#BetaSitosterol and #Stigmasterol Exert Cytototic and Inhibited Effects on #BreastCancer Cells and #Menopausal Symptoms Accordingly

Kyle J. Norton

Estrogen is a primary female sex hormone, responsible for the development and regulation of the female reproductive system and secondary sex characteristics. The estrogenic effect is an action that stimulates the production of the hormone estrogen in case of deficiency.

On the other hand, the anti-estrogenic effect is an action of binding the estrogen receptor(ERα ) and/ERβ in inhibiting the excessive levels of estrogen circulating in the body.

Menopause is a condition in which a woman is in the transition stage of permanent cessation of the ovary's functions in egg production because of less production of estrogen and progesterone, signaling the end of the reproductive phase of a woman's life.

In menopause, menstruation has become irregular and slowly stops over time, but in some women, menstrual flow comes to a sudden halt.

Some researchers suggested that you officially enter the stage of menopause if you have gone 12 months without a menstrual period.

Menopause symptoms are a set of symptoms caused by the gradual decline of levels of estrogen and progesterone due to aging loss of menstrual periods in women.

The reproductive tract, the urinary tract, the heart and blood vessels, bones, breasts, skin, hair, mucous membranes, and pelvic muscles normally regulated by the hormone estrogen also are affected by the declining levels of the hormone.

In the concern of hormone therapy used for the treatment of menopause symptoms, herbal medicine with estrogenic effect have been under intensive investigation, with an aim to find a natural ingredient with similar effects as those used in hormone therapy with no side effects.

The aim of estrogenic therapy is to relieve symptoms of sleep interruption, according to the study of health-related quality of life, fatigue, and hormone-related symptoms in breast cancer survivors, and menopausal symptoms, primarily hot flashes, affect approximately two-thirds of menopausal and postmenopausal women, botanicals and dietary supplement (BDS).

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belonging to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

In finding a potential phytochemical for the prevention and treatment of menopausal symptoms, researchers elucidated the effects of β-sitosterol and stigmasterol in the hydroalcoholic extract of Bambusa bambos as an alternative Hormone Replacement Therapy.

According to the results of the experiment
* The extract containing β-sitosterol and stigmasterol showed an estrogenic effect on the growth of MCF-7 cells in vitro.

* The tested extract showed no cytotoxicity to MCF-7 cells in any of the dilutions, but induced cell proliferation, and all the dilutions except <500μg/ml have significantly increased cell multiplication.

* Furthermore, the doses of 15.6, 31.2, 62.5, and 125μg/ml of HEBB have shown an influence on the proliferation rates similar to the standard 17β-estradiol.

Based on the findings, researchers said, "The results suggest that HEBB might be used as a safe alternative to estrogen replacement therapies".

Beta-sitosterol may be considered a natural estrogenic remedy for the prevention and treatment of menopausal symptoms, pending the confirmation of the larger sample size and multicenter human study.

Intake of beta-sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Black beans Induce Cell Death Programming in #Osteocarcinoma Cells through its Cytototic Activity, in Vitro

Kyle J. Norton

Intake of black beans may reduce risk and delay the progression and treatment of osteosarcoma, an invasive form of bone cancer.

Osteosarcoma is a condition of bone cancer caused by irregular cell growth in bone, affecting mostly adolescents and young adults.

Black beans, small roughly ovoid legumes with glossy black shells, genus Phaseolus, belongings to the family Fabaceae can be bought in most grocery stores all around the year in dried and canned forms. It is believed that the black bean was the first domesticated growth in South America.

According to the Chinese University of Hong Kong, hemagglutinin isolated from an edible legume, namely black bean, showed an anti-proliferative activity against tumor cell lines MG-63 and HepG2 but not the normal cell line.

Malignant osteosarcoma cells treated with the phytochemical also exerted a significant effect in the shrinking of tumor cells as well as inducing apoptosis.

The study also stated that hemagglutinin also induced cell death by interfering with the first stage of tumor cell division in the expression of protein, RNA, and DNA and inducing mitochondrial membrane depolarization in controlling the plasma membrane of a muscle or nerve cell

Furthermore, the study also suggested that the efficacy of hemagglutinin in causing cell death may be attributed to the function of protein in increased regulation of pro-inflammatory expressions.
Dr. Dan X, the lead author said, "A novel hemagglutinin from an edible legume (showed) a growth-inhibitory effect on osteosarcoma and liver cancer cells" and " The hemagglutinating activity of this hemagglutinin was demonstrated to be ion-dependent and stable over a wide range of temperature and pH values".

