer in the advanced stage have been found to induce a higher mortality rate compared to cancer diagnosed in the localized stage.
Head and neck cancer is a group of cancers that starts in the cell on the surface of the inner lining of the mouth, nose, throat, larynx, sinuses, or salivary glands.

Squamous cell carcinoma is cancer arising from the squamous cells found in the outer layer of skin and in the mucous membranes, which are the moist tissues that line body cavities.

Head and neck squamous cell carcinoma (HNSCC) develops in the mucous membranes of the mouth, nose, and throat.

Head and neck squamous cell carcinoma (HNSCC) epidemiologically has been found to spread to the lymph nodes or lungs. More than 90% of the head and neck cancer diagnosed are squamous carcinomas.

Symptoms of head and neck cancer may include a lump or sore that does not heal, a sore throat that does not go away, trouble swallowing, or a change in the voice.

According to the statistics provided by the Western General Hospital, squamous cell cancer of the head and neck is common worldwide accounting for 4% of all cancers in the United States and 5% in the United Kingdom.

The exact causes of head and neck squamous cell carcinoma (HNSCC) are unknown. However, geographically, smoking tobacco, drinking alcohol, and long term poor diet is important to risk factors in the West compared to risk factors of chewing betel or areca nuts, smoking bidis, and taking snuff are important in the Indian subcontinent.

More importantly, the genetic preposition of genome 9p21, which may indicate the loss of a gene that suppresses tumors accounts for a total of 70% of all cases.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a potential ingredient for the treatment of head and neck cancer, researchers examined the effect of curcumin on head and neck squamous cell carcinoma (HNSCC) using a panel of cell lines with and without human papillomavirus (HPV+, HPV-).

According to the tested differentiation, curcumin treatment of HNSCC cell lines not only interfered with RNA of Skp2, which causes cancer tumorigenesis and radiation tolerance but also induced concomitant accumulation of p21 and p27 associated with a cell cycle arrest in HPV+, HPV- cells.

Furthermore, curcumin inhibits cell viability and induces apoptosis in a dose-dependent manner in the tested cell line with and without human papillomavirus (HPV+, HPV-).

Moreover, curcumin treatment of HPV+ and HPV- cells resulted in apoptosis via the mitochondrial pathway of apoptosis and activation of caspases that play a critical role in programmed cell death.

Additionally, the inhibition of HPV+ and HPV- cells by curcumin was also found to suppress the expression of poor prognostic markers such as XIAP, and cancer survival such as cIAP1, and cIAP2.

More importantly, co-treatment of HNSCC cells with curcumin and cisplatin potentiated inhibition of cell viability and apoptotic effects.

Based on the findings, researchers wrote in the final report, "These data suggest an important function for curcumin, acting as a suppressor of oncoprotein Skp2 in squamous cell carcinoma cells in both HPV+ and HPV- cells".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of HNSCC, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Green Peas Extract and Its Phytochemical Lectin Exert Potential Chemopreventive Activity, Researchers Say

Kyle J. Norton

Cancer is a group of cell-mutated diseases caused by the alternation of DNA.
Most cases of cancer begin in the cells on the surface of the inner lining of the tissue of an organ. At early, most people with cancer are asymptomatic.

At the later stage, due to the size and the tumor, patients may experience general cancer symptoms such as unintended weight loss, fatigue, gastrointestinal discomfort, and loss of appetite, and localized symptoms such as severe pain and bleeding.

The exact causes of cancer are not identified. However, the most prevalent factors associated with the cancer onset include age, family history, long-term exposure to certain cancer-induced chemicals, use of tobacco and excessive alcohol drinking,..... the list goes on and on, depending on the type of cancer.

Some researchers suggested that the most common risk factor that causes cancer in the Western world may be associated with widespread obesity.

Dr. Karen Basen-Engquist, in the evaluation of the obesity risk of cancer, said, "In the United States, approximately 85,000 new cancer cases per year are related to obesity. Recent research has found that as the body mass index increases by 5 kg/m2, cancer mortality increases by 10%".

And, "High priorities for future research involve additional work on the underlying mechanisms, and trials to examine the effect of lifestyle behavior change and weight loss interventions on cancer and intermediate biomarkers".

