#Phytochemical 3,3'-Diindolylmethane Inhibits #Overabundance of Pro-Inflammatory Cytokines and Proteins Associated with a Reduced Risk of #CardiovascularDisease (CVD)

Heart muscle or myocardium is one of three types of muscles in humans, that build the walls of the heart.
The most common diseases of heart muscle include angina pectoris myocardial infarction, and other heart muscle diseases caused by restricting blood supply to the muscle.

The most common symptoms of heart muscle diseases include tightness, pain, or swelling of the legs, ankles, and feet

The Genetics Home Reference wrote, " mutations in more than 30 genes have been found to cause familial dilated cardiomyopathy. These genes provide instructions for making proteins that are found in cardiac muscle cells called cardiomyocytes.

And, "Mutations in one gene, TTN, account for approximately 20 percent of cases of familial dilated cardiomyopathy".

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

In finding a natural ingredient that protects the heart muscle against the onset of cardiovascular disease researchers examined the 3, 3'-Diindolylmethane (DIM) effects on cardiomyocytes.

Tested cell lines H9C2 used in the study were evaluated by DIM's influence on inflammation and apoptosis of cardiomyocytes induced by LPS. H9C2 cells incubated with DIM (10, 20 and 30 μM) with or without LPS for 24 h.

Treatment with various concentrations of DIM (10, 20 and 30 μM) inhibited the overabundance of pro-inflammatory cytokines (TNF-α and IL-6 mRNA expression) induced by LPS.


In other words, DIM normalized the levels of impaired TNF-α and IL-6 mRNA expression, in the LPS + DIM30(μM) group.

According to the tested assays, DIM treatment significantly blocked LPS-induced oxidative stress and inhibited LPS-induced apoptosis in H9C2 cardiomyocytes.


Furthermore, DIM significantly inhibited the LPS-induced expression of a protein associated with proapoptosis and enhanced the expression of a protein involved in anti-apoptosis.

Dr. Luo Q, the lead scientist, wrote on the final report, "DIM may have a protective effect for H9C2 cardiomyocytes against LPS-induced inflammatory response and apoptosis. DIM may be a new insight into the treatment of septic cardiomyopathy".

In order to find out more information about DIM anti-cardiovascular disease activity, researchers evaluated the function of DIM on the hypoxia‑induced cardiac injury.

The study included H9c2 cells pretreated with DIM (1, 5, and 10 µM) for 12 h and exposed to hypoxia for 12 h.

DIM with different doses extended markedly attenuated the increased transcription of pro-inflammatory cytokines (interleukin (IL)‑1β, IL‑6 and tumor necrosis factor‑α) induced by hypoxia.

DIM also modulated the genes associated with transcription of autophagy, compared with the hypoxia group.

DIM also protected the cells against apoptosis by inhibiting and exhibiting the proteins associated with anti-apoptosis and pro-apoptosis, respectively.

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the protection of heart muscle against the risk of heart disease, pending to the confirmation of the larger sample size and multicenter human study.

Stroke is a medical condition caused by reduced blood flow to the brain resulting in cell death due to a blocked artery (ischemic stroke) or the leaking or bursting of a blood vessel (hemorrhagic stroke).
In other words, stroke can be caused by either ischemia, due to lack of blood flow, or hemorrhagic, due to bleeding.

A stroke is a medical emergency. Early intervention in most cases can minimize brain damage and potential complications.

Sadly, stroke is one of the most common causes of death worldwide.

In the US, according to statistics, approximately 800,000 people suffer a stroke of which 600,000 of these are first attacks, and 185,000 are recurrent attacks. Nearly three-quarters of all strokes occur in people over the age of 65.

There is a general belief that stroke is an accumulation over a long period of an unhealthy diet with red meat and processed meats in the induction of artery-clogging.

Atherosclerosis associated with the hardening of the arteries caused by the buildup of fatty substances in the arteries can lead to stroke.


In other words, if the blood vessel that feeds the brain is blocked, a stroke occurs.

* D3T suppressed microglia (MG) activation induced by stroke and inhibited peripheral inflammatory immune cell infiltration of CNS in the ischemic brain.

* D3T also exerted a protective effect on ischemic stroke against the induction of oxidative stress by mediating the improved expression of the antioxidant enzyme Nrf2.

Based on the results, researchers said, "D3T confers protection against ischemic stroke, which is mediated through suppression of MG activation and inhibition of CNS peripheral cell infiltration, and that the protective effect of D3T in ischemic stroke is dependent on the activation of Nrf2".

Taken altogether, cruciferous vegetables may be considered functional remedies for protection in ischemic strokes, pending the confirmation of the larger sample size and multicenter human study.