The findings suggested that black beans may be considered a functional food in improved bone health as well as reducing risk and treatment of osteosarcoma, but further studies are necessary to confirm its potency.

#Phytochemical 3,3'-Diindolylmethane Inhibits #Overabundance of Pro-Inflammatory Cytokines and Proteins Associated with a Reduced Risk of #CardiovascularDisease (CVD)

Kyle J. Norton

Heart muscle or myocardium is one of three types of muscles in humans, that build the walls of the heart.
The most common diseases of heart muscle include angina pectoris myocardial infarction, and other heart muscle diseases caused by restricting blood supply to the muscle.

The most common symptoms of heart muscle diseases include tightness, pain, or swelling of the legs, ankles, and feet

Some patients may also experience symptoms of gastrointestinal discomfort including bloating, abdominal pain, and shortness of breath caused by the reduced blood supply.

Most common risk factors associated with heart muscle disease including high blood pressure, heart tissue damage, heart problems, metabolic syndrome, nutritional deficiencies, excessive alcohol drinking.

Some researchers suggested that genetic preposition may have a strong effect on the induction of heart muscle disease.

The Genetics Home Reference wrote, " mutations in more than 30 genes have been found to cause familial dilated cardiomyopathy. These genes provide instructions for making proteins that are found in cardiac muscle cells called cardiomyocytes.

And, "Mutations in one gene, TTN, account for approximately 20 percent of cases of familial dilated cardiomyopathy".

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

In finding a natural ingredient that protects the heart muscle against the onset of cardiovascular disease researchers examined the 3, 3'-Diindolylmethane (DIM) effects on cardiomyocytes.

Tested cell lines H9C2 used in the study were evaluated by DIM's influence on inflammation and apoptosis of cardiomyocytes induced by LPS. H9C2 cells incubated with DIM (10, 20 and 30 μM) with or without LPS for 24 h.

Treatment with various concentrations of DIM (10, 20 and 30 μM) inhibited the overabundance of pro-inflammatory cytokines (TNF-α and IL-6 mRNA expression) induced by LPS.

In other words, DIM normalized the levels of impaired TNF-α and IL-6 mRNA expression, in the LPS + DIM30(μM) group.

According to the tested assays, DIM treatment significantly blocked LPS-induced oxidative stress and inhibited LPS-induced apoptosis in H9C2 cardiomyocytes.

Furthermore, DIM significantly inhibited the LPS-induced expression of a protein associated with proapoptosis and enhanced the expression of a protein involved in anti-apoptosis.

Dr. Luo Q, the lead scientist, wrote on the final report, "DIM may have a protective effect for H9C2 cardiomyocytes against LPS-induced inflammatory response and apoptosis. DIM may be a new insight into the treatment of septic cardiomyopathy".

In order to find out more information about DIM anti-cardiovascular disease activity, researchers evaluated the function of DIM on the hypoxia‑induced cardiac injury.

The study included H9c2 cells pretreated with DIM (1, 5, and 10 µM) for 12 h and exposed to hypoxia for 12 h.

DIM with different doses extended markedly attenuated the increased transcription of pro-inflammatory cytokines (interleukin (IL)‑1β, IL‑6 and tumor necrosis factor‑α) induced by hypoxia.

DIM also modulated the genes associated with transcription of autophagy, compared with the hypoxia group.

DIM also protected the cells against apoptosis by inhibiting and exhibiting the proteins associated with anti-apoptosis and pro-apoptosis, respectively.

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the protection of heart muscle against the risk of heart disease, pending to the confirmation of the larger sample size and multicenter human study.

#CruciferousVegetables Decrease the Damage of the Brain in Post Ischemic Stroke Patients, Researchers Say

Kyle J. Norton

Stroke is a medical condition caused by reduced blood flow to the brain resulting in cell death due to a blocked artery (ischemic stroke) or the leaking or bursting of a blood vessel (hemorrhagic stroke).
In other words, stroke can be caused by either ischemia, due to lack of blood flow, or hemorrhagic, due to bleeding.

A stroke is a medical emergency. Early intervention in most cases can minimize brain damage and potential complications.

Sadly, stroke is one of the most common causes of death worldwide.