The findings strongly indicated the risk of cancer is increased substantially in obese.

Green peas are a species of Pisum Sativum belonging to the family Fabaceae and native to western Asia from the Mediterranean Sea to the Himalayan Mountains. The pea is a green, pod-shaped vegetable and a cool-season crop grown in many parts of the world.

In finding a potent ingredient for the treatment of cancer, researchers investigated the impact of green pea (GP) extracts anti-DNA alternative activity.

According to the studies,
* The extracts of P. sativum have been found to be pharmacologically active inducing anticancer effects.

* Compared to the effect of Bowman-Birk trypsin-chymotrypsin inhibitor, a potential cancer chemopreventive agent, with the protease inhibitors, rTl1B and rTl2B, P. sativum seed showed similar significant activity.

* In human colorectal adenocarcinoma HT-29, P. sativum extracted lectins from Egypt exhibited antiproliferative properties to liver cancer cell line.

* In MCF-7 (breast), HepG-2 (liver), HEP-2 (larynx), and HCT-116 (colon). An extracted lectin from the leaves and buds of P. sativum demonstrated cytotoxic properties.

* Recently, the seed extracts also showed cytotoxic effects on LS174 (colon), MDA-MB-453 (breast), A594 (lung), and K562 (blood) cell lines.

Taken together, green peas may be considered a functional food for the prevention of cancer, pending the confirmation of the larger sample size and multicenter human study.

#Cervicalparavertebral Nerve Block Combined with Mailuoning and Angelica Sinensis in the Treatment of Nerve-Root type #CervicalSpondylosis

Kyle J. Norton

Degenerative disease is a progression that causes degenerative cell changes due to aging, affecting tissues or organs, which will increasingly deteriorate over time. ...

Normal bodily wear and tear or lifestyle choices (such as exercise or eating habits) may worsen degenerative diseases, but this depends on the disease.

Epidemiologically, the most common degenerative diseases include
* Alzheimer's disease (AD).
*Amyotrophic lateral sclerosis (ALS, Lou Gehrig's disease).
* Cancers.
* Cystic fibrosis.

Cervical spondylosis is age-related wear and tear that affects the spinal disks in your neck.

In other words, these age-related wear and tear cause the disks to dehydrate and shrink, leading to osteoarthritis development, including bony projections along the edges of bones (bone spurs).

Cervical spondylosis is very common and worsens with age.

The most common symptoms of cervical spondylosis are neck stiffness pain, and headache that may originate in the neck, pain in the shoulder or arms, inability to fully turn the head or bend the neck, sometimes interfering with driving, and grinding noise or sensation when the neck is turned.

On finding a natural ingredient for the prevention and treatment of degenerative diseases, scientists examined the clinical effect of cervical paravertebral nerve block combined with Mailuoning and Angelica sinensis injection in the treatment of nerve root type cervical spondylosis.

A total of 120 patients with nerve-root type cervical spondylosis were randomly divided into a research group (n=60) that was treated by cervical paravertebral nerve block combined with Mailuoning and Angelica sinensis injection compared to the control group which was applied with glucocorticoid nerve block therapy.

According to the results of the comparison, the total therapeutic response rate of the research group was relatively higher than the control.

By comparing various clinical effect scores between the two groups, the research group also had advantages over the control group.

Furthermore, The treatment satisfaction degree of the research group was also higher than that of the control group.

Based on the findings, researchers concluded, " application of cervical paravertebral nerve block combined with Mailuoning and Angelica sinensis injection can achieve better medical results and achieve higher treatment satisfaction degree".

Taken together, angelica used together with other herbal medicines may be considered an adjunct remedy for the treatment of cervical spondylosis, pending the confirmation of the larger sample size and multicenter human study.

#CranberryJuices (CJ) and #CranberryExtracts (CE) Improve Overall #Neuropsychologic Symptoms, Researchers Suggest

Kyle J. Norton

Neuropsychological assessment provides both general and specific information about current levels of cognitive performance. An average or composite score across multiple ability areas provides an overall index of how well a person functions cognitively at the current time.

Cognition is the process of the brain functioning associated with the attention of working memory, producing and comprehending language, learning, reasoning, problem-solving, and decision-making.

