3,3'-Diindolylmethane Inhibits the Matrix and protein associated with a Refuced Risk of #OvarianCancer in Vitro, Scientists Say

Ovarian cancer is a medical and chronic condition characterized by irregular cell growth in the tissues found either at the tail ends of the fallopian tubes or the ovaries.
According to the statistics provided by the American Cancer Society, ovarian cancer is one of the deadly reproductive cancer in women. Every year, the disease causes more deaths than any other cancer of the female reproductive system.

The lifetime risk of a woman getting ovarian cancer is 1/78 compared to the lifetime dying risk of 1/108.

In seeking a natural product that processes anti-ovarian cancer properties, researchers examined the effects of 3,3'-diindolylmethane (DIM) on ovarian cancer cell viability, invasion, and sensitivity to chemotherapy.

According to the final results obtained by the ovarian cancer SKOV3 and A2780 cell lines treated by the 3,3'-diindolylmethane (DIM), DIM treatment not only potently suppressed the viabilities of ovarian cancer cells but also inhibited xenograft growth in nude mice.

Additionally, in order to obtain more information about DIM anti-ovarian cancer effects, researchers examined the phytochemical activity of Interleukin (IL6) which has been found to be a major contributor to the initiation and progression of ovarian cancer.

During the experiment, human ovarian carcinoma-derived cell lines SKOV3 and A2780 treated with IL6 and/or DIM and subjected to in vitro proliferation, DIM inhibited the increase of ovarian cancer cell adhesion, migration, and invasion caused by the administration of IL-6 through deactivated the expression of protein STAT3 similar to that of the aforementioned study.

Importantly, DIM-induced ovarian cancer cells apoptosis is associated with the inhibition of the expression matrix metalloproteinases MMP-2 and MMP-9 associated with the pathologic relationship to cancer.

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of ovarian cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Serum of Lycopene in the Body Is Associated with a Reduced Level of Oxidativestress, According to Studies

Oxidative stress is characterized by overexpression of free radicals in the body.
In other words, oxidative stress is a result of the imbalance of the ratio of free radicals and antioxidants caused by abundant free radicals and low levels of antioxidant enzymes produced by the body.

A free radical is an unstable atom with a single electron on the outermost ring. Given its nature, free radical readily donates or intercept an electron from another atom, leading to a chain of the free radical cascade that cannot be stopped until all single electron is paired or inhibited by antioxidants.

The antioxidant is a stable atom with a function to inhibit oxidation and comes either from dietary sources or produced by the body.

In contrast to general belief, free radical is also needed in moderated amounts for the function of the heart muscle to pump blood such as NO for vasodilation and increasing blood flow.

According to studies, in cancer, oxidative stress has been found to induce cytotoxicity used in chemotherapy for the treatment of cancer by killing cancer cells.

However, in a healthy person, oxidative stress is harmful because it not only induces cells' lipid and protein oxidation but also causes the change of cell DNA, leading to diseases associated with oxidative stress including cancer, diabetes, and cardiovascular disease.

Ozone, certain pesticides, and cleaners, cigarette smoke, radiation, and pollution are some of the risk factors associated with the increased risk of oxidative stress.

Based on the oxidative stress theory of aging oxidative stress, oxidative stress is also the main culprit contributing to aging in humans.

Dr. Ilaria Liguori, the lead scientist said, "Aging is a process characterized by the progressive loss of tissue and organ function. The oxidative stress theory of aging is based on the hypothesis that age-associated functional losses are due to the accumulation of RONS-induced damages".

With an aim to find a potential compound for the treatment of diseases associated with oxidative stress researchers examined the efficacy of lycopene a carotenoid commonly found in tomatoes and tomato products.

A total of 23 healthy postmenopausal women, 50-60 years old, were provided blood samples at baseline and following a one-month lycopene-depletion period according to the St. Michael's Hospital, Toronto, ON, Canada. PARTICIPANTS AND STUDY DESIGN.

Compared to the blood sample before and after experiment samples, a one-month lycopene-depletion period diet showed a significantly decreased antioxidant profile including serum lycopene, lutein/zeaxanthin, and glutathione peroxidase (GPx).

Furthermore, the diet also depressed levels of antioxidant enzymes superoxide dismutase (SOD), and catalase (CAT), significantly in the participants while increasing levels of lipid and protein oxidation.
In other words, tomatoes restrictive diet increased the risk of diseases associated with oxidative stress in postmenopausal women due to the increase of ROS and depletion of antioxidants in the tissues.

In order to reveal more information about the tomato effect against oxidative stress, researchers conducted a study to determine whether lycopene would act as an antioxidant to decrease oxidative stress parameters.
The study included a total of 60 postmenopausal women, 50-60 years old, following a 1-month washout without lycopene consumption,.

After a month, the participants were randomly divided into groups with 15 members each consuming either : (1) regular tomato juice, (2) lycopene-rich tomato juice, (3) tomato Lyc-O-Mato lycopene capsules, or (4) placebo capsules, twice daily for total lycopene intakes of 30, 70, 30, and 0 mg/day respectively for 4 months.


According to the tested assays, for 4 months significantly increased serum lycopene compared to placebo.

Moreover, the total serum antioxidant capacity (TAC) and decreasing lipid and protein peroxidation increased substantially in the groups treated with consumed juice or lycopene capsules compared to the placebo group.

Taken together, tomatoes and their major bioactive compound lycopene may be considered remedies for the prevention and treatment of diseases associated with oxidative stress, pending the confirmation of the larger sample size and multicenter human study.


Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Cancer is a class of diseases characterized by irregular cell growth on the body tissue, due to the alternation of cell DNA.
Most cases of cancer start in the cell on the surface of the inner lining tissue and do not exert any symptoms at the early stage.

At the later stage, the larger size tumor may press on the nearby blood vessels and nerve cells, causing localized severe bleeding and pain accompanied by the general symptoms of cancers, including unintended weight loss, loss of appetite, fatigue, and tiredness, and gastrointestinal problems.

Most cancer cells at this stage can also travel a distance away from the original site to affect other healthy tissue and organs, leading to secondary metastasis.


Metastasis is the process to induce the development of secondary malignant growths at a distance from primary cancer.

Metastatic cancer also shares the characteristics of primary cancer. Breast cancer that spreads to the lung is called metastatic breast cancer, not lung cancer.

According to statistics, in 2018, approximately over1,700,000 cases of cancer were diagnosed worldwide. The lifetime probability of being diagnosed with cancer is 39.7% for men and 37.6% for women.

Believe it or not, some metastatic cancer may have an unknown primary origin or CUP, secondary cancer without a primary cancer site.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as an anti-oxidant, hypoglycemic, colorant, antiseptic, and wound healing agent, and for the treatment of flatulence, bloating, appetite loss, ulcers, eczema, inflammations, etc.

In the urgency to discover a natural ingredient for the prevention of metastasis in cancer, researchers examined the effects of Furanodiene, a bioactive compound isolated from Curcuma wenyujin Y. H. Chen et C. Ling (C. wenyujin) on tumor metastatic activities.

In a model of carcinogen-induced hepatotoxicity in rats, the selected animals were orally administered with sinigrin on a daily basis for three months before sacrifice.

During the experiment, the injection of sinigrin significantly inhibited the proliferation of liver tumor cells, observed by the reduction of the number of surface tumors in the rat liver.

Sinigrin induced apoptosis of liver cancer cells by exhibiting the expression of a protein associated with tumor suppression in the host rats, namely, the p53.

Furthermore, the application of sinigrin-induced cancer apoptosis by stimulating the production of a protein involved in cell death programming and a gene involved in cellular differentiation.


Interestingly, in the additional analysis of the sinigrin anti-liver cancer activity, researchers showed that the bioactive compound not only inhibits cancer cells from proliferation through cell cycle arrest but also restores liver function without inducing liver toxicity.

The results strongly suggested that sinigrin processed significant anti-liver cancer properties via several genes and pathways.

Dr. Jie M, the lead scientist wrote in the final report, "Sinigrin exerts important anti-proliferative activities in carcinogen-induced hepatocarcinogenesis in rats, and highlights the potential of sinigrin as an anti-cancer agent for liver cancer".

Taken altogether, sinigrin found in Brassica nigra may be considered a functional supplement for the prevention and treatment of liver cancer, pending the confirmation of a large sample size and multicenter human study.

Scientists may have found a bioactive ingredient for the treatment of gastrointestinal cancer with no side effects, according to studies.

Gastrointestinal (GI) cancer is a group of cancers, affecting the digestive system, including esophagus, gallbladder & biliary tract, liver, pancreas, stomach, small intestine, bowel, and anus

The digestive system consists of the gastrointestinal tract and other parts of the accessory organs of digestion including the tongue, salivary glands, pancreas, liver, and gallbladder), involved in the function of breaking down food into smaller and smaller components then absorbed before passing them as nutrients and fluids to the body.

Most cases of gastrointestinal cancer start in the cells on the surface of the inner lining tissues, depending on the location of the affected organs.

At the early stage, most patients are asymptomatic due to the small size of the tumor. However, at the later stage, after penetrating into deeper layers of the organ, cancer cells can travel through blood and fluids circulation to infect other healthy tissues or organs, leading to secondary metastasis.

At this stage, besides sharing the general symptoms of other types of cancer, gastrointestinal cancer also cause symptoms of severe pain and bleeding due to the overgrowth of tumor size that has suppressed the nearby blood vessels and nerve cells.

Some researchers suggested that a poor diet may have a strong implication on the incidence of gastrointestinal cancer.

Dr. Reilly T. Enos, the lead scientist said, "High-fat-diet (HFD) consumption is associated with colon cancer risk".

And, "HFDs with the highest SF content had the greatest concentration of Muc2. Our data suggest that high dietary SF is protective in the AOM/DSS model of colon cancer, which may be due, at least in part, to the ability of SF to maintain intestinal barrier integrity through increased colonic Muc2".

With an aim to find a natural compound for the treatment of gastrointestinal cancers, researchers examined the 3,3'-Diindolylmethane (DIM) activity against effective gastrointestinal cancers.

In vivo and vitro, injection of the DIM acted upon several cellular and molecular processes in gastrointestinal cancer cells.

In cancer cells apoptosis, DIM initiated production reactive oxygen species (ROS) in generating stress, triggering DNA damage by increasing the cytotoxicity.


In angiogenesis, DIM inhibited the protein associated with the generation of a new blood vessel for cell survival and proliferation.

In cell cycle regulation, DIM deactivated the expression of protein kinases which play critical roles in cancer cell division.

Furthermore, DIM triggers cellular endoplasmic reticulum (ER) stress and apoptosis within the cancer cells.

In autophagy, DIM inhibited autophagy that leads to cell death.

In metastasis, DIM inhibited the growth of human cancer cells by interfering with multiple signaling pathways, involved restraining invasion, migration, and promoting apoptosis.


Furthermore, DIM used combined with conventional chemo drugs or therapeutic chemicals increased the efficacy of other drugs for the treatment of gastrointestinal cancer.

Taken together, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of gastrointestinal cancers, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