According to the tested differentiation, curcumin treatment of HNSCC cell lines not only interfered with RNA of Skp2, which causes cancer tumorigenesis and radiation tolerance but also induced concomitant accumulation of p21 and p27 associated with a cell cycle arrest in HPV+, HPV- cells.

Furthermore, curcumin inhibits cell viability and induces apoptosis in a dose-dependent manner in the tested cell line with and without human papillomavirus (HPV+, HPV-).

Moreover, curcumin treatment of HPV+ and HPV- cells resulted in apoptosis via the mitochondrial pathway of apoptosis and activation of caspases that play a critical role in programmed cell death.

Additionally, the inhibition of HPV+ and HPV- cells by curcumin was also found to suppress the expression of poor prognostic markers such as XIAP, and cancer survival such as cIAP1, and cIAP2.

More importantly, co-treatment of HNSCC cells with curcumin and cisplatin potentiated inhibition of cell viability and apoptotic effects.

Based on the findings, researchers wrote in the final report, "These data suggest an important function for curcumin, acting as a suppressor of oncoprotein Skp2 in squamous cell carcinoma cells in both HPV+ and HPV- cells".

Taken together, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of HNSCC, pending to the confirmation of the larger sample size and multicenter human study.


Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Furthermore, the aerosolized honey treatment was also found to reduce the number of airway inflammatory cells present in bronchoalveolar lavage fluid and inhibit goblet cell hyperplasia, an indication of airway remodeling in asthma.

The findings strongly suggested that aerosolized honey has a significant effect on the treatment and management of asthma in animal models.

Additional differentiation of the modulatory effect of honey, royal jelly, and propolis extract on peripheral blood leukocytes and lung inflammation in a mouse conalbumin-induced asthma model in mice model divided into* The mice in group I that were not sensitized or treated; they were kept as controls.

* The mice in group II were sensitized and challenged with conalbumin. Twenty-four hours after the first challenge with antigen.

At the end of the experiment, by comparing the blood samples before and after treatment, researchers showed
* The groups treated with propolis extract exhibited considerable ameliorative effects against asthma, probably due to the expression of flavonoids and phenolics that exert antioxidative effects

* Honey- or royal jelly-treated groups exhibited an increased incidence of asthma cascade events due to increased inflammatory cells, probably due to the antagonist to bronchial asthma cases.


The results clearly indicated that not all honey is the same. Some can be used for asthma while others can not, probably due to types of chemical compounds found in the pollen

Taken together, honey and other bee products may be considered functional remedies for the prevention and treatment of asthma, pending the confirmation of the types of honey in a large and multicenter human study.

#Turmeric #Extract Inhibits Hepatic (Liver) Stress Associated with the Risk of Liver Injury, Researchers Say

The liver is the largest internal organ that not only filters the blood from the digestive system before passing them to other parts of the body but also plays a critical role in the production cholesterol, a waxy substance which aids digestion, builds cell membranes and produces vitamin D and steroid hormones.

Hepatic stress is a condition in which the function of the liver is partly reduced. Hepatic stress can be observed by the levels of liver stress-makers.

Liver function tests (LFTs or LFs) are used to measure the levels of liver transaminases aspartate transaminase (AST) and alanine transaminase (ALT) in patients with liver injury, particularly, in a patient with some degree of intact liver function.

Abnormal high levels of transaminases aspartate transaminase (AST) and alanine transaminase (ALT) are early indications of liver damage or injury.

Oxidative stress in the liver caused by a toxic substance such as an overdose of the supplement has been found to constitute most cases of acute liver damage.

Lauren Cooper, the author in the examine the toxic effects of an overdose of green tea extract and bodybuilding pills wrote, "Many herbal remedies and dietary supplements can also harm the liver, including some that you can easily buy online or over-the-counter in drug or health food stores".

In acute carbon tetrachloride (CCl4)-induced liver stress rats, researchers compared the activity of CLL extract administered once a day for 3 days at three dose levels (100, 200, and 300 mg/kg/day) and curcumin administered once a day at the 200 mg/kg/day.

Before treatment, CCl4 rats showed a significant induced increased serum AST and ALT levels by 2.1- and 1.2-fold, respectively, compared with the control.

CLL- and curcumin-treated rats inhibited the levels of AST but not ALT elevation induced by CCl4.

Furthermore, according to the tested assays, the peroxidation of membrane lipids in the liver was significantly prevented by CLL (100, 200, and 300 mg/kg/day) on liver tissues.

Moreover, both CLL extract and curcumin exhibited significant protection against liver injury by improving the levels of hepatic antioxidant enzymes such as superoxide dismutase and glutathione peroxidase, and glutathione against free radicals expression in the CCl4 treated rats.