Cognitive deficit is a medical condition, that affects a person's thinking, communication, understanding, or memory. The cognitive deficit can be acute or chronic.

Acute cognitive deficit is a short-term health problem caused by drug toxicity and withdrawal, electrolyte disturbance, lack of drugs, liver disease, infection, reduced sensor input, intracranial urinary retention/fecal impaction, or myocardial/metabolic/pulmonary. An acute cognitive deficit may be recovered if the cause is found.

Chronic cognitive deficit is a permanent medical condition caused by the gradual or permanent loss of neurons, leading to problems with language, ideomotor and constructive praxis, visual gnosis, spatial attention, learning abilities, and executive functions.

There is no single cause of the chronic cognitive deficit. An increase in age, genetic preposition, certain medical conditions, such as diabetes, high blood cholesterol, and pressure, being obese, and lack of exercise are some prevalent factors found in patients with chronic cognitive deficits.

Some researchers suggested that depending on the locations of the brain that were affected, patients may be at risk of Alzheimer's disease such as brain change caused by the abnormal clumps of beta-amyloid protein (plaques).

Small strokes or reduced blood flow through the brain can also cause a chronic cognitive deficit.

According to the observation of the patient MRI, the patient's brain with cognitive impairment also showed shrinkage of the hippocampus, a brain region important for memory, enlargement of the brain's fluid-filled spaces (ventricles), and reduced use of glucose, the sugar that's the primary source of energy for cells.

Out of many risk factors associated with the onset of the condition, obesity is also one of the major risk factors for cognitive impairment in the Western world.

Dr. Jason C. D. Nguyen, the lead author on the neurological team wrote in the final report, "The incidence of obesity in middle age is increasing markedly, and in parallel, the prevalence of metabolic disorders including cardiovascular disease and type II diabetes is also rising. Numerous studies have demonstrated that both obesity and metabolic disorders are associated with poorer cognitive performance, cognitive decline, and dementia".

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belonging to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

In finding a potential compound for the treatment of the neurological disorder, researchers examined the cranberry extract (CE) neuropsychologic effect.

The double-blind, placebo-controlled, randomized, parallel-group, clinical trial included 50 community-dwelling, cognitively intact volunteers, > or = 60 years old, who reported no history of dementia or significant neurocognitive impairments.

Participants in the study were randomly assigned to receive either 32 ounces/day of a beverage containing 27% cranberry juice per volume (n = 25) or placebo (n = 25) for 6 weeks.

According to the neuropsychologic tests before and after treatment on participants' self-reported, there was a nonsignificant trend (X2(1) = 2.373, p = 0.123) on overall abilities to remember.

However, more than twice of many participants (37.5%) reported the overall abilities to remember at the end of treatment compared to placebo controls (17.4%).

In other words, cranberry extract (CE) although can not improve overall neuropsychologic symptoms, enhances the short-term memory of dementia or significant neurocognitive impairments.

Taken altogether, cranberry may be considered a remedy for the improvement of memory, pending the confirmation of the larger sample size and multicenter human study.

#Herbal Turmeric's #Curcumin Improves the Function of Hepatic Glucose and Lipid Metabolism in Type 2 #Diabetes, Scientists Say

Kyle J. Norton

Type II diabetes is an acquired chronic and medical condition caused by insufficiency of insulin entering the bloodstream, leading to abnormally high blood glucose.

In other words, type II diabetes is due to the pancreas not producing enough insulin caused by the loss of beta cells or the body not properly using the insulin it makes as a result of insulin receptor sites clogged up by fat or cholesterol.

Although type II diabetes is a disease associated with adult-onset, today, the condition has been diagnosed in children at an alarming rate due to childhood obesity.

The most common symptoms of type II diabetes are increased thirst, frequent urination, unintended weight loss, and fatigue.

In severe cases, patients with type II diabetes also experience symptoms of slow-healing sores, frequent infections, and blurred vision.

Untreated type II diabetes may induce complications of diabetic retinopathy, kidney disease (nephropathy), diabetic neuropathy, and macrovascular problems.