In the US, according to statistics, approximately 800,000 people suffer a stroke of which 600,000 of these are first attacks, and 185,000 are recurrent attacks. Nearly three-quarters of all strokes occur in people over the age of 65.

There is a general belief that stroke is an accumulation over a long period of an unhealthy diet with red meat and processed meats in the induction of artery-clogging.

Atherosclerosis associated with the hardening of the arteries caused by the buildup of fatty substances in the arteries can lead to stroke.

In other words, if the blood vessel that feeds the brain is blocked, a stroke occurs.

Some researchers suggested that increased intake of fruits and vegetables such as carrots accompanied with good fats such as salmon, tuna, and mackerel may have a substantial effect in reduced inflammation in the arteries, improved blood flow, and decreased the chance of blood clots.

Not all strokes can lead to permanent brain damage. Some people with a temporary disruption of blood flow to the brain do not result in permanent damage.

Besides aging, smoking, medical conditions such as unmanaged high blood pressure and cholesterol and diabetes, obesity, and physical inactivity are some of the prevalent factors found in stroke patients.

Cruciferous vegetables are a group of vegetables, belongings to the family Brassicaceae (also called Cruciferae) with many genera, and species, and cultivated all over the world in suitable climates for commercial profits, including cauliflower, cabbage, kale, garden cress, bok choy, broccoli, Brussels sprouts, and green leafy vegetables.

In finding a potential remedy for reducing the mortality rate of cardiovascular diseases, researchers examined the therapeutic potential and protective effect of D3T found in cruciferous vegetables for the treatment of ischemic stroke.

The study included selected mice subjected to transient middle cerebral artery occlusion/reperfusion (tMCAO/R) who were administered with vehicle or D3T to evaluate the effect of D3T in cerebral brain injury.

Based on the results of the experiment,
* D3T reduced infarct size, decreased brain edema, lessened blood-brain barrier disruption, and ameliorated neurological deficits.

* D3T suppressed microglia (MG) activation induced by stroke and inhibited peripheral inflammatory immune cell infiltration of CNS in the ischemic brain.

* D3T also exerted a protective effect on ischemic stroke against the induction of oxidative stress by mediating the improved expression of the antioxidant enzyme Nrf2.

Based on the results, researchers said, "D3T confers protection against ischemic stroke, which is mediated through suppression of MG activation and inhibition of CNS peripheral cell infiltration, and that the protective effect of D3T in ischemic stroke is dependent on the activation of Nrf2".

Taken altogether, cruciferous vegetables may be considered functional remedies for protection in ischemic strokes, pending the confirmation of the larger sample size and multicenter human study.

#Herbal #Turmeric's #Curcumin Inhibits Human Papillomavirus-Positive and Negative Squamous Carcinoma Cells in Vitro, Researchers Say

Kyle J. Norton

Cancer is a group of chronic diseases associated with cell growth disorderly in the tissue of some specific organ. Depending on the grade and stage, most cases of cancer in the advanced stage has been found to induce a higher mortality rate compared to cancer diagnosed in the localized stage.
Head and neck cancer is a group of cancers that starts in the cell on the surface of the inner lining of the mouth, nose, throat, larynx, sinuses, or salivary glands.

Squamous cell carcinoma is cancer arisen from the squamous cells found in the outer layer of skin and in the mucous membranes, which are the moist tissues that line body cavities.

Head and neck squamous cell carcinoma (HNSCC) develops in the mucous membranes of the mouth, nose, and throat.

Head and neck squamous cell carcinoma (HNSCC) epidemiologically ahs have been found to spread to the lymph nodes or lungs. More than 90% of the head and neck cancer diagnosed are squamous carcinomas.

Symptoms for head and neck cancer may include a lump or sore that does not heal, a sore throat that does not go away, trouble swallowing, or a change in the voice.

According to the statistics provided by the Western General Hospital, squamous cell cancer of the head and neck is common worldwide accounting for 4% of all cancers in the United States and 5% in the United Kingdom.

The exact causes of head and neck squamous cell carcinoma (HNSCC) are unknown. However, geographically, smoking tobacco, drinking alcohol, and long-term poor diet is important to risk factors in the West compared to risk factors of chewing betel or areca nuts, smoking bidis, and taking snuff are important in the Indian subcontinent.