More precisely, both CLL extract and curcumin protected the liver cells against the onset of liver cellular stress through reducing the liver damage markers and inhibiting the liver oxidative stress via increased production of liver antioxidant enzymes.

Dr. Lee GH, the lead scientist said, "Our data suggested that CLL extract and curcumin protect the liver from acute CCl4-induced injury in a rodent model by suppressing hepatic oxidative stress. Therefore, CLL extract and curcumin are potential therapeutic antioxidant agents against acute hepatotoxicity".

#Cucumber Potentially Used in the Management of Knee Pain, Stiffness, and Physical Functions Related to #Osteoarthritis

Osteoarthritis (OA) is a chronic condition affecting the joints in the hands, knees, hips, and spine mostly occurring in older adults.
More precisely, OA is a result in which the protective cartilage on the ends of your bones wears down over time due to repetitive movement of the same joints that cause the thinning cartilage, leading to bone rubbing on bone.

With an aim to find a potential compound for the treatment of osteoarthritis, researchers evaluated the efficacy of Q-Actin™, an aqueous extract of Cucumis sativus (cucumber; CSE) against glucosamine-chondroitin (GC) in the management of moderate knee OA.

The randomized double-blind, parallel-group clinical trial took place in three different centers. included 122 patients (56 males and 66 females) aged between 40 and 75 years and diagnosed with moderate knee OA assigned to 2 groups (n= 61).

Among the groups, the GC group received orally the generally prescribed dose of 1,350 mg of GC twice daily and the CSE group received orally 10 mg twice daily of CSE.


According to the assays, on days 30, 60, 90, 120, 150, and 180 compared to the baseline (Day 0), the WOMAC score in the CSE group was decreased by 22.44% and 70.29% on days 30 and 180, respectively, compared to a 14.80% and 32.81% respectively decrease in the GC group.

The pain scores also showed similar trends in flavor to the CSE group. There was no adverse effect on the course of the experiment.

In other words, CSE is more effective for the treatment of OA compared to that of GC.


Dr. the lead scientists wrote, "The use of 10 mg CSE, twice daily, was effective in reducing pain related to moderate knee OA and can be potentially used in the management of knee pain, stiffness, and physical functions related to OA".

Taken together, cucumber extract may be used for the treatment of OA, pending the confirmation of the larger sample size and multicenter human study.

#Carotenoids Reduce the Implication of the Toxic ROS Expression that Causes #CysticFibrosis (CF), Researchers Find

Cystic fibrosis (CF) is the most common fatal genetic disease affecting mostly the lung in Canadian children and young adults, leading to difficulty breathing and coughing up mucus and frequent lung infections.

However, the condition has also been found in the pancreas, liver, kidneys, and intestine.
According to the Cystic Fibrosis Foundation Patient Registry, more than 30,000 people are living with cystic fibrosis in the US.

Believe it or not, most cases of CF are diagnosed by age 2.


The most common symptoms depend on the location of cystic fibrosis including
sinus infections, poor growth, fatty stool, and clubbing of the fingers and toes.

In male adults, the condition also induces infertility as a result of the blockage or absence of the sperm canal.

Conventionally, as of today, there is no cure for CF. Most cases of CF caused by lung infection are treated intravenously, inhaled, or by mouth.

In severe cases, a lung transplant may be the only option.

Although most cases of CF are associated with a genetic preposition, environment, lifestyle, and age are also 2 main factors that increase the risk of CF.

Cystic Fibrosis Canada in the investigation of the risk of CF in older adults wrote, "Today, the estimated median age of survival of CF patients in Canada is over 50 years, and the majority of patients are over 18 years of age. CF patients enjoy an improved quality of life, pursuing careers, and starting families of their own".

In finding a potential compound for the treatment of lung disease researchers examined the effects of carotenoids, the potent antioxidants on singlet oxygen and other toxic oxygen species in patients with cystic fibrosis (CF).

The study included a total of 17 patients with cystic fibrosis (CF) and 10 normal children,

According to the assessment of plasma levels of four carotenoids, beta-carotene, alpha-carotene, lutein, and lycopene, levels of carotenoids in children with CF were significantly lower compared to normal control participants.

There was no significant difference in total cholesterol and total carotene between groups. Furthermore, there was no correlation between fat absorption or measures of adiposity in children with CF compared to control.

However, levels of selected carotenoids were correlated negatively with serum IgG levels associated with the measurement of inflammation.

More importantly, researchers also found that the differences in plasma carotenoid levels between children with CF and normal children were involved in the rapid turnover of carotenoids, by quenching of toxic oxygen species in inflammatory states of CF.

The results strongly suggested that improving the concentration of carotenoids that reduces the implication of the toxic ROS expression may have an impact on the prevention and treatment of CF.