As of today, there is no single cause and cure for type 2 diabetes. However, researchers suggested overweight, family history, aging, race, and medical conditions of prediabetes and gestational diabetes are some prevalent factors associated with the increased risk of the disease.c

Some researchers suggested that type 2 diabetes may be correlated to the promotion of the Western diet over the past few decades.

Dr. David Hefner in the article "Western Diet Linked to Type 2 Diabetes", wrote, "The typical Western diet-red meat, high-fat dairy products, refined grains, and desserts-may increase your chances for Type 2 diabetes".

And, "The news isn't particularly shocking but offers a new view on the health risks of a typical Western diet as a whole, rather than simply attacking elements within the diet such as fatty foods".

In other words, if you are following a high-fat diet, you are at a higher risk of type II diabetes. is

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

In the urgency to find a potential ingredient for the treatment of diabetes, researchers examined the effect of Curcumin (CUR; diferuloylmethane) in the increased oxidative stress and the proinflammatory state that contributes to the development and progression of the disease.

Oxidative stress is an imbalance between the production of reactive oxygen species (ROS) and their clearance by antioxidant defense mechanisms.

Curcumin improved the function of hepatic glucose and lipid metabolism which was dysfunctioned in patients with T2DM.

In vitro, curcumin was found to induce the liver protective effect in the regulation of glucose and lipid metabolism.

Furthermore, curcumin exerted antidiabetic potential by ameliorating skeletal muscle insulin resistance in rodents with diet-induced insulin resistance via an increase in the uptake and oxidation of fatty acids.
Moreover, curcumin was also found to improve insulin sensitivity in muscle tissue of insulin-resistant rodents by increasing cellular glucose uptake by enhancing the GLUT4 translocation.

In adipose tissue analysis, curcumin protected the adipose tissue function, by protecting the normal adipokine production, and FFA release, and preventing inflammation, that is associated with the pathogenesis of T2DM.

In pancreatic β-cells differentiation, curcumin enhanced the pancreatic β-cell function under diabetic conditions.

On metabolic dysfunction, curcumin improved the glycemic status and insulin sensitivity demonstrated in high-fat-diet- (HFD-) fed C57BL/6J mice and C57BL/6J ob/ob mice.

More recently, curcumin exerted a positive effect of CUR on metabolic abnormalities associated with T2DM which has been confirmed in high-fructose-fed Wistar rats by attenuating insulin resistance and glucose intolerance through its antioxidative and anti-inflammatory actions

Taken together, turmeric processed with a high amount of bioactive compound curcumin may be considered a supplement for the prevention and treatment of type II diabetes, pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Methanol Extract of Ashwagandha (MEAG) Gobbles Up Human Head and Neck Cancer Cells Through Its Aopootic Property, Researchers Reveal

Kyle J. Norton

Cancer is a group of diseases associated with cell growth disorderly and uncontrollably in the tissue of a specific organ. Over time cancer can become progressively worse and damage other healthy tissues.
Most cases of cancer start in the cells on the surface of the inner lining tissue of a specific organ, before traveling a distance away to affect other nearby tissues and organs.

Head and neck cancer is a class of cancer including the mouth, nose, throat, larynx, sinuses, or salivary glands.

Most cases of head and neck cancer start in the cells of the inner lining of the affected tissue. Patients with head and neck cancer at the very early stage are asymptomatic due to the very small size of the tumor.

However, at the later stage, the cancer cell can travel a distance away from the original site to infect other healthy tissues and organs, leading to secondary metastasis.

The exact causes of head and neck cancer are not identified. However, researchers found that excessive alcohol intake, smoking, infection by the HPV virus, poor oral health, race, and long-term exposure to wood dust are prevalent factors associated with an increased risk of head and neck cancer.

Dr. Kazuhiro Kobayashi in the investigation of the risk of HPV on head and neck cancer (HNC) wrote, " Infection with high-risk human papillomaviruses (HPVs) has recently been implicated in the pathogenesis of HNSCCs. It is now widely accepted that high-risk HPV is a cause of almost all cervical cancers as well as some forms of HNSCCs. HPV-related HNSCCs are increasing".

Ashwagandha also known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, the Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine for over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered an anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound for the treatment of cancers, researchers examined the apoptotic activity of methanol extract of Ashwagandha (MEAG) in human head and neck squamous cell carcinoma (HNSCC) cells.