More importantly, the genetic preposition of genome 9p21, which may indicate loss of a gene that suppresses tumors accounts for a total of 70% of all cases.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

In finding a potential ingredient for the treatment of head and neck cancer, researchers examined the effect of curcumin on head and neck squamous cell carcinoma (HNSCC) using a panel of cell lines with and without human papillomavirus (HPV+, HPV-).

According to the tested differentiation, curcumin treatment of HNSCC cell lines not only interfered with RNA of Skp2, which causes cancer tumorigenesis and radiation tolerance but also induced concomitant accumulation of p21 and p27 associated with a cell cycle arrest in HPV+, HPV- cells.

Furthermore, curcumin inhibits cell viability and induces apoptosis in a dose-dependent manner in the tested cell line with and without human papillomavirus (HPV+, HPV-).

Moreover, curcumin treatment of HPV+ and HPV- cells resulted in apoptosis via the mitochondrial pathway of apoptosis and activation of caspases that play a critical role in programmed cell death.

Additionally, the inhibition of HPV+ and HPV- cells by curcumin was also found to suppress the expression of poor prognostic markers such as XIAP, and cancer survival such as cIAP1, and cIAP2.

More importantly, co-treatment of HNSCC cells with curcumin and cisplatin potentiated inhibition of cell viability and apoptotic effects.

Based on the findings, researchers wrote in the final report, "These data suggest an important function for curcumin, acting as a suppressor of oncoprotein Skp2 in squamous cell carcinoma cells in both HPV+ and HPV- cells".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of HNSCC, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Honey Restores the Epithelium, Mucosa, and Submucosal Regions of the Airway in Asthmatic Patients, According to Studies

Kyle J. Norton

Scientists may have found a natural alternative for the treatment of asthma, according to studies.
Asthma is a chronic inflammation of the airway of the lung with recurring symptoms. Asthma, attacks triggered by the stimuli can induce airway swelling and inflamed, leading to muscle constriction.

During asthma attacks, patients may experience symptoms of difficulty breathing, severe wheezing in the breath, and coughs that won't stop.

According to the statistics provided by the Centers for Disease Control and Prevention (CDC), 1 in 13 people in the US has asthma.

In other words, more than 26 million Americans have asthma, with half of the patients being children.

If you experience symptoms of a cough which is often worse at night or early in the morning, wheezing in your breath, accompanied by chest tightness and shortness of breath, you may be at the early stage of asthma. Please check with your doctor to rule out the possibility.

Genetic and environmental factors are the most common risk factors associated with the onset of asthma. Some researchers suggested that the widespread obesity over the past few decades may also have a strong implication on the onset of the disease.

Dr. David A. Beuthe r, the lead scientist in the concern of the association of asthma and obesity said, "Cross-sectional and prospective cohort studies of humans have shown a modest overall increase in asthma incidence and prevalence in the obese, although body mass index does not appear to be a significant modifier of asthma severity".

And, "Studying the obesity-asthma relationship in large cohorts, in which self-reports are frequently used to ascertain the diagnosis of asthma, has been complicated by alterations in pulmonary physiology caused by obesity, which may lead to dyspnea or other respiratory symptoms but do not fulfill accepted physiologic criteria for asthma".

These results strongly suggested that if you are obese, your risk of asthma is increased substantially.

Honey is a miraculous product made by bees using nectar from flowers.

The rich golden liquid is considered one of the healthy sweet foods for replacing the use of white sugar and artificial sweeteners many people.

Scientists on finding a natural remedy for the treatment of asthma investigated the effect of aerosolized honey on airway tissues in a rabbit model of ovalbumin (OVA)-induced asthma.

The study included 40 New Zealand white rabbits sensitized twice with a mixture of OVA and aluminum hydroxide on days 1 and 14.

Honey treatments were given from day 23 to day 25 at two different doses (25% (v/v) and 50% (v/v) of honey diluted in sterile phosphate buffer saline.

During the study, the aerosolized honey treatment group restored the epithelium, mucosa, and submucosal regions of the airway infected by the application of OVA.

Furthermore, the aerosolized honey treatment was also found to reduce the number of airway inflammatory cells present in bronchoalveolar lavage fluid and inhibit goblet cell hyperplasia, an indication of airway remodeling in asthma.

The findings strongly suggested that aerosolized honey has a significant effect on the treatment and management of asthma in animal models.

Additional differentiation of the modulatory effect of honey, royal jelly, and propolis extract on peripheral blood leukocytes and lung inflammation in a mouse conalbumin-induced asthma model in mice model divided into* The mice in group I that were not sensitized or treated; they were kept as controls.