In vitro, in human head and neck squamous cell carcinoma (HNSCC) cells treated by MEAG showed dose-dependent growth-inhibitory activity involved in caspase-dependent apoptosis.

The inhibition of human head and neck squamous cell carcinoma (HNSCC) was attributed to induce the loss of mitochondrial membrane, and the release of cytochrome c, accompanied by the activation of caspase 9 the apoptotic pathway.

Furthermore, Bim, truncated Bid (t-Bid), cleaved caspase-8, and death receptor 5 (DR5), associated with pro-apoptotic activity also a significant increase upon the injection of MEAG.

Moreover, the major constituent withaferin A (WA), is a bioactive component of MEAG. also expressed similar effects to those of MEAG.

Based on the findings, researchers said, " MEAG and WA may be potential natural materials for the treatment of HNSCC.".

Taken altogether, Ashwagandha may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of head and neck squamous cell carcinoma (HNSCC), pending to the result of larger sample size and multicenter study

#Berry #nthocyanins-Enriched (ACE) #Extracts Inhibit ROS Associated with the Expression of Advanced Glycation (AD) Expression

Kyle J. Norton

Advanced glycation end is a by produces formed by protein or fat combined with sugar caused by cooking meat at high temperatures.
The products have been found to affect metabolic health.

The most common causes of AGEs are from the diet, particularly in people who follow the Western diet with high saturated and trans fats, a lot of red meats, and processed foods.

Low levels of AGEs may not be a problem, however, overexpression of AGEs can induce oxidative stress, leading to inflammation and damage proteins, lipids, and sometimes alternation of cells DNA.

In other words, AGEs initiates the production and free radicals and free radical a chain reaction that can not be stopped until all electrons in the outer ring paired or inhibited by antioxidants.

Accelerated formation and accumulation of advanced glycation end-products have been found to increased hyperglycemia or oxidative stress, leading to age-related and chronic diseases such as diabetes mellitus, chronic renal failure, neurodegenerative diseases, osteoarthritis, and also non-diabetic atherosclerosis, and chronic heart failure.

Some researchers suggested that modern life byproducts of AGEs are the results of the promotion of the processed food diet.

Dr. Chetan Sharma, and Amarjeet Kaur, the lead scientist in the assessment of AGEs, wrote, "As modern diets are largely heat-processed, they are more prone to contain high levels of advanced glycation end products (AGEs)".

And, "The chemistry of the AGEs formation, absorption and bioavailability and their path-biochemistry particularly in relation to different complications like diabetes and aging,... The emerging evidence about the adverse effects of AGEs makes it necessary to investigate the different therapies to inhibit AGEs".

The results strongly suggested the link of the modern diet with highly processed food is a leading cause of chronic diseases.

In finding a potential compound for the treatment of oxidative stress, researchers examined anti-glycation effects of several polyphenol-rich fruits, including blackberry, black raspberry, blueberry, cranberry, red raspberry, and strawberry extracts, which are rich in anthocyanins.

Berry crude extracts (CE) yielded anthocyanins-enriched (ACE) extracts used in the study, showed a significant effect against oxidative stress by exhibiting the free radical scavenging, reactive carbonyl species trapping, and anti-glycation effects compared to their respective ACFs.

Furthermore, the berry ACEs (at 100 μg/mL) inhibited both thermal- and MGO-induced Aβ fibrillation induced by the overexpression of glycation.

Moreover, the berry ACEs (at 20 μg/mL) also reduced H₂O₂-induced reactive oxygen species production and lipopolysaccharide-induced nitric oxide species in BV-2 microglia by decreasing H₂O₂-induced neuron cytotoxicity and caspase-3/7 activity in BV-2 microglia involved in inflammation.

Based on the findings, researchers said, "The free radical scavenging, reactive carbonyl trapping, anti-glycation, anti-Aβ fibrillation, and microglial neuroprotective effects of these berry extracts warrant further in vivo studies to evaluate their potential neuroprotective effects against AD".

Taken together, cranberry may be considered a remedy for the treatment of AD, pending the confirmation of the larger sample size and multicenter human study.