* The mice in group II were sensitized and challenged with conalbumin. Twenty-four hours after the first challenge with antigen.

* The mice in group III received 0.5 mg/kg of dexamethasone intraperitoneally per day for 18 consecutive days and were kept as positive controls.

* The mice in groups IV, V, and VI received 650, 1000, and 30 mg/kg of honey, royal jelly, and propolis (aqueous and ethanolic extract), respectively, once per day for 18 consecutive days.

At the end of the experiment, by comparing the blood samples before and after treatment, researchers showed
* The groups treated with propolis extract exhibited considerable ameliorative effects against asthma, probably due to the expression of flavonoids and phenolics that exert antioxidative effects

* Honey- or royal jelly-treated groups exhibited an increased incidence of asthma cascade events due to increased inflammatory cells, probably due to the antagonist to bronchial asthma cases.

The results clearly indicated that not all honey is the same. Some can be used for asthma while others can not, probably due to types of chemical compounds found in the pollen

Taken together, honey and other bee products may be considered functional remedies for the prevention and treatment of asthma, pending the confirmation of the types of honey in a large and multicenter human study.

#Turmeric #Extract Inhibits Hepatic (Liver) Stress Associated with the Risk of Liver Injury, Researchers Say

Kyle J. Norton

The liver is the largest internal organ that not only filters the blood from the digestive system before passing them to other parts of the body but also plays a critical role in the production cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Hepatic stress is a condition in which the function of the liver is partly reduced. Hepatic stress can be observed by the levels of liver stress-makers.

Liver function tests (LFTs or LFs) are used to measure the levels of liver transaminases aspartate transaminase (AST) and alanine transaminase (ALT) in patients with liver injury, particularly, in a patient with some degree of intact liver function.

Abnormal high levels of transaminases aspartate transaminase (AST) and alanine transaminase (ALT) are early indications of liver damage or injury.

Oxidative stress in the liver caused by a toxic substance such as an overdose of the supplement has been found to constitute most cases of acute liver damage.

Lauren Cooper, the author in the examine the toxic effects of an overdose of green tea extract and bodybuilding pills wrote, "Many herbal remedies and dietary supplements can also harm the liver, including some that you can easily buy online or over-the-counter in drug or health food stores".

And, "Researchers looked at cases of liver damage reported to the Drug-Induced Liver Injury Network, a program funded by the National Institutes of Health. Roughly 700 cases of liver damage were reported to the program during the period of the study; 130 of those cases were linked to dietary supplements".

The results strongly suggested although green tea extract is considered a functional supplement for the treatment of various diseases, overdose and abusive use of such supplements also have a negative impact on human health, particularly in liver health.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an antioxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

With an aim to find a potential compound for the improvement of liver functioning, researchers examined the effect of Curcuma longa L. (CLL) rhizome on hepatic dysfunction.

In acute carbon tetrachloride (CCl4)-induced liver stress rats, researchers compared the activity of CLL extract administered once a day for 3 days at three dose levels (100, 200, and 300 mg/kg/day) and curcumin administered once a day at the 200 mg/kg/day.

Before treatment, CCl4 rats showed a significant induced increased serum AST and ALT levels by 2.1- and 1.2-fold, respectively, compared with the control.

CLL- and curcumin-treated rats inhibited the levels of AST but not ALT elevation induced by CCl4.

Furthermore, according to the tested assays, the peroxidation of membrane lipids in the liver was significantly prevented by CLL (100, 200, and 300 mg/kg/day) on liver tissues.

Moreover, both CLL extract and curcumin exhibited significant protection against liver injury by improving the levels of hepatic antioxidant enzymes such as superoxide dismutase and glutathione peroxidase, and glutathione against free radicals expression in the CCl4 treated rats.

More precisely, both CLL extract and curcumin protected the liver cells against the onset of liver cellular stress through reducing the liver damage markers and inhibiting the liver oxidative stress via increased production of liver antioxidant enzymes.

Dr. Lee GH, the lead scientist said, "Our data suggested that CLL extract and curcumin protect the liver from acute CCl4-induced injury in a rodent model by suppressing hepatic oxidative stress. Therefore, CLL extract and curcumin are potential therapeutic antioxidant agents against acute hepatotoxicity".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the improvement of liver functioning against liver damage, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
